[Anti-obesity activity of 3-hydroxymethyl N-methyl piperidine 4-chlorophenoxyacetate hydrochloride in mice treated with gold thioglucose].

The 3-hydroxyméthyl N-méthyl piperidine 4-chlorophenoxyacetate, hydrochloride, A, a potent anorectic, reduces weight gain of gold thioglucose obese mice through a reduced body fat and a decrease in metabolic efficiency. Compound A has much less effect in the lean mice than in the obese models. In contrast with pair-fed obese or lean mice, the decreased food consumption cannot account for all the reduced weight gain of the obese controls. Basal lipolytic activity in parametrial adipose tissue is greater in obese mice treated with A then in controls. It seems that the stimulating effect of A on lipolysis could contribute to the weight reduction.

Structure activity studies of a series of 4-(m-OH phenyl) piperidines.

A series of 4-(m-OH phenyl)-piperidine analogs with R4 = methyl or t-butyl and NR = methyl, allyl and phenethyl have been synthesized; and their receptor affinities, in vivo analgetic agonism and antagonism, and energy-conformational profiles determined. These analogs bind selectively and with moderate to high affinity to opioid mu-receptors. Binding in their preferred phenyl axial conformation appears to lead to meperidine-like agonism. In addition, for some R4 = methyl but not t-butyl compounds, binding of an energy-accessible phenyl equatorial conformation produces antagonism.

Alterations in acetylcholine metabolism in rat striatal slices by a 4-methyl piperidine analog of hemicholinium-3.

The in vitro effects of a tertiary amine, 4-methyl piperidine analog of hemicholinium-3 (A-4), were investigated on acetylcholine (ACh) metabolism in rat striatal slices. Rat striatal slices were incubated in the presence of 1.0 microM [3H]-choline in the presence or absence of 0.1 mM A-4. High pressure liquid chromatography with electrochemical detection was utilized to separate and measure total and [3H]-labeled ACh and choline. The effects of A-4 on [3H]-choline uptake, ACh and choline content, ACh release, and specific activity of ACh and choline tissue pools were investigated. Results indicated that A-4 inhibited the uptake of [3H]-choline into the striatal slices. Addition of 0.1 mM A-4 also produced a significant reduction in ACh content and ACh release and reduced the specific activity of the tissue choline and ACh pools. The effects of A-4 were more prominent upon immediate incubation than after a 60-min preincubation. These studies demonstrate that the pharmacological effects exhibited by A-4 are consistent with inhibition of choline uptake, with subsequent reduction in ACh synthesis and release.

Stereoisomers of quaternary and tertiary 4-methyl piperidine analogs of hemicholinium-3.

Previous studies have shown that quaternary and tertiary 4-methyl piperidine derivatives of hemicholinium-3 (A-5 and A-4, respectively) are potent inhibitors of choline uptake. The d-, l-, and mesostereoisomers of A-5 and A-4 were separated and the potency and reversibility were compared. Isomeric forms of each compound were found to be approximately equipotent inhibitors in the following preparations: inhibition of rabbit neuromuscular transmission using the sciatic nerve-gastrocnemius muscle preparation, reductions in acetylcholine content in rat caudate tissue slices and inhibition of choline uptake in neuroblastoma cells, line NB41A3. Because these results show no difference in potency or reversibility for the stereoisomers of A-5 or A-4, these studies indicate that hydroxyl substitutions in these agents do not play a role in their biologic activity. Perhaps only 2-point attachment is required for inhibition of choline transport by hemicholinium-like compounds.

Optical isomers of some piperidine-derived hemicholinium congeners containing secondary alcohol groups: preparation and biological activities.

Previous publications on the title compounds, 1 and 2 ('A-4' and 'A-5'), reported pharmacological data on what were probably mixtures of optical isomers of unknown composition. The compounds can exist as an enantiomeric pair as well as a diastereomeric meso isomer. In the present work, all possible stereoisomers (meso and dextrorotatory and levorotatory isomers) of the piperidine derivatives 'A-4' and 'A-5' have been isolated, identified, and characterized, and pharmacological data have been obtained on all products. In all of the bioassays conducted, optical isomers of each structure exhibited similar qualitative and quantitative effects. We conclude that the stereochemical nature of the chiral centers bearing secondary alcohol groups in these molecules does not play a critical role in interactions with in vivo receptors, and therefore that the alcohol groups in 'A-4' and 'A-5' are not prime sites of interaction(s) with receptor(s).

Activation of energy expenditure in the rat by a new non-amphetaminic compound: the (4-chlorophenoxyacetate) N-methyl 3-hydroxymethyl piperidine, hydrochloride (PM 170).

The (4-chlorophenoxyacetate) N-methyl 3-hydroxymethyl piperidine, hydrochloride (PM 170), chemically unrelated to amphetamine, was studied in normal rats for any thermogenic effect. It was given in graded doses. After an acute treatment PM 170 increases energy expenditure by increasing resting oxygen consumption (VO2) and in the doses used, produced dose-related increases in VO2. Energy expenditure of rats treated for 16 days was chronically elevated, while body weight gain and food intake were reduced. It also stimulated mitochondrial oxygen consumption probably by partial uncoupling of respiration from ATP synthesis. The disruption of oxidative phosphorylation could partially explain the effect of this compound on resting metabolic rate.

[Influence of 3-(3, 4, 5-trimethoxybenzamido) piperidine (KU-54) on gastric mucosal blood flow (author's transl)].

In dogs and rabbits anesthetized with pentobarbital sodium or urethane, respectively, the gastric mucosal blood flow was measured by the thermoelectrical method after intravenous administration of 3-(3, 4, 5-trimethoxybenzamido)piperidine(KU-54), an antiulcer drug. In the gastric corpus of rabbits, the maximal mucosal blood flow (11.9%) was attained within 1 min after KU-54(5 mg/kg i.v.) dosing and there was a prolonged increase during 8 min. Administration of KU-54 produced a transitory depression of the blood pressure by about 20%, and induced a beta-adrenergic-like action, presumably because both the blood pressure depression and the gastric mucosal blood flow increment were simultaneous in rabbits. This phenomenon was however, not abolished by pretreatment with propranolol, a beta-adrenergic blocker, therefore KU-54 may not be a beta-adrenergic blocking agent. Administration of gefarnate, a standard antiulcer drug, produced a slight increase in gastric mucosal blood flow and response to this drug was less than that seen with KU-54. Sulpiride, a standard antiulcer drug, two peaks on the gastric mucosal blood flow increment in rabbits and the pattern of blood flow increment differed from that seen with KU-54. The increment of gastric mucosal blood flow produced by KU-54 (5 mg/kg i.v.) was more successive in the gastric antrum than that in the gastric corpus. In fasting rabbits, the main period of gastric mucosal blood flow increment produced by KU-54 (5 mg/kg i.v.) was more successive than that seen in the non-fasting animals. In dogs, the maximal blood flow (16%) was observed at 7 min after KU-54 (5 mg/KG i.v.) dosing and there was a prolonged increase during 6 min.

Manual and semiautomated spectrophotometric determination of vitamin K3 by reactions wih piperidine and malononitrile.

Simple, rapid, selective, and sensitive spectrophotometric methods are described for manual and semiautomated determination of vitamin K3. These methods are based on reactions with piperidine and malononitrile whereby intense red and blue colors with maximum absorption at 510 nm (E(1%) 1 cm 150) and 580 nm (E(1%) 1 cm 155), respectively, are developed. The reactions are sensitive enough to permit the determination of 2-100 micrograms vitamin K3/mL with an average recovery of 98% and a standard deviation of 1%. There is no interference from vitamin K1 and K2, and the reduced forms of K vitamins, or from many common pharmaceutical diluents and additives.

Cardiac electrophysiologic and antiarrhythmic actions of two long-acting spirobenzopyran piperidine class III agents, L-702,958 and L-706,000 [MK-499].

The cardiac electrophysiologic and antiarrhythmic actions of two Class III ketone- and alcohol-containing spirobenzopyran piperidine analogs, L-702,958 and L-706,000 [MK-499], respectively, were assessed in vitro and in vivo. L-702,958 and L-706,000 [MK-499] selectively blocked the rapidly activating component of the delayed rectifier K+ current in guinea pig isolated ventricular myocytes (IC50 values, 14.6 and 43.9 nM, respectively), and prolonged effective refractory period in ferret isolated papillary muscles (EC25 values, 10.5 and 53.8 nM, respectively). In anesthetized dogs, L-702,958 and L-706,000 [MK-499] increased ventricular refractory periods (ED20 values, 3.3 and 9.2 micrograms/kg i.v., respectively) and concomitantly increased ECG QT interval and left ventricular+dP/dt. Cumulative i.v. administrations of up to 100 micrograms/kg of L-702,958 and 300 micrograms/kg L-706,000 [MK-499] in anesthetized dogs increased atrial and ventricular refractoriness and prolonged the ECG QT interval, but did not alter atrial, atrioventricular nodal, His-Purkinje or ventricular conduction indices. In anesthetized dogs studied chronically (9.2 +/- 1.1 days) after anterior myocardial infarction, the cumulative i.v. administrations of 100 micrograms/kg of L-702,958 and 300 of micrograms/kg L-706,000 [MK-499] suppressed the induction of ventricular tachyarrhythmia by programmed ventricular stimulation (suppression rates: 8 of 10, 80% and 9 of 11, 82%, respectively) and reduced the incidence of lethal ventricular arrhythmias (incidence of lethal ischemic arrhythmias: 4 of 10, 40% and 1 of 11 9%, respectively, compared to 34 of 40, 85%, in vehicle controls. L-702,958 and L-706,000 [MK-499] (cumulative 100 and 300 micrograms/kg i.v., respectively) did not facilitate the induction of arrhythmias by programmed ventricular stimulation in postinfarction dogs. After equivalently effective p.o. doses in conscious dogs, L-702,958 (10 micrograms/kg) and L-706,000 [MK-499] (30 micrograms/kg) increased ECG QT interval with long durations of action of approximately 9 and 14 hr, respectively. L-706,000 [MK-499] elicited a more consistent and sustained prolongation of the QT interval than L-702,958. These findings show that both L-702,958 and L-706,000 [MK-499] are potentially useful agents for the prevention of malignant ventricular arrhythmias in the setting of myocardial ischemic injury.

Antitumor activity of a piperidine phospholipid.

A piperidine phospholipid ((+/-)-2-[hydroxy] [1-octadecyloxycarbonylpiperidin-3-yl]methoxy-phosphinyl] oxy]-N,N,N, trimethylethaniminium hydroxide inner salt, SDZ 62-826) has been prepared that exhibited weak direct cytotoxicity and strong macrophage-induced cytotoxicity in vitro against a variety of murine and one human tumor cell lines. This compound was found to be as effective as ET-18-OCH3 and SRI 62-834, phospholipids with both strong direct and macrophage-induced cytotoxicity, in increasing survivors and reducing tumor volume when given either orally or intravenously in the mouse MethA fibrosarcoma model. These findings suggest that the macrophage-induced cytotoxicity exhibited by ET-18-OCH3 and other phospholipids may play an important role in this tumor model.

[3H]1-(cyclopropylmethyl)-4-(2-(4-fluorophenyl)-2-oxoethyl) piperidine HBr (DuP 734). A selective ligand for sigma receptors in mouse brain in vivo.

1-(Cyclopropylmethyl)-4-(2-(4-fluorophenyl)-2-oxoethyl) piperidine HBr (DuP 734) is a novel sigma receptor ligand which exhibits promise in preclinical animal models as an antipsychotic agent without motor side effects. In vitro and in vivo receptor binding profiles of DuP 734 in mouse brain using [3H]DuP 734 and [3H]N-allylnormetazocine ((+)-SKF 10,047) were studied. The pharmacology and stereospecificity of [3H]DuP 734-labeled sites in mouse brain, both in vitro and in vivo, was consistent with sigma receptor pharmacology. Specific in vivo binding of [3H]DuP 734 in brain peaked 1 hr after i.v. injection and this level of binding was maintained up to 4 hr. On the other hand, plasma concentration of [3H]DuP 734 decreased rapidly within 20 min after injection, indicating different pharmacokinetics between brain and plasma levels. Nonspecific binding, defined using 1 mg/kg (2.66 mumol/kg) of haloperidol, was approximately 30% of total binding 1 hr after injection of radiotracer. Administration of DuP 734 potently antagonized the binding of [3H]DuP 734 and [3H](+)-SKF 10,047 to brain sigma receptors in vivo with ID50 values of 0.02 and 0.07 mg/kg (0.07 and 0.25 mumol/kg), respectively. However, (+)-SKF 10,047, which possess a high affinity (IC50 = 22.5 nM) for [3H]DuP 734 binding in vitro, failed to displace [3H]DuP 734 binding in vivo. Thus in vitro receptor binding data may not predict in vivo receptor occupancy. Overall, the data suggest [3H] DuP 734 is a good ligand for in vivo imaging of sigma receptors.

[Isochroman-4-spiro-4'-piperidine: synthesis and analgesic properties].

The synthesis of some isochroman-4-spiro-4'-piperidines by intramolecular O-methylation of 4-aryl-4-hydroxymethylpiperidines with formaldehyde and their analgesic activity ("hot plate"test in mice) are reported. The introduction of the new ring led to compounds with weak analgesic activity.

Effects of repeated administration of N-[1-(2-benzo(b)-thiophenyl)cyclohexy]piperidine and cocaine on locomotor activity in C57BL/6 mice.

The present study investigated the effects of repeated administration of the dopamine reuptake inhibitors N-[1-(2-benzo(b)thiophenyl)cyclohexyl]piperidine (BTCP) and cocaine on locomotor activity, as well as their ability to induce or express cross-sensitization. Male mice were injected with a fixed dose of BTCP or cocaine (10,20 or 40 mg/kg i.p.) for 3 consecutive days and challenged on the 4th day with one of several doses of BTCP and/or cocaine. After every daily treatment, locomotor activity was assessed. Repeated administration of cocaine produced sensitization to the locomotor activity produced by different challenge doses of both cocaine (2.5-56.6 mg/kg i.p.) and BTCP (2.5-80 mg/kg, i.p.) on day 4. Repeated administration of low and intermediate doses of BTCP did not significantly affect the locomotor activity produced by different challenge doses of BTCP, whereas tolerance-like effects were observed after the higher dose, 40 mg/kg, particularly during the 3-day regimen. Repeated administration of BTCP dose-dependently produced leftward and downward shifts of the cocaine dose-response curve. The results indicate that, under identical treatment conditions, cocaine and BTCP differ markedly with respect to their ability to cause sensitization, but differ less in terms of their ability to elicit locomotor activity in sensitized animals. The demonstration of cross-sensitization between BTCP and cocaine provides evidence for a shared mechanism of action; however, the present results also suggest that the chronic effects of cocaine and BTCP are not identical.

Field evaluation of arthropod repellents, deet and a piperidine compound, AI3-37220, against Anopheles funestus and Anopheles arabiensis in western Kenya.

A field evaluation of the repellents N,N-diethyl-3-methylbenzamide (deet) and 1-(3-cyclohexen-1-yl-carbonyl)-2-methylpiperidine (AI3-37220, a piperidine compound) was conducted against Anopheles funestus and An. arabiensis in Kenya. Both repellents provided significantly more protection (P < 0.001) than the ethanol control. AI3-37220 was significantly more effective (P < 0.001) than deet in repelling both species of mosquitoes. After 9 h, 0.1 mg/cm2 of AI3-37220 provided 89.8% and 71.1% protection against An. arabiensis and An. funestus, respectively. Deet provided > 80% protection for only 3 h, and protection rapidly decreased after this time to 60.2% and 35.1% for An. arabiensis and An. funestus, respectively, after 9 h. Anopheles funestus was significantly less sensitive (P < 0.001) to both repellents than An. arabiensis. The results of this study indicate that AI3-37220 is more effective than deet in repelling anopheline mosquitoes in western Kenya.

Antiarrhythmic action of the new calcium antagonist [1,2,5-trimethyl-4-phenyl-4-beta-(N-cyanoethyl-N-4'-methoxybenzyl) -ethylamino]piperidine dihydrochloride.

Arrhythmias induced by coronary artery ligation in cats, by CaCl2 and epinephrine in rats, and by ouabain in guinea-pigs were used as experimental models for studying the effects of a new calcium antagonist AR-1 ([1,2,5-trimethyl-4-phenyl-4-beta-(N-cyanoethyl-N-4'-methoxybenzyl) -ethylamino]piperidine, calcium channel blocker and calmodulin antagonist) on ventricular arrhythmias. Coronary ligation caused 90% lethality (ventricular fibrillation) with 12.5 min in untreated control cats, which was prevented by administration of AR-1 (4 mg/kg body weight (b.w.) before or after arrhythmia induction. Pretreatment with AR-1 afforded protection in a dose-related fashion. A dose of 1.5 mg/KG b.w. increased survival to 45%, and all cats dosed with 3 to 5 mg/Kg b.w. survived. CaCl2 (180 mg/Kg b.w., i.v.) induced ventricular fibrillation and 100% lethality. These effects were completely prevented by an anti-arrhythmic dose of AR-1 (3 mg/kg b.w.). Epinephrine-induced ventricular arrhythmias were also prevented by the same dose of AR-1. AR-1 (5 mg/kg b.w.) did not prevent ouabain (0.5 mg/kg b.w.)-induced arrhythmias that caused death within 17 +/- 3.7 min, but displayed protective effects during 67 +/- 7.7 min. The results from these animal studies, in conjunction with previously studies demonstrating coronarodilatory and anti-platelet efficacy of this compound, collectively suggest that AR-1 has a potential to become a useful antianginal and antiarrhythmic therapeutic agent.
