Chiral permselectivity in surface-modified nanoporous opal films.

Nanoporous 7 mum thin opal films comprising 35 layers of 200 nm diameter SiO2 spheres were assembled on Pt electrodes and modified with chiral selector moieties on the silica surface. Diffusion of chiral redox species through the opals was studied by cyclic voltammetry. The chiral opal films demonstrate high selectivity for transport of one enantiomer over the other. This chiral permselectivity is attributed to the surface-facilitated transport utilizing noncovalent interactions between the chiral permeant molecules and surface-bound chiral selectors.

22pi smaragdyrin molecular conjugates with aromatic phenylacetylenes and ferrocenes: Syntheses, electrochemical, and photonic properties.

Syntheses, spectroscopic, electrochemical, and third-order nonlinear optical susceptibilities of a series of 22pi smaragdyrins and their corresponding Rh(I) derivatives bearing phenylacetylene substituents and ferrocene-containing substituents are reported. The synthetic strategy involved a [3 + 2] acid-catalyzed oxidative coupling reaction of the appropriate dipyrromethane and oxatripyrrane. The desired meso substituents, such as phenylacetylenylphenyl and the ferrocenes, were incorporated to the dipyrromethane unit prior to the oxidative coupling reaction. The optical absorption, emission characteristics, and the quantum yield of the smaragdyrin conjugates depends on the nature of the substituent, nature of linker group, and the spacer length. Theoretical studies at the DFT level suggest high delocalization of electrons confined to only four of the five available heterocyclic rings for the free bases. However, upon Rh(I) metalation, the pi-electron delocalization is extended to all the heterocyclic rings. The two-photon absorption cross section (TPA) values sigma(2) measured through the open aperture Z-scan method, increases linearly with enhanced pi-electron delocalization for the smaragdyrins containing phenylacetylene substituents. The meta branching of substituents decreases sigma(2) values. Introduction of Rh(I) to the smaragdyrin cavity enhances the sigma(2) values by about 3-10 orders of magnitude, attributed to the increased aromatic character upon Rh(I) insertion. The calculated molecular electrostatic potential (MESP) and harmonic oscillator model of aromaticity (HOMA) for the free bases and the Rh(I) derivatives justifies such a conclusion. A linear correlation observed for the second oxidation potential of Rh(I) derivatives and corresponding free bases also support the increased aromaticity upon Rh(I) insertion. The electrochemical data for ferrocene-containing smaragdyrins reveal easier ring oxidation by about 50-130 mV and harder ferrocene oxidation by 40-180 mV suggesting electron-donating nature of the ferrocene upon linking with the smaragdyrin system. The TPA cross section value of 88782 GM observed for 5g represents one of the highest values known for a metalloexpanded porphyrin derivative.

A self-locking molecule operative with a photoresponsive key.

Rotary host 1, composed of a ferrocene unit as a rotary module, is conformationally locked internally in apolar solvents such as benzene by a double intramolecular Zn-N coordination between the zinc porphyrin and aniline units, attached to each cyclopentadienyl (Cp) ring. Upon addition of the cis form of 1,2-bispyridylethylene (cis-2) to (+)-1 ([cis-2]/[(+)-1] = 5.0), an enantiomer of 1, the intramolecular Zn-N coordination bonds in (+)-1 are readily cleaved to form an externally locked, cyclodimeric one-to-one complex (+)-1 subset cis-2, accompanying a rotation of the ferrocene module, as visualized by CD spectroscopy. In contrast, use of trans-2, in place of cis-2, under otherwise identical conditions to the above, did not result in releasing the internal double lock of (+)-1. Such a large difference between the isomers of 2 in the affinity toward host 1, along with their capabilities of photochemical interconversion, allowed for the demonstration of a reversible self-locking operation of 1. Namely, the externally locked state of 1, as in the form of 1 supersetcis-2, spontaneously retrieves the internally locked state, after the release of 2 from 1 upon cis-to-trans photochemical isomerization of ligating 2, while the backward photochemical isomerization of 2 in the presence of 1 results in switching of 1 to its externally locked state.

Replacement of aromatic or heteroaromatic groups in nonsteroidal antiinflammatory agents with the ferrocene group.

Ferrocene analogues of the antiinflammatory agents tolmetin (1), fenbufen (2), flurbiprofen (3), and fenclofenac (4) were synthesized and tested for biological activity. The derivatives exhibited little or no antiarthritic or platelet antiaggregatory activity, indicating that the ferrocene moiety is a poor bioisostere for aromatic or heteroaromatic groups in nonsteroidal antiinflammatory agents.

Modification of the estrogenic properties of diphenols by the incorporation of ferrocene. Generation of antiproliferative effects in vitro.

We report here the synthesis and the strong and unexpected antiproliferative effect of the organometallic diphenolic compound 1,1-bis(4'-hydroxyphenyl)-2-ferrocenyl-but-1-ene (4) on both hormone-dependent (MCF7) and -independent (MDA-MB231) breast cancer cells (IC(50) = 0.7 and 0.6 microM). Surprisingly, 6 [1,2-bis(4'-hydroxyphenyl)-2-ferrocenyl-but-1-ene], the regioisomer of 4, shows only a modest effect on these cell lines. This pertinent organometallic modification seems to trigger an intracellular oxidation of the structurally favorable compound 4, leading to the generation of a potent cytotoxic compound.

Probing the role of the covalent linkage of ferrocene into a chloroquine template.

A new therapeutic approach to malaria led to the discovery of ferroquine (FQ, SR97276). To assess the importance of the linkage of the ferrocenyl group to a 4-aminoquinoline scaffold, two series of 4-aminoquinolines, structurally related to FQ, were synthesized. Evaluation of antimalarial activity, physicochemical parameters, and the beta-hematin inhibition property indicate that the ferrocene moiety has to be covalently flanked by a 4-aminoquinoline and an alkylamine. Current data reinforced our choice of FQ as a drug candidate.

Synthesis and antimalarial activity in vitro and in vivo of a new ferrocene-chloroquine analogue.

The antimalarial activities of ferrocenic compounds mimicking chloroquine and active upon chloroquine-resistant strains of Plasmodium falciparum were evaluated. Four 7-chloro-4-[[[2-[(N,N-substituted amino)methyl]ferrocenyl]methyl]amino]quinoline derivatives have been synthesized; one of them, 1a, showed high potent antimalarial activity in vivo on mice infected with Plasmodium berghei N. and Plasmodium yoelii NS. and was 22 times more potent against schizontocides than chloroquine in vitro against a drug-resistant strain of P. falciparum.

New ferrocenyl chiral auxiliary substituents for amines: applications to syntheses of mossambine and vinblastine.

(+)-(R)-1,2-(alpha-(R)-Mesyloxy-beta-dimethyltetramethylene)-ferrocene was synthesized and used as a chiral auxiliary for N-alkylation of methyl 1,2,3,4,5,6-hexahydroazepino[4,5-b] indole-5-xi-carboxylates. Condensation with aldehydes then provided tetracyclic products in a diastereomeric ratio of at least 97:3. Gentle cleavage in acetic acid removed the chiral auxiliary to give the corresponding secondary amines in >99% ee. Thus, key intermediates leading to mossambine and vinblastine could be synthesized with high enantioselectivity. The enantioselectivity greatly exceeds that found with other chiral N-auxiliaries developed in our studies.

Synthesis of ferrocene-substituted 2-azetidinones.

The photochemical reaction of alkoxychromium(0)carbene complexes and ferrocene mono- and disubstituted imines formed 2-azetidinones having one or two ferrocene moieties in good yields. Yields decrease when the carbene moiety bears an aminoferrocene moiety attached to the carbene carbon, while complex 9 having the ferrocene directly bonded to the carbene carbon was totally inert in these reactions. Access to beta-lactams with the ferrocene tethered to the C3 position through a methylene group was gained using the lithium enolate derived from ethyl 3-ferrocenylpropanoate. The reaction of this enolate produced two unexpected processes. Thus, 2-azetidinone 15 having an hydroxyl group at the C3 position was obtained together with the expected beta-lactam 14, by reaction of the lithium enolate of ethyl 3-ferrocenylpropenoate and imine 1. Additionally, unsaturated amide 17 was obtained by base-promoted Hoffmann-like breakage of the beta-lactam ring formed in the reaction of the same enolate and imine 2. Oxidation of the anion at the C3 of the 2-azetidinone ring on compound 14, as well as the sterically driven ring-breakage of the C3 anion derived from the nonisolated 2-azetidinone 18, should be responsible for this behavior.

Synthesis and conformational analysis of optically active ferrocene containing macrocyclic peptides.

Two macrocyclic peptides 3 and 4 were formed during lactamization of 1,1'-ferrocenylbis(alanine) 1. Isolation, structure determination, and conformational analysis of 3 and 4 are reported as well as a controlled stepwise synthesis of 4 also including an improved route to 1. On the basis of their (1)H NMR spectra recorded at 300 K in DMSO-d(6), the dimer 3 and trimer 4 were found to be C(2) and C(3) symmetric, respectively. As appeared from computational analysis, the low-energy conformations of the macrocyclic peptides were nonsymmetric. Cyclic voltammetry revealed that the ferrocenyl moieties in 3 or 4 are electrochemically equal to ferrocene.

Molecular triads composed of ferrocene, C60, and nitroaromatic entities: electrochemical, computational, and photochemical investigations.

Synthesis and physicochemical characterization of a series of molecular triads composed of ferrocene, C(60), and nitroaromatic entities are reported. Electrochemical studies revealed multiple redox processes involving all three redox active ferrocene, C(60), and nitrobenzene entities. Up to eight redox couples within the accessible potential window of o-dichlorobenzene containing 0.1 M (TBA)ClO(4) are observed. A comparison between the measured redox potentials with those of the starting compounds revealed absence of any significant electronic interactions between the different redox entities. The geometric and electronic structure of the triads are elucidated by using ab initio B3LYP/3-21G methods. In the energy-optimized structures, as predicted by electrochemical studies, the first HOMO orbitals are found to be located on the ferrocene entity, while the first LUMO orbitals are mainly on the C(60) entity. The coefficients of the subsequent LUMO orbitals track the observed site of electrochemical reductions of the triads. The photochemical events of the triads are probed by both steady-state and time-resolved techniques. The steady-state emission intensities of the triads and the starting dyad, 2-(ferrocenyl)fulleropyrrolidine, are found to be completely quenched compared to fulleropyrrolidine bearing no redox active substituents. The subpicosecond and nanosecond transient absorption spectral studies revealed efficient charge separation (and rapid charge recombination) in the triads, and this has been attributed to the close spacing of the redox entities of the triad to one another.

Synthesis and application of arylmonophosphinoferrocene ligands: ultrafast asymmetric hydrosilylation of styrene.

A short and efficient synthetic route to a novel class of atropisomeric and planar chiral 2-aryl-1-diphenylphosphanylferrocene ligands is presented. The modular design of the ligands allows a synthetic approach in which both the aromatic moiety and the phosphino substituent can be varied easily. This permits fine-tuning of steric and electronic properties. The ligands have been tested in the asymmetric hydrosilylation of styrene where enantioselectivities up to 90% are obtained. Optimization of the palladium-to-ligand ratio resulted in hitherto unparalleled turnover frequencies (TOF) exceeding 180 000 h(-1). The absolute stereochemistry of the ligands was determined from an X-ray structure. 2D NMR experiments in combination with ab initio calculations were used to assign the conformation of the atropisomeric biarylic scaffold.

Ferrocenyl-bearing cyclopseudopeptides as redox-switchable cation receptors.

A family of ferrocenyl-bearing cyclopseudopeptides (1-10) designed for redox-switchable receptors of cations was synthesized. Circular dichroism (CD) and cyclic valtammetry (CV) studies of cation binding properties in both the reduced (K1) and oxidized (K2) forms revealed that the binding preference is mainly governed by the charges and radius of the guest cation as well as by the suitability of the host to accommodate the guest. Particularly worth mentioning is the fact that some synthesized cyclopseudopeptides showed high binding affinity and selectivity toward alkaline-earth ions. For example, the K1 of compound 2 binding with Ca2+ is 4.37 x 10(6) mol(-1) x L and its Ca2+/K+ selectivity is 3.1 x 10(5):1, both values are much greater than those of an excellent natural ionophore, valinomycin (1 x 10(5) mol(-1) x L and 0.33:1, respectively). The linear relationship between the shifts of half-wave potentials (deltaE(1/2)) and the radius/charge [r/(+)] ratios suggests that the sensitivity of electrochemical responses to cation complexation be dominated by repulsion factors between the redox center and the incoming cation guest.

Design and synthesis of a new type of ferrocene-based planar chiral DMAP analogues. A new catalyst system for asymmetric nucleophilic catalysis.

A new first-generation catalyst system for nucleophilic catalysis has been developed. It is based on a planar chiral ferrocene skeleton with either the potent nucleophile 4-(dimethylamino)pyridine (DMAP) or the related 4-nitropyridine N-oxide attached in either the 2- or the 3-position. The syntheses are short, efficient, and enantioselective and X-ray crystal structures of both DMAP-derived catalysts are presented. The DMAP-based catalysts were tested in asymmetric reactions and the 3-derivative 14 showed good activity and a moderate level of enantioselectivity. The sense of induction (selectivity) was studied using molecular modeling and the results pointed at new directions for future generations of catalysts based on this design.

Use of ferrocene scaffolds as pendant groups in hairpin-type pyrrole-imidazole polyamide molecules showing sequence-selective binding to DNA duplexes.

[reaction: see text] The synthesis and properties of new conjugate molecules, Fc-PIA, composed of ferrocene (Fc) and pyrrole-imidazole polyamides (PIA) are reported. As a PIA sequence, we chose Im-Py-Im/Py-Im-Py considering its future application to the SNPs detection of genes having a GCG/CGC sequence. Two types of Fc-containing linkers, i.e., ferrocene-1,1'-dicarboxamide and ferrocenecarboxamide, were designed, and several Fc-PIPA molecules having these linkers were synthesized. Titration studies by use of circular dichroism revealed that the carboxamide-type Fc-PIA could bind to the target DNA with an association constant of 10(7) M(-1). In contrast, ferrocene dicarboxamide-type compounds have slightly weaker affinity for the target DNA. However, the affinity could be recovered by replacing one of the pyrrole residues with beta-alanine. We carried out the CV measurement and observed quasi-irreversible oxidation of the ferrocene moieties in the Fc-PIA compounds. These properties of Fc-PIA indicate the potential usefulness of these molecules in electrochemical detection of genes.

Facile synthesis and detailed characterization of a new ferrocenyl uracil peptide nucleic acid monomer.

A new ferrocenyl uracil peptide nucleic acid (PNA) monomer, tert-butyl-2-(N-(2-(((9H-floren-9-yl)methoxy)carbonylamino)ethyl)-2-(5-(N-ferrocenylmethylbenzamido)-2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)acetamido)acetate (1), has been successfully prepared in good yield by a procedure involving the one-pot reaction of the key synthon, 5-(ferrocenylmethylamino)pyrimidine-2,4(1H,3H)-dione (4), itself prepared from the reaction of (ferrocenylmethyl)trimethylammonium iodide and 5-aminouracil, with benzoyl chloride followed by ethyl bromoacetate. After hydrolysis of the ester, the acid was coupled with a protected PNA backbone to generate 1. NMR spectroscopy showed that 1 hydrogen bonds 9-ethyladenine (EA) in a 1:1 mixture of CD3CN:CDCl3 with an association constant Ka of 70 M(-1) at 30 degrees C. This value is comparable with those observed for model receptors and shows that the ferrocenyl moiety of 1 does not hinder the hydrogen bonding of our new PNA monomer to the complementary DNA base or if it does, not significantly. 1 is oxidized to 1+ with a reversible potential of +538 mV vs the DMFc(0/+) (decamethylferrocene) couple under voltammetric conditions in a 1:1 mixture of CH3CN:CHCl3 (0.1 M Bu4NPF6). For this reversible process, a slightly larger diffusion coefficient of 4.2 x 10(-6) cm(2).s(-1) than usually found for these compounds was determined from these electrochemical studies, which should be analytically useful as it will readily afford submicromolar voltammetric detection limits.

The synthesis of N,O-ferrocenyl pyrrolidine-containing ligands and their application in the diethyl- and diphenylzinc addition to aromatic aldehydes.

A facile route to a series of planar chiral N,O-ferrocenyl pyrrolidine-containing ligands with varying substituents at the nitrogen and oxygen donor atoms is described. The oxygen donor atom was introduced via a diastereoselective ortho-metalation of N-methylpyrrolidinyl and N-allylpyrrolidinyl ferrocene intermediates and was quenched with various ketones. The nitrogen substituent was varied through deallylation and subsequent derivatization of a secondary pyrrolidine. The efficacy of these novel ligands was investigated in the enantioselective addition of diethylzinc and diphenylzinc to aromatic aldehydes. The ligands proved highly effective in the diethylzinc addition to benzaldehyde that resulted in high yields of up to 99% and enantioselectivities (ee's) of up to 95%. The role of planar chirality was explored and the results indicated that the planar chirality, and not the central chirality, of the ferrocenyl ligands was the dominant stereo-controlling element. Employment of a mixed ethyl-phenylzinc reagent in the phenylation of aromatic aldehydes led to a mixture of the two additional products, and the phenylated product was obtained in up to 37% ee.
