Thylakoid-associated polyamines adjust the UV-B sensitivity of the photosynthetic apparatus by means of light-harvesting complex II changes.

The sensitivity of the photosynthetic apparatus to ultraviolet-B (UV-B) irradiation was studied in cultures of unicellular green alga Scenedesmus obliquus incubated in low light (low photosynthetically active radiation intensity [LL]) and high light (high photosynthetically active radiation intensity [HL]) conditions, treated or not with exogenous polyamines. Biochemical and physicochemical measurements showed that UV-B radiation induces a decrease in the thylakoid-associated putrescine (Put) and an increase in spermine (Spm), so that the reduction of Put/Spm ratio leads to the increase of light-harvesting complex II (LHCII) size per active reaction center and, consequently, the amplification of UV-B effects on the photosynthetic apparatus. The separation of oligomeric and monomeric forms of LHCII from isolated thylakoids showed that UV-B induces an increase in the oligomeric forms of LHCII, which was more intense in LL than in HL. By manipulating the LHCII size with exogenous polyamines, the sensitivity degree of the photosynthetic apparatus to UV-B changed significantly. Specifically, the addition of Put decreased highly the sensitivity of LL culture to UV-B because of the inhibitory effect of Put on the LHCII size increasing, whereas the addition of Spm enhanced the UV-B injury induced in HL culture because of the increasing of LHCII size. The ability of the photosynthetic apparatus to recover the UV-B induced changes was also investigated.

Postnatal morphodifferentiation of the subneural apparatuses of the posterior cricoarytenoid muscle in rats: a scanning electron microscopy study.

The subneural apparatus, i. e., the post-synaptic component of the neuromuscular junction, in the posterior cricoarytenoid muscle of the rat was studied by scanning electron microscopy, with special attention given to its postnatal differentiation along with the functional development of the muscle. Primitive synaptic troughs observed in the first postnatal week consisted of single cup-like depressions 5-6 microm in diameter. On the 7th day, low sarcoplasmic ridges appeared in the trough. In the second postnatal week, muscle fibers could be classified into two groups: large (10-15 microm in diameter) and small (less than 10 microm in diameter). In the large muscle fibers, many low ridges became circular and protruded to transform the single trough into numerous cup-like depressions (2-5 microm in diameter). In contrast, the subneural apparatus in the small muscle fibers consisted of a small number of cup-like depressions. The two types of subneural apparatus differentiated into adult forms by the 28th postnatal day, although they remained smaller in size than those of adults. In the large muscle fibers, the number of pit-like or elongated invaginations increased and gradually transformed into slit-like junctional folds by the 28th postnatal day, while the small muscle fibers still possessed a few pit-like or elongated junctional folds at this point in time. The two types of morphodifferentiation of the subneural apparatus are thought to reflect the two types of muscle fibers in the rat posterior cricoarytenoid muscle.

Automated preference testing apparatus for rating palatability of foods.

An electronic preference testing apparatus is described for measuring taste preference of rodents and other small animals with solid or liquid foods. The apparatus is designed on the principle of the two-choice, preference technique. It operates photoelectrically with a sequence of presentations so that whenever a subject eats from a compartmentalized food tray, a standard and a test food are each briefly sampled alone before both foods are presented together (in alternate positions) for preference determination. Preferences are automatically recorded on digital counters. The apparatus is built in two modules (a preference tester and the master control) connected by multiconductor cable. The modular design provides portability and isolation of the animals from the major noise-producing components. Diagrams of the apparatus are given, and test results from a trial that evaluated positional bias and a sucrose-concentration preference experiment are presented to demonstrate its application in research.

[Evaluation of a semi-closed underwater breathing apparatus, the "eOBA"].

Semi-closed underwater breathing apparatus has commonly been used among military and commercial divers, but never available for recreational divers because of complicated operations, difficulties of maintenance, and expensiveness. Nippon Sanso K.K. has newly deployed a semi-closed circuit underwater breathing apparatus called the "eOBA". It is especially designed for divers to enjoy shallow (max. 5 m) and short (10 min.) dives. This eOBA was evaluated from manned testing based on physiological requirements for the apparatus. Four male and four female subjects participated in the three test trials. Subjects maintained their position at the depths of 2 m and 4 m for 10 min. and e exercised on the underwater ergometer at the depth of 2.4 m. The pressure at the mouthpiece, tidal volume, breathing rate, the inspired O2 level, CO2 level were monitored on breath-by-breath basis. The inspired O2 level ranged from 45% (at exercise) to 60% (at rest) and the inspired CO2 level were kept less than 1.0% in the most cases. The P-V loop indicated relatively little external work of breathing (less than 0.1 kg.m/l) and allowable peak pressures (less than 25cmH2O). Results show that the eOBA meets the standards sufficiently under the test conditions and is a safe apparatus for recreational divers if it is properly used.

Computed tomographic study of the effect of a tongue-tie on hyoid apparatus position and nasopharyngeal dimensions in anesthetized horses.

OBJECTIVE: To determine the effect of manual tongue protrusion on the dimensions of the hyoid apparatus, nasopharynx, and oropharynx in anesthetized horses. tongue protrusion on the dimensions of the hyoid. ANIMALS: 5 adult horses. PROCEDURE: Horses were anesthetized and positioned in sternal recumbency for 2 sequential computed tomographic (CT) scans. Images were acquired with the tongue in a natural position inside the mouth. Then, the tongue was pulled rostrally and secured, and a second CT scan was performed. Dorsoventral length of the hyoid apparatus and angles of the basisphenoid, basihyoid, and ceratohyoid were measured on 3-dimensional reconstructed CT images. Cross-sectional diameters and areas of the nasopharynx and oropharynx were determined on reformatted images in the transverse and longitudinal planes, using osseous landmarks for consistency. Results were tested between the 2 groups to determine significant differences. RESULTS: We were unable to detect a significant difference between any of the lengths or angles of the hyoid apparatus measured with or without rostral protrusion of the tongue. Similarly, nasopharyngeal and oropharyngeal diameters and cross-sectional areas were not significantly different with or without rostral protrusion of the tongue. CONCLUSIONS AND CLINICAL RELEVANCE: Tying the tongue rostrally out of a horse's mouth did not influence the position of the hyoid apparatus or dimensions of the nasopharynx or oropharynx in anesthetized horses. Currently, no data suggest that application of a tongue-tie is effective for maintaining stability and patency of the nasopharyngeal or orolaryngeal airways in horses during races.

[Middle ear dynamic characteristics of patients obtained by a newly developed measuring apparatus].

A tympanometer is well known to be a valuable apparatus for the diagnosis of secretory otitis media (SOM). However, it is insufficient for the diagnosis of ossicular chain disorders. Therefore, an attempt was made to develop a new measuring apparatus, which had a function of sweeping both frequency and pressure and was measurable the absolute sound pressure variations at the external auditory canal, owing to the carefully designed probe. In the previous papers, the middle ear dynamical characteristics of normal subjects were measured by this apparatus, and the measurement results were compared with the theoretical analysis. In this paper, the middle ear dynamical characteristics of patients were measured, and the effects of various kinds of middle ear diseases upon its dynamical characteristics were examined. The measurement results of the patients with ossicular chain separation, ossicular chain fixation, SOM and tympanic membrane perforation had their own distinctive patterns, and were also clearly different from those of normal subjects. Therefore, this apparatus seems to have a high degree of the clinical applicability to the diagnosis of these diseases.

Multicomponent collection (MCC) with the latest hemapheresis apparatuses.

BACKGROUND: Collection of multiple products including plasma, packed red blood cells (PBRC) and double units of platelets, is increasingly improving donor utilisation and decreasing blood transfusion costs. Total apheresis is the new trend in blood banking and both industry and physicians are progressively expanding their involvement in this field. During 1998, at our hemapheresis unit, we used three new apparatuses for multicomponent collection (MCC). These apparatuses vary in terms of principle, operation and specific capabilities, and thus knowledge of them is essential to match their characteristics with plateletapheresis needs. STUDY DESIGN: We reviewed all MCCs carried out in 1998 with MCS+, Trima and Excel-Pro cell separators. These apparatuses were evaluated in terms of platelet yield, white blood cell (WBC) contamination, efficiency per minute, platelet activation, and PBRC product quality. RESULTS: The three machines are compared in the following table [table in text]. CONCLUSIONS: MCC with the new apparatuses is well accepted by donors and personnel. There are no risks for the donors and, excluding ACD reactions and/or vaso/vagal attacks at the beginning of procedures, no severe reaction was seen. The quality and quantity of platelet products are good. When PRBC or a second platelets unit are collected, the cost of the kit is covered by this double product.

Blood vessels and excretory apparatus of the kidney in some wild animals.

On 110 preparations of the kidney in some wild animals (hare, fox, wolf, bear, boar and chamois), the blood vessels and the excretory apparatus were studied by dissection, injection-corrosion and microscope. Only the bear has a markedly split kidney, whereas the kidneys of the other animals are unsplit. In the fox there is an obvious split of the renal artery into anterior and posterior branches which supply the anterior and posterior portion, respectively, of the renal parenchyma, being separated from each other, so that we may speak of an anterior and posterior kidney. In the fox, wolf, hare and chamois the interlobar arteries pass through the renal calices in a loop composed of adipose tissue, invested by the epithelium of the renal calyx. The renculi of the bear kidney show complete autonomy in relation to the blood vessels as well as in relation to the excretory apparatus. The relation of the surface of the excretory apparatus to the whole kidney was studied. Thus we have found the fox to have relatively the largest excretory apparatus, whose surface amounts to 31% of the whole kidney. In the remainder of the animals investigated this percentage is considerably less, ranging from 21.7% (boar) to 26.7% (bear).

Exposure to soda-lime dust in closed and semi-closed diving apparatus.

BACKGROUND: Chronic exposure to hyperbaric hyperoxia and venous gas microembolism have been shown to contribute to the long term health effects of diving, especially diver's lung function. Factors related to special diving equipment may add to these effects. This study was conducted to evaluate possible additional hazards for respiratory function of divers employing closed and semi-closed diving apparatus. METHODS: We analyzed soda-lime dust found in the air-intake loop of a closed-circuit oxygen rebreathing diving apparatus which had passed through the filter screen of the diving apparatus's soda-lime cartridge. The geometrical characteristics were evaluated by scanning electron microscopy. The amount of dust that passed through the screen during a normal dive profile was measured by an artificial airflow through a filter membrane. After dives by subjects using these devices, the pH-value of water condensate in the air-intake hose was measured. RESULTS: There was a relevant amount of residual soda-lime dust found in the air-intake loop. The dust particles showed diameters down to 1 micron and a rounded structure. The total amount of dust averaged 9.6 mg.m-3 of breathing mixture. During diving, the mean pH-value of condensate in the hose is estimated at 8.87 (+/- 0.12). CONCLUSION: There is a relevant exposure to soda-lime dust in divers using closed-circuit rebreathing apparatus. This occupational exposure may contribute to chronic airway inflammation and subsequent development of small airway disease in divers.

An apparatus for quantitating experimental spinal cord trauma.

An apparatus is described for producing experimental contusion of the spinal cord and simultaneously monitoring the degree of trauma: velocity of spinal cord deformation, impulse and energy. This is accomplished by modifying an existing trauma apparatus by the addition of a displacement transducer, an induction coil triggering device and oscilloscope. With variability in the mechanics of the trauma apparatus and biological variations in each animal (i.e. differences in the visco-elastic properties of the spinal cord, etc.) and in different kinds of animals, this apparatus will more accurately quantitate the trauma delivered to each spinal cord and, thereby, form a better basis for correlation of amount of injury, degree of neurological dysfunction and efficacy of various methods of treatment.

[Restoration of movement in the joints with the aid of a hinge-distraction apparatus].

The device described presents an external hinge which is fixed by transosseous wires and reproduces as far as possible the normal kinematics of the joint. By removing all load from the joint surfaces and maintaining a constant predetermined space between them, the apparatus eliminates friction, prevents pain and muscle spasm, and minimizes abnormal mobility (or instability). It also permits gradual and controlled reduction of old traumatic or congenital dislocations or contractures. Once the bones are in proper relationship, the apparatus permits graduated, controlled, nearly frictionless motion about the correct axis so that a good range of motion is restored and joint surfaces of the proper shape are formed. Finally, ununited fractures close to the joint surface may be immobilized while joint motion is permitted. Experience gained with the apparatus has shown that the method described is physiological and not difficult, complications associated with its use being rare and not serious. The good results obtained with the use of the apparatus warrant its wider application.

[An experimental study of the surgical trauma on the rabbit's lens suspensory apparatus after cataract surgery].

Extracapsular cataract extraction (ECCE) and posterior chamber intraocular lens (PC-IOL) implantation requires that the posterior capsule and the zonules remain sufficiently intact. To assess the surgical trauma on the lens suspensory apparatus of the enucleated rabbit eye, we developed a new fine pressure gauge that could quantitate its strength. The cornea and the iris of enucleated eyes were removed to allow clear visualization of the anterior surface of the lens to the equator. Afterwards several types of anterior capsulotomy were performed for ECCE and PC-IOL implantation. And then the lens of the pseudophakic eye was vertically pressed with the plastic rod of the fine pressure transducer, and the strength of the lens suspensory apparatus was simultaneously measured. The strength of the lens suspensory apparatus of the pseudophakic eye decreased approximately 35% compared with that of the phakic eye. Moreover the integrity of the lens capsule of animals with circular capsulorhexis with tears or linear capsulotomy decreased approximately 43% compared with that of continuous circular capsulorhexis (CCC) and also zonular rupture and vitreous loss occurred as a result of the expansion of radial tears in the anterior capsule. The present study indicates that the strength of the lens suspensory apparatus decreases with cataract surgery, suggesting that much more attention to cataract surgery is needed if the anterior capsule had radial tears. It is also suggested that CCC is the best type of anterior capsulectomy procedure.

[Pneumo-automatic apparatus for the artificial ventilation of the lungs for small laboratory animals].

We have designed apparatus of artificial ventilation of the lungs (AAV) for small laboratory animals on pneumoelements, examined static and dynamic characteristics of a generator of impulse signals and chose rational parametres of the apparatus. Some advantages of that model as compared with its analogues have been proved. The apparatus was approved at electrophysiological experiments on rats, 200-250 g. Monitoring of the most important parameters of the vital functions showed that external breathing on that apparatus was adequate.

Artificial bloodfeeding of Anopheles sacharovi on a membrane apparatus.

Anopheles sacharovi, the main human malaria vector in Turkey, has been maintained in our laboratory by feeding on anesthetized rabbits for about 20 years but it is a difficult species to colonize and bloodfeed. To eliminate the need for keeping and using live rabbits to supply blood meals, artificial bloodfeeding methods with suitable membrane apparatus were investigated. The feeding apparatus designed by the World Health Organization and 3 other types designed by us (for feeding on preserved human blood) were tested. Artificial membranes (latex and paraffin film) and locally produced and dried calf intestine were used. The calf intestine membrane gave the best feeding results and a modified apparatus designated type III was the most successful. This apparatus was preferable for the artificial feeding of An. sacharovi because it has a small reservoir, is easy to use, is adaptable to different feeding conditions, and supports reasonably high bloodfeeding rates 44.4-50.5% as compared to 35% on live rabbits.

[The use of a hinge distraction apparatus after arthrolysis and arthroplasty (author's transl)].

The authors describe a hinge distraction apparatus which allows physiological movement of joints following surgical procedures such as arthrolysis or arthroplasty. The apparatus maintains some space between the bone ends and provides lateral stability. It is based on the principles of external fixation. The apparatus has been used for ankles, knees, and elbows. Thirty-eight patients were operated on. The technique is fully described. The results on experimental work in the dog are given. At the ankle, the authors have shown that the use of the apparatus allows the formation of fibrous tissue between the bone ends. The indications and results obtained at the level of the knee and elbow are discussed.

Development and evaluation of a new apparatus for lymph drainage: preliminary results.

Due to the lack of a specific treatment apparatus that enhances lymph flow, we developed a simple, easy-to-use, portable device for patients presenting with lower extremity lymphedema. The aims of the present study were to demonstrate the design and operation of the equipment and to test its efficacy. The new apparatus encourages articulation of the ankle and contraction of the muscles of the calves and feet. Eight patients with clinically diagnosed primary or secondary lymphedema were imaged by lymphscintigraphy before and after one week of using the apparatus. The images were evaluated by two experienced nuclear medicine physicians blinded to the other's results. An improvement in the lymphscintigram images was seen in all patients evaluated. This preliminary study suggests that the new apparatus was easily tolerated, enhanced lymph flow as assessed by lymphscintigraphy, and may have benefits as an adjunctive treatment. Further clinical studies are necessary to fully evaluate its use.
