Repercussions of transition out of elite sport on body image.

The purpose of this study was to evaluate the psychological repercussions of transition out of elite sport from a bodily point of view. We hypothesized that the passage from a bodily over-investment to a more sedentary state would have an effect on body satisfaction. 16 Transitioning Athletes following the Sydney Olympic Games were compared with 16 Active Athletes two times during the transition period using the Body-Image Questionnaire of Bruchon-Schweitzer. Qualitative data in the form of interviews provided a complement to the quantitative data. Analysis yielded no significant differences 1 1/2 mo. after career termination between the two groups but showed a decrease in body satisfaction between 1 1/2 and 5 mo. after career termination for Transitioning Athletes, with a significant between-group difference at 5 mo. The Transitioning Athletes initially reported weight gain and uncertainty about their real physical capacities but also a continued social recognition that maintained body satisfaction. Over time, however, they were increasingly aware of this bodily deterioration, as ongoing exercise served as a reality test. Added to this were perceived decreases in social value as well as disturbing somatic manifestations. The result was a substantial effect on body satisfaction.

Meibomian gland phospholipids.

The content of the meibomian gland lipid exprimate is known, but little is known about the phospholipids that comprise the glandular cells. The purpose of the present study is to identify and quantitate the phospholipid complement of the meibomian gland cells that produce the lipid secretion of meibomian oil and which is vital to tear film stability. Eyelids (n = 50) were excised from rabbits, and after surgical removal of surrounding tissues, the tarsal plates with and without expressing meibomian oil were extracted and phospholipids of the plates quantified by 31P nuclear magnetic resonance (NMR). Seventeen phospholipids were quantified from tarsal plates expressed of oil and tarsal plates containing meibomian oil: alkylacylphosphatidylcholine (AAPC), dihydrosphingomyelin (DHSM), dimethylphosphatidylethanolamine, diphosphatidylglycerol (cardiolipin), ethanolamine plasmalogen (EPLAS), lysoethanolamine plasmalogen, lysophosphatidylcholine, lysophosphatidylethanolamine, lysophosphatidylserine, phosphatidic acid, phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylglycerol, phosphatidylinositol, phosphatidylserine, sphingomyelin (SM), sphingosylphosphorylcholine. The six zwitterionic and neutral phospholipids, DHSM, EPLAS, PE, SM, AAPC, and PC together comprise 79.5% of the total meibomian gland phospholipid profile (in meibomian oil this value is 84.2%). The zwitterionic and neutral phospholipids dominate meibomian gland phospholipid profiles. Since the meibomian gland cells undergo holocrine secretion and form the meibomian glad secretion, such a composition is consistent with the hypothesis that a chemically stable lamellar surfactant layer phospholipids bind non-polar meibomian oil to the aqueous layer of the tear film.

Electrophoretic and gel chromatographic analyses of follicle-stimulating hormone in human serum.

The pleomorphism of follicle-stimulating hormone (FSH) in human serum was investigated by gel chromatography on Sephadex G-200 and zone electrophoresis in agarose suspension. The FSH activity was measured by radioimmunoassay. By gel chromatography FSH in sera from 3 males and 5 females of various ages was eluted within a narrow range after albumin and no sex hormone related pleomorphism was found. Electrophoresis in 0.17% agarose suspension was found to be a technique suitable for analysis of differences in charge of FSH in sera from men and women. A total of 31 sera from 25 individuals was investigated. The results strongly indicate that there is in man a sex hormone related pleomorphism of FSH with less acidic forms of FSH in sera of normal women of fertile age than in sera of men and post-menopausal women. Qualitative analysis of FSH in serum by electrophoresis may become an important complement to the quantitative determinations of the FSH concentration for the clinical diagnosis of reproductive disorders and of FSH producing tumours.

Quantitative alleles of CR1: coding sequence analysis and comparison of haplotypes in two ethnic groups.

The quantitative expression of complement receptor type 1 (CR1) on erythrocytes is regulated by two CR1 alleles that differ in having genomic HindIII fragments of either 7.4 or 6.9 kb and that determine high (H allele) or low (L allele) CR1 expression, respectively, across a 10-fold range. To investigate whether the product of the L allele may contain amino acid substitutions that make it more susceptible to proteolysis, cDNA sequence spanning the CR1 coding region was analyzed in two donors who were homozygous for the H and L alleles and differed by 7-fold in their mean numbers of CR1 per erythrocyte. Sequence differences were detected at 10 nucleotide positions, including 6 that would cause amino acid substitutions. The HindIII RFLP and 3 of the latter 6 sites were analyzed in genomic DNA of 85 Caucasians and 75 African Americans; sites encoding the other amino acid substitutions were analyzed less extensively. Two major haplotypes defined prototypic H and L alleles in both ethnic groups, suggesting that these alleles existed before the African and European populations diverged. Decreased erythrocyte CR1 expression is associated with impaired clearance of immune complexes from blood. Persistence of the L allele in all populations that have been analyzed may suggest a compensatory survival advantage, perhaps related to malaria or another infectious disease.

Interactions between smokers and non-smokers in public places: a qualitative study.

Few studies have employed a qualitative approach to better understand how both smokers and non-smokers experience and make sense of no-smoking restrictions in their daily lives. We describe qualitative research involving an examination of self-reported interpersonal interactions between smokers and non-smokers in public places regarding exposure to environmental tobacco smoke (ETS). Five focus group discussions and twenty individual in-depth interviews were conducted with smokers and non-smokers in the Metropolitan Toronto region as a complement to a quantitative survey of the adult population of Ontario. Strategies used by non-smokers in responding to ETS exposures included verbal confrontations, non-verbal cues, use of a third party, and moving away. The perceived consequences of such actions, as described by both smokers and non-smokers, are discussed, together with implications for tobacco control policy and practice.

Number, distribution, and morphologic particularities of encapsulated proprioceptors in pig extraocular muscles.

PURPOSE: To analyze qualitatively and quantitatively the total complement of encapsulated proprioceptors (Golgi tendon organs [GTOs] and neuromuscular spindles) in pig extraocular muscles (EOMs). METHODS: EOMs of four pigs of different ages were prepared for light microscopic histochemical and immunohistochemical analysis and for transmission electron microscopy. RESULTS: GTOs and muscle spindles were numerous in pig EOMs. GTOs were found to be distributed in aponeurotic expansions of the distal and proximal EOM tendons, being more numerous in the distal aponeurosis than in the proximal aponeurosis. The total number of GTOs was higher in the recti EOMs (100-128) than in the oblique EOMs (45-61). Spindles were distributed over the entire muscle length. In each EOM the number of muscle spindles (142-333) exceeded those of GTOs. The morphology of the GTOs was variable. In addition to collagen bundles, approximately one third of the GTOs contained intracapsular muscle fibers that resembled the multiply innervated fiber type. Intracapsular muscle fibers entered the poles of the GTOs and either terminated inside the receptors in collagen bundles or exited the GTOs at the opposite poles. Nerve terminals were numerous in each GTO and established intimate contacts with collagen fibrils. CONCLUSIONS: Most structural particularities formerly observed in GTOs of rhesus monkey and sheep EOMs are also present in GTOs of pig EOMs. The high number of GTOs with their typical nerve terminals indicates functional importance. During muscle activity, afferent signals from GTOs and muscle spindles may provide sufficient information about eye position.

Autoantibodies and related immunity of leprosy patients from leprosarium in Taiwan.

Autoantibodies and related immunological examinations were measured in 60 leprosy patients from a leprosarium in Taiwan. Thirty-one lepromatous type, 24 tuberculoid type and 5 borderline type patients were identified. The measured autoantibodies included antinuclear antibodies, anti-nDNA, anti-cardiolipin and rheumatoid factor. Serum protein electrophoresis and immunofixation were also performed to detect the monoclonal and polyclonal status of immunoglobulins. Circulating immune complex and complements were also quantitated. Delayed type skin tests were performed during patients' visits. A higher frequency of autoantibodies, especially the antinuclear antibodies and anticardiolipin antibodies, were detected in lepromatous type patients. Higher levels of circulating immune complex and frequency of polyclonal and monoclonal gammopathy were also noted in lepromatous type patients. Anergy skin tests were only noted in lepromatous type patients. It was concluded that the more impared cell-mediated immunity in leprosy patients, with lepromatous type in particular, the greater the production of autoantibodies.

Men's role in emergency obstetric care in Osun State of Nigeria.

This study was conducted among the Yoruba of South-West Nigeria to examine the role of men in emergency obstetric care, as men determine whether and when their spouses visit health clinics in most cultures. Simple random sampling was used to select 900 households from three communities in Osun State, south-west Nigeria. Separate interviewers interviewed the man and his wife in each of the households. In polygamous families, two wives of reproductive age were also interviewed. The quantitative survey was complemented with a number of focus group discussions, in-depth interviews and key informant interviews. There was high level of awareness of emergency obstetric conditions by men, particularly in relation to pregnancy signs and labour pains (53.2%). Respondents reported that men play useful roles during their partner's obstetric conditions (89.2%). Women take decisions on health-seeking behaviour during emergency obstetric conditions in the absence of the male partner. Education is found to be the major determinant of this change in male knowledge and behaviour. There is a need to further promote universal basic education in the country especially in areas where the observable change in this study has not been noted. There is also a need to extend the study to other zones in Nigeria in order to have a national picture.

[Serological study of several strains of foot-and-mouth disease virus type "O" isolated in Europe between 1971 and 1975: application of the biomathematical system of classification].

Nine strains of foot-and-mouth disease virus type "O" received in our laboratories since 1971 have been studied serologically by Osler's quantitative method of complement fixation (50% hemolysis). The results, submitted to the biomathematical system of bidimensional classification, allow to conclude that at present in Europe there are two groups of foot-and-mouth disease strains of type "O"; one has reference to our vaccinal strain "O Lausanne 1965" and the other to "O Romania 1972" strain, which has certain points of similarity with subtype "O2". In addition, strain "O Hungary 1975" has been studied on bovines; the immunological tests confirm the serological test.

Blood--artificial surface interactions during cardiopulmonary bypass. A comparative study of four oxygenators.

Evaluation of the biocompatibility of four different types of oxygenator (bubble, membrane, hollow fibre and 'hybrid') was performed on 26 patients undergoing cardiopulmonary bypass during elective coronary surgery. More platelet derangement and an increased degree of hemolysis, revealed by higher plasmatic concentration of beta-thromboglobulin, platelet factor 4 and plasmatic free hemoglobin (p less than 0.05), was seen when using the bubble oxygenator. Damage to blood cells was minimal with the membrane oxygenator while the 'hybrid' and the hollow fibre oxygenators proved to rank at an intermediate level. Complement activation at the beginning of the cardiopulmonary bypass occurred via the alternative pathway as demonstrated by C3ades arg increase (up to nine times) without a concomitant elevation of C4ades arg. Cardiopulmonary bypass complement activation was quantitatively similar with all the oxygenators. A further activation via the classical pathway occurred in all the patients after protamine injection. Consistent differences as far as clinical and biological effects exist among the various commercially available cardiopulmonary bypass apparatus; our study provides guidelines for the evaluation and selection of devices which might reduce postoperative sequelae.

Differential antigenic expression of the DBA/2 lymphoma L1210 and its sublines: cross-reactivity with C3H mammary tumors as defined by syngeneic monoclonal antibodies.

The expression of tumor-associated antigens on the DBA/2 lymphoma L1210 and three L1210 sublines, each resistant to a different antileukemic agent [guanazole, methylglyoxal-bis(guanylhydrazone), and 4,4-diacetyldiphenylurea-bis(guanylhydrazone)] was investigated by the use of monoclonal hybridoma antibodies. Hybridomas were produced by the fusion of spleen cells from DBA/2 mice immunized with irradiated L1210 or L1210 subline cells and cells of a non-immunoglobulin-secreting BALB/c myeloma variant. Three clones producing antibodies reacting with L1210 or L1210 subline cells were used to study the antigenic expression of L1210 and L1210 subline cells. Monoclonal antibodies from anti-L1210 and anti-L1210 subline hybridomas exhibited a greater reactivity with L1210 subline cells than with L1210 cells in complement-dependent cytotoxicity, quantitative absorption, and membrane immunofluorescence experiments, thereby demonstrating a tumor-associated antigen shared by L1210 and the L1210 sublines and an increased expression of this antigen on subline cells. Cross-blocking tests of antibody binding demonstrated that monoclonal antibodies from anti-L1210 and anti-L1210 subline hybridomas recognized the same or very closely situated antigenic determinants on the tumor cell surface. Most syngeneic and allogeneic tumor cells used as controls failed to react with the anti-L1210 and anti-L1210 subline hybridoma antibodies. However, two syngeneic tumors, L5178Y and P388-D1, demonstrated a significant reaction with the monoclonal antibodies. In addition, several spontaneous mammary tumors from C3H/St and DBA/2Ha, both high-frequency mammary tumor strains, reacted in various degrees with anti-L1210 or anti-L1210 subline hybridoma antibodies in absorption tests and in immunofluorescence experiments. On the other hand, liver, kidney, and spleen from normal C3H/St mice, as well as mammary tumors from BALB/c and C3Hf, both low-frequency mammary tumor strains, did not demonstrate significant reactivity in similar experiments. Normal lactating mammary glands from high-frequency mammary tumor mouse strains reacted with the monoclonal antibodies, whereas lactating mammary glands from low-frequency mammary tumor mouse strains were negative by this method. Purified murine mammary tumor virus preparations reacted strongly with the monoclonal antibodies in solid-phase radioimmunoassays, whereas a purified murine leukemia virus preparation failed to do so in similar experiments. These results indicate that the tumor-associated antigen(s), differentially expressed on L1210 and L1210 subline cells, is related to an antigen which is associated with the murine mammary tumor virus.

Immunologic studies in pneumococcal disease.

Many patients die from pneumococcal disease despite the availability of effective antimicrobial agents. Immunologic studies including detection, typing, and quantitation of serum pneumococcal capsular polysaccharide (PCP) antigen by counterimmunoelectrophoresis (CIE), quantitation of PCP antibody by radioimmunoassay (RIA), and quantitation of serum complement components C3, C4, and C3PA and serum immunoglobulins IgG, IgM, and IgA by the radial immunodiffusion technique of Mancini were performed with the sera of 18 patients. Five patients died (group I), and 13 survived (group II) pneumococcal infection. Both groups were comparable in age, underlying disease, and leukopenia on admission. All patients of group I and 10 of 13 (77%) of group II patients were bacteremic. Two patients in each group had an extrapulmonary focus infection. PCP antigen was detected in the sera of all group I and nine of 13 group II patients. PCP antigen levels were larger than or equal to 15 microng/ml in four of five group I and two of 13 group II patients (p = 0.022). Levels of antibody to PCP exceeded 100 ng/ml of antibody nitrogen (AbN) in 10 of 12 group II and one of five group I patients (p = 0.027) during the course of illness. All group I patients and three of 12 group II patients had decreased levels of one or more complement components on admission (p less than 0.01). One or more complement components remained decreased until death in four group I patients but returned to normal or elevated levels in all group II patients. No difference in serum immunoglobulin concentrations were found.

[Detection of IgG fraction of intact rabbit serum interacting with chromatin].

It was shown that the intact rabbit sera interact with chromatin and thymocyte nuclei in a quantitative reaction of complement fixation and immunoluminescence analysis, respectively. Purified immunoglobulins G (IgG) and Fab fragments isolated from them were also found capable of such interactions. The interactions are not caused by Ig aggregate absorption on chromatin and can be due to the presence of natural antibodies against chromatin in the 7S fraction of intact rabbit IgG. These IgG possess heterogeneous charges, presumable due to the presence of antibodies against various antigenic chromatin determinants in rabbit serum.

The impact of health care market changes on local decision making and STD care: experience in three counties.

INTRODUCTION: In 1993 health care reform including universal coverage appeared imminent. Some county health departments elected to discontinue provision of direct services including sexually transmitted disease (STD) care. County A moved in this direction with final clinic closure in 1996. Coincidentally, two other counties elected to continue their STD treatment services. These events have created a "natural experiment" in which to evaluate the contrasting strategies among three counties. METHODS: This report describes the changes in local delivery over a three-year period (1993-1995). Measurements were carried out in three counties in two states. STD program capacity, service delivery, and morbidity rates for STDs (chlamydia and gonorrhea) were monitored in each study county. Quantitative data were complemented by a qualitative patient survey at each site. RESULTS: Capacity changed dramatically in County A as compared with the other two over the three-year period. Major declines in STD clinic visits (-43%) and laboratory testing (-46%) occurred. A major drop in reported STD incidence (-23% for chlamydia and -49% for gonorrhea) also occurred, including a drop in public provider reporting. CONCLUSIONS: In County A, a decision to end delivery of personal health services led to a divestiture in STD service delivery and a decline in all measured parameters. Etiology of concurrent declined in reported STDs is unclear. Possibly decreased public services led to a spurious decline in reporting. Alternatively, an overall strategy of shifting care to private providers has succeeded in reducing disease. Public health surveillance may be less accurate in such settings.

Complement factor adsorption on solid surfaces--an ellipsometric method for investigation of quantitative aspects.

An optical method, ellipsometry, has been used for quantification of organic material adsorbed from complement sufficient sera on antibody coated solid surfaces. Maximal adsorption of organic material from complement sufficient human sera occurred at about 0.5 micrograms/cm2 of IgG. C3 but not C5, C8 or C9 was detected on the antibody surface incubated with complement sufficient sera. This may indicate that IgG adsorbed on methylized silicon surfaces lack binding sites for complement factors beyond C3. A modification of the method was also used for quantification of migration inhibition of human polymorphonuclear leucocytes (PMNL). Locomotion inhibition fell in a sharp interval from 0.2 to 0.5 micrograms/cm2 of IgG on the surface. We believe that the suggested type of measurements is important for understanding the quantitative relationships between humoral effects such as antibody dependent complement activation and cellular effects such as migration of PMNL.

The quantitative genetic basis of male mating behavior in Drosophila melanogaster.

Male mating behavior is an important component of fitness in Drosophila and displays segregating variation in natural populations. However, we know very little about the genes affecting naturally occurring variation in mating behavior, their effects, or their interactions. Here, we have mapped quantitative trait loci (QTL) affecting courtship occurrence, courtship latency, copulation occurrence, and copulation latency that segregate between a D. melanogaster strain selected for reduced male mating propensity (2b) and a standard wild-type strain (Oregon-R). Mating behavior was assessed in a population of 98 recombinant inbred lines derived from these two strains and QTL affecting mating behavior were mapped using composite interval mapping. We found four QTL affecting male mating behavior at cytological locations 1A;3E, 57C;57F, 72A;85F, and 96F;99A. We used deficiency complementation mapping to map the autosomal QTL with much higher resolution to five QTL at 56F5;56F8, 56F9;57A3, 70E1;71F4, 78C5;79A1, and 96F1;97B1. Quantitative complementation tests performed for 45 positional candidate genes within these intervals revealed 7 genes that failed to complement the QTL: eagle, 18 wheeler, Enhancer of split, Polycomb, spermatocyte arrest, l(2)05510, and l(2)k02206. None of these genes have been previously implicated in mating behavior, demonstrating that quantitative analysis of subtle variants can reveal novel pleiotropic effects of key developmental loci on behavior.

Clonal selection and in vivo quantitation of protein interactions with protein-fragment complementation assays.

Two strategies are described for detecting constitutive or induced protein-protein interactions in intact mammalian cells; these strategies are based on oligomerization domain-assisted complementation of rationally designed fragments of the murine enzyme dihydrofolate reductase (DHFR; EC 1.5.1.3). We describe a dominant clonal-selection assay of stably transfected cells expressing partner proteins FKBP (FK506 binding protein) and FRAP (FKBP-rapamycin binding protein) fused to DHFR fragments and show a rapamycin dose-dependent survival of clones that requires approximately 25 molecules of reconstituted DHFR per cell. A fluorescence assay also is described, based on stoichiometric binding of fluorescein-methotrexate to reconstituted DHFR in vivo. Formation of the FKBP-rapamycin-FRAP complex is detected in stably and transiently transfected cells. Quantitative rapamycin dose-dependence of this complex is shown to be consistent with in vitro binding and distinguishable from a known constitutive interaction of FKBP and FRAP. We also show that this strategy can be applied to study membrane protein receptors, demonstrating dose-dependent activation of the erythropoietin receptor by ligands. The combination of these clonal-selection and fluorescence assays in intact mammalian cells makes possible selection by simple survival, flow cytometry, or both. High-throughput drug screening and quantitative analysis of induction or disruption of protein-protein interactions are also made possible.
