Production of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) by high-cell-density cultivation of Aeromonas hydrophila.

The newly screened Aeromonas hydrophila produces copolymer consisting of 3-hydroxybutyrate (3HB) and 3-hydroxyhexanoate (3HHx). The characteristics of cell growth and polymer accumulation were examined using various carbon sources. P(3HB-co-3HHx) was produced from lauric acid and oleic acid only. P(3HB-co-3HHx) content can be increased by limitation of phosphorus. A maximal P(3HB-co-3HHx) content of 28.8 wt% could be obtained in flask culture. By applying the optimally designed nutrient feeding strategy, cell dry weight, P(3HB-co-3HHx) content, and 3HHx fraction obtained over the course of 43 h were 95.7 g/L, 45.2 wt%, and 17 mol%, respectively, resulting in a productivity of 1.01 g polyhydroxyalkanoate (PHA)/L. h.

Comparison of thermal characteristics and degradation properties of epsilon-caprolactone copolymers.

The objective of this study was to enhance the degradation of the homopolymer of epsilon-caprolactone (epsilon-CL) through its copolymerization with a second lactone monomer: delta-valerolactone (delta-VL), gamma-butyrolactone (gamma-BL), gamma-valerolactone (gamma-VL), and gamma-caprolactone (gamma-CL). The influences of the molar ratio and the structure of the second lactone monomers on the molecular weight, melting temperature, crystallinity, and degradation rate of the copolymers were compared and elucidated. The enhancement of degradation of the epsilon-CL polymers was achieved by the addition of a second lactone monomer during the copolymerization process, which prominently depended on the initial molar ratio and the chemical structure of the monomers. The changes in the molecular weight and the melting temperature and the enthalpy of fusion of the copolymers were directly related to the initial molar ratio of two monomers, which further affected the magnitude of the enhancement of the polymer degradation. The degradation rates of copolymers of epsilon-CL/delta-VL with initial molar ratios of 75/25, 80/20, and 85/15% were 7.72, 3.60, and 2.05%/day, respectively. However, the degradation rate of the homopolymer of epsilon-CL was 1. 85%/day. The degradation rates of copolymers of epsilon-CL and three five-member ring lactone monomers were in the order of gamma-BL > gamma-VL > gamma-CL. The number of carbon atoms in the side chain of the lactone monomers determined the hydrophobicity of the polymers, which was not favorable for water penetration causing degradation.

Functional assessment of sciatic nerve reconstruction: biodegradable poly (DLLA-epsilon-CL) nerve guides versus autologous nerve grafts.

The aim of this study was to compare functional nerve recovery after reconstruction with a biodegradable p(DLLA-epsilon-CL) nerve guide filled with modified denatured muscle tissue (MDMT), or an autologous nerve graft. We evaluated nerve recovery using walking track analysis (measurement of the sciatic function index [SFI]) and electrostimulation tests. Functional nerve recovery after reconstruction with a biodegradable p(DLLA-epsilon-CL) nerve guide filled with MDMT was faster when compared with nerve reconstruction using an autologous nerve graft. We conclude that in case of a short nerve gap in the rat, reconstruction can best be carried out using a p(DLLA-epsilon-CL) biodegradable nerve guide filled with MDMT.

Environmental modulation of the inhibitory action of D-mannoheptulose upon D-glucose metabolism in isolated rat pancreatic islets.

In pancreatic islets prepared from fed rats and incubated at a low concentration (1.7 mM) of D-glucose, D-mannoheptulose (10.0 mM) virtually fails to affect the metabolism of the hexose. Likewise, in islets from starved rats, the relative extent of the inhibitory action of D-mannoheptulose upon D-glucose metabolism is much more marked at high (16.7 mM) than low (1.7 mM) hexose concentration. Nevertheless, despite decreasing the metabolism of D-glucose, starvation augments the sensitivity to D-mannoheptulose in the islets incubated at a low concentration of the hexose, D-galactose, but not D-fructose, also augments the inhibitory action of D-mannoheptulose upon D-glucose metabolism in islets prepared from fed rats and exposed to the low concentration of D-glucose. A comparable situation prevails in islets exposed to 2-ketoisocaproate. Forskolin, however, which decreases D-glucose catabolism in the islets from fed rats exposed to 1.7 mM D-glucose, fails to affect significantly the inhibitory action of D-mannoheptulose on D-glucose metabolism. It is proposed that hexoses transported by the same carrier as D-glucose and non-glucidic nutrient secretagogues somehow increase D-mannoheptulose uptake by the islet cells. The latter two conditions may be operative in islets exposed to a high concentration of D-glucose, this accounting for the exquisite sensitivity to D-mannoheptulose of glucose-stimulated islets.

Disorder and twinning in molecular crystals: impurity-induced effects in adipic acid.

The variation in physical properties of crystals grown in the presence of additives or impurities have previously been attributed to lattice disorder developed during crystallization. Adipic acid crystallized in the presence of a variety of stereochemically related impurities typifies such behavior with disorder manifest in variations of dissolution rates and enthalpies of solution and fusion. In this case the most extreme habit, produced by the presence of added monoalkanoic acids, is a rounded dumbbell that was suggested previously to be a twinned crystal. In this contribution such crystals are fully characterized both through their external morphology and by means of single crystal X-ray diffraction. These techniques show that these particles are not twinned but rather are disordered single crystals comprising a small number of slightly misaligned domains. The interaction between additive and substrate is modeled and new additives selected that induce the formation of true mechanical twins in adipic acid.
