A quantitative analysis of stereotyped gnawing induced by apomorphine.

An apparatus was designed and constructed to enable a quantitative analysis of the stereotyped gnawing produced by the dopamine agonist apomorphine. Using this apparatus it was discovered that increasing the subcutaneous dose of apomorphine increased (1) the number of animals that gnawed, and (2) the duration of gnawing in those animals that gnawed at all doses. Other aspects of apomorphine-induced gnawing, in particular the latency to respond and the frequency and duration of individual gnaws, were relatively unaffected. Likely properties of the system responsible for the organization of sterotyped gnawing are discussed.

A quantitative analysis of alterations in the shape of cultured fibroblasts induced by tumour-promoting phorbol ester.

Quantitative analysis of cell size and shape has shown that the tumour promotor 12-o-tetradecanoylphorbol-13-acetate (TPA) alters one of three parameters of cell shape, dispersion. It has no effect on cell area or elongation. This reflects the fact that TPA causes cells to form long narrow processes extending from the cell body. The ability to measure alterations in specific parameters of cell shape is important for correlating changes in the observed behaviour of cells with effects on cytoskeletal organisation.

Quantitative analysis of outflow pathway of corpora cavernosa by pressure flow technique.

Using a pressure flow technique, quantitative analysis of the physiological characteristics of the outflow pathway of the corpus cavernosum was carried out in 19 male dogs weighing 7.5 to 23.0 kg. Pressure flow curves were made on dogs whose pelvic nerve was stimulated electrically and on dogs left unstimulated. When a cyclical change in saline perfusion rate was applied without nerve stimulation, the variable of the intracorporeal pressure showed a large hysteretic loop, indicating that the resistance of the outflow canals to flow was altered by the distension of the sinusoidal space. In dogs whose pelvic nerve was stimulated, the pressure flow curves shifted to the left side in comparison with the outward phase of the pressure flow curve of animals without pelvic nerve stimulation, and this curve piled on the returning phase. No hysteretic relation was observed between the outward and returning phase of the pressure flow curve with pelvic nerve stimulation, but in the detailed analysis, in which the % flow rate was used instead of actual flow rate of saline perfusion, a small hysteretic loop based on the difference of the elasticity of the outlet canals was found. The distension of the corpora cavernosa and the pelvic nerve electrostimulation probably act as the triggers of the same occlusive mechanism in the outflow pathway. The percentage decrease in the blood flow in the outflow canal of the corpus cavernosum induced by the distension of the sinusoidal space or by the pelvic nerve electrostimulation was 69.6 +/- 14.4% (mean +/- SD).

The standardization of hyperventilation on EEG recording in childhood. II. The quantitative analysis of build-up.

In thirty-seven children free of neurological symptoms, we attempted the quantitative analysis of EEG slowing during standarized hyperventilation activation, respiration rate of 30/min, a three-fold elevation of VE and duration of 4 minutes. The degree of build-up gradually became mild with increasing age and no build-up was observed in the subjects older than 14 years old. Fast Fourier Transform analysis of the build-up showed that a big build-up was due to both increasing delta and theta power, and markedly decreasing alpha power with markedly increasing total power, and the mild build-up was due predominantly to increasing theta power. These changes disappeared, the levels before the activation being attained again, within one minute after the hyperventilation in almost all subjects. As to age differences in EEG slowing during hyperventilation, an increase in both delta and theta activity was observed in the younger subjects (6-9 years old), and a mild increase in theta activity was predominantly observed in adolescents. On topographic analysis of EEG slowing, the delta power was found to have expanded to the posterior area and the theta power to have expanded to become diffuse. The standardization of hyperventilation in childhood is important for judgement of the build up because of these age differences, and also may be useful for understanding the development of the central nervous system.

Quantitative analysis of one-dimensional gel electrophoresis profiles.

A general method for the one-dimensional quantitative analysis of band profiles from an autoradiographed or stained gel is described. Curved lanes may be tracked, noise across the width of the band reduced by local averaging of densities within a selected window, and background-corrected density profiles produced. Sections of each profile can be subjected to a choice of three methods of peak-area determination, depending on the type of gel and the requirements of the analysis being carried out. In the first method, simple integration is used to measure the area under the profile between any two positions. The second method integrates a Gaussian distribution fitted to a single peak. Where bands overlap, the third method resolves peaks into their component areas, taking into account the observed increase in the width of the band in the direction of electrophoretic migration. This is incorporated into a least-squares profile-fitting method, which is shown to be both accurate and efficient.

[Quantitative analysis of methoxylated flavonoids in iyo (Citrus iyo) peels by high-performance liquid chromatography].

The quantitative analysis of eight kinds of methoxylated flavonoids in Citrus iyo Hort. ex Tanaka (Rutaceae) was examined by high-performance liquid chromatography. These compounds could be determined within 50 min on C-18 reversed-phase column. Elution system was carried out with a mixture of 1,4-dioxane-water-acetic acid in ratios of 2:7:1 and 7:2:1 in the form of a gradient from 8:2 to 2:8 for 60 min.

Quantitative analysis of dermatoglyphics. (2) Dermatoglyphic patterns.

Using medical students as controls, we devised a method of quantitative analysis of dermatoglyphic patterns. We calculated the frequency of appearance of each of the patterns and obtained the probability of occurrence of the dermatoglyphics of each of the controls. We showed usefulness of this method by presenting some examples of congenital anomalies where this has been put to use. Furthermore, patients with congential heart disease were compared with controls by this method.

Quantitative analysis of the velopharyngeal sphincter function during speech.

OBJECTIVE: This study reports a new concept for quantitative analysis of velopharyngeal function using electromagnetic articulography and simultaneous transoral video endoscopy. PARTICIPANTS: The velopharyngeal closure mechanism was studied in 16 healthy German subjects during production of the vowel-consonant-vowel sequences /afa/ and /apa/. DESIGN: Velar movements in the sagittal and vertical direction were measured by electromagnetic articulography (EMA) while video endoscopy was used for recording in the sagittal and transverse directions. MAIN OUTCOME MEASURE: A transverse sagittal quotient (TSQ) was defined after the measurement of sagittal and transverse diameters of the velopharyngeal port (VPP) in the video-endoscopic pictures on the basis of a frame-by-frame analysis. RESULTS: The sphincter morphology was objectively determined with the TSQ base value. A terminal sphincter deformation (TSD) for closure phases of articulatory movement was revealed by a qualitative shape analysis of the TSQ curves. CONCLUSIONS: The assessment of the TSQ of velopharyngeal port diameters and the TSD in the terminal closure period lead to a new dynamic interpretation of velopharyngeal closure patterns.

[Quantitative analysis of head-shaking nystagmus of peripheral vestibular origin in conjunction with stimulus intensity].

A quantitative analysis of horizontal head-shaking nystagmus (HSN) was made on 48 patients with unilateral peripheral vestibular disorders in conjunction with stimulus intensity (the number of head-shakes). Each patient underwent three head-shaking tests with 10, 30 and 50 horizontal head-shakes, with a rapidity of about 2 Hz, in random order. HSN was recorded by ENG with the eyes open in a completely dark room, and various components of nystagmus were measured manually. 1. HSN appeared in a biphasic or monophasic pattern. In some patients with biphasic HSN, the 2nd phase disappeared as the stimulus intensity diminished. 2. The maximal slow-phase eye velocity (MSV) of the 1st phase of biphasic HSN was significantly greater than that of monophasic HSN. The tendency to increase in proportion to stimulus intensity was also greater in biphasic than in monophasic HSN. The MSV values of the 2nd phase of biphasic HSN were smaller than in the 1st phase (less than one-third) at all stimulus intensities. 3. The duration and total number of HSN beats were greater in the 2nd phase of biphasic HSN and increased markedly in proportion to stimulus intensity. In contrast, in the 1st phase of biphasic HSN and in monophasic HSN, the values of these nystagmus components were smaller, there was no clear relationship with stimulus intensity, and when the stimulus intensity was high, they were instead significantly smaller in the 1st phase of biphasic HSN than in monophasic nystagmus. 4. As the stimulus intensity rose to a high level, the interval between the 1st and 2nd phases of biphasic HSN (2nd phase latency) shortened, and the 2nd phase tended to appear more quickly after head-shaking. 5. The time constant of the decline in slow-phase eye velocity was nearly constant in both the 1st phase of biphasic HSN and monophasic HSN, regardless of stimulus intensity. It was especially noteworthy that in response to an increase in stimulus intensity, both the MSV in the 1st phase and the duration of the 2nd phase of biphasic HSN increased, while the duration of the 1st phase of biphasic HSN was inversely suppressed by the 2nd phase.

[Quantitative analysis of the interaction of low-density lipoproteins with perfused rabbit aorta].

A method for quantitative analysis of interaction between 125I-LDL with perfused rabbit vessels in situ was developed. The method suggested enables the preservation of the integrity of the endothelium in the perfused vascular segment and the study of 125I-LDL incorporation into the affected vessels within the concentration ranges that make possible specific incorporation of LDL into cells. The method can be applied to studying the mechanisms and modulations of LDL uptake by the vascular wall in situ.

Quantitative analysis of left ventricular regional wall motion with color kinesis during abdominal aortic cross-clamping.

OBJECTIVES: The authors aimed to establish a technique for quantitative analysis of regional wall motion abnormality (RWMA) using color kinesis (CK) of transesophageal echocardiography (TEE) in surgical patients. This technique was used to determine whether RWMAs develop de novo after infrarenal aortic cross-clamping in patients undergoing vascular surgery with a preoperative dipyridamole thallium stress test (DTST). DESIGN: An observational study. SETTING: University hospital. PARTICIPANTS: Thirty-eight patients undergoing infrarenal abdominal aortic aneurysm resection or aortofemoral bypass. MEASUREMENTS AND MAIN RESULTS: CK images of the left ventricle (LV) were obtained from the midventricular transgastric short-axis view before and after infrarenal aortic cross-clamping using TEE and analyzed off-line using custom software. The predictive value of the category "reversible perfusion defect" (RD) was also estimated from DTST for predicting new RWMAs with CK. CK analysis is suitable for clinical use based on the comparison with conventional two-dimensional echocardiogram measurements and interobserver variability. CK analysis showed all 7 patients with persistent perfusion defects on DTST had RWMAs. New RWMAs occurred in 2 of 9 patients with RD and in 2 of 15 patients with normal DTST, indicating that there was no significant difference between RD and normal DTST in the incidence of new RWMAs. CONCLUSIONS: A new method is available for clinical use, which is capable of visualizing RWMAs. These results suggest that new RWMAs introduced by aortic cross-clamping occur irrespective of the risk as assessed by preoperative DTST. CK with the new analysis method might be a useful tool to quantitatively evaluate RWMAs during surgery.

Quantitative analysis of interconnectivity of porous biodegradable scaffolds with micro-computed tomography.

Pore interconnectivity within scaffolds is an important parameter influencing cell migration and tissue ingrowth needed to promote tissue regeneration. Methods for assessment of interconnectivity are usually qualitative, restricted to two-dimensional images, or are destructive. Microcomputed tomography nondestructively provides three-dimensional (3D) images of intact specimens at high spatial resolutions. We describe an image analysis technique for quantitative assessment of scaffold interconnectivity. Scaffolds were made via a particulate leaching process with 75%, 80%, 85%, and 88% volumetric porogen fractions. Specimens were scanned and resulting 3D, digital images were analyzed with a custom algorithm. A series of virtual, idealized scaffolds were also created for illustration of the algorithm's analysis approach and for its validation. The program calculated accessible void fractions over a range of minimum connection sizes. In real specimens, nearly 100% of the porous volume was connected with outside air for connections greater than or equal to 20 microm in their smallest dimension. In scaffolds made with 75% porogen, the accessible void fraction decreased to 78% if only those connections greater than or equal to 260 microm were considered. The relationship between accessible void fraction and connection size varied as a function of porogen content. The interconnectivity parameter described here may have implications for cell migration and tissue growth into scaffolds.

A STORM-based protocol for nanoscale imaging and quantitative analysis of protein-associated and phospholipid-associated structures in natural rubber.

Stochastic Optical Reconstruction Microscopy (STORM) enables nanoscale mapping of molecular components beyond the diffraction limit; however, its reproducible implementation in hydrophobic polymer matrices remains challenging because fluorescence-labeling specificity, fluorophore photoswitching, three-dimensional localization, chromatic registration, and quantitative image analysis must be carefully controlled. This protocol presents a standardized experimental workflow for dual-color labeling, astigmatism-based three-dimensional STORM acquisition, and quantitative analysis of protein-associated and phospholipid-associated structures in natural rubber (NR). The workflow covers sample pretreatment, Cy5 NHS ester labeling of protein-associated primary amines, DiI labeling of phospholipid-rich domains, STORM imaging-buffer preparation, three-dimensional single-molecule localization, dual-channel registration, generation of standardized xy projections, aggregate-size analysis, and projected lateral spatial correlation assessment. Reproducibility is supported by defined acquisition and localization criteria, three independent sample preparations with at least five fields of view analyzed per condition, and unlabeled, single-color, dye-only matrix, and processing-associated Cy5 controls. Mean lateral localization precisions of 11.8 ± 2.3 nm for Cy5 and 13.5 ± 2.9 nm for DiI were obtained, while two-dimensional Fourier ring correlation analysis of the xy projections yielded effective lateral image resolutions of approximately 25 and 28 nm, respectively. Image-based particle segmentation and localization-coordinate-based density-based spatial clustering of applications with noise (DBSCAN) were applied to standardized xy projections as complementary quantitative approaches. Application of the protocol to untreated, centrifuged, and protease-treated NR samples demonstrated treatment-associated changes in the detected abundance and projected size distributions of protein- and phospholipid-associated aggregates, together with a non-monotonic change in their projected lateral spatial correlation. These observations describe alterations in nanoscale organization but do not, by themselves, establish stable protein-phospholipid complex formation. Unlike previous studies that primarily demonstrated the feasibility of STORM imaging in rubber materials, the principal contribution of this work is an end-to-end, step-by-step protocol incorporating defined controls, three-dimensional localization, image-quality metrics, chromatic-registration procedures, and complementary quantitative-analysis pipelines for non-expert users. The workflow may be adaptable to other hydrophobic polymers and soft-material systems after appropriate optimization and validation.

[An algorithm for real-time quantitative analysis of remote detection spectrum of chemical vapor with passive Fourier-transform infrared spectroscopy].

The algorithm for quantitative analysis of passive Fourier-transform infrared spectum (FTIR) of a chemical vapor is given. It can be applied to both real-time processing for chemical vapor and quantitative measurements of chemical vapor column density. The algorithm simultaneously achieves quantitation of the vapor spectral transmission, background removal, real-time calibration for FTIR. Finally, we discuss in detail an example of the use of the algorithm to a simulant DMMP.

Quantitative analysis of low-field NMR signals in the time domain.

Two novel methods are described for direct quantitative analysis of NMR free induction decay (FID) signals. The methods use adaptations of the generalized rank annihilation method (GRAM) and the direct exponential curve resolution algorithm (DECRA). With FID-GRAM, the Hankel matrix of the sample signal is compared with that of a reference mixture to obtain quantitative data about the components. With FID-DECRA, a single-sample FID matrix is split into two matrices, allowing quantitative recovery of decay constants and the individual signals in the FID. Inaccurate results were obtained with FID-GRAM when there were differences between the frequency or transverse relaxation time of signals for the reference and test samples. This problem does not arise with FID-DECRA, because comparison with a reference signal is unnecessary. Application of FID-DECRA to 19F NMR data, which contained overlapping signals from three components, gave concentrations comparable to those derived from partial least squares (PLS) analysis of the Fourier transformed spectra. However, the main advantage of FID-DECRA was that accurate (<5% error) and precise (2.3% RSD) results were obtained using only one calibration sample, whereas with PLS, a training set of 10 standard mixtures was used to give comparable accuracy and precision.

Computer-assisted liquid chromatography for automated qualitative and quantitative analysis of toxic drugs.

A computer-assisted liquid chromatographic system (MCASYST) is evaluated for automatic qualitative and quantitative analysis of toxic drugs in poisoned human fluids. The system performance is confirmed by the actual analyses of several cases of poisoning. The results indicate that the accuracy of identification by retention prediction and UV spectral search is very high, and quantitation by the memorized calibration curves can be performed very conveniently. However, it is difficult at present to identify all peaks that appear in chromatograms for human urine and serum because the drugs may be changed to their metabolites in those fluids. Metabolite identification will be the next step to improve the performance of this system.

[The clinical value of quantitative analysis of plasma Epstein-Barr virus DNA in patients with nasopharyngeal carcinoma].

OBJECTIVE: To elucidate the clinical value of quantitative analysis of plasma cell-free EBV-DNA in patients with nasopharyngeal carcinoma (NPC) . METHODS: Using fluorescence quantitative polymerase chain reaction (FQ-PCR), the copies of EBV-DNA in 66 plasma samples of pre- and post-treated NPC patients were examined, and compared with 30 normal controls. The relationships between the copies of free EBV-DNA and tumor stage, metastases load, as well as short-term therapeutic response were analyzed. RESULTS: The positive percentage and copies of plasma EBV-DNA of NPC patients (66.7%, Median 522.0) were significantly higher than those of normal controls (10.0%, Median 0). Among the pre-treated patients, plasma EBV-DNA copies in stage III-IV patients (Median 3130.0) were higher than those were in stage I-II (Median 400.0), and the copies of N2-3 patients (Median 3250.0) were higher than those were in N0-1 patients (Median 400.0). And then the level of post-treated patients (Median 0) was lower than that of pre-treated patients (Median 522.0). Depending on the evaluation of therapeutic response, post-treated patients could be classified as two different categories, patients whose tumor regressed satisfyingly and those with remaining lesions. It was found that EBV-DNA copies of the former (Median 0) were lower than those of the latter (Median 2083.0). CONCLUSIONS: FQ-PCR determination of plasma EBV-DNA may become a promising marker to monitor the course of this disease.
