Fourier transform infrared spectra of aqueous protein mixtures using a novel attenuated total internal reflectance cell with infrared fibers.

A specially designed cell containing a silver halide (AgBr:AgCl) infrared fiber allows convenient and reproducible loading of viscous protein solutions and suspensions. Attenuated total internal reflectance measurements using an FTIR spectrometer were made for bovine serum albumin-water past. Dynamic changes in the protein films are readily followed, a technique which should be generally useful. A band assigned to a secondary structural feature, the alpha-helix, is similar in intensity to that reported (T.M. Fong, M.G. McNamee, submitted for publication).

Hypersensitive teeth. Part II: Treatment.

If the hydrodynamic theory for pain transmission were accepted, occlusion of the patent dentinal tubules would appear to be essential for treatment efficacy. There are many compounds with seemingly varied chemical forms that have been shown to be effective. Their exact mode of action however, is not clearly defined because well-designed, nonbiased, and controlled comparison studies between agents are lacking. The various toothpastes may have ingredients that actually occlude patent tubules or they may cause secondary desensitization by irrational or abrasive action. In any pain study, the nature of the placebo effect and other psychogenic factors play a significant role. Fluoride preparation with and without iontophoresis has been shown to alter tubule structure and form microprecipitates. The natural desensitization process, although slow, is nature's protection, allowing dentinal sclerosis of secondary dentin formation. Although the resin-adhesive systems, especially the new light-cured dentin bonding agents, appear immediately to be effective, the effect on the pulp remains unknown. Perhaps a combination of iontophoresis with sodium fluoride and light-cured dentin bonding material may yield protection and desensitization at a high level of predictability. With the population trends toward a more geriatric society, further research, knowledge, and understanding of dentinal hypersensitivity is of paramount importance. The expected increase in longevity of the dentition suggests that dentin exposure and sensitivity will increase as a clinical problem. There is a clear time-age relationship involved in gingival recession, erosion and attrition of the teeth, and the need for periodontal surgical therapy. For total comprehensive care, patient comfort is important and should be provided along with sound periodontal health and ideal restorative function.

Fracture durability of restored functional cusps on maxillary nonvital premolar teeth.

Palatal cusps of nonvital maxillary premolar teeth were restored with a coronal-radicular restoration of posterior composite resin (Occlusin), amalgam (Tytin), or cermet (Chelon-Silver). Mean fracture strength values were: 370 lb for intact premolar teeth, 266 lb for amalgam, 215 lb for posterior composite resin, and 132 lb for cermet. From this in vitro study it can be suggested that for selected endodontically treated maxillary premolar teeth, the dentist can use acid-etch-retained posterior composite resin or pin-retained high copper amalgam material for a definitive coronal-radicular restoration. The use of cermet for this purpose is contraindicated.

Factors involved in the high radiographic sensitivity of E-speed films.

A microscopic and densitometric analysis of the high speed and fog density of E-speed film was conducted. The grain sizes of silver bromide (AgBr) and developed silver metals were measured, and the optical densities of the film were compared by means of various development procedures. Silver bromide grains were found to be large and accounted for the high speed of the film. Development caused chains of silver metal clusters to form, which contributed to the larger fog density as well as the higher speed.

Convenient synthesis of 18-hydroxylated cortisol and prednisolone.

18,20-Epoxy-11 beta,17 alpha,20 beta,21-tetrahydroxypregn-4-en-3-one was synthesized by the application of hypoiodite reaction to the cortisol acetonide. The intermediary 18-iodo derivative was converted to the 11-oxo steroid by chromic acid prior to silver ion-assisted solvolysis. Removal of the protective group with hydrochloric acid was finally carried out to give the desired 11 beta,17 alpha,18,21-tetrahydroxypregn-4-ene-3,20-dione as the hemiacetal form. 18,20-Epoxy-11 beta-17 alpha,20 beta,21- tetrahydroxypregna-1,4-dien-3-one was also prepared from prednisolone through a similar reaction sequence.

Explosive nephrolithopaxy: reality or fiction?

The use of silver and lead azide explosive charges for the percutaneous distintegration of renal calculi has been investigated. Charges of 10 mg or more reliably reduced calculi to fragments of extractable size; however, the concomitant tissue effects would preclude the use of such charges clinically. Smaller charges require multiple applications. High-speed flash photography demonstrated the unfocussed nature of these discharges. Considerable improvements are anticipated when the shock waves are focused. The use of Nonel tubing is described, and future developments are discussed.

Recent results of the joint ESA-DARA/IBMP experiments Biokosmos "Seeds".

Comparison of experimental data obtained from short (SDEF) and long duration exposure flights (LDEF) have recently led to results which will be significant for longer and/or repeated sojourn of man in space. Under orbital conditions biological stress and damage are induced in test subjects by cosmic radiation, especially the high energetic, densely ionizing component of heavy ions. Plant seeds were successful model systems for a biotest in studying the physiological damages and mutagenic effect caused by ionizing cosmic radiation in particular stem cells. Dosimetrically, the subdivision into charge- and Let-groups reveals the contribution of the intermediate group (LET = 350-1000 MeV/cm) due to the medium heavy ions (Z = 6-10). Their relative contribution increases with the lower inclination of the orbit of LDEF-1; on the other hand, the total fluence becomes higher with longer duration of the flight. The observed endpoints of the biological radiation damage hint at a correlation with particle dose rate rather than with the dose; additionally, data on shielding effects inside and outside the space craft and its exposure were gained from the different SDEF- and LDEF-missions.

Chemical reduction of silver chromate: a procedure for electron microscopical analysis of Golgi-impregnated neurons.

A procedure is described by which the solid silver chromate precipitate present in Golgi-impregnated neurons is reduced by a photographic developer into metallic silver grains or particles. The remaining silver chromate is subsequently removed, and the fine structure and synaptic relationships of the impregnated neurons can be studied with the electron microscope. Three developers, Hydrochinone, Kodalith and D 19 were tested on cortical neurons in Vibratome sections of rapid Golgi-impregnated rat brains. Considerable differences were observed as regards penetration of the developer into the sections, uniformity of development, size and location of the silver particles produced during the development, and the quality of preservation of the ultrastructural details both in the impregnated neurons and the surrounding neuropil. Following the chemical reduction procedure the metallic silver deposit can be converted into metallic gold by a gold toning technique. Of the 3 tested developers, Kodalith and D 19 met the demands: good ultrastructural preservation, high contrast at both light microscopic (LM) and electron microscopic (EM) levels, reliable and easy to perform, minimum of test runs. Because of the high contrast provided by Kodalith and D 19, these substances are well suited for neuroanatomical tracing studies which make use of a transport or degeneration technique combined with Golgi-EM. In contrast to Kodalith, D 19 has little effect on the ultrastructure of the impregnated cells. Therefore, this developer can also be used for ultrastructural studies of Golgi-impregnated, light microscopically identified neurons.

Quantitative electron microscopic analysis of postnatal development of zinc-positive nerve endings in the rat amygdala using Timm's sulphide silver technique.

The distribution and quantitation of zinc during postnatal development of the rat amygdala were investigated by light and electron microscopy with Timm's sulphide silver method. The adult rat amygdala could be divided into basolateral, basomedial, central, cortical, intercalated, lateral and medial subnuclei on the basis of Timm's plus Toluidine blue staining. Only a very weakly positive Timm's reaction could be observed in newborns, and a positive reaction was seen on the 5th postnatal day. The reaction became stronger with development, and reached adult levels by the 30th postnatal day. Electron microscopically, we investigated the basolateral subnucleus of the amygdala. The electron-dense deposits of silver grains from the Timm's reaction were only seen in nerve fibers endings containing many small clear vesicles. The results are similar to those described for mossy fiber endings in the rat hippocampus and in boutons of other telencephalic structures. The number of deposits of silver grains increased with age, and reached that of the adult between the 20th and 30th postnatal days. A significant increase in the number of silver grains per micron 2 area of the Timm's-positive nerve terminals occurred between the 10th and 20th postnatal days. Based on the available literature, these findings suggest that zinc exists in nerve fiber endings in the rat amygdala and that the amount increases with postnatal age. The findings are consistent with an important role for zinc in synaptic transmission.

A silver impregnation method for labeling both Alzheimer paired helical filaments and their polypeptides separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

The Gallyas silver impregnation which is specific to neurofibrillary changes of paired helical filaments (PHF) and 15 nm straight filaments, was adapted to stain polypeptides separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Both PHF and tau polypeptides were readily and consistently stained by the Gallyas stain. This technique stained PHF greater than tau greater than high-molecular-weight microtubule-associated polypeptides (MAPS). Tubulin was stained only weakly. Neurofilament triplet, ubiquitin, bovine serum albumin and histones were unstained. The staining of PHF and tau polypeptides by Gallyas silver stain is consistent with the presence of tau in PHF.

Structure of heavy ion tracks in Ag-Cl detectors.

The microdosimetric lateral structure of tracks of charged nuclear particles in monocrystalline layers of AgCl-detectors has been measured by means of a videoelectronic computer-controlled image analysing system. The lateral optical density profiles, recorded along the track in sequential steps of 0.2 micrometer show compact tracks with a density maximum around the track axis as well as "coreless" tracks with a density minimum towards the axis. This minimum is more pronounced for particles of high effective charge mean q exceeding approximately 25. The effect of fading of the core depends on the charge state mean q of the particle rather than on its (high) LET. This finding points towards an increased atomic displacement by Coulomb-repulsion into the lattice of the detector of Ag(+)-atoms which are needed for the formation of the track. Examples of measurements and of tracks are presented.

Experiment "Seeds" on Biokosmos 9. Dosimetric part.

The aim of the experiment "Seeds" on the Sowjetic satellite Biokosmos 9 was the observation of mutagenic effects caused at special loci of seeds of Arabidopsis thaliana and assigned to particles of the Cosmic radiation. Two types of exposure units were flown: A low-shielding unit Type I, mounted at the surface of the satellite (1.4 g/cm2 shielding) and, for comparison, an identical item inside (16 g/cm2 shielding), using nuclear emulsion as track detectors. A Type II unit, flown inside (18g/cm2 shielding) was mounted with AgCl track detectors. The layout will be briefly described. A first set of dosimetric data from the physical evaluation of the experiment will be presented. The subdivision into charge- and LET-groups shows a rather high contribution of the intermediate LET-group (350-1000 MeV/cm) due to medium heavy particles (Z = 6-10) and to enders of light (p, alpha) particles.

Chloride is preferentially accumulated in a subpopulation of dendrites and periglomerular cells of the main olfactory bulb in adult rats.

GABA is predominantly an inhibitory transmitter. Mediated by GABAA receptors, GABA opens chloride channels, induces a passive flux of chloride ions, which is usually directed from extracellular to intracellular space, and hyperpolarizes postsynaptic neurons. Recent electrophysiological data suggested that GABA may also depolarize neurons and exert excitatory actions. However, it remained unclear whether excitatory GABA effects are based on reversed transmembrane chloride gradient due to modifications in extracellular or intracellular chloride concentrations. Here, the first histochemical evidence is provided for local redistribution of chloride in the CNS of healthy adult rats. Olfactory bulbs were examined using freeze substitution, silver trapping of chloride and intensification techniques at light and electron microscopic level. The chloride content of precipitates was evidenced by electron spectroscopic imaging using a CEM 902 (Zeiss) electron microscope. Chloride concentration was high in a subpopulation of some periglomerular cell bodies and isolated dendritic profiles, while it seemed to be very low in certain parts of the glomerular neuropil including intercellular clefts. Data suggest that reversed chloride gradients can be demonstrated by cytochemical methods, and may be responsible for excitatory GABA effects on selected periglomerular neurons and dendrites in the olfactory glomeruli. Conditions leading to chloride redistribution in the CNS of normal adult rats remain to be determined.

Multiplicities of secondaries in interactions of 1.8 GeV/nucleon 56Fe nuclei with photoemulsion and the cascade evaporation model.

A nuclear photographic emulsion method was used to study the charge-state, ionization, and angular characteristics of secondaries produced in inelastic interactions of 56Fe nuclei at 1.8 GeV/nucleon with H, CNO, and AgBr nuclei. The data obtained are compared with the results of calculations made in terms of the Dubna version of the cascade evaporation model (DCM). The DCM has been shown to satisfactorily describe most of the interaction characteristics for two nuclei in the studied reactions. At the same time, quantitative differences are observed in some cases.
