Circulating immune complexes in bronchial asthma.

Soluble immune complexes were detected by a sold phase Clq binding assay in forty-two out of 106 well studied asthmatic patients (39.6%) and in eleven out of 145 age-matched controls (8%; P < 0.01). Clinical significance of immune complexes has been evaluated by comparing the following parameters in patients with and without such complexes: age, sex, duration of disease, IgE-mediated allergy (RAST, skin test), precipitins (Aspergillus, Candida albicans, Thermoactinomycetes), corticodependency, lung function tests, associated symptoms (hivernal bronchitis, urticaria, eczema,...), desenstization treatment, serum concentration in immunoglobulin in G, A, M and E. Immune complexes were found more frequently in female than in male patients. The prevalence of immune complexes was higher in patients treated by hyposensitization therapy and in cases with precipitating antibodies against thermophilic actinomycetes and Aspergillus.

GAMMA GLOBULIN: UNMASKING OF HIDDEN ANTIGENIC SITES ON LIGHT CHAINS.

Normal light (L) chains cleaved from normal pooled human gamma globulin contain immunologic determinants not overtly present in the native unreduced molecule. These hidden determinants are shared with Bence Jones proteins. Similarly, one of two Bence Jones proteins contained immunologic determinants which were not overtly present on the autologous serum myeloma protein.

GAMMA GLOBULIN ANTIGENIC TYPES DEFINED BY HEAVY CHAIN DETERMINANTS.

Two populations of immunologically distinguishable 7S gamma globulins in normal human serum and two corresponding antigenic types of myeloma 7S gamma globulins have been detected with rabbit antiserums to proteins associated with pathological conditions, the differences being related to the H-chains of 7S gamma globulin. No relationship exists with type I and type II antigenic classification, determined by L-chains. Human serums with various hereditary gamma globulin (Gm) specificities contain both types of 7S gamma globulin.

AGAR-GEL PRECIPITIN-INHIBITION TECHNIQUE FOR C-REACTIVE PROTEIN DETERMINATIONS. 3. QUANTITATION OF C-REACTIVE PROTEIN IN SERUM SPECIMENS.

A quantitative C-reactive protein serological procedure has been developed. By use of this method, which is performed in agar-gel plates, from 2 to 654 mug of C-reactive protein per ml of titrated human serum can be detected. The method is based on the inhibition of a specific C-reactive protein antigen-antibody precipitate formed in agar-gel by the minimal reactive dilutions of each reagent in 48 hr. It is simple, sensitive, and readily reproducible.

Reactivity of alkali-soluble, water-soluble cell wall antigen of Coccidioides immitis with anti-Coccidioides immunoglobulin M precipitin antibody.

The alkali-soluble, water-soluble cell wall antigen of Coccidioides immitis (C-ASWS) mycelia and spherules was shown to react with anti-Coccidioides immunoglobulin M (IgM) precipitin antibody, both in the classical tube precipitin test and in the immunodiffusion assay for tube precipitin antibody (IDTP). The reactions obtained between C-ASWS and reference IgM precipitin antibody were identical to the reaction obtained when reference coccidioidin (CDN) was used. Definitive proof that C-ASWS extracts contain antigenic determinants that are reactive with IgM tube precipitin antibody was obtained by solid-phase immunoadsorption. Elution of reference IDTP antiserum over a column containing mycelium C-ASWS coupled to Sepharose 4B completely adsorbed precipitin antibody; i.e., reactivity in the IDTP was demonstrable in the column eluate but not in the column effluent fraction. The antigenic composition of C-ASWS extracts was evaluated and compared with that of CDN by two-dimensional immunoelectrophoresis against burro anti-CDN. The results established that both mycelium and spherule C-ASWS contain antigenic determinants in common with only one antigen present in CDN. The latter, designated antigen 2, is a large polymer which is predominant among the antigenic components in CDN. On a dry weight comparison, antigen 2 determinants were most concentrated in spherule C-ASWS, followed by mycelium C-ASWS and reference IDTP antigen. The finding that C-ASWS extracts are reactive with IgM tube precipitin antibody and are antigenically identical to antigen 2 in CDN suggests that antigen 2 is the biologically active component of CDN in tube precipitin assays.

Properties of free endotoxin from Pasteurella multocida.

Free endotoxin (FET) from virulent encapsulated Pasteurella multocida or from an avirulent nonencapsulated mutant is capable of inducing active immunity, but the lipopolysaccharide (LPS) moiety of the endotoxin is not. These results suggest that a protein of P multocida is involved in the stimulation of active immunity. The serologic specificity of the FET is associated with the LPS moiety, which is related to a heat extracted antigen that is used for serotyping P multocida. The FET is capable of producing widespread vascular alteration and death. It is present in the vascular system of turkeys with acute fowl cholera, and it can be detected with the "Limulus" test for endotoxins and with the gel diffusion precipitin test.
