Variegated color sensations from rod-cone interactions: flicker-fusion experiments.

Multicolored images were produced by the combination of two different color-separation images, one illuminated with 656 nm light and the other illuminated with 500 nm light. Flicker-fusion frequency measurements were used to identify the conditions in which the 500 nm light was below cone threshold. Under those conditions the multicolored images were shown to be generated by rod and long-wave cone interactions.

Perception of forbidden colors in retinally stabilized equiluminant images: an indication of softwired cortical color opponency?

In color theory and perceptual practice, two color naming combinations are forbidden-reddish greens and bluish yellows-however, when multicolored images are stabilized on the retina, their borders fade and filling-in mechanisms can create forbidden colors. The sole report of such events found that only some observers saw forbidden colors, while others saw illusory multicolored patterns. We found that when colors were equiluminant, subjects saw reddish greens, bluish yellows, or a multistable spatial color exchange (an entirely novel perceptual phenomena); when the colors were nonequiluminant, subjects saw spurious pattern formation. To make sense of color opponency violations, we created a soft-wired model of cortical color opponency (based on winner-take-all competition) whose opponency can be disabled.

Four-color fluorescence correlation spectroscopy realized in a grating-based detection platform.

We have developed a filterless multicolor detection unit for fluorescence correlation spectroscopy (FCS). This grating-based setup is continuously tunable for multicolor separation and is thus a powerful alternative to the classical cascade of dichroic mirrors and filters. Our tailored platform allows for accommodation of up to 15 detection channels covering the entire visible spectral range. As a proof of principle, we successfully demonstrate simultaneous FCS of four distinct fluorescent quantum dot species being mixed in aqueous solution. Grating-based detection allows for spectral high-resolution FCS in a stable and compact setup and is a feasible tool for quantitative investigation of complexbiomolecular dynamics on a single molecule level.

Quantum dot nanocrystals for in vivo molecular and cellular imaging.

Semiconductor quantum dots (QD) are nanometer-sized crystals with unique photochemical and photophysical properties that are not available from either isolated molecules or bulk solids. In comparison with organic dyes and fluorescent proteins, QD are emerging as a new class of fluorescent labels with improved brightness, resistance against photobleaching and multicolor fluorescence emission. These properties could improve the sensitivity of biological detection and imaging by at least 10- to 100-fold. Further development in high-quality near-infrared-emitting QD should allow ultrasensitive and multicolor imaging of molecular targets in deep tissue and living animals. Here, we discuss recent developments in QD synthesis and bioconjugation, applications in molecular and cellular imaging as well as promising directions for future research.

Identification of new fluorescent protein fragments for bimolecular fluorescence complementation analysis under physiological conditions.

Protein-protein interactions play a pivotal role in coordinating many cellular processes. Determination of subcellular localization of interacting proteins and visualization of dynamic interactions in living cells are crucial to elucidate cellular functions of proteins. Using fluorescent proteins, we previously developed a bimolecular fluorescence complementation (BiFC) assay and a multicolor BiFC assay to visualize protein-protein interactions in living cells. However, the sensitivity of chromophore maturation of enhanced yellow fluorescent protein (YFP) to higher temperatures requires preincubation at lower temperatures prior to visualizing the BiFC signal. This could potentially limit their applications for the study of many signaling molecules. Here we report the identification of new fluorescent protein fragments derived from Venus and Cerulean for BiFC and multicolor BiFC assays under physiological culture conditions. More importantly, the newly identified combinations exhibit a 13-fold higher BiFC efficiency than originally identified fragments derived from YFP. Furthermore, the use of new combinations reduces the amount of plasmid required for transfection and shortens the incubation time, leading to a 2-fold increase in specific BiFC signals. These newly identified fluorescent protein fragments will facilitate the study of protein-protein interactions in living cells and whole animals under physiological conditions.

Semiautomated clone verification by real-time PCR using molecular beacons.

Conventional, high-throughput PCR analysis of common elements utilizing numerous primer sets and template DNA requires multiple rounds of PCR to ensure optimal conditions. Laborious gel electrophoresis and staining is then necessary to visualize amplification products. We propose novel multicolor molecular beacons, to establish a high-throughput, PCR-based sequence tagged site (STS) detection system that swiftly and accurately confirms marker content in template containing common repeat elements. A simple, one-tube, real-time PCR assay system was developed to specifically detect regions containing CA and GATA repeats. Ninety-six samples can be confirmed for marker content in a closed-tube format in 3 h, eliminating product confirmation on agarose gels and avoiding crossover contamination. Multiple STSs can be detected simultaneously in the same reaction tube by utilizing molecular beacons labeled with multicolor fluorophores. Template DNA from 260 RPCI-11 bacterial artificial chromosome (BAC) clones was examined for the presence of CA and/or GATA repeats using molecular beacon PCR and compared with conventional PCR results of the same clones. Of the 205 clones containing CA and GATA repeats, we were able to identify 129 clones (CA, n = 99; GATA, n = 30) by using molecular beacons and only 121 clones (CA, n = 92; GATA, n = 29) by conventional PCR amplification. As anticipated, 55 clones that contained sequences other than CA or GATA failed molecular beacon detection. Molecular beacon PCR, employing beacons specific for tandem repeat elements, provides a fast, accurate, and sensitive multiplex detection assay that will expedite verification of marker content in a multitude of template containing these repeats.

Detection of cell-surface antigens using antibody-conjugated fluorospheres (ACF): application for six-color immunofluorescence.

Antibody-conjugated fluorospheres (ACF) were used to phenotype murine leukocytes by flow cytometric analysis. Multicolor immunofluoresence (beyond simultaneous 4-color analysis) is limited by the availability of specific antibody-fluorochrome chrome conjugates and even further restricted by the spectral emission overlap of many of the fluorochromes when used in combination. Fluorospheres possessing unique excitation/emission spectra can provide much needed versatility to existing protocols of multicolor fluorescence. SKY BLUE (647 nm excitation, 730 nm emission) fluorospheres conjugated to CD11b monoclonal antibody were used in combination with the monoclonal antibodies IAd-FITC, L3T4 (CD4)-PE, LYT2 (CD8)-APC, THY1.2 (CD90)-biotin and B220 (CD45R)-RED613 for the simultaneous detection of six distinct cell-surface antigens in a mixed cell population. All fluorescence signals were resolved, and comparison of results from five-, six- and single-color samples indicated that the percentages of cells positive for specific surface antigens were equivalent.

Laser removal of tattoos.

Tattoos are placed for different reasons. A technique for tattoo removal which produces selective removal of each tattoo pigment, with minimal risk of scarring, is needed. Nonspecific methods have a high incidence of scarring, textural, and pigmentary alterations compared with the use of Q-switched lasers. With new advances in Q-switched laser technology, tattoo removal can be achieved with minimal risk of scarring and permanent pigmentary alteration. There are five types of tattoos: amateur, professional, cosmetic, medicinal, and traumatic. Amateur tattoos require less treatment sessions than professional multicolored tattoos. Other factors to consider when evaluating tattoos for removal are: location, age and the skin type of the patient. Treatment should begin by obtaining a pre-operative history. Since treatment with the Q-switched lasers is painful, use of a local injection with lidocaine or topical anaesthesia cream may be used prior to laser treatment. Topical broad-spectrum antibacterial ointment is applied immediately following the procedure. Three types of lasers are currently used for tattoo removal: Q-switched ruby laser (694 nm), Q-switched Nd:YAG laser (532 nm, 1064 nm), and Q-switched alexandrite laser (755 nm). The Q-switched ruby and alexandrite lasers are useful for removing black, blue and green pigments. The Q-switched 532 nm Nd:YAG laser can be used to remove red pigments and the 1064 nm Nd:YAG laser is used for removal of black and blue pigments. The most common adverse effects following laser tattoo treatment with the Q-switched ruby laser include textural change, scarring, and pigmentary alteration. Transient hypopigmentation and textural changes have been reported in up to 50 and 12%, respectively, of patients treated with the Q-switched alexandrite laser. Hyperpigmentation and textural changes are infrequent adverse effects of the Q-switched Nd:YAG laser and the incidence of hypopigmentary changes is much lower than with the ruby laser. The development of localized and generalized allergic reactions is an unusual complication following tattoo removal with the Q-switched ruby and Nd:YAG lasers. Since many wavelengths are needed to treat multicolored tattoos, not one laser system can be used alone to remove all the available inks and combination of inks. While laser tattoo removal is not perfect, we have come a long way since the advent of Q-switched lasers. Current research is focusing on newer picosecond lasers, which may be more successful than the Q-switched lasers in the removal of the new vibrant tattoo links.

[Emission model of volatile organic compounds from materials used indoors].

Various materials, such as wall-paper, floor-wax, paint, multicolor wall-coat, air freshener and mothball were experimented in a simulated test chamber under constant selected temperature, humidity and air exchange rate. The relation between the total VOCs concentration and time was regressed by four emission models and the surface emission rate was calculated. The regressed results indicated the similarity among four emission models for the liquid materials with volatile-solvent such as paint and multicolor wall-coat. But for low volatile solid materials, such as wall-paper, floor-wax, mothball, the sink model and the empirical model were better than the dilution model and vapor pressure model. Only for air freshener, it was improper to the total VOCs concentration as a parameter.

Mosquito species and their densities in some Egyptian governorates.

The present work studied the mosquito's identification, distribution and densities in fourteen Egyptian Governorates. The abundance and distribution of mosquito species monitored by three phases. The first was carried out in 1999 in five governorates, Qalyobia, Menoufia, Behaira, Fayium and Assuit. The second was in the year 2000 in Kafr El Sheikh, Giza, Sharkia, Menia and Aswan. The third was in the year 2001 in Kena, El Wady El Gadeed, Dakahlia and South Sinai. Culex species were the commonest mainly C. pipiens, C. antennatus and C. univittatus. C. thelerei was found only in El Kharga Oasis. Culiseta sp. was found in Qalyobia, Menofia, Behaira, Fayium, El Wady El Gadeed, Dakahlia and South Sinai and as larvae in Kafr El Sheikh, Giza, and Menia. Aedes detritus was found in Assiut, Fayium, Giza, Aswan, El Wady El Gadeed and South Sinai. Ae. caspius was found in Assiut and Aswan and as larvae in Kena and El Wady El Gadeed. Anopheles pharoensis was found in Behaira and Fayium, while A. algeriensis in Aswan. A. multicolor and A. sergentii were found in Fayium, Aswan and El Wady El Gadeed; but in Kena A. sergentii was found as larvae and A. multicolor as adults.

[Nipple discharge: personal experience with 2,818 cases].

After mastodynia, nipple discharge is the second most frequent condition that brings women to the attention of breast clinics. Seven types of nipple discharge exist: milky, multicolored, purulent, clear-watery, serous, pink or serosanguineous, brown or reddish-brown. From January 1982 to January 2003 we observed 2818 patients with nipple discharge (range: 16-83 years). Amongst these, 805 patients with nipple discharge were submitted to cytological examination of the secretions. One hundred and seventy-six had bilateral discharge, and 629 unilateral discharge. All patients with positive C3, C4 or C5 cytology and with unilateral discharge (227) were referred for surgical treatment. In 92 of these 227 cases (41%) the secretion was serous, in 59 cases (26%) bloody, in 45 cases (20%) purulent and in 31 cases (13%) multicolored. We performed duct galactophorectomy in 89 cases (39%), resection with reconstruction of the nipple-areola complex in 48 cases (21%), microdochectomy in 42 cases (18.5%), segmentectomy or quadrantectomy in 41 cases (18%), and mastectomy in 6 cases (3%); we also removed a papilloma from the ductal orifice in 1 case (0.44%). Histological specimens showed papilloma in 98 cases (43%), breast cancer in 39 cases (17%), galactophoritis in 36 cases (16%), fibrocystic disease in 46 cases (20%), including 31 (14%) with florid adenosis, and papillomatosis in 8 cases (4%). An increased probability of cancer is therefore associated with serous, bloody, reddish brown and watery secretions. This is particularly true when the secretion is unilateral and originates from a single duct, when there are cytological alterations, whether mammographic or galactographic, and when the patient is aged over 50 years.

Prevalence and distribution of anopheline mosquitoes in malaria endemic areas of Asir region, Saudi Arabia.

To study the prevalence of anopheline mosquitoes, over 180 sites were sampled in malaria-endemic areas of Asir region, Saudi Arabia, during June 1999-April 2001. A total of 7085 larval and 754 adult female Anopheles spp. specimens were collected. Seven species were identified: An. dthali, An. rupicolus, An. sergentii, An. arabiensis, An. multicolor, An. turkhudi and An. pretoriensis. Both An. arabiensis and An. sergentii are known vectors of malaria in the region. An. dthali occurred in all sites and was the most abundant species. An. turkhudi was collected in low numbers as larvae only. An. multicolor and An. pretoriensis were recorded for the first time in Asir region. An. sergentii is a species of the northern areas of the region, whereas An. arabiensis was more prevalent in the south.

Management of nipple discharge.

In a series of 8,703 breast operations, nipple discharge was the presenting symptom in 7.4% of cases. It is even more common in the office and clinic since many discharges can be treated medically. To be significant, a discharge should be true, spontaneous, persistent, and nonlactational. Of the 7 basic types, i.e., milky, multicolored and sticky, purulent, clear (watery), yellow (serous), pink (serosanguineous), and bloody (sanguineous), the last 4 are the surgically significant ones. Of the 586 patients operated on for one of these types of discharge, the majority had a benign etiology, i.e., intraductal papillomata (48.1%) and fibrocystic changes (32.9%), but 14.3% were due to cancer and another 7.3% to precancerous mastopathy. In the 84 patients with cancers, the false-negative rate for mammography was 9.5% and was 17.8% for cytology. There was no palpable mass in 13.1% of patients. There was an increasing likelihood of the discharge being due to cancer when the discharge was, in order of increasing frequency, yellow, pink, bloody, or watery, when it was accompanied by a lump, when it was unilateral and from a single duct, when the mammogram or galactogram and the cytology were positive, and when the patient was over 50 years of age. Milky discharges are usually treated medically unless they are due to a pituitary adenoma. If the cause cannot be found and eradicated, bromocriptine is the drug of choice. Multicolored sticky discharges are also treated medically, chiefly by nipple hygiene, except when advanced. Purulent discharges are treated with appropriate antibiotics but abscesses need drainage and a biopsy of the wall. Except in women under 35 years of age or in those anxious to have children, surgically significant discharges are treated by central duct excision. Good cosmetic results can be obtained with careful technique and the danger of a recurrent discharge is eliminated.

Color constancy in the honeybee.

A multicolored display was illuminated by 3 bands of wavelengths corresponding to the maxima of the spectral sensitivities of the 3 types of photoreceptors found in the bee retina. The intensity of each band could be varied individually. The light fluxes emitted by the colored areas of the multicolored display were determined quantitatively. Free-flying honeybees were trained with sugar solution to choose one of the colored areas. The illumination was then changed in such a way that the light fluxes formerly emitted by the training area were now measured on another area. When the trained bees were tested under those conditions, they still chose the training area. The relative positions of the colored areas were changed in order to exclude learning of position. It is concluded that color vision in bees is, in a certain range, independent of the spectral content of the illumination. Model calculations show that the behavior observed in bees is consistent with the retinex theory (Land, 1977), i.e., an algorithm using long-range interactions is required to explain color constancy.

Laparoscopical and histological abnormalities of the liver in chronic HBs ag (hepatitis B surface antigen) carriers.

Previously, I have reported the laparoscopical and histological findings of 16 chronic HBs Ag carriers in whom the time of HBV infection could be assumed, HBs Ag in the sera persisted for at least 6 months, and laparoscopy with liver biopsy was performed. In these carriers, no abnormal results of liver function tests were found and early history of hepatitis was unknown. Besides these 16 HBs Ag carriers, I have encountered 15 chronic HBs Ag carriers in whom the time of HBV infection could be assumed, HBs Ag in the sera persisted for at least six months, and laparoscopy with liver biopsy was performed. In these carriers, no early history of hepatitis could be found and normal results of liver function tests were observed at least at the time of the first laparoscopy with liver biopsy, but blood chemical analyses resulted in slightly abnormal values at rare intervals during the clinical observations after the first laparoscopy with liver biopsy. In six cases of 15 chronic HBs Ag carriers, laparoscopy revealed a white liver; in four cases, a multicolored liver and in the remaining five cases, a nodulated liver. Histology of the white liver in six cases showed chronic persistent hepatitis (CPH) in three cases, chronic aggressive hepatitis (CAH) in one case and chronic persistent hepatitis with fatty metamorphisis in two cass. Histology of the nodulated liver showed liver cirrhosis in five cases. Histology of the multicolored livers showed CAH in all four cases. The question remains unanswered as to how such morphological alterations of the liver developed in these chronic HBs Ag carriers with normal liver chemistries.

[Criticism of sacroiliitis staging in ankylosing spondylitis].

Information-theoretical considerations demonstrate the uselessness, in terms of understanding and diagnosis, of seven different systems for grading sacroiliitis in ankylosing spondylitis used by German, French, Canadian, Norwegian, Swedish, and Swiss authors. The illustrations prove that it is not possible to differentiate between a normal and abnormal (inflammatory) x-ray of the sacroiliac joints without the application of tomography. A grading system promises diagnostic and communicative success only when based upon tomography results (6 standard sections in adults), and when it takes the "multicolored picture" into consideration ("multicolored picture": no = sacroiliitis stage 0; yes = stage 1; complete bilateral ankylosis: yes = stage 2).

Laparoscopic an histologic abnormalities of the liver in chronic asymptomatic hepatitis B surface antigen carriers.

In most cases, Hepatitis B surface antigen (HBs Ag) in sera can be incidentally detected in patients with liver diseases at the time of medical examinations. It is rather difficult to assume how long HBs Ag has persisted in the sera of these patients before the medical examination. However, we have encountered 16 asymptomatic HBs Ag carriers in whom the time of HBV infection can be estimated, HBs Ag in sera persisted for at least 6 months (actually up to 5 years), and laparoscopy with liver biopsy was performed. In 11 out of these 16 asymptomatic HBs Ag carriers, laparoscopy revealed a white liver and in the remaining five cases, a multicolored liver. Histology of the white liver showed minimal inflammatory changes of the liver in two cases, chronic persistent hepatitis in eight cases and chronic aggressive hepatitis in one case. Histology of the multicolored liver including transitional change to the nodulated liver in five case showed chronic aggressive hepatitis in two cases and liver cirrhosis in three cases. The question remains open as to how such morphological alterations of the liver developed in these asymptomatic HBs Ag carriers with normal liver chemistries although none of these 16 cases developed overt acute hepatitis, either of type A or non A non B, 6 to 21 months after the assumed HBV infection.

Studies on the bionomics and vector competence of adult anopheline mosquitoes in El Faiyum Governorate, Egypt.

Malaria is by far the most important parasitic tropical disease, causing great suffering and loss of money and life. Its prevalence and incidence varies throughout the world. In Egypt, it is an ancient disease. Nowadays, it causes sporadic cases and sometimes limited outbreaks, mainly in El Faiyum Governorate where it has been endemic since several decades. It seems that the environmental and climatic conditions e.g. population habits and the fauna and flora favour the indigenous transmission in this governorate. To carry out feasible anti-vectorial measures, all vectorial activities should be critically known. This paper aimed to throw some light on the parasitological aspects of adult anopheline mosquitoes in El Faiyum Governorate. The results showed the presence of four species of Anopheles; (1) A. (C.) Pharoensis, (2) A. (C.) sergenti, (3) A. (C.) multicolor and (4) A. (A.) tenebrosus (= coustani). The most common species was A. pharoensis, followed by A. sergenti and A. multicolor. The least common was A. tenebrosus. The highly infested area with Anopheles species was Sinouris and the least was El Siliyien. The bionomics and the vector competence of the adults of the four species were studied.
