Accessibility of colloidal gold and horseradish peroxidase to cytosolic spaces in Limulus ventral photoreceptors.

Physiological studies of intracellular messengers frequently employ intracellular injections of large molecules that either monitor or modulate the metabolism of the messenger cascade. Injected molecules have unknown mobility in the cytosol and unknown accessibility to various cytosolic compartments, including those postuiated to be traversed by intracellular messenger molecules. In order to determine whether injected molecules have access to the confined spaces through which messenger molecules must diffuse, we injected 5-nm colloidal gold or horseradish peroxidase, or both, into Limulus ventral photoreceptors. Injections were made by applying pressure pulses to the back of an intracellular micropipette that also monitored membrane voltage. The tissue was fixed at varying times after injection and processed for electron microscopy by conventional techniques. Cells fixed 1-3 min after injection contained HRP reaction product only in the cell body. HRP reaction product was found at varying distances down axons in direct relation to the interval between injection and fixation. Colloidal gold particles were found throughout the cell body but not in axons of tissue fixed 1-3 min after injection. Both HRP reaction product and 5-nm colloidal gold particles were observed within the microvillar projections of internal and external rhabdomere, as well as within the extracisternal spaces of endoplasmic reticulum. We conclude that large molecules injected from an intracellular micropipette into an arbitary locus of ventral photoreceptor cells have access to all of the presumed sites of the phototranduction cascade.

Gram-scale synthesis of monodisperse gold colloids by the solvated metal atom dispersion method and digestive ripening and their organization into two- and three-dimensional structures.

We describe a synthetic procedure for preparation of large quantities of monodisperse thiol-stabilized gold colloids in toluene solution. The method is based on the solvated metal atom dispersion technique (SMAD), which is very suitable for preparation of large amounts of metal colloidal solutions, as well as of metal sulfide, metal oxide, and other types of dispersed compounds in different solvents. A combination of two different solvents like acetone and toluene is used for the preparation of the gold colloids. The necessity of initially carrying out the SMAD reaction in acetone comes from its high degree of solvation of gold particles. Acetone acts as a preliminary stabilizing agent. After its removal from the system, the particles are stabilized by dodecanethiol molecules, which enable their very good dispersion in toluene solution. A digestive ripening procedure is carried out with the gold-toluene colloid, and for this purpose pure toluene as solvent is necessary. This has a dramatic effect on the narrowing of particle size distribution and almost monodisperse colloids are obtained (some discussion of the probable mechanism of this remarkable digestive ripening step is given). These colloidal solutions have a great tendency to organize in two- and three-dimensional structures (nanocrystal superlattices, NCSs). We believe that this procedure provides a real opportunity to synthesize large amounts of gold nanocrystals as well as NCSs.

A rapid procedure suitable to assess quantitatively the endocytosis of colloidal gold and its conjugates in cultured cells.

We measured the endocytic uptake of low-density lipoproteins (LDLs) conjugated to colloidal gold in cultured cells, either by counting gold particles on electron micrographs or by inductively coupled plasma (ICP) mass spectrometry (MS). Both procedures are comparable but the latter requires a considerably shorter time and allows analysis of a much larger sample. In addition, ICP MS, compared to alternative radioactive or fluorescent procedures, offers the major advantage of using the same probe to quantify the endocytic uptake and to follow it by electron microscopy. Therefore, ICP MS analysis provides an easy, rapid, and sensitive quantification of endocytosis that complements the electron microscopic studies.

Colloidal gold probes in immunocytochemistry. An optimization of their application in light microscopy by use of silver intensification procedures.

Colloidal gold particles are the markers of choice for ultrastructural localization of antigens. By reducing gold chloride with tannic acid and trisodium citrate, a broad range of narrowly determined particle sizes can be obtained. Such particles can easily be coupled to a number of proteins and the resulting conjugates are conveniently purified on a gel-chromatography column. Their application in light microscopy requires an amplification step with a silver physical developer. Silver-intensified colloidal gold probes can advantageously be used for immunostaining of cryostat, paraffin and plastic sections. Moreover, permeabilized cultured cells and whole-mount preparations can also be stained with gold-silver techniques. Silver intensification does not affect reactivity of a number of tissue antigens, thus permitting double staining combinations with immunoperoxidase or immunofluorescence methods.

Colloidal gold particles as a new in vivo marker of early acute lung injury.

Permeability of the endothelial barrier to large molecules plays a pivotal role in the manifestation of early acute lung injury. We present a novel and sensitive technique that brings microanatomical visualization and quantification of microvascular permeability in line. White New Zealand rabbits were anesthetized and ventilated mechanically. Rabbit serum albumin (RSA) was labeled with colloidal gold particles. We quantified macromolecular leakage of gold-labeled RSA and thickening of the gas exchange distance by electron microscopy, taking into account morphology of microvessels. The control group receiving a saline solution represented a normal gas exchange barrier without extravasation of gold-labeled albumin. Infusion of lipopolysaccharide (LPS) resulted in a significant displacement of gold-labeled albumin into pulmonary cells, the lung interstitium, and even the alveolar space. Correspondingly, intravital fluorescence microscopy and digital image analysis indicated thickening of width of alveolar septa. The findings were accompanied by a deterioration of alveolo-arterial oxygen difference, whereas wet/dry ratio and albumin concentration in the bronchoalveolar lavage fluid failed to detect that early stage of pulmonary edema. Inhibition of the nuclear enzyme poly(ADP-ribose) synthetase by 3-aminobenzamide prevented LPS-induced microvascular injury. To summarize: colloidal gold particles visualized by standard electron microscopy are a new and very sensitive in vivo marker of microvascular permeability in early acute lung injury. This technique enabling detailed microanatomical and quantitative pathophysiological characterization of edema formation can form the basis for evaluating novel treatment strategies against acute lung injury.

THE ENTRY AND DISTRIBUTION OF HERPES VIRUS AND COLLOIDAL GOLD IN HELA CELLS AFTER CONTACT IN SUSPENSION.

The way in which herpes virus of a well adapted strain penetrates susceptible HeLa cells has been investigated using thin sectioning techniques for electron microscopy. Mature virus particles and cells were mixed together in suspension cultures for 15, 30, 60, or 120 minutes so that the stages in virus uptake could be followed in sequence. The ingestion of particles of colloidal gold by HeLa cells under similar conditions was studied for comparison in parallel experiments. After 15 minutes' contact, the mature virus was found adsorbed on the surface of the cells but separated from them by a narrow gap in which phosphotungstic acid staining was sometimes able to reveal an extraneous coat which appeared as an amorphous layer on the outer aspect of the plasma membrane. When mixing continued for longer the particles were present in deep invaginations or actual cytoplasmic vacuoles, with their outer layers in various stages of stripping and digestion. The stripped, naked, central portion of the virus was occasionally found in these vacuoles but was more commonly free in the cytoplasmic matrix; the mode of transition between these sites could not be determined. Where contact continued for 2 hours these phenomena were much less frequently observed. The larger particles of colloidal gold were ingested in the same way as the virus, but smaller ones were taken up in micropinocytosis vesicles. The gold passed through membrane-bounded cytoplasmic spaces to accumulate in vacuoles from which, in contrast to herpes particles, it did not escape. These findings are discussed, and considered with particular reference to their bearing on the initiation of infection, the uptake and disposal of particles by cells, and the influence on the latter of virus morphology.

Formation of spherical nanostructures by the controlled aggregation of gold colloids.

Stable suspensions of size-uniform spherical assemblies of 5-8 nm gold colloids in toluene are readily obtained by cross linking the colloidal particles using alkanedithiols within a defined range of gold-dithiol molar ratios. The assemblies are very stable and remain suspended in toluene for several months without significant aggregation. These porous gold spheres can be further organized into hierarchically assembled relatively linear chains by the addition of ethanol.

Detection of Norwalk-like virus and specific antibody by immune-electron microscopy with colloidal gold immune complexes.

Direct electron-microscopy (DEM), immune electron microscopy (IEM) and four different procedures of immune electron microscopy with colloidal gold immune complexes were evaluated for the detection of Norwalk-like virus and specific antibody. A solid-phase immune electron microscopy with colloidal gold immune complexes-triple layer method (SPIEMGIC-TLM) is developed for screening patients' specimens for the detection of Norwalk-like virus and its specific antibody. The method demonstrates low non-specific background labelling and is simple, sensitive and easy to perform. A quadruple layer method (SPIEMGIC-QLM), which is a modification of the triple layer method, has been established by adding a cross-linking anti-IgG layer to amplify the reaction and to provide a more sensitive test which is suitable for screening monoclonal antibodies prepared against 32-34-nm Norwalk-like virus isolated in our laboratory.

Zonal heterogeneity of rat hepatocytes in the in vivo uptake of 17 nm colloidal gold granules.

The in vivo uptake in hepatocytes of intravenously injected colloidal gold granules with a diameter of 17 nm or 79 nm and coated with bovine serum albumin or with polyvinyl-pyrrolidone was studied. Irrespective of coating only the 17 nm granules were taken up in hepatocytes. Perivenous hepatocytes did take up much more gold granules than periportal hepatocytes. The gold granules were found in lysosomes around bile canaliculi. Two hours after injection hepatocytes contained the maximal amount of granules. At least a portion of the granules was discharged into the bile. The observed zonal gradient in the uptake of 17 nm gold granules might be caused by the greater supply of granules to the perivenous hepatocytes as a combined result of the higher porosity of the endothelial lining and the smaller number of Kupffer cells with a low endocytic activity in this zone.

Lectin from Codium fragile ssp. tomentosoides conjugated to colloidal gold: a new histochemical reagent.

A new histochemical reagent has been developed utilising a lectin from a marine alga for the first time. Colloidal gold was coupled to the N-acetyl-alpha-D-galactosamine specific lectin from the green alga Codium fragile ssp. tomentosoides. The lectin--gold conjugate bound to the membranes of blood-group A1 human erythrocytes which were used as a model system. The bound complex could be detected, readily, by transmission electron microscopy. This novel reagent incorporating a lectin of low molecular weight (15 kDa) has potential value for studies of cell-surface topography of a variety of tissues.

Demonstration of fluid-phase endocytosis in epithelial cells of the male reproductive system by means of horseradish peroxidase-colloidal gold complex.

The occurrence of fluid-phase endocytosis in Sertoli cells as well as epithelial cells of the rete testis and ductuli efferentes was demonstrated by use of horseradish peroxidase combined with colloidal gold (HRP-G) as a tracer. The characteristic electron dense spherical colloidal gold particles facilitate the localization of the HRP-G complex in pinocytotic vesicles, multivesicular bodies, and lysosomes of these epithelial cells. With this method, which does not require the use of diaminobenzidine, a clear distinction can be made between peroxidases from endogenous and exogenous origins.

[The detection and nature of the labelling of F-actin using a phalloidin-colloidal gold marker].

On the basis of a conjugate of phalloidin-colloidal gold a marker has been obtained for electronmicroscopic identification and study of marking character of filament actins. Using this marker the filamentous form of actins from rabbit skeletal muscles and chicken gizzard was revealed. The marking characters of the two actins appeared to differ at the level of electron microscopy. A periodicity of the marker setting with a step of 105.4 +/- 1.7 nm on the rabbit F-actin was revealed. A possibility of differential evaluation of the filamentous and globular forms of the actins was shown when using the same marker in dot-blot analysis procedure with the sensitivity from 15 to 50 ng.

[Detection of an antigen related to systemic Candida infection using a monoclonal antibody conjugated to colloidal gold].

Previously we showed it was possible to detect antigenemia associated with systemic candidiasis using an anti-C. albicans monoclonal antibody conjugated to colloidal gold. The technique being used, known as Immuno-Gold-Silver staining (IGSS), is applied to serum dots on cellulose nitrate. It is very simple in practice, the results of the reaction being visible with the naked eye. The diagnostic value of IGSS has been compared, on the one hand, with that of the anti-Candida antibody detection by co-counterimmunoelectrophoresis and indirect immunofluorescence, on the other hand with that of the antigen detection using the Cand-Tec commercial test. The specificity and sensitivity of these methods have been established in relation to sera of 79 subjects shared out into 4 groups: sound-subjects, patients having developed systemic candidiasis following surgery, leukemic patients apparently uninfected with Candida and leukemic patients suffering from systemic candidiasis. The IGSS which is slightly less specific than the Cand Tec makes it possible to diagnose a much greater number of infections. Selected bioclinical observations show that there exists complementarity between the detection tests of antibodies and those of antigens and that it is possible to attribute a prognosis value to antigenemia detected with the IGSS dot method.

[Use of specific antibodies, labeled with colloidal gold particles, for the detection of Brucella antigens using dot-immunoassay].

The method of dot immunoassay with the use specific antigens, labeled with colloidal gold particles, for the detection of brucellar antigens was developed and tested under laboratory and field conditions. In this work soluble antigens isolated from different Brucella species and corpuscular antigens (13 strains belonging to 7 species of the genus Brucella, most pathogenic for humans and animals in the S- and R-forms) were used. The method was tested in the study of pathological material obtained from sick animals and humans in a farm with unfavorable situation for brucellosis in the Irkutsk Region. The sensitivity of the proposed assay system was found to be high and constituted 10 pg/ml to 1 ng/ml for soluble brucellar antigens and 200 CFU/ml to 13.5 x 10(6) CFU/ml for corpuscular antigens of Brucella S- and R-forms. The specificity of the method was tested with the use of 10 heterologous microorganisms. False positive results were observed only with Yersinia enterocolitica 0:9 at a concentration of 1 x 10(6) CFU/ml due to similarity of their polysaccharide-containing surface antigens. The newly developed dot immunoassay is simple for use, rapid and does not require expensive reagents and equipment.

Postembedding colloidal-gold immunocytochemistry of noncollagenous extracellular matrix proteins in mineralized tissues.

Immunocytochemistry is a powerful tool for investigating protein secretion, extracellular matrix assembly, and cell-matrix and matrix-matrix/mineral relationships. When applied to the tissues of bones (bone and calcified cartilage) and teeth (dentin, cementum, and enamel), where calcium phosphate-containing extracellular matrices are the predominant structural component related to their weight-bearing and masticatory roles, respectively, data from immunocytochemical studies have been prominent in advancing our understanding of mineralized tissue modeling and remodeling. The present review on the application of postembedding, colloidal-gold immunocytochemistry to mineralized tissues focuses on the advantages of this approach and relates them to conceptual, theoretical, and experimental data currently available discussing matrix-mineral interactions and extracellular matrix formation and turnover in these tissues. More specifically, data are summarized regarding the distribution and role of noncollagenous proteins in different mineralized tissues, particularly in the context of how they interface with mineral, and how this relationship might be affected by the various tissue-processing steps and immunocytochemical strategies commonly implemented to examine the distribution and function of tissue proteins. Furthermore, a technical discussion is presented that outlines several different possibilities for epitope exposure in mineralized tissues during preparation of thin sections for transmission electron microscopy. Cell biological concepts of protein secretion by cells of the mineralized tissues, and subsequent extracellular matrix assembly and organization, are illustrated by examples of high-resolution, colloidal-gold immunolabeling for osteopontin, bone sialoprotein, and osteocalcin in the collagen-based mineralized tissues and for enamel protein (amelogenin) in enamel.

A novel immunosensor based on immobilization of hepatitis B surface antibody on platinum electrode modified colloidal gold and polyvinyl butyral as matrices via electrochemical impedance spectroscopy.

Hepatitis B surface antibody (HBsAb) was immobilized to the surface of platinum electrode modified with colloidal gold and polyvinyl butyral (PVB) as matrices to detect hepatitis B surface antigen (HBsAg) via electrochemical impedance spectroscopy (EIS). The electrochemical measurements of cyclic voltammetry and impedance spectroscopy showed that K(4)[Fe(CN)(6)]/K(3)[Fe(CN)(6)] reactions on the platinum electrode surface were blocked due to the procedures of self-assembly of HBsAb-Au-PVB. The binding of a specific HBsAb to HBsAg recognition layer could be detected by measurements of the impedance change. A new strategy was introduced for improving the sensitivity of impedance measurements via the large specific surface area and high surface free energy of Au nanoparticles and the encapsulated effect of polyvinyl butyral. The results showed that this strategy caused dramatic improvement of the detection sensitivity of HBsAg and had good linear response to detect HBsAg in the range of 20-160 ng.ml(-1) with a detection limit of 7.8 ng.ml(-1). Moreover, the studied immunosensor exhibited high sensitivity and long-term stability.
