Vaccination against schistosomiasis and fascioliasis with the new recombinant antigen Sm14: potential basis of a multi-valent anti-helminth vaccine?

Molecular cloning of components of protective antigenic preparations have suggested that related parasite fatty acid binding proteins could form the basis of the well documented protective, immune cross reactivity between the parasitic trematode worms Fasciola hepatica and Schistosoma mansoni. We have now confirmed the cross protective potential of parasite fatty acid binding proteins and suggest that it may be possible to produce a single vaccine that would be effective against at least two parasites, F. hepatica and S. mansoni of veterinary and human importance respectively.

Decrease in albendazole sulphonation during experimental fascioliasis in sheep.

1. The in vivo S-oxidation of albendazole was measured from the pharmacokinetic profile of albendazole sulphoxide and sulphone determined in young male sheep receiving oral albendazole (1.9 mg/kg). Studies were carried out before, and each month after, oral infestation by 150 metacercariae of Fasciola hepatica. 2. Parasitic pathology was ascertained by clinical observation of animals, and the increase in plasma antibodies directed against liver flukes. 3. Rate of conversion of sulphoxide to sulphone and rate of sulphone elimination, were respectively decreased by 47% and 87% at week 8 post-infection, whereas significant increases in the area under plasma sulphone concentrations versus time curve and mean residence time, occurred 4-12 weeks following the infestation. 4. A 58% decrease in albendazole sulphonation was demonstrated in liver microsomal preparations obtained from 8-week-infected sheep, while there was no change in the FAD-directed sulphoxidation of albendazole. 5. The transient impairment of albendazole sulphonation could be related to the decrease in liver microsomal cytochrome P450-dependent monooxygenases observed in sheep with a similar parasitic pathology.

Fasciola gigantica-specific antigens: purification by a continuous-elution method and its evaluation for the diagnosis of human fascioliasis.

Immunodominant antigens of an approximate molecular mass of 27 kD were obtained from an excretory-secretory product of adult Fasciola gigantica by a continuous-elution method. An indirect ELISA using the antigens obtained by this relatively simple procedure was developed for detecting specific antibodies from patients infected with F. gigantica. Sera from patients with other parasitic infections, healthy volunteers, and cholangiocarcinoma were also analyzed. The sensitivity, specificity, and positive and negative predictive values for this ELISA using the fractionated antigens were 100%. The data indicated a possible correlation of antibodies to F. gigantica with cholangiocarcinoma.

Fascioliasis causing hepatitis in two eaters of water cress.

Fasciola hepatica infection of the liver in a husband and wife occurred after they had eaten water cress exposed to pollution by cattle. Diagnosis was a problem. Both responded to treatment with chloroquine and emetine. The epidemilology and pathology of infection of the liver by Fasciola hepatica are described. The danger of eating water cress is emphasized for this momentary delight may lead to a chronic debilitating illness.

Studies on heterologous resistance between schistosomiasis mansoni and fascioliasis.

S. mansoni and F. gigantica GSTs were purified from adult worm homogenates by affinity chromatography. Assessment of both SmGST and FgGST as candidate vaccines was done by comparing vaccinated mice groups challenged with S. mansoni cercariae with control ones in terms of worm load, tissue egg number viability and maturity, and the survival rate of the animals. SmGST 5 ug vaccination generally appeared to confer best protection against homologous challenge followed by FgGST 5 ug while vaccination with both antigens using low doses (2 ug) appeared to have no specific role in decreasing worm load but had significant effects on egg production.
