Abscisic acid studied by fluorescent techniques.

Crude analytical abscisic acid (ABA) exhibited fluorescence (lambdaex 340, lambdaem 425 nm). The results demonstrated a relationship between fluorescence response and ABA concentration, in the range of 0.04-0.38 microgram/ml. Chromatographic purification increased the emission signal, and oxidation induced by adding a sulphuric acid: ethanol (1:1) mixture also increased and displaced it (lambdaex 450, lambdaem 495 nm). Using these properties, a fluorimetric method for identification and evaluation of ABA, had been developed, and used for inhibitor recognition in P. pinea seed coats.

Effect of structure on tumor specificity of alicyclic alpha-amino acids.

The selective affinity of [carboxyl-11C]-1-aminocyclopentanecarboxylic acid (ACPC) for tumor tissue has led us to study the tumor-localizing characteristics of a series of alicyclic alpha-amino acid analogs of ACPC. The tissue distributions of [14C]-1-aminocyclopropanecarboxylic acid, 1-aminocyclobutanecarboxylic acid (ACBC), 1-aminocyclohexanecarboxylic acid, 1-amino-2-methylcyclopentanecarboxylic acid were compared with that of ACPC in Buffalo rats bearing Morris 5123C hepatomas. ACPC and ACBC were found to have significantly higher tumor-to-nontumor concentration ratios than the other four amino acids. ACBC generally had higher tumor-to-nontumor ratios than did ACPC, significantly so for muscle, kidney, and testis and marginally so for blood. These results suggest that [carboxyl-11C]ACBC may be a better agent than [carboxyl-11C]ACPC for tumor imaging by positron tomography.

Short-term re-epithelialization of the alkali-burned rabbit cornea.

Pfister and associates have shown that treatment with citrate solution reduces the incidence of stromal ulceration after alkali burn of the cornea. In the course of study of the effect of inhibitors of plasminogen activation on corneal re-epithelialization following alkali burns, we noted that when citrate was used in the medication, the healing process proceeded rapidly for three days; however, the denuded area became abruptly larger on the fourth day, and then resumed healing on the fifth day.

[Blakeslea trispora mutants with a disrupted sexual process].

Three groups of Blakeslea trispora (+) and (-) mutants were obtained and their phenotypical characteristics were studied as well as biochemical changes in the course of mating and the ability to synthesize carotenoids when the sexual process of these mutants was disordered. The first group of mutants synthesized carotenoids at the control level, the second group produced them below the control level, and the third group accumulated less than 1% of carotenoids as compared to the control. The difference in the yields of carotenoids among the three groups of mutants in the mated culture is attributed to the presence (or absence) of the ability to synthesize trisporic acids in them.

Germination inhibitors in embryos and endosperms from Pinus pinea seeds.

Inhibitors from Pinus pinea L. seed embryos and endosperms were separated using thin layer chromatography and a Sephadex G-10 column. The inhibitory activity was resolved into several fractions. One of these behaved similarly to abscisic acid in thin layer chromatography and Sephadex G-10 chromatography and in UV absorption and fluorescence spectra. In endosperm another potent inhibitor was detected, which showed a characteristic syndrome similar to that of phosphon D. These germination inhibitors are involved in the regulation of P. pinea seed germination.
