Coumarin compounds in Ammi majus L. callus cultures.

Callus cultures of Ammi majus L. (bishop's weed), Apiaceae, were maintained on variants of Linsmaier-Skoog's (L-S) medium differing in the content of the phytohormones, alpha-naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP) (0.1-10.0 mg/l). The increments of callus tissue fresh weight showed considerable differences (1.4 bis 4.4-fold) during 4-week subcultures on the media tested. HPLC analysis revealed the presence of six compounds in the extracts of callus tissues, which are known metabolites in plants growing under natural conditions: the linear furanocoumarins psoralen, bergapten, xanthotoxin, isopimpinellin, imperatorin and their precursor umbelliferone. The total contents of coumarin compounds under examination showed marked differences dependent on the phytohormone concentrations in the medium. They ranged from 40.95 to 871.05 mg/100 g of dry weight. Imperatorin was the metabolite dominating among the furanocoumarins in calli from almost all tested variants of L-S medium (maximum content of 169.27 mg/100 g). Callus tissue cultured on one of the variants (0.1 mg/l NAA, 0.1 mg/l BAP) yielded in high content of xanthotoxin (145.33 mg/100 g). On the majority of media, umbelliferone was also accumulated at high quantities (maximum content of 536.29 mg/100 g). The medium containing 0.1 mg/l NAA and 0.1 mg/l BAP was the best for the accumulation of analysed coumarins. This medium favoured the formation of embryogenic callus. Xanthotoxin also dominated quantitatively among the furanocoumarins in the tested vegetative organs and fruits of the plant grown under natural conditions (leaves: 26.10 mg, roots: 5.55 mg, fruits: 3010.41 mg/100 g). Maximum contents of this metabolite in in vitro culture were many times higher than those found in vegetative plant organs but manifold lower than in fruits. On the other hand, maximum contents of imperatorin obtained in in vitro culture were many times higher in comparison with those detected in vegetative organs and fruits (leaves: 14.10 mg, roots: 3.30 mg, fruits: 94.70 mg/100 g of dry weight.). The A. majus L. callus culture, established in the course of the present experiments, can be considered a valuable model for studies of the biosynthesis of coumarin compounds, and a potential source of the psoralen 8-alkoxy derivatives imperatorin and xanthotoxin and their precursor umbelliferone.

[Influence of sulphur smoking on the content of coumarins in the root of Angelica dahurica (Fisch. ex Hoffm.) Benth. et Hook.f.var. formosana (Boiss) Shan et Yuan].

The content of coumarins in the root of Angelica dahurica (Ad) treated in different time spans with smoke of sulphur was determined by HPLC. The result showed that in nine hours before Ad was smoked thoroughly the content of coumarins reduced rapidly with the increase of time, and there after, the reduction was not obvious. After being treated for nine hours, the total coumarins were reduced by 75.3% and after forty hours the ratio reached 77.9%.

Coumarins during pregnancy: long-term effects on growth and development of school-age children.

Anticoagulation during pregnancy is complicated because of potential risks for mother and foetus. Unfractionated or low-molecular-weight heparin is used for most anticoagulant indications. Its efficacy, however, in pregnant women with prosthetic heart valves is questioned, therefore coumarins are preferred for this indication. We studied long-term effects of prenatal coumarin-exposure on growth and on neurological, behavioural and cognitive development in 274 school-age children in comparison with 231 age-matched non-exposed controls. No major abnormalities were found. The exposed children had an increased risk for minor neurological dysfunction and for a low intelligence quotient (IQ below 80). The risk for a combination of two or more (minor) abnormalities was higher for the exposed children, RR = 7.6. We conclude that prenatal exposure to coumarins is associated with an increased risk for disturbances in development in school-age children. However, for the vast majority of children there is no clinical significant effect on growth and long-term development.

[Effect of precursors on the production of coumarins in a suspension culture of Angelica archangelica L].

The effect of potential precursors (cinnamic acid, phenylalanine, tyrosine) in three concentrations (0.01; 0.1; 1 mmol/l) on the growth of the culture and coumarin production was investigated in the suspension culture of Angelica archangelica L. The cultures were cultivated under constant illumination (3500 lux) and in the dark. The growth of the culture in the dark was not affected by the precursors, cinnamic acid in a concentration of 1 mmol/l exerted toxic action. The growth of the culture cultivated under constant illumination was stimulated by tyrosine in a concentration of 0.1 and 1 mmol/l, and phenylalanine in a concentration of 0.1 mmol/l; phenylalanine and cinnamic acid in a concentration of 1 mmol/l inhibited the growth of the culture. In the cultivation in the dark, coumarin production was increased by phenylalanine in a concentration 0.01 mmol/l and by tyrosine in a concentration of 1 mmol/l. In the cultivation under constant illumination, coumarin production was decreased by the action of cinnamic acid, was not affected by tyrosine, and phenylalanine (0.01 and 0.1 mmol/l) increased it in comparison with the culture without precursors.

[Effect of auxins on production of coumarin in a suspension culture of Angelica archangelica L].

The paper examined the effect of selected auxins (2,4-dichlorophenoxyacetic acid, alpha-naphthalene-acetic acid, beta-indoleacetic acid, beta-indoleburytic acid; each in four concentrations--0.2, 2, 10, and 20 mg/l) on the production of coumarins in the suspension culture of Angelica archangelica L. cultinated in the dark and under permanent lighting(3500 lux). The effect of the light regimen is, in comparison with auxins, less marked--the content of coumarins is mostly comparable both under permanent lighting and in the dark. The highest coumarin content was achieved with the use of alpha-naphthalene-acetic acid in a concentration of 0.2 mg/l with cultivation in the dark.

Structure-cytotoxic activity relationships of simple hydroxylated coumarins.

Several hydroxylated and/or methoxylated coumarin derivatives were tested for their relative cytotoxicity on four human tumor cell lines (oral squamous cell carcinoma HSC-2, HSC-3, melanoma A-375 and promyelocytic HL-60) and three normal human cells (gingival fibroblast HGF, periodontal ligament fibroblast HPLF and pulp cell HPC). Tumor cell-specific cytotoxicity was detected in all 6,7-dihydroxy-substituted coumarins only. The observations indicate that the tumor-specific cytotoxicity of the naturally occurring coumarin esculetin can be further enhanced by proper substitutions at 3- and/or 4-position(s) of the molecule. Agarose gel electrophoresis revealed that esculetin and its derivatives with tumor-specific cytotoxicity induce internucleosomal DNA fragmentation in HL-60 cells.

[Antiradical activity of complex copper compounds (II) on coumarin ligand base].

The antioxidant capacity of copper chelates with coumarins has been studied by the method of iron-induced chemiluminescence. All substrates studied were potent antioxidants, comparable to butylated hydroxytoluene. The mechanism of the antioxidant action of these copper-coumarin chelates was similar to that of Cu-Mn-superoxide dismutase, with a coumarin part of the complex being involved as a free radicals trap.

Inhibitory effect of eight simple coumarins on the lucigenin enhanced chemiluminescence of rabbit neutrophils.

The generation of superoxide anion (O2*-) and other reactive oxygen species (ROS), such as HO*, HOCl, NO, 1O2 and H2O2, by stimulated polymorphonuclear leukocytes during phagocytosis is a major mechanism of host defense against invading microorganisms. However, large amounts of ROS are suggested to be responsible for many diseases. In this work we studied the inhibitory effect of eight simple coumarins on O2*- generation by rabbit neutrophil upon stimulation with opsonized zymosan, using lucigenin enhanced chemiluminescence assay. We observed that coumarins containing hydroxy or acetoxy substituents at position 7 of the benzopyrone ring were the most active ones (IC50 values ranging from 6.0 +/- 2.8 to 18.6 +/- 2.6 micromol/L). Substitution of these groups by allyloxy or metoxy groups decreased the activity and unsubstituted coumarin had no effect. Cell damage after exposure to 200 micromol/L of each compound was determined by measurement of the activity of the released cytosolic lactate dehydrogenase and by Trypan Blue dye exclusion test. None of the drugs affected significantly the cellular viability.

Heterozygous protein C deficiency and coumarin necrosis of the skin.

An obese female patient aged 47 with a personal and familial history of recurrent venous thrombosis, who developed a coumarin-induced skin necrosis is presented. Laboratory investigations, performed three months after the acute event and in absence of coumarin therapy, emphasized a decreased anticoagulant activity of her plasma protein C (50% of the normal). These results as well as the high incidence of thrombotic disease in her relatives point to a familial heterozygous protein C deficiency. The antithrombotic role of the protein C system and the mechanism of coumarin induced necrosis of the skin are briefly discussed.

Management of coumarin-associated coagulopathy in the non-bleeding patient: a systematic review.

BACKGROUND AND OBJECTIVES: Excessive anticoagulation is a frequent complication of anticoagulant therapy. The risk of hemorrhage approximately doubles for each one point increase in the International Normalized Ratio (INR) above 3.0. Reducing a prolonged INR to within the desired therapeutic range requires that oral anticoagulants be withheld. In addition, vitamin K may be administered. Since this latter treatment can produce rapid reductions in the INR, it must be carefully tailored to meet individual needs, balancing the risk of bleeding against the potential risk of causing thromboembolism. METHODS: To review available literature on the management of coumarin-associated coagulopathy in asymptomatic patients, a Medline search was carried out and papers published in English from 1966 and 2003 were identified. All available information on the management of asymptomatic patients presenting with coumarin-associated coagulopathy was analyzed. RESULTS: Following the results of clinical studies that only used an elevated INR as a surrogate end-point for the risk of bleeding, low dose oral vitamin K appears as the preferable strategy for rapidly restoring therapeutic INR levels in asymptomatic patients who present with an excessively prolonged INR due to warfarin therapy. For the treatment of patients with asymptomatic acenocoumarol-induced coagulopathy, vitamin K does not add any benefit to the strategy of simply withholding oral anticoagulant treatment. INTERPRETATION AND CONCLUSIONS: Large randomized trials using clinical end-points are now required to provide evidence-based treatment recommendations for patients with coumarin-associated coagulopathy.

[Location and relative quantity of coumarins in the stem of Dendrobium thyrsiflorum].

AIM: To determine the location and relative quantity of coumarins in the stem of Dendrobium thyrsiflorum Rchb. f. , and to provide a scientific basis for evaluating and utilizing the famous medicinal plant. METHODS: The stems of one, two and three years old, separately, were collected in February. Location and relative quantity of coumarins in the top, middle and basal parts of each stem sample were determined by using laser scanning confocal microscopy (LSCM). ANOVA and Tukey's test were employed in the statistical analysis. RESULTS: The coumarins located mainly in vascular bundles, especially on the walls of the outer fiber cells. There was significant difference or great significant difference exist in every part of the stem from different ages. The values of significant difference were as follows: basal part P = 0.004 (< 0.01); middle part P = 0.009 (< 0.01); top part P = 0.036( < 0.05). CONCLUSION: Two years old stem could be the best choice when collecting Dendrobium thyrsiflorum in Feburary.

[Influence of coumarin on the lipolytic activity in the hyperlipaemic plasma of cocks].

Two groups of cocks were kept for 84 days on a diet rich in fat (group D) or on a similar diet with addition of coumarin (group DK). In the plasma of these birds there occurred a distinct increase of the concentration of total fat (TC) and cholesterol (CH), in comparison with the concentration of these substances of the plasma of control birds. The concentration of TC in the plasma of group DK was higher than that observed in group D. The concentration of CH was similar in both experimental groups. The level of the lipolytic activity of plasma (ALpO) changed during the experiment and was higher in the plasma of the cocks of group DK than in that of group D. Confrontation of the results of the determinations of ALpO, TC and CH in groups D and DK demonstrates that the symptoms of a disturbance of the metabolism of fats, which occur in cocks kept on a diet rich in fats, are stimulated by coumarin. This influence is demonstrated by a speeding up, by about 42 days, of the occurrence of these symptoms, i.e. of the increase of the TC concentration and of the ALpO level. The values of TC and ALpO, determined on the 21st and 42nd day of the experiment in group DK, corresponded to the relevant values found in group D on the 63rd and 84th day of the experiment. It is highly probable that coumarin acts on the mechanisms which control the ALpO level indirectly, through the carbohydrate metabolism.

[Antiradical activity of 3-substituted coumarins and their effect on iron-dependent chemiluminescence].

The antioxidant capacity of 3-aminocoumarin, 3-oxycoumarin, 3-acetylaminocoumarin, and 3-coumarin carbonic acid has been investigated with chemiluminescence measurement and by the accumulation of TBA-active products. All coumarins were found to be antioxidants, with 3-oxy-, 3-amino- and 3-acetylamino coumarins being capable of amplifying chemiluminescence at early stages of the process.

[The antiradical activity of coumarin reductones].

The antiradical activity of coumarine reductones was investigated by the method of inhibition of Fe2+ induced chemiluminescence of egg-yolk lipoproteins. All coumarines studied exhibited high antioxidant activity. The dependence of chemiluminescence intensity on the antioxidant concentration shows that coumarines reductions resemble their chemical analog--ascorbic acid rather than the lipid antioxidant butilated hydroxitoluene (ionol).

Percutaneous absorption of coumarin, griseofulvin and propranolol across human scalp and abdominal skin.

The objective of this study was to investigate the role of the transfollicular pathway in the diffusion process of chemicals through excised human skin in vitro. Skin was obtained from 5 cadavers (3 males, 3 females) within 24 h of death. The age of the subjects varied between 18-77 years. Three radiolabelled drugs, namely 14C-coumarin, 3H-propranolol and 3H-griseofulvin, were studied. The permeation parameters such as flux, lag time, diffusion coefficient and permeability constant were determined across scalp and abdominal skin using the Thomas Diffusion Cell. For all tested substances the flux through scalp skin was higher than across abdominal skin and the lag time was decreased. The differences were statistically significant at p less than 0.05 for coumarin and propranolol. These data suggest that the transfollicular pathway in permeation might have a significant impact on the diffusion parameters for some drugs. Also, in the case of coumarin, permeability seems to be epidermis/dermis-controlled, whereas for griseofulvin and propranolol the Stratum corneum apparently is the permeability limiting barrier.

Prevention of early recurrence of high risk malignant melanoma by coumarin. Irish Melanoma Group.

Coumarin (1,2-benzopyrone) as a daily oral dose of 50 mg was evaluated in a multicentre prospectively randomized, double-blind, placebo-controlled trial to prevent early recurrence of malignant melanoma TNM Stage IB (Breslow thickness greater than 1.70 mm) and Stage II. Intake for the trial started in 1984 and was stopped prematurely, after review, in 1987. There were two recurrences in 13 treated patients and 10 in 14 controls which was significant (P 0.01). The sites of the metastases differed in each group, being local and in bone in the treated group, and in lymph nodes, skin and lung in the control group. There were no toxic effects associated with coumarin treatment and all patients without recurrence are now receiving coumarin.
