A squash preparation method for root meristem field studies.

We present a field method for squash preparation of root apical meristem that does not require dry ice for cell adhesion to a slide. The method is based on the use of a weak gelatin solution as a stabilizing agent to prevent cell loss. Alcian blue 8GX was used as a counterstain for better recognition of cell shapes. Root meristems of Sonoran Desert succulents were used to illustrate the method. Fixed root tips were stained by Feulgen and squashes were prepared in a 1:2 mixture of solution A (45% acetic acid) and solution B (3:1 mixture of 0.5% gelatin in water:0.02% of alcian blue 8GX in 45% acetic acid). After squashing, the material was lightly dried for 5-6 min, then dehydrated and mounted in Canada balsam. Our method permits observation of chromosomes, recognition of meristematic and nonmeristematic cells, and estimation of mitotic indices and thus recognition of growing, dormant, and dead roots. The method can be used for estimation of cell division cycle time by colchicine and other methods where squash preparation is required.

Resistance of two planarian species to UV-irradiation.

The aim of this work was to determine the effects of 20, 25 and 30 minute UV-irradiation periods lambda = 253.5 nm to two planarian species Dugesia tigrina (Gir.) and Polycelis felina (Daly.). In vivo, UV light effects have been reported to affect intracellular receptors and disrupt simple behaviour. The effects of UV-rays on mortality and behavior as well as morphological, cytological and histological changes in the two planarian species were assessed, and the course and the dynamics of regenerative processes were compared between them. Experimental populations of Dugesia tigrina and Polycelis felina species were maintained in laboratory conditions at room temperature. Mortality, behavioral and morphological changes were monitored daily by means of a light stereomicroscope. For cytological and histopathological analysis, planarians were fixed in Bouine fixative on the first, second, third, fifth and seventh day after exposure to UV-irradiation, respectively. They were embedded in paraffin, cut on a microtome, stained with toluidin blue and embedded in Canada-balsam. UV-rays caused mortality, behavioral, morphological, cytological and histological changes in each planarian species. In regeneration of damaged body parts reticular cells and neoblasts played the main role. Neoblasts as totipotent cells extremely increased in number in the area of damaged tissue, immediately after UV-exposure. Dugesia tigrina was more sensitive to UV-rays than Polycelis felina due to possession of less pigmented cells. The course of regeneration in both species was similar. Most individuals of both species regenerated in 5 to 12 days after UV-irradiation.

Ocular linguatuliasis in Ecuador: case report and morphometric study of the larva of Linguatula serrata.

Linguatula serrata is a pentastomid, a cosmopolitan parasite belonging to the Phylum Pentastomida. Humans may act as an intermediate or accidental definitive host of this parasite, manifesting the nasopharyngeal or visceral form, with the latter having been described more frequently. The occurrence of ocular linguatuliasis is extremely rare, but it has been reported in the United States and Israel. The objective of the present paper was to report the first case of ocular linguatuliasis in Ecuador and to extend the morphologic study of L. serrata by morphometric analysis. The patient studied was a 34-year old woman from Guayaquil, Ecuador who complained of ocular pain with conjunctivitis and visual difficulties of two-months duration. Biomicroscopic examination revealed a mobile body in the anterior chamber of the eye. The mobile body was surgically removed. The specimen was fixed in alcohol, cleared using the technique of Loos, stained with acetic carmine, and mounted on balsam between a slide and a coverslip. It was observed with stereoscopic and common light microscopes in combination with an automatic system for image analysis and processing. The morphologic and morphometric characteristics corresponded to the third-instar larval form of L. serrata. To our knowledge, ocular linguatuliasis has not been previously described in South America, with this being the first report for Ecuador and South America. The present study shows that computer morphometry can adequately contribute both to the morphologic study and to the systematic classification of Pentastomids, and L. serrata in particular.

Determination of ochratoxin A in red wine and vinegar by immunoaffinity high-pressure liquid chromatography.

A method is described for the determination of ochratoxin A (OTA) in red wine and vinegar using an acidic chloroform extraction, an immmunoaffinity clean-up step, and a high-performance liquid chromatographic determination with fluorescence detection. The detection limit was estimated at 0.002 microg/liter. The mean recovery factors were found at 91.3 and 96.6% for wine and vinegar, respectively. Thirty-one samples of red wine originating from Mediterranean sea countries and 15 samples of vinegar were examined for the presence of OTA. All red wine samples contained OTA. Seventy-two percent of these samples were found to be contaminated over 0.1 microg/liter. Among them, nine samples contained ochratoxin A in the range of 0.5 to 3.4 microg/liter, 12 samples in the range of 0.10 to 0.50 microg/liter (median: 0.176 microg/liter), and 9 samples in the range of 0.010 to 0.100 microg/liter (median: 0.041 microg/liter). All 15 vinegar samples showed the presence of OTA. The most contaminated ones were three balsamic vinegar samples containing 0.156 microg/liter, 0.102 microg/liter, and 0.252 microg/liter of OTA. In the remaining 12 samples, ochratoxin A levels ranged from 0.008 microg/liter to 0.046 microg/liter (median: 0.012 microg/liter). These data are in good agreement with the hypothesis that wine originating from Southern countries might contain significant OTA concentration and showed the possible occurrence of traces of OTA in vinegar.

Differences in urinary chemistry profiles of moose on Isle Royale during winter.

During winters 1987-1988 (I) and 1988-1989 (II), we assessed the physiological status of moose (Alces alces) residing on the west and east ends of Isle Royale (Michigan, USA) by collecting and chemically analyzing urine deposited in snow (snow-urine) from January to early March. Samples were assayed for urea nitrogen (U), sodium (Na), potassium (K), calcium (Ca), phosphorus (P) and creatinine (C). Throughout both winters, elevated urinary U:C ratios in snow-urine samples collected from east-end moose compared to west-end moose indicated greater dietary energy deprivation and accelerated net catabolism of endogenous protein. Sodium: C ratios were low throughout the study and were similar between moose from both ends of the island, except during the middle of winter I. Greater K:C, P:C and Ca:C ratios in east-end moose compared to west-end moose throughout winter I, and increases in these ratios and U:C in east-end moose from middle to late winter during the second year provided additional evidence of a greater deterioration in condition in east-end moose. The superior nutrition provided to moose on the west end of the island was associated with more developed soils and diverse vegetation and a lower stem density of balsam fir compared to the east end.

[Leg ulcers. Allergologic studies of 359 cases].

INTRODUCTION: Leg ulcers are a common disease in dermatology. Their social and economic implications are important, especially when contact dermatitis complicates these wounds. PATIENTS AND METHODS: In this study a total of 359 patients had contact dermatitis investigations over a period of 5 years. Patch-tests were performed on 116 men and 243 women hospitalized with venous and/or arterial leg ulcers, with or without clinical appearance of periulcerous contact dermatitis. Standard patch-tests of the ICDRG (International Contact Dermatitis Research Group) were systematically performed as well as a specific series of 40 tests. RESULTS AND DISCUSSION: Positive patch tests were observed in 82.5 p. 100 of the patients, indicating a very high rate of contact allergy in patients with leg ulcers. Peru balsam, lanolin and neomycin were the most frequent culprits of positive patch-tests in this population. Five percent of patch-tests with eosin were positive in our study. This is not common in the literature. Moreover, we highlight the high rate of contact sensitization to corticosteroids in this population. According to these results and to the literature, a new series of patch-tests for leg ulcers is suggested. CONCLUSION: Polysensitization in patients with chronic wounds is very frequent. A series of patch-tests in leg ulcers may lead to some interesting conclusions but a good questioning of the patient is always necessary to complete this series.

Significance-prevalence index number: a reinterpretation and enhancement of data from the North American contact dermatitis group.

BACKGROUND: The North American Contact Dermatitis Group (NACDG) periodically reports allergen reaction rates and clinical significance. These results are not formally interpreted combining both components into one index. OBJECTIVE: The purpose of this study is to introduce the Significance-Prevalence Index Number (SPIN), which is a parameter that assesses the relative importance of different allergens. METHODS: Data was collected from published studies reported by NACDG from 1984 through 1996. The SPIN for each tested allergen was calculated as the product of the proportion of the population allergic by the weighted clinician-assessed likelihood of relevance of the reaction. RESULTS: In the studies done in 1992 through 1996, fragrance mix, quaternium-15, balsam Peru, formaldehyde, thiuram mix, nickel sulfate, neomycin, bacitracin, carba mix, and paraphenylenediamine were the 10 leading allergens in clinical importance as measured by SPIN. Our SPIN calculations suggest that ethylenediamine and benzocaine have markedly decreased in clinical importance over the past decade, whereas methylchloroisothiazolinone/methylisothiazolinone has become a leading allergen. A number of allergens demonstrate high to intermediate SPIN, yet are not included in the T.R.U.E. test. These clinically important allergens include bacitracin, diazolidinyl urea, glyceryl thioglycate, ethyleneurea melamine-formaldehyde, and imidazolidinyl urea. CONCLUSION: SPIN allows clinicians to incorporate both the allergenicity and the relevance of the allergen into one measure.

[Standard epicutaneous tests in ambulatory care of patients at the dermatovenereology clinic].

BACKGROUND: Allergic diseases have a rising trend worldwide. Allergic contact eczema is the most frequent occupational disease. The spectrum of allergens is influenced by a number of factors. The basic method for detection of contact allergy and detection of the allergen are epicutaneous tests. The objective of the work was to evaluate the results of these tests in patients with contact dermatitis. METHODS AND RESULTS: A total of 437 patients were tested (163 men, 274 women). For testing an extended standard set of medicamentous and non-medicamentous allergens was used. In women more often positive reactions were obtained (39.5%) than men (27.5%). The mean age of women was 40 years, that of men 47 years. Both groups responded on average to 1.6 and 1.7 allergens resp. The most frequent allergen in women was nickel (17.9%) in men chromium (12.9%). Other frequent allergens in women included chromium (11.3%), Peru balsam (3.6%), HgCl2 and cobalt. In men it was nickel, cobalt (6.1%) and HgCl2. There were also concurrent allergies. (The results are not statistically significant). In youngest age group the number of women was more than sevenfold: the most frequent allergen in this group was nickel. In the group of patients above 60 years of age women also predominated (2.5x): most frequently allergy to chromium was found (9.1%). The allergic reaction to camomile and tar was significantly higher. CONCLUSIONS: The most frequent allergen in standard epicutaneous tests in women is nickel, in men chromium. Other frequent allergens are cobalt, mercury and Perubalsam. Women have a higher ratio of positive reactions, in the group under 30 years women predominate 7x, in the whole group 2.4x.

Salivary gland neoplasms: a descriptive analysis of the pattern seen in Enugu.

The objective of this study is to describe the pattern of salivary gland neoplasms as experienced in Enugu-Nigeria. Patients are selected from those attending the Otorhinolaryngology units of the author at the University of Nigeria Teaching Hospital and Balsam Clinics both in Enugu from January 1992 to December 1997, with tumours of salivary gland origin. Non-neoplastic lesions were excluded. There were forty-one (41) patients with salivary gland neoplastic growths in the following sites: parotids (25) submandibular gland (SMG) (10) and minor (6). There were 15 males, 26 females; age range 10-74 year, with mean of 41 years. Seven of the parotid tumours were recurrent/residual. Salivary gland neoplasms are important surgical disease in this region with clinical manifestations similar to what obtains in other parts of the world. Surgical excision of salivary neoplasms is beneficial, but misadventure is still rampant.

How to test for fragrance allergy.

Fragrance is the most common cause of cosmetic allergic contact dermatitis. Fragrance allergy detection is best accomplished by testing with the fragrance mixture, balsam of Peru, and either jasmine synthetic or absolute. It would be desirable to have common fragrance allergens listed on cosmetic labels so that patients could avoid the allergens to which they are allergic.

Spectral reflectance of Picea rubens (Pinaceae) and Abies balsamea (Pinaceae) needles along an elevational gradient, Mt. Moosilauke, New Hampshire, USA.

Relationships among elevation, foliar morphology, spectral reflectance, and chlorophyll fluorescence of two co-occurring montane conifers, red spruce (Picea rubens Sarg.) and balsam fir (Abies balsamea [L.] Mill.), were investigated along two transects from 460 to 1460 m on Mt. Moosilauke in the White Mountains of New Hampshire, USA. Spectral reflectance (300-1100 nm wavelengths) and the chlorophyll fluorescence F(v)/F(m) ratio were measured on dark-adapted needles. Foliar morphology (needle size, shape, and mass) and nitrogen concentrations were measured in the laboratory. Reflectance spectra varied between species and with elevation. Two chlorophyll measures, red edge position and a chlorophyll-based difference index (Chl NDI = R750 - R705/R750 + R705), indicated more chlorophyll in fir than in spruce and decreasing chlorophyll with increasing elevation in both species. The structure-independent pigment index (SIPI = R800 - R445/R800 - R680) increased with elevation, indicating an increasing carotenoid : chlorophyll ratio. The photochemical reflectance index (PRI = R531 - R570/R531 + R570), a measure of photosynthetic radiation use efficiency, decreased with increasing elevation up to 1370 m. In the highest elevation site, within the stunted alpine krummholz at 1460 m, PRI was higher than at 1370 m, but still lower than at 1070 m. This same pattern was evident in the chlorophyll fluorescence F(v)/F(m) measurements. These independent indices indicate higher stress in spruce than fir, which may be related to the "spruce decline" reported in the northeastern USA. Results also indicate progressively increasing stress with increasing elevation up to 1370 m. Stress appears to be lower at 1460 m than at 1370 m, despite the harsher conditions at the very summit of Mt. Moosilauke. This may be a consequence of stress-tolerant physiology and/or prostrate architecture.

[Not Available].

In 1686 the Zeeland Chamber of the West Indian Company undertook a serious effort to establish a colony on the Wild Coast at the Pomeroon. The Wild Coast, a territory described as stretching from the Amazon to the Orinoco river, was of growing significance for the trade in pharmaceutical and technical products of the Guyana country: dyes, letterwood, balsam of copaiba, tobacco, sugar, vanilla beans and carape oil. The expedition consisted of the ship 'De Vrijheyt' which was dispatched from Flushing with the new commander Jacob de Jongh, his family, some soldiers and the surgeon David van Cassel aboard. The latter could rely on a surgeon chest with 103 medicines. The new Pomeroon colony however quickly collapsed. Mortality due to dysentery and malaria was high and the lack of leadership led to faulty discipline. The list of medicines used to combat diseases is reviewed in this article. Antimony takes an important place in the assortment and theriac was used for its anti-inflammatory activity.

[Eugenol in root canal cement:possible cause for urticarial reactions].

A case is reported of a patient with chronic urticaria. The correlation between the symptoms and dental treatment gave rise to the supposition that a root canal cement was causing the trouble. This hypothesis has been tested with epicutaneous patch tests and intradermal tests in cooperation with the Utrecht University Hospital (the Netherlands), Institute of Dermatology, Clinical Allergy Section. A causal relationship with eugenol, a cement constituent, could only be established through provocative oral ingestion. Nevertheless, caution is needed when dealing with chronic urticaria in view of the risk of anaphylactic reaction. Consequently, the article highlights the allergenic aspects of eugenol, as a substance associated with balsam of Peru. Attention is focused on the fact that eugenol, used in dental materials, is able to elicit and maintain urticaria protractedly.

[Allergic contact dermatitis to the Comfeel hydrocolloid dressing].

INTRODUCTION: Allergic contact dermatitis is frequent in patients with chronic leg ulcers. However, it rarely occurs with modern wound dressings and is exceptional with hydrocolloids. CASE REPORT: A 66-year-old woman was treated for a leg ulcer with the Comfeel plus(R) transparent hydrocolloid dressing for two months. She developed a pruriginous, erythematous and vesiculous dermatitis under the hydrocolloid plaques. Patch tests for the Comfeel plus(R) transparent hydrocolloid, the Comfeel plus(R) hydrocolloid, balsam of Peru and epoxy resin were positive. Only the positive test for the Comfeel plus(R) transparent hydrocolloid was clinically pertinent. The histological examination of the positive test was suggestive of eczema. DISCUSSION: To our knowledge, allergic contact dermatitis to Comfeel plus(R) hydrocolloid dressings has not been reported. Most previous studies which included systematic patch-testings in patients with leg ulcers showed high sensitization rates for various allergens, but no allergy to hydrocolloids. Only isolated cases of allergic contact dermatitis to another hydrocolloid (Duoderm E(R)) have been reported. Our case report shows that allergic contact dermatitis is a possible side-effect of Comfeel plus(R) hydrocolloid dressings. However, it seems exceptional. Since the patch-tests failed to identify the constituent responsible for this allergy in our observation, comprehensive allergologic investigations should be repeated in further cases.

Interference of the position of substances in an epicutaneous patch test battery with the occurrence of false-positive results.

BACKGROUND: Epicutaneous patch tests represent a practical and objective method that help in the etiologic diagnosis of allergic contact dermatitis. The technique of patch test application is an important factor in obtaining good results. OBJECTIVES: The aims of this study were (1) to determine whether the substances that form the test battery interfere with the patch test result and (2) to establish a rule for positioning the substances during patch test application. METHODS: Two hundred patients were studied. The standard patch test battery was applied in 3 versions. The original, with the substances applied in alphabetical order was called version 1 (V1) and tested on the left back in all patients In 100 patients, on the right side, the same substances were tested but applied at different positions, avoiding the proximity of elements with a tendency to cross reaction and/or cosensitizing. This version of the battery was named version 2 (V2). Another 100 patients had V1 applied to the left back, and, on the right side, the version 3 of the battery (V3) was applied, consisting of the same allergens but placed close to those with a tendency for cross reaction and/or cosensitization. RESULTS: In the group V1-V2, 163 results were positive in V1 and 124 in V2, with the difference being statistically significant (P <.05). In the group V1-V3, 134 results were positive in V1 and 207 in V3, with the difference also being significant (P <.005). The substances with the largest number of positive results, when tested close to other elements with a tendency for chemical affinity, were parabens, fragrance-mix, thimerosal, balsam of Peru, potassium dichromate, cobalt chloride, mercapto mix, and propylene glycol. CONCLUSIONS: (1) The substances of a patch test battery eliciting positive responses may interfere with test positivity to neighboring substances. (2) In addition to the already established techniques, the position of the substances forming the test battery needs to be determined; substances with a tendency to cross reaction or cosensitizing substances should be tested distant from one another, thus preventing the occurrence of false-positive results.

[Optimization model of spatial population structure: example of poplar moth laying eggs on leaves].

The authors analyze spatial distribution and survival of populations of poplar moth Litchcolletis populifoliella Tr. on its feeding plant--balsam poplar Populus balsamifera. Imago of the moth glue its eggs on the leaves thus determining the future location of their offspring on the host plant. Spatial distribution of eggs on leaf surface and distribution of leaves according egg numbers are not random. On the short distance from each egg the average number of eggs is less, than it should be in case of random distribution. While this distance increases up to some particular value the occurrence of eggs is higher than random. Thus, the eggs of moth are located by groups on the leaf surface. Within each group eggs are situated not very close to each other, this allowing larvae to lower competition for common resource. It is suggested that on the same feeding plant individuals have different interactions: competition, caused by limited quantity of resource and cooperation that is necessary to resist leaf defensive (antibiosis) reaction.
