Studies of the infection of the cockroaches (Blattella germanica[L.]) with Bac. thuringensis.

The results of an experimental infection of the cockroaches Blattella germanica (L.) with some trade preparations containing Bac. thuringensis spores are presented. Biological studies supported by bacteriological and histological examinations of the excised intestines of the above insects have shown that the microorganisms studied are pathogenic for the cockroaches. This effect might be eventually applied as a biological method of eradication of the cockroaches in such environments, where chemical preparations can not be used or this disinfestation method would be complementary to the other method of disinfestation.

[Serine proteases from Bac. subtilis].

Using biospecific adsorbent and subsequent gel-filtration of Sephadex G-75 three fractions of serine proteases (I--III) having different physicochemical properties were isolated from Bac. subtilis. The first protease had molecular weight of 23000--24000 (pH optimum 6,5, activation energy 16,6 ccal/mol. The second one had molecular weight of 29000, pH optimum 11,0, activation energy 14,4 ccal/mol. The third protease was a mixture of proteases with average molecular weights 26000 and pH optima at 7,0, 8,5 and 11,0.

Allogeneic bone marrow transplantation in high-risk myeloid disorders using busulfan, cytosine arabinoside and cyclophosphamide (BAC).

Twenty-one patients (median age = 34, range = 10-49; F:M = 7:14) received a preparative regimen consisting of busulfan 4 mg/kg/day x 4, cytosine arabinoside 2 g/m2/12 h x 4 and cyclophosphamide 60 mg/kg/day x 2 ('BAC' regimen) for allogeneic bone marrow transplantation. Out of 12 patients with acute myeloid leukemia (AML), two were in first remission, six were in second remission and four had resistant, relapsed disease or prolonged marrow aplasia after induction chemotherapy. Five of the 12 patients with AML had secondary AML. Four patients had transfusion-dependent myelodysplastic syndrome. Three patients with chronic myeloid leukemia were in the accelerated phase and two were in the blastic phase. Organ toxicities related to the preparative regimen were graded. Liver toxicity occurred in 11 patients, two of these were fatal veno-occlusive disease (VOD) (10%). Nineteen of the 21 patients had grade 2 or less diarrhea, and 13 also had mucositis. One patient developed grade 3 cardiac toxicity, and one other patient had grade 1 skin toxicity. Four patients had gross hematuria related to treatment (19%). No renal, pulmonary or CNS toxicities were encountered. Ten patients have died, two from regimen-related hepatic VOD. Of the remaining eight deaths, four were from respiratory failure in four patients (one case each of Pneumocystis pneumonia, CMV pneumonia, bronchiolitis obliterans associated with chronic graft-versus-host disease, and interstitial pneumonitis complicated pulmonary emboli), and one patient each from GI bleeding, cardiac arrhythmia, sepsis and CNS bleeding. Thus far, only one patient transplanted for secondary AML in second remission relapsed at day 230.(ABSTRACT TRUNCATED AT 250 WORDS)

[Determination for optimal ozone dose in O3-BAC].

The ozonation can increase biodegradability of water and the biofilter after ozonation can increase biostability of water. The result shows that with different doses of ozone between 2 and 8 mg/L, ozonation can increase AOC-P17, AOC-NOX and BDOC by 20.9%-85.5%, 42.1%-158.2% and 21.4%-84.4%, respectively. The optimum ozone dose for maximum AOC and BDOC formation is 3 mg/L. In the test, the AOC of effluent from BAC is lower than 50 microg/L (acetate-C). The AOC concentration is between 35.9 and 46.6 microg/L (acetate-C), and the effluent belongs to biostability.

A Survey of the Brassica rapa genome by BAC-end sequence analysis and comparison with Arabidopsis thaliana.

Brassica rapa ssp. pekinensis (Chinese cabbage) is an economically important crop and a model plant for studies on polyploidization and phenotypic evolution. To gain an insight into the structure of the B. rapa genome we analyzed 12,017 BAC-end sequences for the presence of transposable elements (TEs), SSRs, centromeric satellite repeats and genes, and similarity to the closely related genome of Arabidopsis thaliana. TEs were estimated to occupy 14% of the genome, with 12.3% of the genome represented by retrotransposons. It was estimated that the B. rapa genome contains 43,000 genes, 1.6 times greater than the genome of A. thaliana. A number of centromeric satellite sequences, representing variations of a 176-bp consensus sequence, were identified. This sequence has undergone rapid evolution within the B. rapa genome and has diverged among the related species of Brassicaceae. A study of SSRs demonstrated a non-random distribution with a greater abundance within predicted intergenic regions. Our results provide an initial characterization of the genome of B. rapa and provide the basis for detailed analysis through whole-genome sequencing.

[Effect of different salts and their concentration on polymyxin M biosynthesis by Bac. polymyxa Ross].

The effect of various concentrations of ammonium sulfate, sodium and potassium chlorides and sulfates on biosynthesis of polymyxin M by Bac. polymyxa Ross. on media containing wheet meal as the carbon source was studied. The culture growth, antibiotic accumulation and consumption of the main nutrients was studied as dependent on the level of the salts in the medium. The highest productivity was observed on media with the salt concentrations amounting to 1--2 per cent except sodium sulfate: changes in the concentration of the latter had practically no effect on the activity of the fermentation broth.

[Mutant Bac. subtilis A2 with an enhanced capacity to induce an adaptive response].

Mutant A2 with increased ability to induce adaptive response was isolated in Bac. subtilis and its properties studied. Mutant A2 was shown to be more resistant to mutagenic action of MNNG, EMS, UV light. It was also discovered that A2 was more sensitive to the lethal action of MNNG and UV light than parent strain 103. It was shown by clonal analysis of mutant colonies, formed by mutant cells A2 and 103 that A2 strain had increased ability to form complete mutants. Properties of A2 mutant suggest that in the process adaptive response induction were is expression of both adaptive response enzymes and some other which are necessary for reparation of premutagenic UV lesions.

[Effect of Bac. intermedius RNAse on a cell population of ascitic lympholeukemia NKLy].

RNAase of Bac. intermedius and a modification of this enzyme containing a histidine-inactivated active site were studied for their influence on the activity of proliferation of murine ascitic lympholeukemia cells (NKLy) using a 3H thymidine label. Each mouse received a single intraperitoneal injection (dosage 1 mg per kg) of the normal or inactivated enzyme. It was shown that irrespective of the catalytic activity both enzyme preparations decreased the proliferative activity of the cells, blocked G2-M, slowed the course of the prophase and metaphase of meiosis, diminished the number of cells synthetizing DNA and lowered the intensity of labelling. After 24 hours all these characteristics returned to normal.

[Change in the activity of tumor cell antigens as affected by treatment of the cells with the metabolic products of Bac. mesentericus AB-56].

Tumour cells (ascitic A/Sn mouse lymphosarcoma ON-2), treated by the Bac. mesentericus AB-56 metabolism products used for the production of antitumour autovaccines were analyzed for their antigenic properties. The reactions of precipitation in gel, zonal and oncoming immunoelectrophoresis, passive hemagglutination as well as cytotoxic, absorption and transplantation tests were used. A considerable number of soluble and insoluble antigens inherent in intact tumour cells were inactivated by the products of microbial metabolism in the process of antitumour vaccine production, and modified antigens which induce synthesis of humoral antibodies in the syngenic system appeared.

[Modification of 3-methylcholanthrene-induced blastomogenesis in BALB/c mice by using the subcellular components of Bac. megaterium H].

It is shown that the cytoplasm of Bac. megaterium H possessing the antigenic affinity with the malignant tumour cells, being injected 7 days before the 3-methylcholanthrene injection promotes intensification of its blastomogenic effect, reduces both the period of tumour appearance and longevity of the tumour-bearing animals. On the contrary the cell wall preparation under the similar conditions, produces a therapeutic effect. An assumption is advanced that the effect of these preparations is associated with their different influence on the immune system.

BACS: evolution of an integrated library system toward information management.

The evolution of the Washington University School of Medicine BACS integrated library system toward information management functions is outlined. The creation of a machine-readable database and its extension through telecommunications have consequences that reach beyond the functions of the library as we have perceived them. It is argued that libraries are flexible institutions that, with automation, are likely to enlarge rather than to diminish.

[Clinico-epidemiologic characteristics of group illness caused by Bac. cereus food poisoning].

The authors describe an outbreak of food poisoning caused by Bac. cereus embracing 80% of persons who catered at the same catering establishment. The poisoning occurred in the course of 15 hours after the moment of eating a sauce stored in warm place. In the majority of cases the course of the disease was mild; 77.8% of the patients displayed phenomena of enteritis, 20.2%--of entercolitis, and 2.0%--of gastroenterocolitis. All the patients recovered without any treatment in the course of 1--2 days.

[On the utilization different organic and anorganic compounds by some strains of Bac. cereus var. mycoides (Flügge) (author's transl)].

In order to investigate the decomposition of 46 organic and anorganic compounds by Bac. mycoides, the behaviour of four strains was tested by means of plate counting technique (after Koch) and the Warburg method. Mostly intensive substances containing protein were decomposed by all strains. Furthermore amino acid, sugar, alcohols, and ammonium salts are used by the bacteria, but there were large differences between strains tested. Urea and nitrates have not been decomposed.

[Effect of K+ ions on Bac. polymyxa 153 growth and the biosynthesis of polymyxin B].

The growth of Bac. polymyxa, 153 producing polymyxin B did not change under the effect of various concentrations of potassium ions, while the antibiotic productivity of the cells significantly decreased. A great amount of polymyxin is excreted by the bacteria into the fermentation broth and a certain portion of the antibiotic remains in the cells. Under the experimental conditions the potassium salts slightly changed the pH value of the fermentation broth and probably did not affect polymyxin secretion from the cells into the fermentation broth.

[Proteolytic enzymes bound to Bac. thuringiensis crystals].

It was demonstrated that crystals of entomopathogenic protein from Bac. thuringiensis contain admixture of proteinase either adhered to their surface on inconponated into crystal lattice defects. A proteolytic action, particularly when enhanced by crystal dissolution, causes progressive degradation of crystal proteins with molecular weights of 140 000--129 000 down to the components with smaller molecular weights. This may, at least, partially account for the contradictions in the literature data on crystal composition. Using synthetic peptide substrates and specific inhibitors, it was shown that the enzymes incorporated into crystals belong to serine and metalloproteases. The presence of leucine aminopeptidase was also noted. A method for enzyme separation from crystal has been developed.

[Effect of MEA on the accumulation of DNA breaks in Bac. stearothermophilus irradiated with gamma and ultraviolet rays and treated with nitrosomethylurea].

beta-Mercaptoethylamine (MEA) decreased the accumulation of enzymatic single- and double-strand breaks in DNA of thermophilic bacteria exposed to gamma- and UV-radiation and treated with N-nitroso-N-methylurea. The protective effect of MEA, as registered according to accumulation of single-strand and double-strand breaks in DNA of Bac. stearothermophilus immediately after irradiation and after 30 min postirradiation incubation, was similar.

Technology at Washington University School of Medicine Library: BACS, PHILSOM, and OCTANET.

A brief overview of the Bibliographic Access and Control System developed by the Washington University School of Medicine Library is presented. Because the system has been described in two previous reports, this paper focuses on its relationship to other automated programs (i.e., PHILSOM and OCTANET), education of users, evaluation of the system, and outreach to the medical center. In operation for more than two years, BACS represents the computerization of much of the managerial and operational functions of the library, and marks the completion of stage 1 of the three stages of library evolution described in the AAMC report Academic Information in the Academic Health Sciences Center: Roles for the Library in Information Management.
