Inhibition of type 5 adenovirus infectivity by periodate oxidation.

Periodate oxidation of purified type 5 Adenovirus (Ad5) led to a mean loss of infectivity of 6.84 logs. There were no significant differences in adsorption and penetration between oxidized and mock-oxidized virus. However, after infection with oxidized virus, no synthesis of viral structural proteins could be detected and a 78.5% inhibition of viral DNA synthesis was observed. Labelling experiments performed by treating oxidized and mock-oxidized virus with tritiated sodium borohydride revealed that the fiber glycoprotein was one of the proteins labelled in oxidized virus whereas no labelled proteins were detected in non oxidized virus. In addition, it was found that one mol of formaldehyde generated during oxidation of sugar residues was bound per 500 base pairs in oxidized virus. One consequence of this in situ generation of formaldehyde is the formation of DNA-protein crosslinks. The DNA so crosslinked showed different patterns of restriction fragments with endonucleases such as Hpa I, Hind III and Kpn I but not with Xho I.

Accelerated onset of viral transcription in adenovirus-infected HeLa cells treated with the tumor promoter 12-O-tetradecanoyl-phorbol-13-acetate.

When adenovirus type 5-infected HeLa cells were exposed to the tumor promoter 12-O-tetradecanoyl-phorbol-13-acetate, short pulse-labeling with [3H]uridine in vivo and [3H]UTP incorporation by isolated nuclei in vitro were both consistent with a decreased latent period before initiation by RNA polymerase at early viral promoters. Acceleration was not dependent upon concurrent protein synthesis and could not be attributed to rapid entry of virus into the cell nucleus. 12-O-tetradecanoyl-phorbol-13-acetate suppressed the transcription-delay phenotype of the E1a mutant, hr1, without restoring its ability to replicate.

A 60 kd cdc2-associated polypeptide complexes with the E1A proteins in adenovirus-infected cells.

p60 is a cellular protein that binds to the adenovirus E1A protein complex in virally infected or transformed human cells. In both infected and uninfected cells, p60 was found in a complex with the cdc2 protein kinase. Immune complexes containing p60 and cdc2 display a cell cycle-dependent histone H1 kinase activity that is most active in interphase. The previously described cdc2-p62/cyclin complex also acts as a histone H1 kinase but is maximally active in mitotic metaphase. The shift in the timing of activation of different cdc2-containing complexes suggests that each might play a distinct role in regulation of the cell cycle.

Pathogenesis of guinea pig adenovirus infection.

BACKGROUND AND PURPOSE: The existence of guinea pig adenovirus (GPAdV) has been suspected on the basis of histopathologic findings, but the virus has not yet been isolated. In susceptible animals, it may cause severe bronchopneumonia and death. Adenovirus-like inclusion bodies have been observed in the lungs of animals with clinical disease. Prevalence of the infection is unknown. Recently, a polymerase chain reaction (PCR) assay was described that was able to selectively detect GPAdV. METHODS: To investigate the pathogenesis of GPAdV, we inoculated eight guinea pigs with GPAdV; eight control animals were sham inoculated. The PCR assay was used to trace the infection. In a second experiment, transmission of GPAdV from an experimentally infected animal to five immune-naive cohorts was examined. RESULTS: None of the infected animals developed clinical disease. The GPAdV could be detected by PCR analysis of nasal-swab specimens on days 6 through 15 after infection. Infective virus could be recovered from the nasal mucosa during this period (as determined by inoculation of immune-naive animals). The virus was transmitted from an experimentally infected animal to two of five immune-naive cage mates. CONCLUSION: The GPAdV may cause transient subclinical upper respiratory tract infection that may descend to the lungs.

[Adenovirus infections of cattle].

Results are given of studies on the adenoviral infections in cattle in the region of Southeast Bulgaria. Serologic methods were used to elucidate some moments of the enzootic process and the part played by bovine adenoviruses in the etiology of the respiratory diseases of calves. Demonstrated was the participation of adenovirus strains of group II. It was established that the technologic moments influence the course of the adenoviral enzootics. A successful attempt was made to uncode the type picture and the circulation of types 1, 2, 3, and 8 adenoviruses within the cattle herds. The method of direct immunofluorescence was successfully employed in the diagnostics of adenoviral diseases in calves when alive. Demonstrated was also the presence of specific skin supersensitivity in cattle infected with adenoviruses. Data are given on the specificity of the reaction, the quality and standardization of the allergen, the age-associated dynamics of the process, etc.

Adenovirus infections in human immunodeficiency virus-positive patients: clinical features and molecular epidemiology.

Prospective surveillance of 63 human immunodeficiency virus (HIV)-positive patients and 9 HIV-negative partners over 5-27 months yielded 51 adenoviruses from 18 HIV-positive patients. These were serotyped and compared by restriction enzyme analysis (REA) together with 24 isolates from 19 other HIV-positive patients. The actuarial risk of infection at 1 year in HIV-positive patients was 28% (17% with entry CD4 cell count of > 200/mm3 and 38% with CD4 cell count of < or = 200/mm3, P = .03). The most frequent site of infection was gastrointestinal (17/18 patients) with mainly subgenus D adenoviruses, while urinary infection was caused by subgenus B or D. Prolonged fecal excretion (2-27 months) was associated with CD4 cell counts < 150/mm3. Identical strains were seen in 2 HIV-positive partners and 2 unrelated patients. Gastrointestinal infection was temporally associated with diarrhea in only 7 (41%) of 17 cases. The remainder (59%) were asymptomatic or minimally symptomatic, and diarrhea was often caused by other opportunistic pathogens.

Disseminated adenovirus infection with respiratory failure in pediatric liver transplant recipients: impact of intravenous cidofovir and inhaled nitric oxide.

Adenoviruses (AdV) are opportunistic pathogens that can lead to severe infections and respiratory failure (acute respiratory distress syndrome, ARDS) with high mortality in immunosuppressed patients. Cidofovir (CDV) has been used in adenoviral disease in bone marrow transplant recipients. Two pediatric liver transplant recipients with disseminated adenoviral disease and ARDS were treated with reduction of immunosuppression, CDV, and inhaled nitric oxide (iNO). CDV 1 mg/kg was given three times per week intravenously with intravenous hydration and oral probenecid. Viral suppression and clinical improvement was achieved. AdV hepatitis did not occur, and graft function was preserved, although acute rejection occurred in both patients. Adverse effects were mild and transient not requiring dose modification. Severe hypoxemia was reversed with iNO 10-20 p.p.m. CDV treatment of AdV infections in organ transplant recipients and other immunocompromised patients should be further investigated in prospective studies.

Molecular epidemiology of adenovirus infection among pediatric population with diarrhea in Asia.

A total of 3,577 fecal specimens from infants and children with acute gastroenteritis in Japan, Korea and Vietnam during 1998 and 2001 were tested for adenovirus by the ELISA method. Of these, adenovirus was detected in 158 (4.4%). The detection rate of adenovirus was highest in Korea (8.7%, 20/231) followed by 5.0% (100/1,991) in Japan and 2.8% (38/1,355) in Vietnam. All adenoviruses were further serotyped by PCR-RFLP. The diversity of adenovirus serotypes including Ad2, 3, 5, 8, 31, 40 and 41 was demonstrated. Worth of note was a decrease in the rate of isolation of Ad40 (7.6%, 12/158) and a concomitant increase of Ad41 (63.9%, 101/158) to become the predominant serotype. Another interesting feature of the study was the presence of Ad3 (4.0% and 30%) and Ad8 (8.0% and 20%) in Japan and Korea, respectively, which is mainly associated with keratoconjunctivitis worldwide. Our result underscored the importance of adenovirus in association with acute gastroenteritis in Asian countries.
