Switchable surfactants.

Many industrial applications that rely on emulsions would benefit from an efficient, rapid method of breaking these emulsions at a specific desired stage. We report that long-chain alkyl amidine compounds can be reversibly transformed into charged surfactants by exposure to an atmosphere of carbon dioxide, thereby stabilizing water/alkane emulsions or, for the purpose of microsuspension polymerization, styrene-in-water emulsions. Bubbling nitrogen, argon, or air through the amidinium bicarbonate solutions at 65 degrees C reverses the reaction, releasing carbon dioxide and breaking the emulsion. We also find that the neutral amidines function as switchable demulsifiers of an aqueous crude oil emulsion, enhancing their practical potential.

Anti-Pneumocystis activities of aromatic diamidoxime prodrugs.

Aromatic dicationic compounds, such as pentamidine, have potent antimicrobial activities. Clinical use of these compounds has been restricted, however, by their toxicity and limited oral activity. A novel approach, using amidoxime derivatives as prodrugs, has recently been proposed to overcome these limitations. Although results were presented for amidoxime derivatives of only one diamidine, pentamidine, the authors in the original proposal claimed that amidoxime derivatives would work as effective prodrugs for all pharmacologically active diamidines. Nine novel amidoxime derivatives were synthesized and tested in the present study for activity against Pneumocystis carinii in corticosteroid-suppressed rats. Only three of the nine compounds had significant oral anti-Pneumocystis activity. The bisbenzamidoxime derivatives of three direct pentamidine analogs had excellent oral and intravenous activities and reduced acute host toxicity. These compounds are not likely candidates for future drug development, however, because they have chronic toxic effects and the active amidine compounds have multiple sites susceptible to oxidative metabolism, which complicates their pharmacology and toxicology. Novel diamidoximes from three other structural classes, containing different groups linking the cationic moieties, lacked significant oral or intravenous anti-Pneumocystis activity, even though the corresponding diamidines were very active intravenously. Both active and inactive amidoximes were readily metabolized to the corresponding amidines by cell-free liver homogenates. Thus, the amidoxime prodrug approach may provide a strategy to exploit the potent antimicrobial and other pharmacological activities of selected, but certainly not all, aromatic diamidines.

Antileishmanial activities of several classes of aromatic dications.

Aromatic dicationic molecules possess impressive activity against a broad spectrum of microbial pathogens, including Pneumocystis carinii, Cryptosporidium parvum, and Candida albicans. In this work, 58 aromatic cations were examined for inhibitory activity against axenic amastigote-like Leishmania donovani parasites. In general, the most potent of the compounds were substituted diphenyl furan and thiophene dications. 2,5-Bis-(4-amidinophenyl)thiophene was the most active compound. This agent displayed a 50% inhibitory concentration (IC50) of 0.42 +/- 0.08 microM against L. donovani and an in vitro antileishmanial potency 6.2-fold greater than that of the clinical antileishmanial dication pentamidine and was 155-fold more toxic to the parasites than to a mouse macrophage cell line. 2,4-Bis-(4-amidinopheny)furan was twice as active as pentamidine (IC50), 1.30 +/- 0.21 microM), while 2,5-bis-(4-amidinopheny)furan and pentamidine were essentially equipotent in our in vitro antileishmanial assay. Carbazoles, dibenzofurans, dibenzothiophenes, and benzimidazoles containing amidine or substituted amidine groups were generally less active than the diphenyl furans and thiophenes. In all cases, aromatic dications possessing strong antileishmanial activity were severalfold more toxic to the parasites than to a cultured mouse macrophage cell line. These structure-activity relationships demonstrate the potent antileishmanial activity of several aromatic dications and provide valuable information for the future design and synthesis of more potent antiparasitic agents.

Some products of the bacterial dissimilation of streptomycin.

Klein, Dolph (Rutgers, The State University, New Brunswick, N. J.) and David Pramer. Some products of the bacterial dissimilation of streptomycin. J. Bacteriol. 83:309-313. 1962.-Bacterial dissimilation of streptomycin resulted in products detectable with Folin-Ciocalteu's phenol reagent, ninhydrin, and Ehrlich's reagent; these products absorbed ultraviolet light, with a maximum at 265 mmu. Evidence was also obtained for the production of a volatile base, possibly methylamine. Urea and streptamine were formed by growing cultures and washed-cell suspensions supplied with streptomycin. More than 90% of the amidine-nitrogen of the antibiotic was recovered as urea-nitrogen in culture filtrates. It was evident that the bacterial dissimilation of streptomycin involved hydrolysis of the guanido groups of the streptidine moiety of the antibiotic molecule. The site of action was the bond between the amidine group and the secondary amino-nitrogen of each guanido group, and the products of the reaction were urea and streptamine.

Microparticulate uptake mechanisms of in-vitro cell culture models of the respiratory epithelium.

The objective of this study was to examine the uptake mechanisms of fluorescent polystyrene microspheres of various diameters and surface chemistry by two human cell lines derived from the respiratory epithelium, A549 and Calu-3. Briefly, A549 and Calu-3 cells were grown to confluence in 12-well cluster plates and the uptake of fluorescent microspheres by the cells was determined at various time points. The amount of microspheres internalized by the cells was determined by correcting for non-specific binding to the cell surface. The data showed that A549 cells appeared to have more phagocytic activity than Calu-3 cells. Albumin-coated microspheres as large as 3 microm diameter can be internalized by A549 cells. The amount of internalization by A549 cells observed for 0.5-microm diameter albumin-coated microspheres was approximately 10-times greater than that observed for 1-microm diameter spheres and approximately 100-times greater than values observed for 2- and 3-microm diameter beads. Transmission electron micrographs confirmed that the microspheres were internalized by the cells. Uptake experiments conducted with Calu-3 cells indicated that albumin-coated microspheres were neither bound nor internalized by the cells. The effect of microsphere surface chemistry on the uptake mechanism indicated that amidine microspheres were internalized more rapidly and to a greater extent by both A549 and Calu-3 cells than carboxylate microspheres and non-coated microspheres. This phenomenon is thought to be attributed to masking of the negative polystyrene core by the positive amidine functional group; this effect was less marked for the carboxylate microspheres. These results suggest that A549 and Calu-3 cells can internalize microspheres and that size and effective charge played an important role in the uptake process.

[2-Amino-oxazoles as potential bonding agents in virostatic research. 1. N-unsubstituted and n-substituted 2-amino-oxazoles].

2-Amino-oxazoles with the semicyclic amidine structure are able in principle to form hydrogen bonds with native components for example with viral nucleic acids or viral enzymes. In this way the inhibition of viral replication seems to be possible. The syntheses of 39 compounds with amidine structure are described and the results of the antiviral agar diffusion plaque inhibition test are discussed in relation to the chemical structures.

Reassembly of a regularly arranged protein in the cell wall of Lactobacillus buchneri and its reattachment to cell walls: chemical modification studies.

Regularly arranged protein (RA protein) isolated from the cell wall of Lactobacillus buchneri was chemically modified by amidination, acetylation, succinylation, and amidation. The modified RA proteins were examined for their ability to reassemble into a regular array and to reattach to the cell walls from which the regular array had been detached. Only amidinated RA protein could be either reassembled into a regular array or reattached to the cell walls; RA proteins modified by the other methods lost the ability for both reassembly and reattachment. The unmodified RA protein could be reattached to periodate-oxidized cell walls, but not to methylated ones. These results suggest that the positive charge of the amino group as well as the negative charge of the carboxyl group of RA protein plays an important role(s) in morphogenesis of the hexagonal array and in its attachment to the underlying cell wall layer. The periodate-insensitive lone hydroxyl groups of the neutral polysaccharide molecule in the cell wall seem to be the receptor sites for RA protein in the attachment to the cell wall.

[Synthetic serine proteinase inhibitors. 30. Synthesis of alpha-benzoylamino-4-amidinocinnamic acid and alpha-(4-amidinobenzoylamino) cinnamic acid amides, and their inhibitory effect on trypsin-like enzymes].

To test their inhibitory activity against enzymes, the authors synthetized the compounds named in the title. The synthesis started from the corresponding azlactones 1 and 9. Subsequent aminolysis led to the alpha-benzoyl-aminocinnamic acid amides 2 and 10 which contain a cyano function. The acid amides were converted, in different ways, to the desired amidine salts 4, 7, 12 and 15. The amidines obtained showed but little inhibitory activity against the serine proteinases thrombin, trypsin and plasmin.

A chemical procedure for determining the sidedness of the NH2 terminus in a membrane protein. The small intestinal sucrase-isomaltase.

We have developed a chemical procedure for determining the sidedness of the NH2 termini of sucrase-isomaltase complex in the small intestinal brush-border membrane. The methodology involves amidination of sealed right-side-out brush-border membrane vesicles with the impermeant imidoester 3-[dimethyl-2-[( 3H]acetimidoxyethyl)ammonio]propanesulfonate with subsequent quantitation of this reaction at the NH2 termini of sucrase-isomaltase. It was found that amidination yields were every similar for the reaction of 3-[dimethyl-2-[( 3H]acetimidoxyethyl)-ammonio] propanesulfonate with intact membrane vesicles and with leaky, deoxycholate-extracted membrane fragments. This demonstrates that the NH2 termini were equally accessible to the impermeant reagent in both systems and, hence, are exposed on the outside of the sealed membrane vesicles. The methodology developed does not involve proteolysis and should be of wide applicability.

DNA-binding properties of nitroarene oligopeptides designed as hypoxia-selective agents.

The DNA-binding properties of six candidate nitroarene-functionalized oligopeptides 8-13 (i.e. 5-nitrofuran, 2-nitroimidazole and 4-nitrobenzene derivatives) with potential hypoxia-selective activity, developed as analogues of the archetypal minor groove-binding ligands, netropsin 1 and distamycin 2, have been examined using an extended molecular mechanics and dynamics (MM/MD) modelling approach. The energies calculated for interaction with the d(CGCGAATTCGCG) duplex, or d(CGCAAATTTGCG)2 in the case of the elongated nitrobenzene 13, correlate with fluorimetric data obtained by indirect competitive displacement of ethidium bromide from double-stranded calf thymus DNA. The results suggest that the mode of interaction for these agents resembles the behavior of structurally unrelated bis(amidine) derivatives that also bind to AT-rich stretches of duplex B-DNA via the minor groove. Analysis of the binding behaviour indicates that the interactions are determined by enthalpic rather than entropic energy terms, suggesting that specific or differential hydration effects for the DNA and ligand may be unimportant. A rational structure-activity relationship is established for the interaction of this family of oligopeptides with DNA that also encompass the bis(amidine) class of ligands.

Benzo[d]isothiazol-3-yl-benzamidines: a class of protective agents on culture of human cartilage and chondrocytes stimulated by IL-1beta.

New derivatives of N-benzo[d]isothiazol-3-yl-benzamidine 6 a were synthesized as nonacidic anti-inflammatory/antidegenerative agents. We investigated the influence of the amidines 6 a-j on the production of NO, PGE(2), MMP-3, COX-2, ROS, and GAGs, key molecules involved in cartilage destruction in osteoarthritic diseases. The antidegenerative properties of the novel designed derivatives 6 b-j were improved with respect to N-benzo[d]isothiazol-3-yl-benzamidine 6 a. All of the compounds 6 a-j promoted the reduction of most of the IL-1beta-induced harmful effects. Derivatives 6 d, 6 h, and 6 j were the most potent of all the tested compounds, particularly in the human chondrocyte culture model.

Monovalent amidiniums block calcium channels in chick sensory neurons.

The effect of amidiniums on high-threshold Ca2+ channel currents (ICa) was studied in chick dorsal root ganglion neurons. Guanidinium reduced ICa in a dose-dependent fashion. The block was relieved by increasing the concentration of the permeant ions, Ba2+ or Ca2+, suggesting a competition for a common binding site within the channel. Formamidinium and methyl-guanidinium suppressed ICa with similar potencies, whereas L-arginine had no effect. A neutral amidine, urea, increased ICa. In Ca(2+)-free solutions guanidinium and Na+ permeated through the Ca2+ channel equally well. Structure-activity relationship obtained for blocking efficacies of different amidiniums are used to discuss possible configurations of the selectivity filter in the Ca2+ channel.

The solid-phase synthesis of 2'-5'-linked oligoriboadenylates containing 8-bromoadenine.

To increase the accessibility of 8-bromo-2',5'-oligoadenylates, we developed a synthesis of 2'-5'-linked oligoriboadenylates containing varying numbers of 8-bromoadenosine residues based on the use of a CPG-LCA solid support and the phosphoramidite approach. Although N6-benzoyl protection was satisfactory for incorporation of nonmodified adenine residues into 2',5'-oligonucleotides, the effective incorporation of 8-bromoadenine into such 2',5'-linked oligomers required use of a non acyl protecting group. Amidine protection of the purine exocyclic amino function proved compatible with all aspects of the phophoramidite approach and with the hydroxyl protection groups employed.

Differential effects of benzamidine derivatives on the expression of c-myc and HLA-DR alpha genes in a human B-lymphoid tumor cell line.

In this paper, we report the effects of aromatic tetra amidines (TAPP-H) on cell growth and gene expression of a B-lymphoid human tumor cell line, WI-L2. The results obtained give evidence (a) for inhibition of cell proliferation by TAPP-H; (b) for stronger antiproliferative activity of TAPP-halo derivatives; (c) for TAPP-mediated inhibition of accumulation of c-myc RNA sequences but not of HLA-DR alpha mRNA and DR antigens. These results suggest that this class of antiproliferative compounds exhibit differential effects on cell-cycle specific and differentiation specific genes. In addition, also TAPP-related compounds containing 2 (DAPP) or 3 (TAPB) benzamidine residues retain inhibitory activity on the proliferation of WI-L2 cells. These latter compounds might be proposed as useful substrate in the synthesis of drug-conjugated monoclonal antibodies or growth factors.

Radiosynthesis of (E)-N-(2-[11C]methoxybenzyl)-3-phenyl-acrylamidine, a novel subnanomolar NR2B subtype-selective NMDA receptor antagonist.

Recently, a novel series of amidines has been described, exhibiting high NR2B-subtype selective N-methyl-D-aspartate (NMDA) antagonist activity with nanomolar or subnanomolar affinity. Within the styrylamidine subclass, (E)-N-(2-methoxybenzyl)-3-phenyl-acrylamidine (1), displayed the highest affinity (Ki=0.7 nM versus [(3)H]ifenprodil) and was considered an appropriate candidate for isotopic labelling with carbon-11 (T(1/2): 20.38 min) at its methoxy group for imaging of NMDA receptors with PET. Derivative 1 has been labelled from the corresponding nor-analogue using [(11)C]methyl triflate and the following experimental conditions : (1) trapping at -10 degrees C of [(11)C]methyl triflate in 300 microL of acetone containing 0.6-0.8 mg of precursor 5 (2.4-3.2 micromol) and 5 microL of a 3M solution of NaOH in water (about 5 eq.); (2) concentration to dryness of the reaction mixture (at 110 degrees C, using a helium stream for 1-2 min); (3) taking up the residue with 0.5 mL of the HPLC mobile phase and (4) purification using semi-preparative HPLC (SymmetryPrep) C-18, Waters, 300 x 7.8 mm). Typically, starting from a 1.5 Ci (55.5 GBq) [(11)C]CO(2) production batch, 120-240 m Ci (4.44-8.88 GBq) of [(11)C]-1 (20-40% decay-corrected radiochemical yield, n=5) was obtained within a total synthesis time of 25-30 min. Specific radioactivities ranged from 0.8 to 1.2 Ci/micromol (29.6-44.4 GBq/micromol) at the end of radiosynthesis. No attempts were made to further optimise these reactions, as sufficient material was obtained to allow for preliminary pharmacological characterisation.
