Primary composite angiogenic leiomyosarcoma-epithelioid angiosarcoma of the brain.

The authors describe a primary sarcoma of the brain with immunohistochemical and ultrastructural features of leiomyosarcoma as well as epithelioid hemangiosarcoma. The leiomyosarcomatous component consisted of spindle cells with well-developed external lamina, subsarcolemmal densities composed of microfilaments, pinocytic vesicles, and abundant intermediate filaments, and showed immunohistochemical reactions for smooth muscle actin. The epithelioid part of the tumor contained scattered cells reactive for alkaline phosphatase as well as CD31 and factor VIII. Many epithelioid cells were lipidized and remarkably similar to "stromal cells" of a hemangioblastoma. Occasional Weibel-Palade bodies, indicating endothelial differentiation, were present in scattered neoplastic cells. There were also cells with features intermediate between endothelium, pericytes and smooth muscle cells, and undifferentiated mesenchymal cells. The brain at the periphery of sarcoma showed conglomerates of well-differentiated capillaries, telangiectasias and small dysplastic arteries, features that raise the possibility of origin of this tumor from a preexisting vascular developmental abnormality.

[Remote sequelae of combined influence of vinyl chloride and dichloroethane (experimental study)].

Exposure to concentrations of vinyl chloride (VC) + dichloroethane (DCE) 50 + 50 mg/m3, VC + DCE 50 + 500 mg/m3 and VC + DCE 500 + 50 mg/m3 during pregnancy induced higher occurrence of fetal resorption, increased incidence of preimplantational and postimplantational fetal death. Carcinogenic activity of the same levels of VC and DCE was seen in various organs: high concentrations of DCE increased incidence of lung tumors, high levels of VC--of breast neoplasms, mesenterial lymphosarcoma and liver hemangiosarcoma.

Electrochemotherapy: potentiation of local antitumour effectiveness of cisplatin in dogs and cats.

The aim of this study was to introduce electrochemotherapy with cisplatin into veterinary medicine, where there is a need for inexpensive and effective treatment of cutaneous and subcutaneous tumours of various histological types. The response to treatment was assessed on tumour nodules in 3 cats with mammary adenocarcinoma and fibrosarcoma, and in 7 dogs with mammary adenocarcinoma, cutaneous mast cell tumour, hemangioma, hemangiosarcoma, adenocarcinoma glandulae paranalis and neurofibroma. Twenty-four tumour nodules of different size were treated; 5 with cisplatin injected intratumourally and 19 with electrochemotherapy, i.e. intratumoural administration of cisplatin followed by delivery of electric pulses to the tumour nodule. Electrochemotherapy with cisplatin had a good antitumour effect on all tumours treated. Their average size 4 weeks after treatment was also greatly reduced (0.01 cm3) compared to those treated by intratumoural cisplatin injection alone (3.0 cm3). Altogether, electrochemotherapy- treated tumours responded with 84% objective responses, whereas only one tumourpartially responded to cisplatin treatment alone. Evaluated by contingency table, the response to treatment with electrochemotherapy was significantly better than that of the cisplatin treated group (p=0.014). Furthermore, there was a significant prolongation of the duration of response in electrochemotherapy treated tumours (p = 0.046). This study showed that electrochemotherapy with cisplatin is an effective, safe and simple local treatment of different histological types of cutaneous and subcutaneous tumours in cats and dogs.

Hepatic angiosarcoma mimicking cavernous hemangioma on angiography.

A 60-year-old woman was admitted to our department for evaluation of a hepatic mass. The mass was diagnosed as a hemangioma of the liver by abdominal angiography because of typical cotton wool appearance and stretched arterial vessels and no peripheral staining. However, one month later, the mass was surgically removed because of extravasation. Histological findings of a specimen of the mass revealed that it entirely contained abundant necrotic tissue, and a small residual part after transcatheter arterial embolization was consistent with hemangioma. However, she complained of hemoptysis and thigh pain after several weeks. Computed tomography revealed multiple lung masses and a mass of right musculus gluteus medius. Reexamined histological findings of the liver tumor showed hemangiosarcoma. We should pay attention to the fact that it is sometimes difficult to differentiate cavernous hemangioma from angiosarcoma by angiography.

Vascular tumors of the female genital tract: a clinicopathological study of nine cases.

UNLABELLED: The aim of this study was to investigate the clinicopathological features of nine unusual vascular tumors of the female genital tract and to investigate the problems in the differential diagnosis from other genital lesions. MATERIAL-METHODS: In a review of 15,000 specimens of the female genital system examined in our Laboratory during the last decade, nine cases of vascular tumors were found. RESULTS: A capillary hemangioma of the endometrium, one capillary and one cavernous hemangioma of the cervix, six vulva lesions (one capillary, and one cavernous hemangioma and four angiokeratomas) and one hemangiosarcoma of the mons pubis in a 76-year-old patient were diagnosed. CONCLUSIONS: The differential diagnosis of the vascular tumors must be made mainly from endometriotic lesions and melanomas. Immunohistochemistry (S-100, CD31, CD34, CKS, EMA) aid in the diagnosis. Local excision is adequate for the benign vascular lesions.

A study of the carcinogenicity of glycidol in Syrian hamsters.

The industrial chemical glycidol is a directly acting mutagen and a broadly acting carcinogen in rats. It was administered to Syrian golden hamsters (20 male and 20 female) by gavage of 12 mg twice a week for 60 weeks. The total dose per animal was 1.45 g or 20 mmol. Survival was not different from control hamsters treated with corn oil/ethyl acetate. Of the treated males, 9 had tumors and 13 of the treated females had tumors, some of which were adrenal cortex tumors seen in controls. More tumors were seen in the glycidol-treated hamsters than in controls, but the spleen was the only notable target organ and the number of animals with spleen hemangiosarcomas was small. Glycidol appeared to be less carcinogenic in hamsters than in rats or mice.

Activation of protooncogenes in spontaneously occurring non-liver tumors from C57BL/6 x C3H F1 mice.

The C57BL/6 x C3H F1 (hereafter called B6C3F1) mouse is an important animal model for long-term carcinogenesis studies. Maintained under normal laboratory conditions, these mice develop various types of spontaneous tumors during their lifetime. Activated Ha-ras genes have been detected in 66% of spontaneous hepatocellular tumors in the B6C3F1 mouse [Reynolds et al., Science (Washington DC), 237:1309, 1988]. In this study 49 spontaneous non-liver tumors were investigated for oncogene activation by DNA transfection techniques. Of the 49 tumor DNAs analyzed, only 5 yielded multiple foci in the NIH 3T3 focus assay: 2 of 10 pulmonary adenocarcinomas; 0 of 25 lymphomas; 2 of 2 Harderian gland adenomas; 0 of 1 adenocarcinoma of the small intestine; 1 of 6 malignant skin tumors; 0 of 4 hemangiosarcomas; and 0 of 1 lung metastasis of a hepatocellular carcinoma. DNA from six lymphomas which were negative in the NIH 3T3 focus assay were further analyzed for transforming genes by the nude mouse tumorigenicity assay. One of the five lymphomas tested positive with this assay. Southern blot analysis identified five activated ras genes: H-ras in two Harderian gland adenomas; K-ras in one pulmonary adenocarcinoma and in one s.c. adenocarcinoma; and N-ras in one lymphoma. The mutations involved were CG to AT and AT to TA in codon 61 of the Ha-ras genes, GC to AT or TA in codon 12 of the K-ras genes, and a GC to AT mutation in codon 12 of the N-ras gene. Transformant DNA from a pulmonary adenocarcinoma which yielded multiple foci in the transfection assay did not hybridize to DNA probes specific for the K-, H-, and N-ras, raf, neu, and met genes. Thirteen additional tumor DNAs yielded a single focus in the NIH 3T3 transfection assay. The transformant DNAs retransmitted in a second cycle transfection assay. Rearranged and/or amplified raf genes were detected in six of the transformant DNAs. At present we do not know whether these activated raf genes were present in the original tumor DNA. The other seven transformant DNAs did not hybridize with any of the above mentioned specific DNA probes utilized in Southern blot analysis. Unlike liver tumors, the activation of ras protooncogenes is not a frequent event in the development of spontaneous non-liver tumors of the B6C3F1 mouse. The results from this study should aid in understanding the neoplastic development associated with exposure to chemical carcinogens in the B6C3F1 mouse.

Evaluation of hemostatic defects secondary to vascular tumors in dogs: 11 cases (1983-1988).

Two populations of dogs with cutaneous hemangiomas and hemangiosarcomas were evaluated retrospectively. One population consisted of 96 dogs seen at the Veterinary Medical Teaching Hospital at the University of California, Davis. The second population consisted of 116 dogs that had skin biopsy specimens submitted to a private veterinary diagnostic laboratory for histologic diagnosis. Nine dogs from the teaching hospital and 2 dogs, from which samples had been submitted to the veterinary diagnostic laboratory, developed hemostatic defects in association with the tumors. Hemostatic defects included hemorrhage directly from the tumor, thrombocytopenia, hypofibrinogenemia, and findings associated with disseminated intravascular coagulation. Because bleeding during surgery can develop in animals with hemostatic defects, dogs with one or more tumors suspected of being vascular in origin should have platelet numbers and hemostatic analytes evaluated prior to surgery, especially if petechiae or ecchymoses are evident.

Carcinogenicity of 1,3-butadiene in C57BL/6 x C3H F1 mice at low exposure concentrations.

The carcinogenicity of inhaled 1,3-butadiene was evaluated in C57BL/6 x C3H F1 mice exposed to concentrations of this gas ranging from 6.25 to 625 ppm. Butadiene is a high production volume chemical, used mainly in the manufacture of synthetic rubber. In these 2-yr inhalation studies, a potent multisite carcinogenic response was observed, including neoplasms of the lung at concentrations as low as 6.25 ppm. Early occurrence and extensive development of lethal lymphocytic lymphomas in mice exposed to 625 ppm of butadiene reduced the number of animals at risk for the expression of later developing neoplasms at other sites; at lower exposure concentrations, dose responses were demonstrated for hemangiosarcomas of the heart and neoplasms of the lung, forestomach, Harderian gland, preputial gland, liver, mammary gland, and ovary. So far, no long-term studies on butadiene have been conducted at exposure concentrations that have not shown a carcinogenic response. In separate experiments with reduced exposure durations, butadiene induced neoplastic responses at multiple organ sites even after only 13 wk of exposure. Because of the correspondence between these animal data and recent epidemiology findings, there is a worldwide public health need to reevaluate current workplace exposure standards for 1,3-butadiene.

[Angioblastoma (hypertrophic hemangioma). Preliminary report].

We are presenting herewith the case of a rare vascular tumor called angioblastoma which histopathological and clinical aspects may lead into errors of diagnosis and therapy. Its fast development, tendency to ulceration and bleeding and dark colour, are signs that may be confused with those of hemangiosarcomas and, inclusive, melanomas, because, from a clinical point of view, its pleomorphism and cellular atypical nature, due to immaturity, may induce diagnostic and histological doubts. In order to simplify diagnosis and therapy of an angioblastoma, we have treated it within the complex subject of vascular hyperplasias in general. Thereby, we have made three synopses of interest for such a purpose. Out of the consulted bibliography, its limited frequency is stated, as relative cases don't amount to a hundred worldwide and none of them was found in our country. We have reported the most accurate data for its correct clinical and histopathological diagnosis to avoid extremely aggressive therapies, since it is a lesion of benign biological behaviour.

Thoracic neoplasia in equids: 35 cases (1967-1987).

Thirty-two horses and 3 ponies had neoplasia involving the thoracic cavity. Lymphosarcoma of the thorax was found in 19 (54.2%) of the equids. The other 16 equids had metastatic adenocarcinoma (7 horses; 20%), metastatic squamous cell carcinoma (5 equids; 14.3%), metastatic hemangiosarcoma (3 equids; 8.6%) or undifferentiated sarcoma (1 horse; 2.9%). Thoracic neoplasia was diagnosed before death in 15 equids (42.9%).

Nasal cavity neoplasia in F344/N rats and (C57BL/6 x C3H)F1 mice inhaling propylene oxide for up to two years.

Propylene oxide (CAS: 75-56-9) was studied for potential carcinogenicity and chronic toxicity by inhalation in F344/N rats and (C57BL/6 x C3H)F1 mice. Groups of 50 animals of each sex were exposed to 0, 200, or 400 ppm propylene oxide for 6 hours/day, 5 days/week, for up to 103 weeks. Survival decreased in mice exposed to propylene oxide; the decrease was significant (P less than .005) in mice exposed to 400 ppm. Survival of exposed rats was comparable to that of controls. Mean body weight of rats and mice exposed to 400 ppm propylene oxide decreased, when compared to that of controls, during the 2d year of exposure. Exposure to propylene oxide for up to 2 years induced inflammatory and proliferative responses in nasal cavity of both species. There was clear evidence of carcinogenicity in mice exposed to 400 ppm propylene oxide; 10 of 50 males and 5 of 50 females had hemangiomas or hemangiosarcomas of the nasal submucosa. Papillary adenomas involving the nasal respiratory epithelium and underlying submucosal glands were observed in 3 female rats and 2 male rats exposed to 400 ppm propylene oxide.

[Primary sarcoma of the middle ear. Apropos of 3 cases].

Authors present three cases: one hemangiosarcoma, one reticulum-cell sarcoma and one rhabdomyosarcoma of the middle ear. Histopathology slides were reviewed, as well as the Western medical literature the authors could obtain on the subject. The rare frequency of these neoplasms in the middle ear was made evident. Aspects relative to diagnosis and treatment were discussed. Long term prognosis remains unfavourable.

Angiosarcoma with pulmonary siderosis and persistent reticulocytosis. Steroid responsiveness suggests an immune basis.

A 35-year old man with cough, hemoptysis, and dyspnea was found to have diffuse pulmonary infiltrates and iron-laden macrophages in the sputum. Pulmonary siderosis was confirmed by transbronchial biopsy. An associated hypochromic anemia required frequent transfusion. Though marrow iron stores were absent, reticulocytosis was maintained. Corticosteroid therapy resulted in cessation of hemoptysis, clearing of the pulmonary infiltration, and a substantial reduction in transfusion requirement. Splenectomy was of no benefit. The patient developed cerebral symptoms with seizures, and rapid deterioration led to cerebral symptoms with seizures, and rapid deterioration led to death. Disseminated hemangiosarcoma was found at autopsy. Steroid responsiveness of the associated pulmonary siderosis suggests that it had an immune basis.

Tumor and tumor-like lesions of perilimbal conjunctiva in laboratory dogs.

Review of records of 1,680 research colony Beagles revealed proliferative and neoplastic lesions in the nasal or temporal limbal conjunctiva of 8 male and 6 famale dogs. The mean age at observation was 5.2 years. The lesions ranged from acanthosis and squamous cell carcinoma of the conjunctival epithelium to vascular ectasia, hemangioma, and invasive hemangiosarcoma of the underlying lamina propria. The lesions developed under circumstances that suggested solar radiation was involved in the pathogenesis.

[Growth of human tumor transplanted into nude mice with various genetic backgrounds].

Nude mice with different genetic backgrounds were used for the experiment to evaluate the growth of human and murine tumors in the different hosts. Twelve human tumors including 5 gastric carcinomas (St-4, St-10, St-15, St-40, KS-1), 2 colon carcinomas (Co-3, Exp-42), 1 breast carcinoma (MX-1), 1 bile duct carcinoma (Ch-1), 1 hepatocellular hepatoma (Li-4) and 2 hemangiosarcomas (LC-27-R, LC-27-UR), and one murine tumor spontaneously arisen from C3H/HeN nu/+ mouse were transplanted into the subcutaneous tissue of nude mice with genetic backgrounds of BALB/cA, DDD and ICR (CD-1). BALB/cA-nu was originated from the Central Institute for Experimental Animals, DDD-nu was supplied from the Institute of Medical Science, University of Tokyo and ICR (CD-1)-nu was supplied from Charles River Co., Ltd. Tumors were measured by the width (W) and length (L) three times a week by the same person. The tumor volume (V) was estimated by the formula V = W2 X L/2. Tumor doubling time (Td) during the exponential growth phase was compared between the nude mice with different genetic backgrounds. It was found that Td of MX-1 in DDD-nu mice and Td of St-40 and Exp-42 in BALB/cA-nu mice were shortened in comparison with those of tumors transplanted into nude mice with the other strains. However, no statistically significant differences were observed when t-test of matched pairs and coefficients of correlation were applied to compare Td in the other kinds of nude mice. Nude mice with genetic backgrounds of DDD and ICR (CD-1) were thought to be also useful for various kinds of therapeutic experiments in human tumor xenografts/nude mice system.

Angiosarcoma in primary lymphedema of the lower extremity--Stewart-Treves syndrome.

After a 20-year latent period an angiosarcoma developed in the edematous leg of a 74-year-old woman with primary lymphedema. A deep venous thrombosis of the leg which further aggravated tissue swelling preceded the appearance of angiosarcoma. Histogenetic classification of the tumor as hemangiosarcoma rather than lymphangiosarcoma was favored by positive immunohistochemical staining for Factor VIII. Despite high amputation and isolated perfusion with hyperthermal cytostatic infusion, she developed local recurrence and distant metastases and died 16 months after operation. Patients with chronic primary or secondary lymphedema are susceptible to angiosarcoma although the overall risk is small.
