Spatially distributed dominant excitation frequencies reveal hidden organization in atrial fibrillation in the Langendorff-perfused sheep heart.

INTRODUCTION: Atrial fibrillation (AF) is characterized by complex wave propagation, yet periodic excitation suggesting a high degree of organization may be revealed during sustained AF. We provide a systematic quantification of the spatial distribution of dominant frequencies (DFs) of local excitation on the epicardium of the right atrial (RA) free wall and left atrial (LA) appendage of the isolated sheep heart during AF. The data reveal, for the first time, hidden organization, independent of the activation sequences or nature of electrograms. METHODS AND RESULTS: In 13 Langendorff-perfused sheep hearts, AF was induced in presence of 0.1 to 0.6 microM acetylcholine. Video movies (potentiometric dye di-4-ANEPPS) of the RA and LA (>30,000 and >20,000 pixels, respectively) were obtained at 120 frames/sec and a biatrial electrogram was recorded. Spectral analyses were performed on movies with DF maps constructed. During AF, the activity formed stable discrete domains with uniform DFs within each domain. Acceleration of AF increased the number of domains (R = 0.81, P < 0.0001) and the DF variance (R = 0.63, P < 0.001), indicating a decrease in organization. Also, the LA was faster and more homogeneous, with smaller number of DF domains, compared to the RA (P < 0.00001). CONCLUSION: In this model, AF is characterized by multiple domains with distinct DFs on the atrial epicardium. The decrease in domain area with increased rate suggests that AF results from high-frequency impulses that undergo spectral transformations. The LA is generally faster and more organized than the RA, suggesting that the sources for the impulses are localized to the LA.

An electrophoretic approach to the assessment of the spatial distribution of DNA double-strand breaks in mammalian cells.

An approach is presented making it possible to investigate whether breaks in fragmented mammalian chromosomal DNA were induced randomly and independently from each other. Genomic DNA isolated from mammalian cells irradiated with gamma-rays or restriction enzyme-treated human DNA was resolved according to size using pulsed field gel electrophoresis, and the resulting DNA mass distributions were measured in ethidium bromide-stained gels. The DNA profiles thus obtained were compared to the predictions on DNA fragment size distribution which follow from a so-called random breakage model to test whether the experimental outcome is compatible with the assumption of a random localization of breaks. Comparisons of fragment distributions may be performed utilizing two equivalent representations that are linked by an adequate transformation. Considering either directly measurable DNA mass profiles in units of migration distances along a gel lane or transformed distributions in units of molecular length, we show for gamma-irradiated samples that the predictions derived from the employed models agree well with the observed data, thus allowing an immediate quantification of double-strand breaks (DSB). Using restriction enzyme-treated DNA as a paradigm, the disagreement of predicted and observed data shows the applicability of our approach to the detection of a non-random distribution of DSB. Therefore, we suppose that our approach may also be useful to reveal a clustering of DSB, which is postulated to occur after damage induction by densely ionizing radiation. Furthermore, investigations on the spatial distribution of chemically or endogenously produced DSB, as well as residual DSB after repair, may be attempted.

Spatial distribution of hypoxic pulmonary vasoconstriction in the supine pig.

Hypoxic pulmonary vasoconstriction (HPV) serves to maintain optimal gas exchange by decreasing perfusion to hypoxic regions. However, global hypoxia and nonuniform HPV may result in overperfusion of poorly constricted regions leading to local edema seen in high-altitude pulmonary edema. To quantify the spatial distribution of HPV and its response to regional Po2 (Pr(O2)) among small lung regions, five pigs were anesthetized and mechanically ventilated in the supine posture. The animals were ventilated with an inspired O2 fraction (Fi(O2)) of 0.50 and 0.21 and then (in random order) 0.15, 0.12, and 0.09. Regional blood flow (Q) and alveolar ventilation (Va) were measured by using intravenous infusion of 15 microm and inhalation of 1-microm fluorescent microspheres, respectively. Pr(O2) was calculated for each piece at each Fi(O2). Lung pieces differed in their Q response to hypoxia in a manner related to their initial Va/Q with Fi(O2) = 0.21. Reducing Fi(O2) < 0.15 decreased Q to the initially high Va/Q (higher Pr(O2)) regions and forced Q into the low Va/Q (dorsal-caudal) regions. Resistance increased in most lung pieces as Pr(O2) decreased, reaching a maximum resistance when Pr(O2) is between 40 and 50 Torr. Local resistance decreased at PrO2 < 40 Torr. Pieces were statistically clustered with respect to their relative Q response pattern to each Fi(O2). Some clusters were shown to be spatially organized. We conclude that HPV is spatially heterogeneous. The heterogeneity of Q response may be related, in part, to the heterogeneity of baseline Va/Q.

Spatial distribution patterns of the sporocyst infrapopulation of Schistosoma mansoni within its mollusc host (Biomphalaria glabrata): an unusual phenotype of aggregation.

A study carried out on the intramolluscan development of the larval stages of Schistosoma mansoni from Venezuela in its intermediate host Biomphalaria glabrata shows the existence of 2 patterns for the spatial distribution of daughter sporocysts within the digestive gland of the snail. Besides the normal pattern, which is a random distribution of daughter sporocysts within the gland, 11% of the infected snails harbored an infection phenotype with daughter sporocysts strongly aggregated in only the posterior half of the digestive gland. These data suggest a possible strategy allowing a better nutritional efficiency of the infected digestive gland, a process to block the supply of hemolymph toward the gonad, resulting in castration or both.

Spatial distribution of trace metals in sediments from urban streams of Semarang, Central Java, Indonesia.

Elevated environmental concentrations of metals are usually associated with the impact of urbanization. The present study is focused on metal contamination in urban sediments. A field survey was carried out to determine the distribution of four metals, i.e., cadmium (Cd), lead (Pb), copper (Cu), and zinc (Zn), in the coastal urban area of Semarang, Central Java, Indonesia. Sediment samples were collected from 101 grids of 2x2 km. To map the spatial distribution of these metals, concentrations of each metal were plotted against the corresponding grid coordinate. Cd was below the detection limit (<0.03 microg/g) in all samples, whereas concentrations of Pb, Zn, and Cu fell into a wide range. Frequency distributions of Pb, Zn, and Cu concentrations indicated a similar pattern, in which the major proportion of the sites had a low metal concentration. Some sites, however, had extremely high metal concentrations, Zn up to 1257 microg/g, Pb up to 2666 microg/g, and Cu up to 448 microg/g. The data were used to define background concentrations for sediments in coastal zones of Indonesia ("reference values"). The proposed reference values are 25.6 microg/g, 132.2 microg/g, and 40.7 microg/g, respectively, for Pb, Zn, and Cu. The degree of metal contamination of each individual site was classified according to the calculated value of a combined pollution index, W. Four categories of the degree of metal contamination were proposed, i.e., unpolluted, slightly polluted, polluted, and heavily polluted. Based on this classification, from 101 sites investigated in the greater Semarang area, 51 are unpolluted, 36 slightly polluted, 9 polluted, and 5 heavily polluted.

Spatial distribution of microorganisms and measurements of oxygen uptake rate and ammonia uptake rate activity in a drinking water biofilter.

The biofilm characteristics (population dynamics and biofilm composition) in a biological filter for the removal of iron, manganese and ammonium were studied in a drinking water treatment plant. The objective was to examine the spatial distribution and biological composition of active biomass that grows in a biological filter and to verify the effect of the backwashing on the quantity of fixed biomass and on the density and activity of the biological population. Heterotrophic microorganisms activity was higher in the upper layer of the filter. Nitrifying microorganisms colonized the biofilter in a stratified manner and their activity was higher in the second layer of the filter. A total of 14 species of ciliated protozoa and 7 species of filamentous microorganisms were found in the biofilters. Ciliates were concentrated in the filterbed layer in which the heterotrophic activity was higher. The grazing activity of ciliates on heterotrophic bacteria reduced the competition pressure on nitrifying microorganisms, supporting their growth and thus raising the ammonium removal efficiency. In general, filamentous microorganisms appeared to be indifferent to operating changes in the plant such as backwashing and filtering cycles. Crenothrix was the prevalent filamentous microorganism in terms of both frequency and abundance; it was found prevalently in the first layer where the oxidisation of iron and manganese occurred.

Effects of pressure and volume of the receiving chamber on the spatial distribution of regurgitant jets as imaged by color Doppler flow mapping. An in vitro study.

Regurgitant jet dimensions imaged by color Doppler flow mapping have been used to evaluate the severity of valvular insufficiency in clinical studies. To study the effect of pressure and volume within the receiving chamber on the magnitude of spatial distribution of regurgitant jets assessed by color Doppler techniques, we designed a simple constant-flow model in which a jet was driven through a known orifice (1.5 mm2) into a compliant receiving chamber by a steady-flow pump. A distal tube at the outflow closed the system and maintained the volume of the chamber constant during pump operation. We varied flow rate from 60 to 270 ml/min into elastic balloons with different static compliances of 1, 2, 4.5, and 9 ml/mm Hg (pressures of 57, 28, 18, and 8 mm Hg, respectively); the balloons served as receiving chambers at the constant volume of 150 ml. We also evaluated the effect of different volumes of a receiving chamber (110, 130, and 150 ml and pressures of 5, 15, and 24 mm Hg) with a static compliance of 2 ml/mm Hg over the same range of flow rates. For each of the different balloons, jet area correlated linearly with the jet velocity across the orifice (r = 0.98, 0.99, 0.98, and 0.97) and also with flow rate (r = 0.97, 0.99, 0.98, and 0.99). At the same flow rate and volume of receiving chamber, however, the jet area imaged by color Doppler decreased as the pressure in the receiving chamber increased, although receiving-chamber volume was constant.(ABSTRACT TRUNCATED AT 250 WORDS)

The number and spatial distribution of IP3 receptors underlying calcium puffs in Xenopus oocytes.

Calcium puffs are local Ca(2+) release events that arise from a cluster of inositol 1,4,5-trisphosphate receptor channels (IP(3)Rs) and serve as a basic "building block" from which global Ca(2+) waves are generated. Important questions remain as to the number of IP(3)Rs that open during a puff, their spatial distribution within a cluster, and how much Ca(2+) current flows through each channel. The recent discovery of "trigger" events-small Ca(2+) signals that immediately precede puffs and are interpreted to arise through opening of single IP(3)R channels-now provides a useful yardstick by which to calibrate the Ca(2+) flux underlying puffs. Here, we describe a deterministic numerical model to simulate puffs and trigger events. Based on confocal linescan imaging in Xenopus oocytes, we simulated Ca(2+) release in two sequential stages; representing the trigger by the opening of a single IP(3)R in the center of a cluster for 12 ms, followed by the concerted opening of some number of IP(3)Rs for 19 ms, representing the rising phase of the puff. The diffusion of Ca(2+) and Ca(2+)-bound indicator dye were modeled in a three-dimensional cytosolic volume in the presence of immobile and mobile Ca(2+) buffers, and were used to predict the observed fluorescence signal after blurring by the microscope point-spread function. Optimal correspondence with experimental measurements of puff spatial width and puff/trigger amplitude ratio was obtained assuming that puffs arise from the synchronous opening of 25-35 IP(3)Rs, each carrying a Ca(2+) current of approximately 0.4 pA, with the channels distributed through a cluster 300-800 nm in diameter.

Spatial distribution of mercury in the sediments and riparian environment of the River Yare, Norfolk, UK.

Concentrations of total mercury (T-Hg) were determined in sediments and riparian (bankside) soils from the River Yare, Norfolk, UK to assess the current extent of contamination arising from a historical point source discharge. The results demonstrate that the spatial distribution pattern in surficial sediments and soils follows that of a distinct pollution plume with an initial increase 2-3 km downstream from the point source discharge at Whitlingham Sewage Treatment Works (STW) outfall. Average T-Hg concentrations in the surficial sediments ranged from 0.1 to 8.13 mg kg(-1); bankside soil concentrations ranged from 0.1 to 2.63 mg kg(-1). There has been a decline in downstream background sediment concentrations of Hg over time. This is likely to be the result of burial by fresh relatively uncontaminated sediments and possibly in the lower reach as a consequence of the influence of the freshwater-saline interface occurring near Cantley. Channel morphology was also shown to be an important factor in determining the large variations of Hg concentrations between sample points within transects. The predominant source of Hg to the soils appears to be due to dredging and the deposition of sediments during flooding.

Larval spatial distributions and other early life-history characteristics predict genetic differentiation in eastern Pacific blennioid fishes.

In marine organisms, a pelagic larval stage increases the opportunities for long-distance dispersal and is often associated with little genetic differentiation over large geographical distances. Here we test the hypothesis that early life-history characteristics, including larval spatial distributions, affect the rates of dispersal and, therefore, the levels of genetic partitioning among three Gulf of California reef fishes: Axoclinus nigricaudus, Malacoctenus hubbsi and Ophioblennius steindachneri. These three blennioid fishes have markedly different early life histories: A. nigricaudus has a short larval duration (18 days) and develops inshore, M. hubbsi has an intermediate larval duration (24 days) and most individuals develop inshore and O. steindachneri has a long larval life (50 days) and disperses offshore. Estimates of genetic partitioning from mtDNA control region sequences differed greatly between these species and were in the same rank order as predicted by their early life-history characteristics (A. nigricaudus N(ST)=0.536, M. hubbsi N(ST)=0.261 and O. steindachneri N(ST)=0.000). These results indicate that larval strategies may be good predictors of population genetic structure in some marine fishes.

Size-correlated division of labour and spatial distribution of workers in the driver ant, Dorylus molestus.

Driver ants ( Dorylus spp.) show a high degree of worker polymorphism. Previous reports suggest that large Dorylus workers are specialised for defensive tasks. In this study, we first quantitatively tested whether there is a size-correlated division of defensive labour among workers. Second, we determined whether the spatial distribution of workers outside the nest can be predicted based on such size-specific differences in task allocation. We show that the division of defensive behaviour among different-sized workers is not strict. However, there is a significant correlation between worker size and the tendency to carry out defensive tasks. First, workers of larger size were more likely than smaller workers to participate in colony defence. Second, larger workers were more frequent near the nest containing the reproducing individuals and the brood. Finally, large workers were more common in open sections of the trail than in covered sections, which are likely to be less exposed to predators.

Multiple spatially distributed stimulators and timing programs for entrainment of activation during ventricular fibrillation.

Activation sequences during ventricular fibrillation (VF) display complex pattern and fast rate. Recent evidence suggests that even during VF an excitable gap may exist. Existence of excitable gap lead us to hypothesize that it should be possible to entrain activation patterns during VF by using spatially distributed and temporally phased pacing strength stimuli. We describe here the electronics hardware and software that were developed to test our hypothesis. Eight biphasic stimulators were designed and fabricated, each addressable via a TTL input and thus independently triggered. To minimize electrical interference from stimulus pulses the stimulators were optically isolated. A program written in C was used to deliver TTL inputs to time the sequence of stimulation. The parameters that could be varied were, pulse intensity, polarity, and duration, inter pulse interval and activation pattern cycle length. Restitution's of action potential duration, conduction velocity, and complex activation patterns make the timing of stimulators complex. To aid in optimal timing of these stimulators, we used a Luo-Rudy ionic model of cellular activation to simulate VF in a matrix of 400 x 400 cells. Entrainment of activation was verified using animated displays of activation sequences. Using results of simulation we verified the function of our stimulators experimentally using electrically induced VF in canines and by using electrograms recorded from 121-electrode patch. Our results show that the developed hardware and software can be used to deliver distributed stimuli in a flexible and effective pattern, which may aid in development of approaches for treatment of VF.

Spatial distribution of polychlorinated biphenyls in coastal marine sediments receiving industrial effluents in Kuwait.

The concentrations of polychlorinated biphenyls (PCBs) were measured in surficial sediments receiving industrial and municipal effluents in Kuwait. The SigmaPCB concentrations varied by two orders of magnitude ranging from 0.4 to 84 microg kg(-1) dw. The homologue distribution in the study favored the more chlorinated congeners and generally followed the order: penta-PCBs > hexa-PCBs > tetra-PCBs approximately hepta-PCBs, with the dominant congeners being 138, 101, 110, 180, 153, 132, 149, and 118. The spatial distribution revealed significant intersite difference in concentration, with high levels encountered close to a harbor and several wastewater outlets suggesting that point source input is the primary delivery mechanism of PCBs to the sediment. This study suggests that atmospheric deposition of PCBs may not be a significant delivery mechanism to sediments in Kuwait possibly due to low annual precipitation and high annual temperatures that are experienced in the Arabian Gulf. The implication of this observation is that PCBs in air are likely to remain in the gas phase long enough to be subject to long-range atmospheric transport to other regions.

Expression time course and spatial distribution of activated caspase-3 after experimental status epilepticus: contribution of delayed neuronal cell death to seizure-induced neuronal injury.

Pilocarpine-induced status epilepticus (PCSE) is a widely used model to study neurodegeneration in limbic structures after prolonged epileptic seizures. However, mechanisms mediating neuronal cell death in this model require further characterization. Examining the expression time course and spatial distribution of activated caspase-3, we sought to determine the role of apoptosis in PCSE-mediated neuronal cell death. Expression of activated caspase-3, predominantly located in neurons, was detected 24 h (amygdala; piriform and temporal cortex) and 7 days (hippocampus; amygdala; piriform, temporal and parietal cortex; thalamus) after PCSE with strongest induction being observed in the amygdala, the piriform cortex, and the hippocampus. Further analysis revealed TUNEL positivity (24 h and 7 days after SE) and a significant, progressive neuronal cell loss in all brain regions displaying caspase-3 activation. Corresponding to high levels of activated caspase-3 expression, neuronal cell loss was most pronounced in the amygdala, piriform cortex, and dorsal CA-1 hippocampus. These results demonstrate that apoptosis contributes significantly to PCSE-induced neuronal cell death.

Spatial distribution of tissue inhibitor of metalloproteinase-1 mRNA in chronic liver disease.

BACKGROUND/AIMS: Tissue inhibitor of metalloproteinase-1, a specific inhibitor of matrix metalloproteinases, plays an important role in the pathogenesis of fibrosis and tumor progression. However, the precise expression of tissue inhibitor of metalloproteinase-1 messenger RNA in human hepatic fibrosis has not yet been defined. We investigated the spatial distribution of tissue inhibitor of metalloproteinase-1 messenger RNA in chronic human liver disease. METHODS: Northern and in situ hybridization of probes to tissue inhibitor of metalloproteinase-1 messenger RNA were performed in specimens from 16 surgically resected human livers. Immunohistochemical staining of sections for tissue inhibitor of metalloproteinase-1 and immunoelectron microscopy were also performed. RESULTS: Northern hybridization demonstrated that expression of tissue inhibitor of metalloproteinase-1 messenger RNA was increased 3.9-fold in mild chronic hepatitis, 6.8-fold in moderate chronic hepatitis, and 6.4-fold in cirrhosis, compared with control liver. In situ hybridization showed the expression of tissue inhibitor of metalloproteinase-1 messenger RNA in spindle-shaped cells in the fibrous septa and lobules in chronic hepatitis and cirrhosis; these cells were immunohistochemically positive for a-smooth muscle actin. Immunoelectron microscopy revealed localization of tissue inhibitor of metalloproteinase-1 in between fibers, to the rough endoplasmic reticula of stellate cells located in the lobules and periportal areas, and to fibroblasts in the fibrous septa. These results indicate that tissue inhibitor of metalloproteinase-1 was produced mainly by stellate cells in the specimens of chronic liver diseases. CONCLUSIONS: Expression of tissue inhibitor of metalloproteinase-1 messenger RNA is increased in hepatic fibrosis and stellate cells are involved primarily in its expression.

Spatial distribution of pollution in an urban stormwater infiltration basin.

Infiltration basins are frequently used for stormwater drainage. Because stormwater is polluted in highly toxic compounds, assessment of pollution retention by infiltration basins is necessary. Indeed, if basins are not effective in trapping pollution, deep soil and groundwater may be contaminated. This study's objective is to investigate soil pollution in infiltration basins: spatial distribution of soil pollution, optimisation of the number of soil samples and a contamination indicator are presented. It is part of a global project on long-term impact of stormwater infiltration on groundwater. Soil sampling was done on a basin in suburban Lyon (France). Samples were collected at different depths and analysed for nutrients, heavy metals, hydrocarbons and grain size. Pollutant concentrations decrease rapidly with depth while pH, mineralisation and grain size increase. Sustainable metal concentrations are reached at a 30-cm depth, even after 14 years of operation; hydrocarbon pollution is deeper. Principal component analysis shows how pollutants affect each level. The topsoil is different from other levels. Three specifically located points are enough to estimate the mass of pollution trapped by the basin with a 26% error. The proposed contamination indicator is calculated using either average level concentrations or maximum level concentrations. In both cases, the topsoil layer appears polluted but evaluation of lower levels is dependent on the choice of input concentrations.

Changes in perimicrovascular protein spatial distribution due to superfusate.

OBJECTIVE: To determine superfusate-induced changes in the distribution of plasma proteins in the perimicrovascular interstitial matrix. METHODS: Rats were anesthetized with sodium pentobarbital and a mesenteric loop was exteriorized. Intravital video microspectrophotometry was performed using wavelengths of 280, 320, and 700 nm. The images were analyzed to give protein and collagen spatial distributions in vascular regions of rat mesenteric tissue. Perimicrovascular protein concentrations were fitted to an exponential decay model ci + cr exp (-x/k), where ci is distal protein concentration, ci + cr is the protein concentration proximal to the vessel, x is the distance from the vessel wall, and k is the decay constant indicating protein gradient slope. RESULTS: Before superfusion with 0.5-ml normal saline, ci = 1.45 +/- 0.13 g/dl, ci + cr = 4.56 +/- 0.23 g/dl. After the first superfusion, ci decreased (p < 0.01) to 0.53 +/- 0.06 g/dl; following a second superfusion, cr decreased to 0.4 +/- 0.03 g/dl; an additional final superfusion caused a further decrease to 0.33 +/- 0.02 g/dl. ci + cr also decreased significantly during repeated superfusions to 2.92 +/- 0.15, 2.35 +/- 0.25, and 2.1 +/- 0.12 g/dl, respectively. CONCLUSIONS: Superfusion produced changes in perivascular and distal interstitial matrix protein distribution. Protein concentration proximal to the microvessel remained higher than distal concentrations. This could be due to increased gel concentrations inhibiting protein mobility.
