Vicia cytogenetic tests for environmental mutagens. A report of the U.S. Environmental Protection Agency Gene-Tox Program.

Vicia root-tip mitotic and pollen mother-cell meiotic tests are two major kinds of cytogenetic tests for environmental mutagens. According to the present review, 81 of 85 earlier studies used mitotic tests to determine the frequencies of chromosome or chromatid aberrations and/or sister-chromatid exchange from root-tip meristematic cells; only 4 used meiotic tests to determine the frequencies of chromosome aberration from pollen mother cells. Treatment of root-tip meristem can be done by allowing the newly germinated roots to absorb the chemical mutagens from a water solution. Pollen mother cells can be treated by spraying the solution or pipetting the liquid over the flower buds. After an appropriate recovery time, the samples are fixed and stained, and the slides are prepared for metaphase or anaphase figures for scoring aberration frequencies. Slides for meiotic tests are prepared for metaphase I and/or Anaphase I stages for scoring chromosome aberration frequencies. Results of both cytogenetic tests should be expressed in terms of number of breaks per cell or per 100 cells. Test results of 76 chemicals from 32 classes in this review indicate that the Vicia root-tip mitotic test is reliable, efficient, and relatively inexpensive. These results also reveal that antibiotics are most frequently studied, followed by alkyl sulfones, pyrimidine, and purine derivatives. Of all the agents studied through root-tip mitotic tests, about 90% gave positive responses; antibiotics (phleomycin and bleomycin) had very high mutagenicity (less than 1 ppm gave positive response).

Micro-BIA, a colorimetric microtiter assay of lambda prophage induction.

Escherichia coli that are lysogenic for a lambda-lacZ fusion phage produce beta-galactosidase, product of the lacZ gene, upon induction of the prophage by DNA-damaging agents. The miniaturization of a quantitative, colorimetric beta-galactosidase (prophage) induction assay (BIA) is presented. Induction assays are performed in microtiter wells with the aid of multichannel pipetting devices. Results are shown with screening strain BR513 (uvrB delta envA) and a strain, BR339 (uvrB delta lexA3ind-) which exhibits enhanced induction. A method developed for strain BR339 utilizes bacteria stored frozen in log phase, permeabilized in vitro, and used immediately; with this method, 2 consecutive assays may be completed in 1 working day. Mutagens utilized for the model studies included 4NQO, ENNG, daunorubicin, bleomycin, acetoxy-AAF, B[a]P, DMBA, and DEN (the last three in the presence of liver S9). Induced levels of beta-galactosidase were monitored using a vertical light path photometer that measured color absorbance in each microtiter well. Alternatively, color intensity could be determined by using a color chart prepared for this assay. These values were then plotted to generate dose-response curves. Considerable savings in labor and materials are achieved with the method described, one which may be used as a screen for DNA-damaging chemicals. Automated equipment and computers may be used to advantage with this assay, but they are not required.

A cytogenetic approach for detecting the selective toxicity of drugs in avian embryonic B and T lymphocytes.

The developing immune system of late stage embryos and neonates may be particularly susceptible to the toxicity of drugs and environmental contaminants due to high rates of cell proliferation and ongoing processes of differentiation. We have developed a cytogenetic assay to study the mechanisms of the selective targeting of cyclophosphamide (CP) to B lymphocytes compared to T lymphocytes in chicken embryos at days 18-19 of incubation. 5-Bromo-2'-deoxyuridine (BrdU; 3 mg/200 microliters PBS; two doses; 3-h interval) was pipetted onto the inner shell membrane in order to label DNA of replicating lymphoid cells. CP (1.25-40 mg/kg) was injected 1 h after the initial BrdU dose, and the embryos were exposed to colcemid (10 micrograms/100 microliters H2O) at hour 17. Three hours later, the bursa and thymus were removed, and the lymphocytes were swollen in hypotonic solution, fixed, and processed through a fluorescence-plus-Giemsa technique to differentiate sister chromatids. Based on reductions in mitotic indices, B cells were approximately 213 times more susceptible than T cells to the cytotoxicity of CP. Because the mitotic indices of B and T cells were comparable (21.3 +/- 3.7%, vs. 25.5 +/- 6.9%), the differential toxicity cannot be ascribed to greater numbers of B cells being in mitosis. CP induced a dose-related increase in the sister-chromatid exchange frequency in B cells of up to 10.4-fold above controls, representing one of the most sensitive vertebrate systems for detecting the genotoxicity of CP. The average generation time was slowed from 9.8 +/- 0.3 h in control B cells to 19.4 +/- 0.9 h in embryos exposed to 10 mg CP/kg. Furthermore, an analysis of control SCE data from 56 embryos indicated that there was a significant overdispersion of B cells exhibiting relatively high SCE frequencies compared to a Poisson distribution. Our data indicate that the chicken embryo in the late developmental stage is a good model for detecting the presence and selective toxicity of drugs and environmental toxins in differentiating B and T lymphocytes in vivo.

The use of red cells with fused Semliki Forest virus envelope proteins in antibody determinations by hemolysis in gel.

Chicken red blood cells with fused Semliki Forest virus (SFV) proteins on the cell membrane were used in the hemolysis-in-gel (HIG) plates. Optimally the plate contained a 1.5 mm thick gel of 1% agarose with 1% red cells and 1 unit/ml gel of complement. 400 ng of SFV was fused to the red cells in 1 ml of the gel (about 20 virions fused to one red cell). Five microliters of inactivated (56 degrees C, 30 min) serum samples were pipetted into wells in the gel. After 20 h of incubation at 37 degrees C the diameters of the hemolytic zones were directly proportional to the logarithm of the serum dilution. This made it possible to calculate the antibody titers for the samples using an experimental formula T = 10 phi / kappa (T is the titer, phi the diameter in mm and kappa a variable coefficient, which had to be determined for each batch of the plates using a standard serum). Using regression and residual analyses, the formula was shown to fit the experimental results. The fusion-based HIG could be read after as early as 2 h of incubation. The specificity of the test was studied using antisera against Western equine encephalomyelitis, Eastern equine encephalomyelitis, Chikungunya and Uruma viruses, which all gave negative results in the SVF HIG test. Antisera against SFV E1 and E2 proteins were positive, but anti-E3 serum was negative when measured in the SVF HIG test.

Dot-ELISA for serodiagnosis of human infections due to Western equine encephalitis virus.

A standard dot-ELISA (enzyme-linked immunosorbent assay) was modified for use in detecting IgM and IgG class antibodies to Western equine encephalitis (WEE) virus in serum samples from humans infected with this virus. Nitrocellulose membranes were soaked in supernatant fluid from WEE virus-infected cell cultures, air dried, and blocked with bovine protein. Serum samples were pipetted onto sections of the nitrocellulose, incubated, and washed. Addition of antibody to human immunoglobulin conjugated to alkaline phosphatase and enzyme substrate were used to detect the antibodies. Of 13 samples positive for IgM antibody to WEE virus by IgM antibody capture ELISA, 12 were positive by IgM dot-ELISA. IgG antibody to WEE virus was detected by dot-ELISA in 7/8, 10/14 and 7/10 samples with neutralizing, hemagglutination-inhibiting, or complement-fixing antibodies, respectively.

Rapid sampling of multiple enzyme reactions.

A simple method of initiating and sampling six simultaneous reactions was devised. A commercially available vial rack was fitted with a Plexiglas overlaying sheet to stabilize the vials for the addition and sampling procedures. Glass vials were routinely used because of their thermal conductivity advantages. Samples were added and removed, and the reactions were mixed with a multichannel pipet using every other channel. The data showing six simultaneous progress curves for the rapid inactivation of herpes simplex virus ribonucleotide reductase were presented and analyzed. In addition, the time course of 12 reactions catalyzed by varicella zoster virus thymidine kinase were assayed at one min intervals generating 96 data points within 8.5 min. A second experiment generated data points every 30 s for six simultaneous replicate thymidine kinase reactions. The ease of use and high reproducibility of the method are demonstrated by these data.

In vivo study of developmental changes in carbamoyl-phosphate synthetase I in rat liver. Repression of the enzyme synthesis immediately after birth.

The regulatory mechanism of the developmental increase of carbamoyl-phosphate synthetase I in fetal and neonatal rat liver was studied in vivo. The appearance and rapid increase of the enzyme in late fetal period were caused by de novo synthesis of the enzyme protein. The amount of the enzyme protein analyzed by SDS-polyacrylamide gel electrophoresis was proportional to the enzyme activity throughout the period of development. No indication was observed for preexisting protein which could be converted into the active protein. A novel system for the in vivo study of carbamoyl-phosphate synthetase I synthesis was developed. Hepatocytes, mechanically dispersed by repeated passage of the tissue through a pipet, incorporated [35S]methionine into the enzyme. Taking advantage of this system, the regulation of the enzyme synthesis was studied. In vivo synthesis of the enzyme was detected at 4 days before birth and rapidly increased until 1 day before birth. However, the enzyme synthesis was markedly repressed after birth, when the amount of carmamoyl-phosphate synthetase I itself reached the adult level. This result was in a clear contrast with the constant level of the translatable mRNA (Raymond, Y. and Shore, G.C. (1981) Biochim. Biophys. Acta 656, 111-119) and suggested that post-transcriptional regulation is important in addition to the level of mRNA for the regulation of the carbamoyl-phosphate synthetase I level.

Mechanical and enzymatic isolation of mammalian CNS neurons.

Neurons were isolated from various regions of the central nervous system (CNS), by both enzymatic and mechanical isolation. These neurons were isolated from the brains of 3-21-day-old and adult rats, guinea pigs and kittens. Particular attention was directed to the thickness of the slice, the enzymes used and the treatment time, the ionic composition, pH and buffer of the bathing medium, the pipetting of brain slices after enzyme treatment, and the preservation of the isolated cells. The isolated neurons had long dendrites with numerous branchings and a typical morphological shape: e.g. the pyramidal neurons of the rat hippocampus had apical dendrites over 250 micron long; those of the rat neocortex had apical dendrites longer than 300 micron; the Purkinje cells of the rat and kitten also maintained dendrites with fifth branchings. All isolated neurons had well-preserved electrical and chemical membrane properties of the voltage-dependent channels and the receptor-ionophore complexes respectively, even after the enzyme treatment. Thus, these isolated CNS neurons having a well-preserved morphological state can be used extensively for electrophysiological and pharmacological studies on voltage-dependent and drug-gated ionic channels.

Preparative separation of human T cells reactive with the OKT4 monoclonal antibody.

A cell separation technique is described which can provide large numbers (i.e. greater than 5 x 10(7)) of human lymphoid cells which are positively and negatively selected for their ability to react with the OKT4 monoclonal antibody. Peripheral blood lymphocytes are depleted of plastic-adherent cells and then exposed to OKT4 antibody. Unbound OKT4 antibody is removed by washing and then this OKT4-exposed cell population is placed on plastic Petri dishes that are coated with affinity-purified goat anti-mouse immunoglobulin. Cells which do not bind the OKT4 antibody (OKT4- cells) can be obtained by gently washing the nonadherent cells from the plates, while the adherent (OKT4+) cells can be recovered by vigorous pipetting. This procedure yields functional OKT4+ and OKT4- cell populations with a purity which approaches that obtained by separation using the fluorescence-activated cell sorter. The principal advantages of this plate separation technique are that: (1) large numbers of functional cell populations can be obtained; (2) very small quantities of monoclonal antibody (e.g. 1:10(4) dilution of ascites) are required; and (3) sterility of the cell preparations can be maintained quite easily.

Flow cytometric analysis of cell-surface binding elements for fibronectin on mouse lung cell isolates.

Fresh lung cell isolates from LAF1 mice were examined for the presence of fibronectin-binding elements using flow cytometric analysis. Thoroughly perfused lungs from adult male mice were dissociated using an elastase-trypsin digestion, gentle pipetting and filtering. The resulting heterogeneous cell suspension was incubated with fibronectin coated 0.5 micron fluorescent beads. Subsequent flow cytometric analysis indicated the presence of two species of specific fibronectin-binding populations; one of higher binding affinity which can be blocked with exogenous plasma fibronectin and one of lower binding affinity. We tentatively identify the lower affinity binding element with the fibronectin adhesion receptor and the higher affinity element with the putative matrix assembly receptor.

Growth and morphogenesis of mouse prostate epithelial cells in collagen gel matrix culture.

Epithelial cells were isolated from the ventral prostate gland of the mouse after prolonged incubation in a mixture of collagenase, Dispase and hyaluronidase followed by extensive pipetting. The isolated epithelial cells were then embedded in collagen gels. After cultivation in Dulbecco's modified Eagle's medium supplemented with fetal bovine serum, epidermal growth factor, 5 alpha-dihydrotestosterone and cortisol, stimulation of growth and branching morphogenesis of the epithelial cells were observed. Under these culture conditions, growth of contaminating fibroblastic cells was rarely seen. These observations suggest that hormones including androgen directly stimulate the growth and morphogenesis of mouse prostate epithelial cells in culture.

A method for the study of fungal growth inhibition by plant proteins.

A bioassay is described for the study of inhibitory activity of plant proteins on fungal growth. Fungal spores were germinated in liquid growth medium and pipetted into wells of a microtitre plate. Fungal growth was followed spectrophotometrically. The bioassay was tested using crude protein extracts from plant tissues known to have high activities of chitinase and beta-1,3-glucanase, and with purified enzymes. Crude protein preparations and combinations of the purified enzymes produced a temporary reduction of growth but no permanent growth inhibition.

Screening test of stimulants in human urine utilizing headspace gas chromatography for field test.

An accurate and simple screening method of stimulants in human urine using headspace gas chromatography utilizing heat of dissolution of potassium carbonate was developed. A 4.9-g portion of potassium carbonate was put into the vial prior to sending to the field, and a 5-ml aliquot of urine, suspected of containing stimulants and internal standard components was pipetted. After the vial was sealed and shaken by hand, 1 ml of its headspace gas was taken by disposable syringe and injected into the gas chromatograph. A compact gas chromatograph device with flame ionization detector and fused silica capillary column was developed for this experiment. Detection limits of methamphetamine and amphetamine were 1.0 micrograms/ml and 1.5 micrograms/ml, respectively.

Rapid, non-separation electrochemiluminescent DNA hybridization assays for PCR products, using 3'-labelled oligonucleotide probes.

Described are rapid assays for the analysis of PCR products in a one step, non-separation assay based on the use of electrochemiluminescence generated from a tris-bipyridine ruthenium (II) label. The assay uses PCR incorporation of a biotinylated oligonucleotide as a primer, with the inclusion of a labelled oligonucleotide. Oligonucleotides were labelled with an N-hydroxy succinimide ester of tris-bipyridine ruthenium (II) dihexafluorophosphate (Origen-label) by modifying the 3' and 3' 5' ends of the oligonucleotide probes. The assay makes use of the inherent thermal stability and absence of polymerase activity on such probes to allow the PCR and probe hybridization to be completed automatically on the thermocycler. The assay is concluded by the addition of PCR samples to streptavidin beads on an electrochemiluminescence analyser for binding and analysis. Target genes evaluated were the HIV-1 gag gene, and cystic fibrosis delta F-508 deletion mutation. The results obtained from these assays demonstrated the detection of 10 copies of the HIV-1 gag gene, and cystic fibrosis delta F-508 mutation in 1 ng of human DNA within 15 min. This assay format allows a rapid and simple determination of specific amplified DNA sequences, reducing the contamination risks due to washes and multiple pipetting.

Ethanol inhibition of L-type Ca2+ channels in PC12 cells: role of permeant ions.

The effects of acute ethanol exposure on voltage-activated Ca2+ channels in undifferentiated pheochromocytoma (PC12) cells were investigated using whole-cell patch clamp techniques. Concentrations of ethanol (5, 25 and 50 mM), at or below blood alcohol levels which constitute legal intoxication significantly reduced Ca2+ currents evoked from a holding potential of -30 mV. Ethanol-induced inhibition of current was voltage-dependent in some cases, but this was not consistently observed. Inhibition of currents was reversible and was not due to an osmotic effect. The non-inactivating nature of the current, the inhibition of the current by nifedipine, and the lack of inhibition by omega-conotoxin, indicated that the current was carried through high-voltage activated, L-type Ca2+ channels. Since intracellular Ca2+ levels were highly buffered by exchanged with the contents of the patch pipet, ethanol-induced inhibition of currents in PC12 cells is not likely to involve either a change in driving force due to a change in intracellular Ca2+ levels or potentiation of Ca(2+)-dependent Ca2+ channel inactivation by the influx of Ca2+. The degree of inhibition by 25 mM ethanol was the same when either Ca2+, Ba2+ or Na+ was used as the current-carrying ion. This equivalency suggest that the channel's ion selectivity filter is not a site of action for ethanol.

Technical evaluation of thymidine kinase assay in cytosols from breast cancers. EORTC Receptor Study Group Report.

Pilot retrospective studies have pointed to the prognostic value of thymidine kinase (TK) in breast cancer. We studied the Prolifigen TK-REA assay (Sangtec Medical, Sweden), usually applied to serum, for TK analysis in breast cancer cytosols. Reproducibility was good, provided that small volume pipetting was performed carefully. The TK assay was not influenced by the short-term storage of cytosols in liquid nitrogen or at -80 degrees C. However, some steps appeared critical for good laboratory practice. The TK level was affected by thawing the cytosols more than twice. Tumour storage in liquid nitrogen should be preferred over storage at -80 degrees C. The components of the homogenisation buffer, especially sodium molybdate and KCl can have a marked influence on results. Finally, linearity problems arose with some cytosols. Thus, although assay of TK in cytosols is apparently simple, care must be taken in practice. The TK-REA kit should be standardised before widespread use in breast cancer.

Plasma input function determination for PET using a commercial laboratory robot.

A commercial laboratory robot system (Zymate PyTechnology II Laboratory Automation System) was interfaced to standard and custom laboratory equipment and programmed to perform rapid radiochemical assays necessary for plasma input function determination in quantitative PET studies in humans and baboons. A Zymark XP robot arm was used to carry out two assays: (1) the determination of total plasma radioactivity concentrations in a series of small-volume whole blood samples and (2) the determination of unchanged (parent) radiotracer in plasma using only solid phase extraction methods. Steady state robotic throughput for determination of total plasma radioactivity in whole blood samples (0.350 mL) is 14.3 samples/h, which includes automated centrifugation, pipetting, weighing and radioactivity counting. Robotic throughput for the assay of parent radiotracer in plasma is 4-6 samples/h depending on the radiotracer. Percents of total radioactivities present as parent radiotracers at 60 min, postinjection of 25 +/- 5.0 (N = 25), 26 +/- 6.8 (N = 68), 13 +/- 4.4 (N = 30), 32 +/- 7.2 (N = 18), 16 +/- 4.9 (N = 20), were obtained for carbon-11 labeled benztropine, raclopride, methylphenidate, SR 46349B (trans, 4-[(3Z)3-(2-dimethylamino-ethyl) oxyimino-3 (2-fluorophenyl)propen-1-yl]phenol), and cocaine respectively in baboon plasma and 84 +/- 6.4 (N = 9), 18 +/- 11 (N = 10), 74 +/- 5.7 (N = 118) and 16 +/- 3.7 (N = 18) for carbon-11 labeled benztropine, deprenyl, raclopride, and methylphenidate respectively in human plasma. The automated system has been used for more than 4 years for all plasma analyses for 7 different C-11 labeled compounds used routinely in our laboratory. The robotic radiotracer assay runs unattended and includes automated cleanup procedures that eliminates all human contact with plasma-contaminated containers.
