The phagotrophic origin of eukaryotes and phylogenetic classification of Protozoa.

Eukaryotes and archaebacteria form the clade neomura and are sisters, as shown decisively by genes fragmented only in archaebacteria and by many sequence trees. This sisterhood refutes all theories that eukaryotes originated by merging an archaebacterium and an alpha-proteobacterium, which also fail to account for numerous features shared specifically by eukaryotes and actinobacteria. I revise the phagotrophy theory of eukaryote origins by arguing that the essentially autogenous origins of most eukaryotic cell properties (phagotrophy, endomembrane system including peroxisomes, cytoskeleton, nucleus, mitosis and sex) partially overlapped and were synergistic with the symbiogenetic origin of mitochondria from an alpha-proteobacterium. These radical innovations occurred in a derivative of the neomuran common ancestor, which itself had evolved immediately prior to the divergence of eukaryotes and archaebacteria by drastic alterations to its eubacterial ancestor, an actinobacterial posibacterium able to make sterols, by replacing murein peptidoglycan by N-linked glycoproteins and a multitude of other shared neomuran novelties. The conversion of the rigid neomuran wall into a flexible surface coat and the associated origin of phagotrophy were instrumental in the evolution of the endomembrane system, cytoskeleton, nuclear organization and division and sexual life-cycles. Cilia evolved not by symbiogenesis but by autogenous specialization of the cytoskeleton. I argue that the ancestral eukaryote was uniciliate with a single centriole (unikont) and a simple centrosomal cone of microtubules, as in the aerobic amoebozoan zooflagellate Phalansterium. I infer the root of the eukaryote tree at the divergence between opisthokonts (animals, Choanozoa, fungi) with a single posterior cilium and all other eukaryotes, designated 'anterokonts' because of the ancestral presence of an anterior cilium. Anterokonts comprise the Amoebozoa, which may be ancestrally unikont, and a vast ancestrally biciliate clade, named 'bikonts'. The apparently conflicting rRNA and protein trees can be reconciled with each other and this ultrastructural interpretation if long-branch distortions, some mechanistically explicable, are allowed for. Bikonts comprise two groups: corticoflagellates, with a younger anterior cilium, no centrosomal cone and ancestrally a semi-rigid cell cortex with a microtubular band on either side of the posterior mature centriole; and Rhizaria [a new infrakingdom comprising Cercozoa (now including Ascetosporea classis nov.), Retaria phylum nov., Heliozoa and Apusozoa phylum nov.], having a centrosomal cone or radiating microtubules and two microtubular roots and a soft surface, frequently with reticulopodia. Corticoflagellates comprise photokaryotes (Plantae and chromalveolates, both ancestrally with cortical alveoli) and Excavata (a new protozoan infrakingdom comprising Loukozoa, Discicristata and Archezoa, ancestrally with three microtubular roots). All basal eukaryotic radiations were of mitochondrial aerobes; hydrogenosomes evolved polyphyletically from mitochondria long afterwards, the persistence of their double envelope long after their genomes disappeared being a striking instance of membrane heredity. I discuss the relationship between the 13 protozoan phyla recognized here and revise higher protozoan classification by updating as subkingdoms Lankester's 1878 division of Protozoa into Corticata (Excavata, Alveolata; with prominent cortical microtubules and ancestrally localized cytostome--the Parabasalia probably secondarily internalized the cytoskeleton) and Gymnomyxa [infrakingdoms Sarcomastigota (Choanozoa, Amoebozoa) and Rhizaria; both ancestrally with a non-cortical cytoskeleton of radiating singlet microtubules and a relatively soft cell surface with diffused feeding]. As the eukaryote root almost certainly lies within Gymnomyxa, probably among the Sarcomastigota, Corticata are derived. Following the single symbiogenetic origin of chloroplasts in a corticoflagellate host with cortical alveoli, this ancestral plant radiated rapidly into glaucophytes, green plants and red algae. Secondary symbiogeneses subsequently transferred plastids laterally into different hosts, making yet more complex cell chimaeras--probably only thrice: from a red alga to the corticoflagellate ancestor of chromalveolates (Chromista plus Alveolata), from green algae to a secondarily uniciliate cercozoan to form chlorarachneans and independently to a biciliate excavate to yield photosynthetic euglenoids. Tertiary symbiogenesis involving eukaryotic algal symbionts replaced peridinin-containing plastids in two or three dinoflagellate lineages, but yielded no major novel groups. The origin and well-resolved primary bifurcation of eukaryotes probably occurred in the Cryogenian Period, about 850 million years ago, much more recently than suggested by unwarranted backward extrapolations of molecular 'clocks' or dubious interpretations as 'eukaryotic' of earlier large microbial fossils or still more ancient steranes. The origin of chloroplasts and the symbiogenetic incorporation of a red alga into a corticoflagellate to create chromalveolates may both have occurred in a big bang after the Varangerian snowball Earth melted about 580 million years ago, thereby stimulating the ensuing Cambrian explosion of animals and protists in the form of simultaneous, poorly resolved opisthokont and anterokont radiations.

Recovering the geographic origin of early modern humans by realistic and spatially explicit simulations.

Most genetic and archeological evidence argue in favor of a recent and unique origin of modern humans in sub-Saharan Africa, but no attempt has ever been made at quantifying the likelihood of this model, relative to alternative hypotheses of human evolution. In this paper, we investigate the possibility of using multilocus genetic data to correctly infer the geographic origin of humans, and to distinguish between a unique origin (UO) and a multiregional evolution (ME) model. We introduce here an approach based on realistic simulations of the genetic diversity expected after an expansion process of modern humans into the Old World from different possible areas and their comparison to observed data. We find that the geographic origin of the expansion can be correctly recovered provided that a large number of independent markers are used, and that precise information on past demography and potential places of origins is available. In that case, it is also possible to unambiguously distinguish between a unique origin and a multiregional model of human evolution. Application to a real human data set of 377 STR markers tested in 22 populations points toward a unique but surprising North African origin of modern humans. We show that this result could be due to ascertainment bias in favor of markers selected to be polymorphic in Europeans. A new estimation modeling this bias explicitly reveals that East Africa is the most likely place of origin for modern humans.

Geminivirus replication origins have a modular organization.

Tomato golden mosaic virus (TGMV) and bean golden mosaic virus (BGMV) are closely related geminiviruses with bipartite genomes. The A and B DNA components of each virus have cis-acting sequences necessary for replication, and their A components encode trans-acting factors are required for this process. We showed that virus-specific interactions between the cis- and trans-acting functions are required for TGMV and BGMV replication in tobacco protoplasts. We also demonstrated that, similar to the essential TGMV AL1 replication protein, BGMV AL1 binds specifically to its origin in vitro and that neither TGMV nor BGMV AL1 proteins bind to the heterologous origin. The in vitro AL1 binding specificities of the B components were exchanged by site-directed mutagenesis, but the resulting mutants were not replicated by either A component. These results showed that the high-affinity AL1 binding site is necessary but not sufficient for virus-specific origin activity in vivo. Geminivirus genomes also contain a stem-loop sequence that is required for origin function. A BGMV B mutant with the TGMV stem-loop sequence was replicated by BGMV A, indicating that BGMV AL1 does not discriminate between the two sequences. A BGMV B double mutant, with the TGMV AL1 binding site and stem-loop sequences, was not replicated by either A component, indicating that an additional element in the TGMV origin is required for productive interaction with TGMV AL1. These results suggested that geminivirus replication origins are composed of at least three functional modules: (1) a putative stem-loop structure that is required for replication but does not contribute to virus-specific recognition of the origin, (2) a specific high-affinity binding site for the AL1 protein, and (3) at least one additional element that contributes to specific origin recognition by viral trans-acting factors.

Effects of position and orientation of the 72-base-pair-repeat transcriptional enhancer on replication from the simian virus 40 core origin.

A number of recent studies have reported that in papovaviruses such as simian virus 40 (SV40) and polyomavirus, the replication of the viral DNA in vivo is activated by the viral transcriptional enhancer or promoter sequences. Both viral and cellular transcriptional enhancers are well known for their ability to activate transcription in a position- and orientation-independent manner. In the present study, we investigated the effect of the position and orientation of the SV40 72-base-pair (bp) repeat enhancer on its replication activation function. We constructed plasmids containing one copy each of the SV40 core origin and enhancer placed in either order and orientation and at different distances from each other. We assayed the replication efficiencies of these plasmids in the presence of an internal control plasmid in COS-1 monkey kidney cells producing the SV40 T antigen required for replication. We found that the 72-bp repeat was capable of activating replication equally well in either orientation when placed 8 or 9 bp from the core origin. The activation of replication was totally abolished, and replication efficiencies in most instances were found to be lower than that obtained with the core origin alone, when the 72-bp repeat was separated from the core origin by distances of 99 bp or more. This was in direct contrast to the situation with polyomavirus, in which activation of replication by the homologous enhancer or by the SV40 72-bp repeat enhancer is known to be position independent. We also found that when the SV40 core origin and the 72-bp repeat enhancer were adjacent to each other, efficient activation of replication was obtained only if the end of the core origin containing the 17-bp A + T block was linked with the enhancer. In the other orientation of the core origin, activation of replication was either diminished or abolished. Hypotheses such as alteration of chromatin structure by the enhancer and interaction between trans-acting factors binding to the enhancer and the core origin mediating the activation effect are discussed.

Analysis of an origin of DNA replication located at the L terminus of the genome of pseudorabies virus.

We have localized an origin of DNA replication at the L terminus of the pseudorabies virus genome. This origin differs in location as well as in general structure from the origins of replication of other herpesviruses that have been identified. The 600 leftmost nucleotides of the genome that were found to include origin function have been analyzed. This sequence is composed of an 82-bp palindrome whose center of symmetry is separated by 352 unique bp (UL2). Within the UL2, a sequence that fits the consensus sequence of the NF1 binding site, as well as one that has partial homology to the binding site of UL9 of herpes simplex virus, is present. Using truncated fragments of DNA, sequences essential for minimal origin function were delimited to within a fragment that includes the terminal 104 bp of the left end of the genome. Within these 104 bp, two elements essential to origin function have been identified. One of these elements is present within the terminal 64 bp of the L component (within one of the palindromic arms). The other is present within the 22 bp of the UL2 adjacent to this palindromic arm. Other auxiliary elements, although not essential for origin function, contribute to more efficient replication. The NF1 and UL9 binding site homologies were found to be nonessential to origin function.

Mutation of the cyclin-dependent kinase phosphorylation site in simian virus 40 (SV40) large T antigen specifically blocks SV40 origin DNA unwinding.

A mutant simian virus 40 (SV40) large tumor (T) antigen bearing alanine instead of threonine at residue 124 (T124A) failed to replicate SV40 DNA in infected monkey cells (J. Schneider and E. Fanning, J. Virol. 62:1598-1605, 1988). We investigated the biochemical properties of T124A T antigen in greater detail by using purified protein from a baculovirus expression system. Purified T124A is defective in SV40 DNA replication in vitro, but does bind specifically to the viral origin under the conditions normally used for DNA replication. The mutant protein forms double-hexamer complexes at the origin in an ATP-dependent fashion, although the binding reaction requires somewhat higher protein concentrations than the wild-type protein. Binding of T124A protein results in local distortion of the origin DNA similar to that observed with the wild-type protein. These findings indicate that the replication defect of T124A protein is not due to failure to recognize and occupy the origin. Under some conditions T124A is capable of unwinding short origin DNA fragments. However, the mutant protein is almost completely defective in unwinding of circular plasmid DNA molecules containing the SV40 origin. Since the helicase activity of T124A is essentially identical to that of the wild-type protein, we conclude that the mutant is defective in the initial opening of the duplex at the origin, possibly as a result of altered hexamer-hexamer interactions. The phenotype of T124A suggests a possible role for phosphorylation of threonine 124 by cyclin-dependent kinases in controlling the origin unwinding activity of T antigen in infected cells.

[Bibliometric study of the original articles published in the Revista Española de Salud Pública (1991-2000). I. General indicators].

BACKGROUND: For some time, the most of reports have been being disseminated by way of scientific journals, bibliometric studies therefore being fundamental to the characterization and evaluation thereof. The purpose of this study is that of characterizing the Revista Española de Salud Pública based on the original articles published therein throughout the 1991-2000 period. METHODS: Original articles published in the Revista Española de Salud Pública throughout the 1991-2000 period, all inclusive. A study has been made of the following variables: number of original articles, collaboration index or number of signing authors per study, productivity index, geographical spread and main subject. RESULTS: Throughout the 1991-2000 period, 290 original studies (52.3%) of a total of 555 studies were published. The number of originals averaged 29 originals/year A 4.5 degree of collaboration was found to exist for this journal (number signing authors/number originals) for the period under study. The annual of originals by Autonomous Community reveals in the Autonomous Community of Madrid (20.7%), Autonomous Community of Valencia (16.4%), Andalusia (16.1%) and Catalunya (10.0%) have published studies every year throughout the ten-year period under study. The most prevalent subject of all was that related to "Communicable disease" (86 originals), Primary Health Care" (34) and "Environmental pollution" (21). CONCLUSIONS: Generally speaking, it apparently follows that the Revista Española de Salud Pública continues to fall within the output-related indicators of other Spanish and foreign journals and that it has also evolved in keeping with the trend proper of scientific output in the biomedical field. Although "Communicable diseases" are not the main cause of mortality, they continue being the main subject more frequently studied.

[A tentative plan of measuring the amount of the stretch of the skin using origin on body surface].

The present study was made to search for the origin on human dorsal surface, which is available for a correct grasp of a change of body surface (skin surface) with human activities and to try to re-evaluate, using the above origin, quantitative skin deformation of human dorsal surface on raising arms. The following results were obtained: 1) X. examination in standing (static) and arm-raising (dynamic) positions revealed a site with inter-positional difference, as zero, in slippage on dorsal median line. This site was supposed to be the origin and was confirmed to be Th10 and Th12. The palpation of the site of, Th10 and Th12 on dorsal median line was thought to be simple and effective in searching for a site with the least bone and skin slippage, thus in presuming the origin. 2) Based upon the origins on dorsal median line and scapular line parallel thereto in sitting position, 5 cm phi-correct circles were stamped up and down, then arm raising action was taken. The utilization of the origin enabled us to determine the length, directional angle, theta, and area, S, of the slippage between correct and distorted circles as obtained by these actions. 3) For arm raising, the skin slipped up and down on median line for lower and higher sites than the origin, respectively, with the angle theta on scapular line indicating the + direction, while and theta appeared greater and smaller for a nearer site to the origin, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Was the historic contribution of Spain to the Mexican gene pool partially responsible for the higher prevalence of type 2 diabetes in mexican-origin populations? The Spanish Insulin Resistance Study Group, the San Antonio Heart Study, and the Mexico City Diabetes Study.

OBJECTIVE: Mexican-American populations in San Antonio, Texas (SA-MA) and Mexico have a higher prevalence of type 2 diabetes than non-Hispanic whites in San Antonio (SA-NHW). However, the higher prevalence of type 2 diabetes in Mexican-origin populations might be related, in part, not to Native American genetic admixture but to Spanish genetic admixture. RESEARCH DESIGN AND METHODS: Four population-based epidemiological surveys conducted with Mexican-origin and European-origin samples provided data relevant to this question. In all four surveys, type 2 diabetes was defined as fasting plasma glucose > or =7.0 mmol/l or 2-h glucose > or =11.1 mmol/l or use of antidiabetic agents. RESULTS: A comparison of the two Mexican-origin populations showed that the age- and sex-adjusted prevalence of type 2 diabetes was lower in Mexico than in SA-MA (15.1 vs. 17.9%, P = 0.032). Between the two European-origin populations, the prevalence of type 2 diabetes was lower in SA-NHW than in Spain (6.2 vs. 9.1%, P < 0.0001), but differences were attenuated by adjustment for BMI or after stratification by education. In logistic regression analyses, type 2 diabetes was associated with Mexican ethnic origin after adjusting for age, education, BMI, and waist-to-hip ratio. CONCLUSIONS: The prevalence of type 2 diabetes in Spain was intermediate between that in Mexican-origin populations and SA-NHW. Although the higher degree of Native American admixture is a major contributor to the higher rates of type 2 diabetes, we cannot completely rule out a partial contribution of Spanish admixture to diabetes susceptibility among Mexican- origin populations.

Different pattern of chromosomal allele loss in multiple hepatocellular carcinomas as evidence of their multifocal origin.

One of the most problematic aspects of surgery for hepatocellular carcinoma (HCC) is the frequent development of multiple tumors. Determination of the origin of multiple tumors, i.e., multifocal or metastatic, is important for predicting the clinical course of the disease after surgery. In order to clarify the origin of multiple tumors of HCC genetically, we examined patterns of loss of heterozygosity (LOH) on chromosome 16 for DNA isolated from 43 HCCs resected from 19 patients by analysis of restriction fragment length polymorphism. The cases were classified macro- and microscopically into 3 groups: multifocal origin; metastatic origin; and undetermined. Classification based on morphological features was shown to be well correlated with patterns of LOH in multiple tumors of HCC. Different patterns of LOH on chromosome 16 were detected in 8 of 11 patients with tumors of morphologically multifocal origin, whereas they were detected in none of 5 patients with tumors of morphologically metastatic origin. Among five patients with tumors of morphologically undetermined origin, a difference of LOH pattern among the tumors was detected in two, whereas in the other three, the pattern was identical between the tumors. A different pattern of LOH among HCCs arising in situ showed that they were composed of different clones, strongly suggesting their independent clonal origin and multifocal development. These results show that not only appropriate morphological observation but also examination of the LOH pattern on a particular chromosome is useful in diagnosis of multifocal HCC.

[Education level and origin as predictors of hospitalization among Jewish adults in Israel: a population-based study].

BACKGROUND: Age, sex, origin and level of education are recognized as predictors of healthcare utilization. However, the joint effects of these variables on the risk of hospitalization have yet to be described for specific diagnostic categories, such as diseases of the circulatory, respiratory, genitourinary, digestive and nervous systems, infectious and parasitic diseases, malignant neoplasms, injury and poisoning. OBJECTIVES: To evaluate the association between origin, education, sex, age and the risk of hospitalization due to major diagnostic categories. METHODS: Cross-sectional study linking population-based databases, including Israeli census and hospitalization records. RESULTS: The study encompassed 35,413 hospitalized subjects and 386,317 non-hospitalized subjects. Origin other than Israel was a risk factor for hospitalization, with an odds ratio of 1.31 (95% CI 1.25-1.36) for Asia, 1.43 (1.37-1.50) for Africa, and 1.24 (1.20-1.29) for Europe-America (P for all origins <0.0001). Compared to subjects with post-high school educations, those with high school educations were 1.18 times more likely to be hospitalized (1.14-1.21), while those with elementary school educations experienced a risk of 1.32 (1.28-1.37) (P < 0.0001). Origin was a marked risk factor for hospitalization due to malignancy, with an odds ratio of 2.19 (1.71-2.79) for European-American origin relative to Israeli origin (P < 0.0001). The inverse association between education level and risk of hospitalization was constant across all diagnostic categories other than malignancy. CONCLUSIONS: Lower education was found to be a significant risk factor for hospitalization in most diagnostic categories. Origin was associated with increased risk for hospitalization due to malignant neoplasms and diseases of the respiratory, nervous, genitourinary and circulatory systems.

Anatomical variation in origin and course of the thoracoacromial trunk and its branches.

The variations in origin and course of the thoracoacromial trunk (TAT) and its branches were studied in 178 cadavers during the routine dissection from the year 1982 to 2002. The TAT originated from the first part of the axillary artery (AA) in 13.4% cases of the right and 10.6% of the left axilla. The variations in origin of the branches of TAT were divided into three groups. First variation group showed deltoacromial (DA) and clavipectoral (CP) subtrunks of the TAT originating directly from the AA in majority of cases. Second group revealed clavicular branch of the TAT originating from the AA, whereas in the third group all classical branches originated directly from the AA and there was no existence of the TAT. The superior thoracic artery (STA) originated from the TAT in 16.8% (Confidence Interval, CI: 11.12-21.89) cases of the right and 6.1% (CI: 2.59-9.53) of the left axilla and the lateral thoracic artery in 39.8% (CI: 32.01-46.10) cases of the right and 29.3% (CI: 15.01-26.80) of the left axilla. The incidence of variations in origin of the TAT and its branches was found higher on right side. The knowledge of these variations is of anatomical and surgical interest. This information is useful for the surgeons dealing with the axillary region especially in case of reconstructive surgery.

[Value of multislice spiral CT in the diagnosis of anomalous origin of the coronary arteries].

OBJECTIVE: To investigate the value of multislice spiral CT (MSCT) coronary angiography in the diagnosis of anomalous origin of coronary arteries. METHODS: 5000 patients diagnosed or suspected as with diseases of coronary artery underwent MSCT coronary angiography (retrospectively ECG-gating; 0.5 s rotation; one-sector or two-sector reconstruction algorithm; intravenous contrast agent) 2001-2005. RESULTS: Anomalous origin of coronary artery was incidentally found with multislice spiral CT in 39 patients (0.78%), including anomalously high origin of the right coronary artery (n = 20), anomalous origin of the right coronary artery from left coronary sinus (n = 11), aberrant circumflex artery arises from the right coronary sinus or right coronary (n = 3), anomalous origin of the right coronary artery from the posterior coronary sinus (n = 1), anomalous origin of the left coronary artery from the posterior coronary sinus (n = 1), single coronary artery (n = 2), separate origin of circumflex and left anterior descending arteries from left coronary sinus (n = 1). These signs were better shown by MSCT coronary angiography. CONCLUSION: MSCT coronary angiography is a useful adjunctive technique to invasive coronary angiography in the diagnosis of anomalous origin of coronary arteries.

Conformational changes induced by integration host factor at origin gamma of R6K and copy number control.

We have investigated the role of integration host factor (IHF) in the replication of plasmid R6K by studying the maintainance of the plasmid in a strain of Escherichia coli that lacks both subunits of IHF and in an isogenic wild type strain and found that all three origins, alpha, beta, and gamma, were functional in the absence of IHF; however, loss of IHF reduced the copy number of those replicons initiating solely from ori gamma by 5-fold. Concomitant loss of direct repeats within the origin that bind the R6K replication initiator protein, Pi, resulted in a further reduction in copy number. Using gel mobility shift analysis, we showed that IHF bound specifically only to one site within the A/T rich region of the minimal origin adjacent to the Pi binding sites. The origin region possessed no intrinsic DNA curvature although IHF induced a strong bend upon binding. Combination footprinting with different orders of addition of Pi and IHF suggested that there was no cooperativity between the two proteins with regard to DNA binding. Hydroxyl-radical footprinting revealed hypersensitive asymmetric periodic cleavage sites within the origin region in the presence of IHF that extended over 200 base pairs and a localized perturbation of cleavage chemistry. The presence of periodic cleavages was dependent upon the presence of the wild type R6K origin sequence and was not observed when the IHF binding site was positioned adjacent to a heterologous sequence. We observed that the conformational changes induced by IHF upon binding to the R6K origin were negatively correlated with the observed decrease in copy number, and therefore, origin conformation altered by protein-DNA interaction may play an important role in the regulation of replication initiation.

[Transfection efficiency of adenoviral vector AD5/F35 to malignant hematopoietic cells of different origins].

This study was aimed to investigate the transfection efficiency of adenoviral vector AD5/F35 to hematopoietic malignant cells lines of various origins and AD5/F35 cytotoxicity. The hematologic malignant cell lines of various origins were transfected by AD5/F35-EGFP at different multiple of infection (MOI) and AD5-EGFP was used as control; the proportion of fluorescence positive cells was detected by flow cytometry; the killing effect of virus on infective target cells was assayed by MTT and observed by fluorescence microscopy. The results showed that the transfection efficiency of AD5/F35 vector to cell line of myeloid origin was > 99% at MOI = 30, the transfective efficiency of AD5 vector was 26.4% at MOI = 1,000; the transfection efficiency of AD5/F35 vector and AD5 vector to cell line of B cell origin were 11.7% and 5.7%, respectively, at MOI = 1,000. AD5/F35 and AD5 vectors could not effectively transfect cells of T cell origin, no fluorescence positive cells were detected at MOI = 1,000; no significant killing effect of AD5/F35 vector on infective target cells was observed at MOI = 1,000. It is concluded that AD5/F35 vector infection has definite selectivity to hematologic malignant cells of various origin, the infection ability of AD5/F35 vector to cells of myeloid origin is stronger than that to cells of B cell origin, the cytotoxicity of AD5/F35 vector to infective target cells is small. The AD5/F35 vector is preferable to AD5 vector in respect of infection ability and offers good prospects of application in gene therapy for myeloid leukemia cells as target cells.

[Liver metastases of unknown origin.].

OBJECTIVE: Approximately 5% of cancer patients are diagnosed with tumour of unknown origin (3-4% in Iceland). Of those 10-30% have liver metastases. Liver metastases of unknown origin is thus not an uncommon problem. In the present study information about the origin and histology of liver metastases of unknown origin was compiled. MATERIAL AND METHODS: Records of all biopsies from liver metastases performed in the years 1987-1996 were retrieved from the medical database of the Department of Pathology at the University of Iceland. The biopsies came from a group of 176 patients. Ninety-two cases, in which the origin of the primary tumour was suspected or known, were excluded from the study, leaving 84 cases where the primary was completely unknown. The database of the Icelandic Cancer Society was used to gather data about the final tissue diagnosis and the location of the primary tumour when known. RESULTS: The Cancer Society data revealed the location of the primary tumour in 55 of the 84 cases of liver metastases of unknown origin. The most prevalent (75%) primary tumours were cancers of the pancreas (15), lung (13) and colon/rectum (12). The tissue diagnosis was adenocarcinoma in 33 of the 55 cases. In the male patients 83% of the adenocarcinoma metastases came from the colon/rectum or pancreas. The corresponding figure for the female patients was 67%, while 20% of the tumours in females originated in the gallbladder and biliary tree. CONCLUSIONS: In two thirds of the cases of liver metastases of unknown origin the primary tumour was later discovered. The most prevalent tumours were cancers in the pancreas, lung and colon/rectum. Adenocarcinoma was the tissue diagnosis in 60% of cases.

The origin of the osteoclast.

The origin of the osteoclast has long been controversial. Various approaches such as traditional light microscopy, electron microscopy, microcinephotography, autoradiography, parabiosis, quail-chick nuclear marker, etc, have been used in attempts to resolve the problem. There are 2 major concepts: the view of skeletal (local) origin vs, that of extraskeletal origin (from blood-borne cells), and recently the latter view appears to have prevailed. Electron micrographic studies revealed that osteocytes and preosteoblasts can merge with pre-existing osteoclasts. Several studies by autoradiography and by quail-chick nuclear marker strongly supported the concept of extraskeletal origin. Although an alternative interpretation of the experimental results of Crelin and Koch supports the concept of skeletal origin, the majority of various experimental results by other authors would be in accordance with the following supposition: the osteoclast originates from a hematopoietic cell line (mononuclear phagocyte system) of the bone marrow which is essentially of extrinsic origin, but in the process of increasing the number of nuclei of the osteoclast, osteoblastic cells as well as osteoclastic cells can merge with the preosteoclasts or osteoclast. However, several problems regarding the origin of the osteoclast including the relation between the osteoclast and other giant cells, still remain unsolved.

[Studies on phenotype, development, and viability of human spontaneous abortuses with acrocentric trisomies and polyploidies: with reference to the relationship of the viability to the origin of extrachromosomes (author's transl)].

The specimens to be presented consist of 65 trisomics and 37 polyploids which were obtained from a cytogenetic survey of human spontaneous abortion with various chromosome banding techniques. The phenotype of the 102 abortuses was analysed macroscopically before and after fixation of the tissues. The developmental age was estimated with the crown-rump measurement and from stage of Streeter's horizon on embryos and fetuses, or from the size of the placenta when an embryo was lost. The degree of phenotypic abnormality and the developmental age were used on determining the viability of abortuses. This viability was examined in each type of chromosomal anomlies. Three of 4 13-trisomics and all of 4 21-trisomics showed normal phenotype and advanced development, while most of 14-trisomics, 15-trisomics and 22-trisomics had the various severe phenotypic anomalies such as blighted ovum and disorganized embryo, and less developed. These findings suggest that abortuses with the chromosomal anomalies which can be found in the newborns longer survive than those with other anomalies. In other words, these anomalies have a better viability among abortuses. Eleven trisomics and 11 polyploids were ascertained for the origin of extra chromosomes by sequential QFQ- and RFA-banding. Of 7 abortuses with trisomy 21 studied, 4 originated at the maternal meiotic non-disjunction and none was of paternal origin. On the other hand, several published studies have shown that the paternal origin is not uncommon in Down children. Therefore, as far as trisomy 21 is concerned, it is concluded that the trisomy 21 of paternal origin has much better survival rate than that of maternal origin. In triploidy, the present study could not define the relationship between the viability and the origin, since 9 of 10 abortuses studied and all of 4 triploid newborns so far reported are of paternal origin.
