Synthesis of enantiopure 3-quinuclidinone analogues with three stereogenic centers: (1S,2R,4S)- and (1S,2S, 4S)-2-(Hydroxymethyl)-1-azabicyclo[2.2.2]octan-5-one and stereocontrol of nucleophilic addition to the carbonyl group.

The pseudoenantiomeric title compounds have been prepared from quincorine (QCI) and quincoridine (QCD), respectively, in enantiopure form following an efficient six-step pathway. Nucleophilic attack at the carbonyl group proceeds preferentially from the supposedly more hindered endo pi-face, giving quinuclidinols with natural configuration at C5 (up to 97%). pi-Face selectivity is highest in the QCD series with bulky O-protecting groups, involving an unprecedented 1,7-stereoinduction.

Photocyclization of 2-azabicyclo[3.3.0]octane-3-carboxylate derivatives: induced and noninduced diastereoselectivity.

[reaction: see text] The decarboxylative photocyclization of potassium 2-azabicyclo[3.3.0]octanoate ethylene-linked to phthalimide leads to a mixture of two diastereoisomeric products. The overall noninduced diastereoselectivity is remarkably high, indicating a preference of spin-orbit coupling controlled 1,7-biradical combination process. Memory of chirality vanished due to free rotation about the connecting spacer bonds.

Reversal of stereoselectivity in [5 + 2] pyrone--alkene cycloadditons using a sulfoxide-to-sulfoximine switch. Enantiodivergent synthesis of 8-oxabicyclo[3.2.1]octane systems.

[reaction: see text] Switching from a sulfinyl to a sulfonimidoyl group allows the reversal of the sense of asymmetric induction in thermal [5C + 2C] intramolecular pyrone-alkene cycloadditions. Removal of the sulfoximine unit from the resulting cycloadducts yields optically active oxabicyclic systems that are enantiomeric to those obtained using the sulfinyl chiral auxiliary.

Oxabicyclo[3.2.1]octane derivatives as highly reactive dienophiles: synthesis of bicyclo[5.n.0] systems.

[reaction: see text] We have developed highly versatile, homochiral oxabicyclo[3.2.1]octadiene building blocks for the synthesis of natural products. We have found that these bridged alkenes undergo exceptionally facile Diels-Alder reactions and react faster than several well studied bicyclo[2.2.1]heptene dienophiles. The reaction proceeds with high levels of stereochemical control and in very good to excellent yields, providing access to bicyclo[5.4.0]undecane and bicyclo[5.3.0]decane systems. This reactivity is attributed to strain and homoconjugation effects.

Efficient bioreduction of bicyclo[2.2.2]octane-2,5-dione and bicyclo[2.2.2]oct-7-ene-2,5-dione by genetically engineered Saccharomyces cerevisiae.

A screening of non-conventional yeast species and several Saccharomyces cerevisiae (baker's yeast) strains overexpressing known carbonyl reductases revealed the S. cerevisiae reductase encoded by YMR226c as highly efficient for the reduction of the diketones 1 and 2 to their corresponding hydroxyketones 3-6 (Scheme 1) in excellent enantiomeric excesses. Bioreduction of 1 using the genetically engineered yeast TMB4100, overexpressing YMR226c, resulted in >99% ee for hydroxyketone (+)-4 and 84-98% ee for (-)-3, depending on the degree of conversion. Baker's yeast reduction of diketone 2 resulted in >98% ee for the hydroxyketones (+)-5 and (+)-6. However, TMB4100 led to significantly higher conversion rates (over 40 fold faster) and also a minor improvement of the enantiomeric excesses (>99%).
