Single-bubble sonochemiluminescence in aqueous luminol solutions.

Sonochemiluminescence (SCL) of luminol due to a single bubble is studied through spectral measurement. No SCL was observed from a stable single bubble that emitted high-intensity sonoluminescence (SL). In contrast, SCL was observed under conditions of an unstable dancing bubble, where a bubble grows and ejects tiny bubbles, making it "dance" by counteraction. Furthermore, SCL was observed from dancing bubbles even when SL was not observed, depending on the dissolved gas content. The instability of bubble collapse is the key parameter governing SCL.

Evaluation of ferryl formation by electrocatalytic oxidation of alkene using luminol chemiluminescence.

Electrochemical formation of ferryl porphyrin was examined by electrocatalytic oxidation of alkene by measuring luminol chemiluminescence using a flow-injection method. Emission was observed both below the reduction potential of Fe(III)TMPyP (-0.08 V at pH 11, -0.02 V at pH 7 and 0.15 V at pH 3) and above the oxidation potential (0.6 V at pH 11, 0.75 V at pH 7 and 1.1 V at pH 3). However, both anodic and cathodic emissions were inhibited significantly by the addition of alkene (cyclopent-2-ene-1-acetic acid) solutions downstream of the working electrode. Further, the spectra at both anodic and cathodic sides shifted to the longer wavelength (>424 nm) compared to the original spectrum of Fe(III)TMPyP (422 nm), which was not observed with the addition of alkene solution. Therefore, the results suggest that the electrochemically generated oxo-ferryl species have been engaged in catalytic oxidation of alkene before the flow reaches the observation cell.

[Flow injection chemiluminescence method for the determination of phenol with N-bromosuccinimide-luminol system].

A novel flow injection chemiluminescence method was presented for the determination of phenol. The method is based on the quenching effect of phenols on the chemiluminescence reaction between luminol and N-bromosuccinimide. The linear range for the determination of phenol is 1.0 x 10(-5) -9.0 x 10(-4) mg x mL(-1), and the detection limit is 1.81 x 10(-7) mg x mL(-1). This method was used for the determination of phenol in water samples with satisfactory results. The mechanism of the reaction was also expounded.

[Effect of cigarette-smoke tar on oxygenation activity of granulocytes detected by luminol- and lucigenin-enhanced chemiluminescence response of rabbit whole blood].

Both luminol- and lucigenin-dependent rabbit whole blood chemiluminescence showed a concentration-dependent inhibition response to cigarette-smoke tar of 0.26 to 2.6 mg/ml. Tar showed no cytotoxicity to granulocytes by Evans blue exclusion. It is suggested that oxygenation activity of granulocytes in smoker's blood could be depressed by tar, resulting in harmful effects on the host defense mechanism with increase of susceptibility to infection and incidence of tumorigenicity.

[Luminol-dependent chemiluminescence of peripheral neutrophils in asthmatic patients].

To evaluate the role of the neutrophil in the pathophysiology of bronchial asthma, we examined the luminol-dependent chemiluminescence of peripheral neutrophils (CL) in steroid-dependent asthmatic patients during an acute attack (n = 18), during stable period (n = 10), and in normal controls (n = 21). When stimulated with formyl-methionyl-leucyl-phenylalanine (fMLP 0.1 microM), the CL of patients in acute attack was lower than that of normal controls. (5.29 +/- 1.25 vs. 9.29 +/- 0.84 relative light units p less than 0.01) Among 18 patients, we could examine 10 patients during the stable period after receiving steroids. The CL of those 10 patients during the stable period was significantly higher than their CL during acute attacks, when evaluated by the paired t-test (p less than 0.05), and was not very different than in normal controls. When stimulated with opsonized zymosan (5 mg/ml), CL was similar in the three groups. We also examined the CL of peripheral neutrophils of normal persons after the preincubation with serum of patients during acute attack and stable period. When stimulated with fMLP, CL after preincubation with serum of patients during acute attack was lower than CL after preincubation with the serum of patients during the stable period. It was suggested that the respiratory-burst activity of peripheral neutrophils in acute asthmatic attack was reduced to the chemotactic stimulus of fMLP.

Effect of neutrophil activating substances on intracellular generation of phagocyte chemiluminescence by means of luminol-bound microspheres.

The intracellular chemiluminescence of granulocytes was measured by luminol-bound microspheres. After one minute of incubation, which was necessary for the granulocytes to phagocytize microspheres, intracellular chemiluminescence was generated. The intracellular chemiluminescence was enhanced by the introduction of granulocyte stimuli including concanavalin A, phorbol-myristate acetate, and formyl peptides. This incremental effect was the result of certain metabolic changes inside the cells. This newly discovered chemiluminescent reagent promises to be a useful tool for examining metabolic changes inside phagocytic cells.

Determination of phagocyte activity in whole blood of carp (Cyprinus carpio) by luminol-enhanced chemiluminescence.

In this study, the phagocytic capacity of fish peripheral blood leucocytes was studied using the luminol-enhanced chemiluminescence (CL) method. Suspensions of rice starch (1%) or opsonized zymosan particles-OZP (0.2%) were used as activators. The level of CL was detected at 25 degrees C in various whole blood volumes (100, 50, 20 a 10 microliters) during the mixed or unmixed measurement mode. The peak of CL in all starch-activated samples was between 2.0-4.0 mV, regardless of blood volume or a mode of measurement. Similar results were obtained for OZP activated CL when measured in the mixed mode. On the other hand, the peak of OZP activated CL in unmixed samples was much higher and it showed positive correlation with the increasing volume of blood in a sample. In contrast, no significant differences between the reaction curve areas were found for different blood volumes. After correction for phagocyte number the highest CL activity per cell was observed in the samples containing 10 microliters of blood. It can be concluded that 10-20 microliters of blood are optimal volumes for the measurements of respiratory burst activity in the whole blood of carp and opsonized zymosan particles should be used as an activator. At the same time, the unmixed mode of measurement seems to give better results than the mixed one.

Distinct patterns of granulocyte luminol-dependent chemiluminescence response to lectins WGA and RCA-I.

THe chemiluminescence response was measured in human polymorphonuclear leukocytes (PMNLs) after stimulation with different concentrations of the lectins Triticum vulgaris agglutinin (wheat germ agglutinin, WGA) and Ricinus communis agglutinin I (RCA-I). The two lectins achieved distinct patterns of chemiluminescence; RCA-I evoked a dose-dependent response (0-15 micrograms/ml) with respect to the initial rate of light emission, but the peak value was reached after different lengths of time. A maximum response was obtained after about 25 min with a concentration of 2-4 micrograms/ml. By contrast, WGA caused a bimodal reaction after stimulus addition. In this case the first peak occurred after 15-20 min, and the second peak after around 60 min with maximum effect for 0.5 and 0.25-0.50 micrograms/ml WGA, respectively. The PMNL response is discussed in relation to the regulation of the production of oxygen metabolites and in relation to the pathophysiologic consequences of lectin-mediated activation of phagocytic cells in intestinal epithelium.

Specificity of oxygen radical scavengers and assessment of free radical scavenger efficiency using luminol enhanced chemiluminescence.

The selective scavenging capacities of 19 important oxygen radical scavengers were determined by adding them individually to each of the four oxy radical standards, (superoxide, hydroxy, alkoxy and hydroperoxy, and singlet O2), calculating the percent chemiluminescence inhibited, and extrapolating O2 equivalents neutralized from baseline. The sensitivity (0.01 nm/ml) and selectivity of this method not only allows identification of individual oxygen free radical species but also quantitates the efficiency of free radical scavengers.
