Credibility judgements of detectives: the impact of nonverbal behavior, social skills, and physical characteristics on impression formation.

Ninety-one detectives watched videofragments of 20 confederates who had been instructed either to tell the truth or to lie. After each videofragment the detectives indicated whether the confederate was lying. Although the study took place in a police context, and thus had higher ecological validity, the results strongly corresponded with those of previous studies: The accuracy rate was low (49%). Like the students in other deception studies, the detectives used the wrong cues to detect deception, and the actual situation (whether the confederates were telling the truth) did not predict suspiciousness.

The radiation exposure of black-headed gulls (Larus ridibundus) in the Ravenglass Estuary, Cumbria, U.K.: a preliminary assessment.

A variety of fission-product and transuranic radionuclides originating from the marine discharges from the fuel reprocessing plant at Sellafield, Cumbria are detectable in the bodies of black-headed gulls and their environment in the Ravenglass Estuary approximately 10 km to the south-west of the plant. The maximum concentrations of 95Nb, 137Cs and 239/240Pu detected in body tissues lie in the range 2 X 10(-3) to 1.5 X 10(-2) Bqg-1 wet weight. Many more radionuclides are detected in the regurgitated pellets and faeces produced by the gulls and have higher concentrations in the range 7 X 10(-3) to 1.7 Bqg-1 wet weight. The radionuclide contamination of the sediment produces a source of enhanced external gamma-ray exposure which can be measured directly. The available data on distributions and concentrations of radionuclides have been combined with simple dosimetry models to provide estimates of the radiation exposure of the birds. The total whole body dose rate to the adult birds from the contaminant radionuclides is approximately ten times that from the natural radiation background, while that to the developing eggs is approximately four times the natural background. The potential radiation exposure of the cells lining the alimentary tract could be much higher when contaminated food is in transit, but the long-term average exposure in this case is subject to uncertainty. The limited data on the effects of radiation exposure on birds do not indicate any response at dose rates below one hundred times that from the natural radiation background.(ABSTRACT TRUNCATED AT 250 WORDS)

[Significance of detection of HBV-DNA and CMV-DNA by polymerase chain reaction in screening mothers' milk].

Hepatitis B virus(HBV) DNA and cytomegalovirus DNA in the colostrum of lying-in women were detected by polymerase chain reaction for the safety of mothers' milk in the milk bank. The results showed that: 1. HBV-DNA was not found in the milk of women with negative serological markers of the virus, while in the milk of women with positive marker of HBsAg, HBeAg and HBcAb, the positive rate of HBV-DNA was 81.25%, in the milk of mothers with positive marker of HBsAg, HBcAb and HBeAb, the positive rate was 45.24%, in the milk of women only with HBcAb and HBeAb positive, the rate was 23.08%. 2. The specific amplified products of CMV DNA in the milk were obtained in 42.80%-70.00% women whose CMV IgM antibody was positive. These findings suggest that the excretion of viruses would increase in mothers with active virus replication, and the viruses would be transmitted to newborns by milk. The authors think that only the milk from the mothers with both negative serological markers and negative PCR of the viruses can be collected and stored in the milk bank. PCR is a specific and sensitive technique which can be used to monitor the safety of mothers' milk. In this paper, the authors have presented some suggestions about how to give breast-feeding and protect newborns whose mothers are various status of virus secretion in the milk.

Detection of A t-complex antigen by secondary cell-mediated lymphocytotoxicity.

Because of the inconsistency in published results concerning the serological detection of cell surface antigens coded for by the t-complex, a cell-mediated lymphocytotoxicity (CML) assay, secondary CML, was used in a search for t-antigens. By sensitizing C3H.Ttf (C3H.Brachyury, tufted) with the congenic strain C3H.Ttf/tw18 splenic cells, a response against lipopolysaccharide (LPS) stimulated splenic cells from C3H.Ttf/tw18 mice is obtained. The locus coding for the antigen detected by this reaction lies to the left of tf on the murine seventeenth chromosome. The secondary CML response to this antigen is H-2 restricted and detects an antigen on all t-haplotypes tested: tw18, tw18tf, t12, t6, th2tf, and tw5.

Evidence that the Saethre-Chotzen syndrome locus lies between D7S664 and D7S507, by genetic analysis and detection of a microdeletion in a patient.

The locus for Saethre-Chotzen syndrome, a common autosomal dominant disorder of craniosynostosis and digital anomalies, was previously mapped to chromosome 7p between D7S513 and D7S516. We used linkage and haplotype analyses to narrow the disease locus to an 8-cM region between D7S664 and D7S507. The tightest linkage was to locus D7S664 (Z = 7.16, theta = .00). Chromosomes from a Saethre-Chotzen syndrome patient with t(2;7) (p23;p22) were used for in situ hybridization with YAC clones containing D7S664 and D7S507. The D7S664 locus was found to lie distal to the 7p22 breakpoint, and the D7S507 locus was deleted from the translocation chromosomes. These genetic and physical mapping data independently show that the disease locus resides in this interval.

Radioimmunoscintigraphy and radioimmunotherapy in cancer: principles and application.

Radioimmunoscintigraphy (RIS) and radioimmunotherapy (RIT) are recent approaches in the diagnosis and treatment of cancer. They take advantage of the antibody specificity of tumor surface antigens and of the emitted radiation from suitable radioisotopes, as a means of imaging (RIS) or therapy (RIT). Research into RIS and RIT radiolabelled agents remains an ongoing process. Principles governing the choice of radionuclides, labelling protocols, antibody suitability, and optimization of "tumor to normal tissue ratios" are the same for both RIS and RIT. The investigational stages of the labelled product, prior to clinical application, are also the same. These stages include radiochemical and radiobiological evaluation as well as determination of immunoreactivity. Furthermore, RIS may be considered as the first stage in development, before progressing on to RIT. Differences between RIS and RIT are associated with the application of each technique, that is, the type of radiation emitted by the isotope, dosage regimens, haematopoetic toxicity and the appearance of human antimurine antibody response (HAMA). RIS has found widespread clinical application, detecting a variety of tumors. However, its potential lies in patient management and in detecting metastases. On the other hand RIT is still in its infancy. It appears promising, and for the moment is used as a complementary technique to surgery and/or chemotherapy in clinical trials on cancer treatment. Finally, incorporation of these basic principles arising from past experiences, into the design of RIT trials improve responses.

NaCl detection thresholds: comparison of Fischer 344 and Wistar rats.

Adult Fischer 344 (F344) rats fail to display any preference for NaCl solutions at concentrations typically preferred by other rat strains. To determine whether this behavior is due to a strain difference in NaCl detection threshold, a conditioned taste aversion (CTA) was first established to a suprathreshold concentration of NaCl (0.1 M). Then, a series of dilute NaCl solutions, ranging from 0.0 to 0.011 M NaCl, were presented to F344 (n = 16) and Wistar (n = 16) rats. The lowest concentration at which there was a reliable difference in the preference scores of conditioned and control rats was defined as the detection threshold. Results indicate that the detection threshold for NaCl lies between 0.001 and 0.002 M NaCl for both F344 and Wistar rats. The addition of the sodium channel blocker amiloride to the NaCl solutions raised the detection threshold 10-fold to 0.03-0.04 M NaCl for both strains of rats. These results suggest that the NaCl detection thresholds of F344 and Wistar rats are similar and that these strains do not differ in the degree to which amiloride raises this threshold.

[Effectiveness of differential diagnosis of benign and malignant ulcers at endoscopy].

AIM: Assessment of efficacy of differential diagnosis between benign and malignant lesions in gastric mucosa by means of endoscopy. MATERIALS AND METHODS: Morphological verdicts upon investigation of operative specimens were compared to findings at preoperative duodenofibrogastroscopy with spot biopsy in 326 patients operated for gastric ulcer. The number of discrepancies between the endoscopic and morphological diagnoses enabled estimation of endoscopy + biopsy efficiency in identification of the ulcer malignancy. RESULTS: The number of diagnostic errors made on malignant or benign nature of ulcer at endoscopic and biopsy investigations is rather big, i.e. at least 12% for both false negative and false positive hypophyses. The overall diagnostic efficacy of endoscopy in detection of the disease lies within 65-80%. CONCLUSION: Reliability of endoscopic identification including biopsy of malignant nature of gastric ulcers is not very high. Real sensitivity of this method in detection of ulcers undergoing malignant transformation reaches 76-78%.

Seminal vesicle biopsy: accuracy and implications for staging of prostate cancer.

OBJECTIVES: Seminal vesicle biopsy (SVB) is a new technique for detecting the spread of prostate cancer to the seminal vesicles. A comparison of findings following SVB in patients undergoing radiation therapy with pathologic findings following radical retropubic prostatectomy (RRP) was made to evaluate the accuracy of this test and its use in the staging of prostate cancer. METHODS: Four hundred nine patients with clinically localized adenocarcinoma of the prostate gland were evaluated for treatment: 222 patients underwent SVB prior to radiation therapy and 187 patients underwent RRP. Clinical stages in patients undergoing SVB included T1a (1 patient), T1b (4), T1c (35), T2a (49), T2b (96), and T2c (37); RRP clinical stages included T1b (3 patients), T1c (48), T2a (57), T2b (66), and T2c (13). The Gleason scores in patients undergoing SVB were 2 to 4 in 50 men, 5 to 6 in 110 men, and 7 and greater in 62 men; the Gleason scores in patients undergoing RRP were 2 to 4 in 53 men, 5 to 6 in 94 men, and 7 and greater in 40 men. Prostate-specific antigen (PSA) values ranged from 1.3 to 190 ng/mL (median 10.75) in men undergoing SVB and ranged from 0.5 to 140.6 ng/mL (median 9.0) in men undergoing RRP. RESULTS: The overall incidence of seminal vesicle involvement as determined by the two techniques was the same. Seminal vesicle involvement was found in 33 of 222 patients (15%) undergoing SVB and in 27 of 187 (14%), of the RRP specimens (P = 0.9). When the two groups were further divided by three prognostic categories (clinical stage, PSA level, and grade), there was no difference in the incidence of seminal vesicle involvement between the two methods, except in the patients with Gleason score of 4 or less. In these patients, 5 of 53 (9%) had seminal vesicle involvement in the RRP group, compared with none of the 50 men in the SVB group (P = 0.02). Disease that was not organ confined was found in 69 of 187 prostatectomy specimens (37%). Of these patients, 27 of 69 (39%) had seminal vesicle involvement. CONCLUSIONS: SVB is an accurate method of detecting seminal vesicle invasion based on comparisons with radical prostatectomy findings. Its importance lies in its ability to detect a large percentage of patients with non-organ-confined disease and in its use in modifying treatment planning accordingly.

A finite element model of remote palpation of breast lesions using radiation force: factors affecting tissue displacement.

The early detection of breast cancer reduces patient mortality. The most common method of breast cancer detection is palpation. However, lesions that lie deep within the breast are difficult to palpate when they are small. Thus, a method of remote palpation, which may allow the detection of small lesions lying deep within the breast, is currently under investigation. In this method, acoustic radiation force is used to apply localized forces within tissue (to tissue volumes on the order of 2 mm3) and the resulting tissue displacements are mapped using ultrasonic correlation based methods. A volume of tissue that is stiffer than the surrounding medium (i.e., a lesion) distributes the force throughout the tissue beneath it, resulting in larger regions of displacement, and smaller maximum displacements. The resulting displacement maps may be used to image tissue stiffness. A finite-element-model (FEM) of acoustic remote palpation is presented in this paper. Using this model, a parametric analysis of the affect of varying tissue and acoustic beam characteristics on radiation force induced tissue displacements is performed. The results are used to evaluate the potential of acoustic remote palpation to provide useful diagnostic information in a clinical setting. The potential for using a single diagnostic transducer to both generate radiation force and track the resulting displacements is investigated.

Ambulatory system for human motion analysis using a kinematic sensor: monitoring of daily physical activity in the elderly.

A new method of physical activity monitoring is presented, which is able to detect body postures (sitting, standing, and lying) and periods of walking in elderly persons using only one kinematic sensor attached to the chest. The wavelet transform, in conjunction with a simple kinematics model, was used to detect different postural transitions (PTs) and walking periods during daily physical activity. To evaluate the system, three studies were performed. The method was first tested on 11 community-dwelling elderly subjects in a gait laboratory where an optical motion system (Vicon) was used as a reference system. In the second study, the system was tested for classifying PTs (i.e., lying-to-sitting, sitting-to-lying, and turning the body in bed) in 24 hospitalized elderly persons. Finally, in a third study monitoring was performed on nine elderly persons for 45-60 min during their daily physical activity. Moreover, the possibility-to-perform long-term monitoring over 12 h has been shown. The first study revealed a close concordance between the ambulatory and reference systems. Overall, subjects performed 349 PTs during this study. Compared with the reference system, the ambulatory system had an overall sensitivity of 99% for detection of the different PTs. Sensitivities and specificities were 93% and 82% in sit-to-stand, and 82% and 94% in stand-to-sit, respectively. In both first and second studies, the ambulatory system also showed a very high accuracy (> 99%) in identifying the 62 transfers or rolling out of bed, as well as 144 different posture changes to the back, ventral, right and left sides. Relatively high sensitivity (> 90%) was obtained for the classification of usual physical activities in the third study in comparison with visual observation. Sensitivities and specificities were, respectively, 90.2% and 93.4% in sitting, 92.2% and 92.1% in "standing + walking," and, finally, 98.4% and 99.7% in lying. Overall detection errors (as percent of range) were 3.9% for "standing + walking," 4.1% for sitting, and 0.3% for lying. Finally, overall symmetric mean average errors were 12% for "standing + walking," 8.2% for sitting, and 1.3% for lying.

Overlapping submicroscopic deletions in Xq28 in two unrelated boys with developmental disorders: identification of a gene near FRAXE.

Two unrelated boys are described with delay in development and submicroscopic deletions in Xq28, near FRAXE. Molecular diagnosis to exclude the fragile X (FRAXA) syndrome used the direct probe pfxa3, together with a control probe pS8 (DXS296), against PstI restriction digests of DNA. Deletions were detected initially by the control probe pS8, which is an anonymous fragment subcloned from YAC 539, within 1 Mb distal to FRAXA. Further molecular analyses determined that the maximum size of the deletion is < 100 kb in one boy (MK) and is wholly overlapped by the deletion of up to approximately 200 kb in the other (CB). These deletions lie between the sequences detected by the probe VK21C (DXS296) and a dinucleotide repeat VK18AC (DXS295). The patient MK had only speech delay with otherwise normal development, while patient CB had global developmental delay that included speech delay. Detection of overlapping deletions in these two cases led to speculation that coding sequences of a gene(s) important in language development may be affected. Hybridization of the pS8 and VK21A probes to zooblots revealed cross-species homology. This conservation during evolution suggested that this region contains sequences with functional significance in normal development. The VK21A probe detected a 9.5-kb transcript in placenta and brain and a smaller, 2.5-kb, transcript in other tissues analyzed.

[Sonographic findings in interstitial lung diseases].

UNLABELLED: Up to now, sonography of the thorax has not been taken into consideration in cases of interstitial lung disease because of its technical limitations. This study was aimed at examining pleural and subpleural alterations in cases of interstitial lung disease and comparing the sonographic method with the usual imaging procedures such as x-ray and computer tomography. PATIENTS AND METHODS: 24 patients, aged 25 to 70 who were diagnosed as suffering from an interstitial lung disease and underwent sonography of the thorax, were analysed with regard to the following criteria: 1 pleural effusions, 2 pleural fragmentations, 3 subpleural infiltrations > 2 mm, 4 pleural nodules. RESULTS: Six patients were diagnosed to have small pleural effusions which had not been visible on x-ray-scans. 14 patients showed pleural fragmentations, 10 patients had subpleural infiltrations, and in one patient pleural nodules could be detected. Out of the 24 patients suffering interstitial lung disease, sonographic alterations were diagnosed in 17 cases (= 71%). Among the 9 patients definitively suffering from sarcoidosis, five had had a completely inconspicuous ultrasonic result, four of them with sarcoidosis stage I. CONCLUSION: Sonography of the thorax has proved to be an excellent complementing examination method in cases of interstitial lung disease. The advantages lie in the possible detection of small pleural effusions and small subpleural infiltrations, where this method can also be used to monitor therapy.

Clinical correlates of high lie scale elevations among forensic patients.

The MMPI Lie scale serves as an indicator of test validity, primarily intended to detect efforts by psychologically unsophisticated subjects to present themselves in an unrealistically positive fashion. The unexpected occurrence of Lie scale elevations among paranoid patients not considered to be unsophisticated or naive prompted previous researchers to speculate that the measure might have other interpretive utility. The current study examined the potential relationship between high Lie scores and paranoid psychopathology in a forensic population. All subjects who met the inclusion criteria, Lie greater than raw score of six and no clinical scale elevations, were found to have some type of psychotic disorder with prominent paranoid features. The possible implications of the findings for clinicians responsible for determinations of dangerousness resulting from mental illness were discussed.

Optimizing low-light microscopy with back-illuminated electron multiplying charge-coupled device: enhanced sensitivity, speed, and resolution.

The back-illuminated electron multiplying charge-coupled device (EMCCD) camera is having a profound influence on the field of low-light dynamic cellular microscopy, combining highest possible photon collection efficiency with the ability to virtually eliminate the readout noise detection limit. We report here the use of this camera, in 512 x 512 frame-transfer chip format at 10-MHz pixel readout speed, in optimizing a demanding ultra-low-light intracellular calcium flux microscopy setup. The arrangement employed includes a spinning confocal Nipkow disk, which, while facilitating the need to both generate images at very rapid frame rates and minimize background photons, yields very weak signals. The challenge for the camera lies not just in detecting as many of these scarce photons as possible, but also in operating at a frame rate that meets the temporal resolution requirements of many low-light microscopy approaches, a particular demand of smooth muscle calcium flux microscopy. Results presented illustrate both the significant sensitivity improvement offered by this technology over the previous standard in ultra-low-light CCD detection, the GenIII+intensified charge-coupled device (ICCD), and also portray the advanced temporal and spatial resolution capabilities of the EMCCD.

The "lying down" adrenal sign: a sonographic indicator of renal agenesis or ectopia in fetuses and neonates.

An inability to image the kidneys, severe oligohydramnios, and an empty (nonvisualized) urinary bladder are the most common sonographic findings in fetuses with renal agenesis. The same features, except for the oligohydramnios, are seen in neonates with renal agenesis. Failure to visualize a kidney in the usual flank location with or without demonstration of the kidney in an ectopic location may also be a feature in fetuses or neonates with renal ectopia. The adrenal gland has an unusual but distinctive appearance on longitudinal sonograms in these patients. Sonograms of 23 fetuses and six neonates referred for presumed bilateral or unilateral renal agenesis or ectopia were reviewed retrospectively for the presence of a flattened ("lying down") adrenal gland. At the time of the examination, the adrenal gland was not actively sought as a part of the fetal survey or postnatal examination but was found retrospectively in 48% of these subjects. Although this finding has been described previously, the emphasis was on this observation as a potentially confusing sonographic feature. This manuscript emphasizes the characteristic appearance of the adrenal gland which instead serves as a simple means of confirming that the kidney did not develop in the flank. Detection of a flattened ("lying down") adrenal gland is an observation that should be actively sought in all fetuses and neonates with a presumed diagnosis of renal agenesis or ectopia.

Asymmetric distribution of exogenous thymidine among pyrimidine isostichs suggests compartmentalization of replicative DNA synthesis during regeneration in rat liver.

The distribution of radioactivity among pyrimidine isostichs (or isoplyths) of DNA from 24-h regenerating rat liver was studied with [3H]Thd, [14C]orotate or with inorganic 32Pi. Expression of incorporated radioactivity as log10% of total radioactivity recovered for each of the 11 pyrimidine isostichs detected showed that radioactivity from [3H]Thd was asymmetrically distributed among the isostichs, i.e., 3H radioactivity failed to access regions of DNA yielding lower molecular weight pyrimidine isostichs as efficiently as it accessed regions yielding higher molecular weight pyrimidine isostichs. The thymine (T) content of isostichs exceeded that of cytosine (C), i.e., T/C ratios for the first 10 isostichs averaged 1.43 +/- 0.08 and 1.28 +/- 0.05, depending on the method of analysis; furthermore, the T/C ratio for isostich 1 was significantly higher than ratios for isostichs 2 through 10. Asymmetric distributions of [3H]Thd radioactivity also were seen at 18 or 30 h post-partial hepatectomy. Thus, radioactivity from [3H]Thd, a DNA precursor from the salvage pathway, failed to efficiently access lower molecular weight isostichs despite thymine enrichment, suggesting that thymine moieties were supplied from additional sources. Radioactivity from [14C]orotate accessed lower molecular weight pyrimidine tracts more efficiently than [3H]Thd, but less efficiently than it accessed higher molecular weight isostichs, resulting in an asymmetric distribution of 14C radioactivity. This result suggested that appreciable quantities of thymine and cytosine moieties utilized for DNA synthesis were supplied de novo, but other sources also were utilized. Radioactivity from 32Pi, a de novo precursor, was distributed symmetrically, i.e., the slope among lower molecular weight isostichs increased enough that it was indistinguishable from slopes for intermediate and higher molecular weight isostichs. Since 32P radioactivity among lower molecular weight isostichs reflects appreciable contributions of de novo phosphate moieties from both pyrimidine- and purine-containing deoxynucleoside triphosphates, opportunities for observing contributions of 32P radioactivity from pathways other than the de novo pathways appeared to lie beyond limits of detectability. The distribution of radioactivity from labeled DNA precursors among lower molecular weight pyrimidine tracts (a) indicate that thymine moieties are contributed by both salvage and de novo pathways; (b) support the possibility that cytosine moieties also are contributed by both pathways; and (c) support the 'replitase' concept for channeling dNTPs to replicating forks.
