Evidence for suppression of immune function by insulin-like growth factor-1 in dwarf rats in vivo.

These studies were undertaken to investigate the effects of increasing or decreasing IGF-1 levels on aspects of immune function in rats. Female dwarf rats were treated with recombinant human IGF-1 or with a potent sheep anti-IGF-serum. Body weight, thymus weight and spleen weight increased with IGF-1 treatment (p < 0.001), while there was no effect of anti-IGF-1 treatment when compared with the appropriate normal sheep serum (NSS) treated controls. IGF-1 treatment significantly decreased WBC and RBC counts, but increased the ratio of CD4+:CD8+ T-cells. Anti-IGF-1 serum had no effect on these parameters compared with NSS. However anti-IGF-1 was associated with increased T-cell numbers, decreased natural killer cells, and enhancement of the animals' ability to produce specific IgG in response to injection of keyhole limpet haemocyanin (KLH). These results indicate that IGF-1 may suppress immune function although increasing the size of immune organs such as spleen.

[Effects of medium and long-chain triglyceride on the immune function of burn patients during early postburn stage].

OBJECTIVE: To explore the effects of medium and long-chain triglyceride on the immune function of burn patients during early postburn stage and its possible mechanism. METHODS: Thirty burn patients with TBSA exceeding 30% were enrolled in this study and were randomly divided into two groups (n = 15 in each group). The patients in F group were supplemented orally with enteral nutrient Fresubin 750MCT containing medium (MCT) and long-chain triglyceride (LCT), and those in N group with Nutrition containing only LCT. All the patients were treated with nutritional support beginning from 24 PBH, continued for 10 days. The changes in plasma levels of IL-2, IL-4, PGE(2) and T lymphocyte transformation rate (LTR) in peripheral blood of patients in the two groups were determined on 1, 4, 7 and 10 postburn days (PBDs). RESULTS: The plasma IL-2 level in all time points exhibited no difference in all patients (P > 0.05). The plasma level of PGE(2) on PBD 4 in F group was obviously lower than that in N group (P < 0.01). The plasma level of IL-4 on PBDs 4, 7 and 10 in F group were evidently higher than that in N group (P < 0.01). The T LTR in peripheral blood on PBDs 4 and 7 in F group was much higher than that in N group (P < 0.05 - 0.01). CONCLUSION: Enteral nutrient containing MCT/LCT seemed to be superior to that containing only LCT in the improvement of postburn immune function in burn patients.

Thymoma and late onset mucocutaneous candidiasis associated with a plasma inhibitor of cell-mediated immune function.

A case is described of chronic mucocutaneous candidiasis in association with thymoma. A plasma inhibitor of certain in vitro tests of cell mediated immune function, both of autologous and of normal cells was isolated from the patient's blood and shown to be stable on heating to 56 degrees C for 1 hour and to freezing and thawing. Isolation of the active fraction of Sephadex G-200 suggested a m.w. of 50,000-65,000 daltons. Despite disappearance of the inhibitor after apparently curative radiotherapy, suggesting the tumour was the origin, the chronic mucocutaneous candidiasis persists. The association of plasma inhibitors of immune function, with and without thymoma and chronic mucocutaneous candidiasis is reviewed and the possible nature of the substance discussed.

[Immune functional changes in patients of acute Henoch-Schonlein purpura and regulatory effect of integrated Traditional Chinese and Western medicine on it].

OBJECTIVE: To observe the changes of immune function in children with acute Henoch-Schonlein purpura (AHSP) and the effects of integrated traditional Chinese and western medicine (TCM-WM) on it. METHODS: Immunological criteria in TCM-WM group (n = 35) treated by Yinfu Decoction (YFD) combined with transfer factor and 35 patients in the control group (n = 35) treated by conventional treatment were observed and compared before and after treatment. Also the criteria were compared with those of 40 healthy subjects. RESULTS: Before treatment, the levels of IgA, IgM, CD4, CD4/CD8 ratio, rosette forming rate of RBC-C3b receptor in patients were higher than those in the healthy subjects, but the levels of CD8 was obviously lower, the difference was significant (P < 0.01). The above-mentioned criteria were all improved in the two treated groups after treatment, and the improvement was more significant in the TCM-WM group than that in the control group (P < 0.05). Besides, the cure-markedly effective rate in the former was better than that in the latter significantly (P < 0.01). CONCLUSION: There exist multiple immune functional disturbance in AHSP patients. Combined treatment of YFD and transfer factor has obvious immune regulatory effect and is an effective therapy with few side-effects and low recurrence rate.

Dietary effect of EPA-rich and DHA-rich fish oils on the immune function of Sprague-Dawley rats.

The dietary effect of fish oils (FOs) rich in eicosapentaenoic acid (EPA) or docosahexaenoic acid (DHA) on the immune function of Sprague-Dawley rats was compared with that of safflower oil. After 3 weeks of feeding at the 10% level of a dietary fat, the IgG and IgM production by splenocytes and IgG production by mesenteric lymph node (MLN) lymphocytes were significantly higher in the FO-fed rats, while no significant difference was found in IgA or IgE productivity by both the spleen and MLN lymphocytes. In the FO-fed rats, peritoneal exudate cells released a lower amount of LTB4, reflecting their lower arachidonic acid level, and a higher amount of LTB5, reflecting their higher EPA level in phospholipids. On these EPA-rich FO exerted a stronger effect than DHA-rich FO immune functions.

Intravenous hyperalimentation with high arginine levels improves wound healing and immune function.

The purpose of this study was to evaluate the effect of increased arginine levels in intravenous hyperalimentation (IVH) therapy on wound healing and thymic immune function. Groups of SD rats, 275-325 g, underwent placement of internal jugular catheter, 7-cm dorsal skin wounding, insertion of polyvinyl alcohol sponges subcutaneously, and closure of wounds with stainless-steel sutures. Twenty-four hours later, rats were started on IVH at a rate of 0.8-1 ml/100 g body wt/hr. All IVH solutions contained 20% dextrose, adequate amounts of minerals and vitamins, and two different amino acid mixtures: (A) Fre III (4.05 g ARG/liter) (n = 13); (B) experimental (7.50 g ARG/liter) (n = 11). Solutions were isonitrogenous, and contained similar amounts of essential amino acids. After 7 days of IVH, weight gain did not differ between the two groups; however, cumulative N balance was superior in group A. Wound healing was improved in group B as assessed by fresh wound strip breaking strength, fixed breaking strength, and the amount of reparative collagen deposition as assessed by the hydroxyproline content of the implanted sponges. Group B animals also had improved thymic function as assessed by thymic weight, the total number of thymic lymphocytes/gland and mitogenic reactivity of thymic lymphocytes to PHA and Con A. The experiments indicate that high arginine levels in IVH solutions improve wound healing and thymic immune function following injury.

Structured intervention in family caregivers of the demented elderly and changes in their immune function.

The aim of the present study was to investigate the effects of a group structured intervention on the mental and physical discomfort and immune function of 20 family caregivers. A structured intervention for caregivers consists of five sessions, each of which lasts 90 min. This was a modified version of the program that had been originally developed for cancer patients. All the family caregivers were female and ranged in age from 47 to 66 years (mean: 54.7 +/- 4.4). The period of care at home ranged from 1 to 12 years (mean: 5.8 +/- 2.7). Concerning the original diseases of the care-receivers, 10 had vascular dementia and eight had Alzheimer's disease. Nine out of 20 caregivers had no care support, and seven utilized no public resources such as day-care centers. Only five caregivers felt that they were healthy. Two psychometries, that is, Profile of Mood States (POMS) and General Health Questionnaire-30 (GHQ-30) were administered and blood samples were drawn before and after intervention. Comparison of results showed that there was significant improvement (P < 0.05) in the scores of depression, anger-hostility, fatigue and confusion in the POMS, and physical symptoms, anxiety-mood disorder, suicidality-depression in the GHQ-30. Also, there was significant (P = 0.0325) augmentation of natural-killer cell activity. The present study suggests that this kind of intervention was effective for relieving emotional and physical discomfort, and also for improving immune function.

The effects of an instant haw beverage on lipid levels, antioxidant enzyme and immune function in hyperlipidemia patients.

OBJECTIVE: To determine the effectiveness of an instant haw beverage in regulating lipid disturbance, enhancing antioxidant enzyme activity and immune function. METHODS: Data was collected from 60 hyperlipidemic subjects. In this crossover design, each subject randomly received either the instant haw beverage (100 ml corresponding to 3 g of haw powder or 30 g of fresh haw fruit plus the carrier-guar gum plus some starch) or placebo (guar gum 1.5 g plus some starch as the carrier of the beverage) twice daily. Each supplementation lasted 31 days with a 28-day washout period between treatments. RESULTS: The instant haw beverage significantly reduced total serum cholesterol (9.6%), triglyceride (12.1%), LDLC (18%) while significantly increased SOD activities (7.5%). The placebo was shown to have positive results in some of the lipid profiles, though the effects of the instant haw beverage demonstrated greater significance. Serum triglyceride levels were significantly decreased and SOD activity significantly increased only as subjects were supplemented with the instant haw beverage while no significant changes were seen with placebo. CONCLUSION: Supplementation with the instant haw beverage positively affects blood lipid profile, antioxidant status and immune function in individuals with hyperlipidemia.

Iron status and resting immune function in female collegiate swimmers.

Iron deficiency may lead to anemia and may result in compromised endurance exercise performance. Iron deficiency has also been reported to adversely affect the immune system and has been associated with attenuation of natural killer cell (NK) activity. This study was conducted to examine the relationship between iron status and NK activity in highly conditioned female athletes. Ten collegiate female swimmers (SWM) and 9 inactive females (SED) participated in this investigation. Resting blood samples were obtained and analyzed for serum iron and ferritin. NK activity (% lysis) was determined using a whole blood method (51Cr release assay). No significant relationship was found between iron and NK activity (r = 0.55, p =.09), nor between serum ferritin and NK activity (r = 0.33, p =.35) for SWM. ANOVA revealed significantly greater NK activity for SWM (51.63 +/- 15.79%) versus SED (30.34 +/- 13.67%). Serum ferritin levels were not significantly different between SWM (20.38 +/- 8.62 hg á ml-1) and SED (16.79 +/- 10.53 hg á ml-1), nor were iron values different between groups (16.54 +/- 2. 17 mmol á L-1 SWM; 11.92 +/- 2.61 mmol á L-1 SED). A significant relationship between iron status and resting immune function could not be established. Exercise training may affect NK activity; however, the influence of iron status on immune function requires further evaluation.

Immune function, stress response, and body condition in arctic-breeding common eiders in relation to cadmium, mercury, and selenium concentrations.

We examined relationships between trace metal concentrations in tissues of common eider ducks (cadmium, mercury, and selenium) and selected biomarkers of health (stress response, immune function, and body condition). This study was conducted at an eider nesting colony in the Canadian arctic in 1998 and 1999. Capture-induced stress, measured as the rise in corticosterone concentrations following capture, was positively related (P=0.03) to renal cadmium concentration in 1998 when incubating eiders were sampled, but not in 1999 when prenesting eiders were sampled. Stress response was inversely related (P=0.02) to selenium concentrations in 1999. Following capture and blood sampling in 1999, eiders were placed in a flight pen on-site for eight days in order to examine immune function. Cell-mediated immunity, measured as the skin-swelling response to an intradermal injection of phytohemagglutinin-P, (PHA-P), was positively related (P=0.003) to hepatic selenium. The heterophil:lymphocyte ratio was inversely related (P=0.08) to hepatic selenium. In 1998, selenium was positively related to body mass (P=0.01), abdominal fat mass (P=0.07), kidney mass (P=0.03), and liver mass (P=0.07). In 1999, hepatic mercury was negatively related to abdominal fat mass (P=0.01), spleen mass (P=0.07) and body mass at capture (P=0.09) in prenesting eiders.

[The effect of melatonin on enhancing immune function and inhibiting the ability of NO over-release in morphine dependent mice].

AIM: To observe the effects and mechanism of melatonin (MT) on the immune function of morphine dependent mice. METHODS: A physical dependent mice model was established by repeated subcutaneous injection of morphine. The intensity of morphine withdrawal syndrome was evaluated according to the weight of immune organs, the proliferation reaction of stimulated splenic lymphocytes by Con A, the phagoindex of blood primed macrophages and the content of NO induced in the peritoneal macrophage (pM phi). RESULTS: MT reversed the inhibitory effect of morphine on the proliferation ability of splenic lymphocytes and enhanced the phagocytosis of macrophages of morphine dependent mice obviously and prevented the over-release of NO from pM phi. The enhancing effects of MT on the phagocytosis can be prevented by naloxon. CONCLUSION: MT can significantly enhance the immune function of morphine dependent mice and inhibit NO excessive release from pM phi.

Multiple sclerosis: in relapsing patients, immune functions vary with disease activity as assessed by MRI.

We have serially studied immunoglobulin G secretion in vitro, natural killer cell function, and concanavalin A-induced suppression in a group of seven patients with relapsing-remitting multiple sclerosis. In two patients, the development of a large clinically asymptomatic MRI lesion was accompanied by reductions in natural killer cell function, immunoglobulin G secretion in vitro (after pokeweed mitogen stimulation), and concanavalin A-induced suppression without parallel change in lymphocyte markers. We did not see this type of change in matched controls nor in stable multiple sclerosis studied serially. When clinical attacks appeared, there was no significant change in immune function. We conclude that changes in immune function correlate well with the activity of the disease as recognized by MRI. We suspect that decreased natural killer cell function, immunoglobulin G secretion in vitro, and concanavalin A-induced suppression are secondary to the large lesions recognized by MRI.

The role of immune function in schizophrenia: an overview.

Immune alterations in schizophrenia have been described for decades. However, modern immunological methods and new insights into the highly developed and functionally differentiated immune system allows an integrative view of both, the older and also recent findings of immunological abnormalities in schizophrenia. Both, the unspecific and the specific arm of the immune system seem to be involved in the dysfunction of the immune system in schizophrenia. The unspecific "innate" immune system shows signs of an overactivation in unmedicated schizophrenic patients, as increased monocytes and gamma delta-cells point to. Increased levels of Interleukin-6 (IL-6) and the activation of the IL-6 system in schizophrenia might also be the result of the activation of monocytes/macrophages. On the contrary, several parameters of the specific cellular immune system are blunted, e.g. the decreased T-helper-1 (TH-1) related immune parameters in schizophrenic patients both, in vitro and in vivo. It seems that a TH-1-TH-2 imbalance with a shift to the TH-2 system is associated with schizophrenia. During antipsychotic therapy with neuroleptics, the specific TH-1 related immune answer becomes activated, but also the B-cell system and the antibody production increases.

Immune function testing of human pharmaceuticals: regulatory overshoot?

Suppression of the activity of the immune system can result in decreased resistance to (opportunistic) infection and cancer. Experimental animal studies have identified many immunotoxic agents, but data on unintended immuno-toxicity of human pharmaceuticals are limited. To enhance the sensitivity of the non-clinical screening of human pharmaceuticals, immunotoxicity endpoints in standard toxicity studies, as well as an immune function study, are requested by the EU, incorporating a T-cell-dependent antigen response in the first screening phase. Authorities from other areas in the world follow similar lines for immunotoxicity testing, but differ in whether or not the functional assay is seen as a primary requisite or can be secondary to other factors or observations. Based on a limited survey in the framework of the International Conference on Harmonization, the European position has been reconsidered to negotiate a broad cause-for-concern approach. The relevance of functional testing of the immune system to show effects of human pharmaceuticals is shown in this review. There is certainly a need for more in-depth testing of the integrity of the immune system and a cause-for-concern approach is, therefore, needed. A broad category is the number of compounds that might interact via binding to receptor sites at the broadly taken population of leukocytes (e.g., lymphocytes, macrophages). Criteria to differentiate among compounds in this category have yet to be ascertained.

[The effects of pcDNA3.1-IL-15 transfection on the surface molecules and immune function of murine bone marrow-derived dendritic cells].

AIM: To explore the effects of pcDNA3.1-IL-15 transfected on co-stimulatory molecule expression and immune function on murine bone marrow-derived dendritic cells (DCs). METHODS: Recombinant plasmid pcDNA3.1-IL-15 was constructed and used to transfect DCs. The expression of CD40, CD80 and CD86 on the transfected DCs was analyzed by flow cytometry. Murine splenocytes were stimulated with the transfected DCs. CD4+ and CD8+ T cell subsets in the splenocytes were analyzed by flow cytometry. The proliferation of splenocytes was detected by MTT colorimetry. The IFN-gamma in the culture supernatant of the splenocytes was detected by ELISA. RESULTS: pcDNA3.1-IL-15-transfected DCs expressed higher level of CD40, CD80 and CD86, and induced proliferation of CD4+ T cells and CD8+ T cells in the splenocytes. But the ratio of CD4+/CD8+ T cells was lower than that in the spleen cells stimulated by untransfected DCs or DCs transfected with pcDNA3.1. CONCLUSION: pcDNA3.1-IL-15 can improve the expression of co-stimulatory molecules on DCs and enhance their immune function.

Effect of CLA supplementation on immune function in young healthy volunteers.

OBJECTIVES: This study investigated the effect of dietary CLA supplementation (3g/day; 50:50 mix of the two major isomers) on the immune system and plasma lipids and glucose of healthy human (male and female) volunteers. DESIGN: Double-blind, randomized, reference-controlled study. SUBJECT AND INTERVENTION: A total of 28 healthy male and female participants aged 25-50 y received either high oleic sunflower oil (reference) or 50% CLA 9-11 and 50% CLA 10-12 CLA isomers (50:50 CLA-triglyceride form). The treatments were given as supplements in soft-gel capsules providing a total 3 g (6 x 500 mg capsules) per day in treatment groups for 12 weeks. A 12-week washout period followed the intervention period. RESULTS: Levels of plasma IgA and IgM were increased (P < 0.05 and 0.01 respectively), while plasma IgE levels were decreased (P < 0.05). CLA supplementation also decreased the levels of the proinflammatory cytokines, TNF-alpha and IL-1beta (P < 0.05), but increased the levels of the anti-inflammatory cytokine, IL-10 (P < 0.05). Another aspect of immune function, delayed type hypersensitivity (DTH) response, was decreased during and after CLA supplementation (P < 0.05). However, plasma glucose, lipids, lymphocyte phenotypic results were not affected significantly by CLA. CONCLUSION: This is the first study to show that CLA, a fatty acid naturally found in dairy and meat products, can beneficially affect immune function in healthy human volunteers. [Effects of early intestinal application of sijunzi decoction on immune function in post-operational patients of gastrointestinal tumor].

OBJECTIVE: To assess the effects of early intestinal application of sijunzi decoction (SJZD) on the immune function in post-operational patients of gastrointestinal tumor. METHODS: Ninety-two patients with malignant gastrointestinal tumor were randomly divided into two groups. Patients in both groups were given the isocaloric and isonitrogenous enteral nutritional support starting from the first day after operation for 1 week, but to the tested group, SJZD was given additionally. The concentration of serum cytokines such as interleukin-1 (IL-1), interleukin-2 (IL-2), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha), the peripheral blood cell counts of total lymphocyte, T-lymphocyte, B lymphocyte, and T lymphocyte subsets (CD3, CD4, CD8, CD4/CD8) as well as the levels of IgA, IgG, IGM and C-reactive protein (CRP) were measured on the day before operation, the first morning after operation and at the end of study. RESULTS: At the end of the study, the concentration of IgA, IgG, 1gM, number of total lymphocyte, CD3, CD4 and CD4 lCD8, and serum IL-2 were obviously higher (P < 0.05), and levels of IL-6, TNF-alpha and CRP were obviously lower in the tested group than those in the control group (P < 0.05). CONCLUSION: Early application of SJZD on the base of enteral nutritional therapy can lessen the degree of post-operational stress and inflammatory response, and enhance the immune function of patients.

Effect of 2,4-dicholorophenoxyacetic acid, trifluralin and triallate herbicides on immune function.

The commercial formulations of 3 commonly used herbicides (the amine salt of 2,4-dichlorophenoxyacetic acid, trifluralin and triallate) were evaluated for effects on immune function in male Fisher 344 rats. The herbicides were prepared in an olive oil vehicle and administered by oral gavage twice weekly for 28 d at the following doses: 10.0 mg 2,4-D/kg; 17.5 mg trifluralin/kg; 5.0 mg triallate/kg/treatment. Normal body weight and organ/body weight ratios indicated the rats tolerated the herbicide treatments without difficulty. Exposure to 2,4-D did not alter lymphocyte blastogenesis, 1 gm antibody production (anti-sheep red blood cell), lymphocyte cell surface marker expression or phagocytic function of peritoneal macrophages. Trifluralin acted as a weak mitogen, but impaired T-lymphocyte blastogenesis induced by phytohemagglutinin and concanavalin A. Other immunological measurements were unaffected by trifluralin exposure. Triallate exposure reduced peritoneal macrophage phagocytosis by 33%, showed weak mitogenic properties and impaired T-lymphocyte blastogenesis in the presence of phytohemagglutin. Triallate also increased the anti-sheep red blood cell response expressed/spleen by 43%, a phenomenon suggestive of a compensatory response to minimize the impact on overall immune function. The changes in lymphocyte or macrophage function due to the herbicide treatments were not associated with changes in lymphocyte cell surface antigen expression.
