Rats were treated intraperitoneally with the gold-containing compounds sodium aurothiomalate (Myocrisin), sodium aurothiosulfate (Sanocrysin), and aurothioglucose. Using stem energy dispersive X-ray analysis, gold and silver were shown to be located at the same point in lysosomes of proximal tubular cells of the kidney, in hepatocytes and in macrophages of lymph glands, spleen and liver. This result indicates that, after exposure to ultraviolet radiation, chemically bound tissue gold is transformed to metallic gold that subsequently can catalyze the reduction of silver ions to silver when subjected to physical development, i.e. exposed to a photographic developer containing silver ions in addition to the reducing molecules.
The pharmacokinetics of gold sodium thiomalate (GST) and triethylphosphine gold (auranofin; AF) are different. Gold sodium thiomalate (GST) is completely bioavailable while only 15-25% of auranofin (AF) is absorbed. Protein binding of AF occurs to a larger extent to macroglobulins than does GST and total body retention of GST is much greater than AF at six months (30% versus approximately 1%). While terminal serum half-lives are approximately equal, total body half-lives are 250 days for GST and 69 days for AF. In addition, excretory pathways contrast markedly, with 85% of AF appearing in the feces while only 30% of GST is excreted by this route; 15% of AF gold appears in the urine and approximately 70% of GST gold is excreted via this route. With all the above differences one would expect that organ and cellular distribution of these compounds would differ. While gold from both drugs is concentrated in kidney, the percent of the dose found in the kidneys is less for AF than GST, at least in animals (0.4% vs 4.8%). Minute quantities are found in other organs but more study is needed to more clearly define organ distribution of these gold compounds, particularly in man.
Nitrofluoren-9-one and nitrofluoren-9-dicyanomethylene electron acceptors 7, 8, and 11 functionalized with a terminal thioctic acid unit have been synthesized from 2,4,5,7-tetranitrofluorenone. The self-assembled monolayers (SAMs) of these compounds on gold, formed via gold-sulfur interaction, have been fully characterized by electrochemical, FTIR, ellipsometry, and contact angle measurements. Cyclic voltammetry of SAMs reveals two reversible single-electron reduction waves for fluorenone derivatives 7a,b and 11, and three single-electron reductions for the dicyanomethylene-fluorene 8b, providing the first observation of a radical trianion species in SAMs. The tendency of the thioctic anchor to form multilayers via disulfide links is noted.
Well-ordered self-assembled monolayers (SAMs) were prepared on gold from helical peptides carrying a ferrocene (Fc) moiety at the N- or C-terminal end, and long-range electron transfer (ET) from Fc to gold was investigated. Electrochemical studies revealed that an inelastic hopping mechanism dominated over the superexchange mechanism in the ET reactions in the present SAMs and the dipole moment of the helix accelerated the ET reactions probably due to the lowering of the barrier height between the gold surface and peptide layer.
The rate of electron transfer is measured to two ferrocene and one iron tetraphenylporphyrin redox species coupled through terminal acetylenes to azide-terminated thiol monolayers by the Cu(I)-catalyzed azide-alkyne cycloaddition (a Sharpless "click" reaction) to form the 1,2,3-triazole linkage. The high yield, chemoselectivity, convenience, and broad applicability of this triazole formation reaction make such a modular assembly strategy very attractive. Electron-transfer rate constants from greater than 60,000 to 1 s(-1) are obtained by varying the length and conjugation of the electron-transfer bridge and by varying the surrounding diluent thiols in the monolayer. Triazole and the triazole carbonyl linkages provide similar electronic coupling for electron transfer as esters. The ability to vary the rate of electron transfer to many different redox species over many orders of magnitude by using modular coupling chemistry provides a convenient way to study and control the delivery of electrons to multielectron redox catalysts and similar interfacial systems that require controlled delivery of electrons.

The carbohydrate compounds of the mucus of flask cells in the kidney of claw-frogs (Xenopus laevis) were studied by gold marked lectins (WGA, RCA, L, LCA, HPA, PNA). We used a post-embedding technique. Seminthin or ultrathin sections of Lowieryl K H M-embedded kidney tissue were incubated. For light microscopy, a gold-silver technique was used. The mucus of the flask cells reacted strongly with WGA, RCA L and HPA, whereas LCA and PNA showed no binding. The Golgi apparatus and small cytoplasmatic vesicles reacted also positively with WGA, RCA L and HPA. The autoradiographically detected secretion routes of the glycosaminoglycan-rich secretion of flask cells are also demonstrable by lectins.
A sensitive method for the determination of gold-based drugs auranofin, myochrysine, and their metabolites has been developed. These gold-containing compounds were separated by reversed-phase ion-pair chromatography with tetrabutylammonium chloride as the ion-pairing agent. Gold-specific on-line detection utilized inductively coupled plasma mass spectrometry (ICP-MS). The separation conditions of pH, methanol content, concentration of the ion-pairing agent and ionic strength have been investigated. The detection limit for auranofin, the last peak in the chromatogram, was 0.3 ng. These methods were applied to the analysis of gold-containing species in urine from arthritis patients on auranofin, myochrysine or solganol therapy. The recovery of the total gold-containing species from urine was greater than 90%. Dicyanogold(I) anion, [Au(CN)2]-, was detected in the urine of several patients.
The emission of CdSe quantum dots linked to the 5'-end of a DNA sequence is efficiently quenched by hybridisation with a complementary DNA strand with a gold nanoparticle attached at the 3'-end; contact of the quantum dot and gold nanoparticle occurs.
