11C-labelling of dimethylphenethylamine in two different positions and biodistribution studies.

Dimethylphenethylamine (DMPA), a substrate for the B-form of the monoamine oxidase enzyme (MAO, EC 1.4.3.4), was labelled with 11C in two different positions; in the methyl group (M-DMPA) and in the phenethyl group (P-DMPA). M-DMPA was prepared by N-alkylation of methylphenethylamine with [11C]methyl iodide and P-DMPA was prepared by N-alkylation of dimethylamine with [1-11C]phenethyl iodide. The radiochemical yields were 30-35% (M-DMPA) and 10% (P-DMPA), based on [11C]carbon dioxide, with overall synthesis times of 30-35 min (M-DMPA) and 50 min (P-DMPA). The compounds were isolated by semi-preparative HPLC and the radiochemical purity was in both cases greater than 99%. The biodistributions of 11C-labelled M-DMPA [correction of M-DPMA] and P-DMPA were studied in rat brain by dissection and in the brain of a Rhesus monkey by positron emission tomography (PET).

High resolution proton magnetic resonance spectroscopy of cyst fluids from patients with polycystic kidney disease.

High field 1H-NMR spectra of fluid collected from the cysts of six renal transplant recipients with autosomal dominant polycystic kidney disease (ADPKD) have been measured and the major metabolite signals assigned. Quantitative NMR measurements have revealed a combination of unusual biochemical features of the cystic fluids that shows them to be distinct from both blood plasma and urine. Isoleucine, lysine, threonine and valine were present at mM concentrations, in cyst fluid and in some cases levels up to 2 orders of magnitude higher than normal plasma or urine were recorded. Mean glucose concentrations in the cyst fluids ranged from 3.4-9.6 mM and a number of organic acids and bases, including acetate, lactate, succinate, creatinine and dimethylamine were also present at high concentration and in different ratios to those found in either plasma or urine. The majority of cyst fluids examined also contained significant quantities of glycoproteins with characteristic 1H-NMR signals from N-acetyl groups of amino-sugar and sialic acid side chains which had a high degree of molecular mobility (as indicated by their relatively long T2 relaxation times, greater than 120 ms). High levels of ethanol (0.5-12.6 mM/l) were found in all fluid samples from the six transplanted patients (confirmed by conventional analysis). In general there was little variation in the 1H-NMR spectral patterns of either the intra- or interpatient cyst fluids, although the contribution of the protein macromolecules to individual spectra was lower in a few cysts. This constancy of biochemical composition probably reflects the chronic nature of the accumulation of cyst fluid and a long turnover of the cystic fluid components which has the effect of averaging composition. These findings suggest that the dynamic composition of cyst fluid from ADPKD patients is unique among the other body fluids and that the unusual composition may be related to epithelial polarity reversal of the cystic epithelium which could also contribute to the growth of the cysts.

A microbial biosensor for trimethylamine using Pseudomonas aminovorans cells.

A biosensor system based on the difference in the oxygen uptake response of two microbial electrodes was developed to monitor trimethylamine (TMA). The first electrode, constructed using Pseudomonas aminovorans grown on TMA, was sensitive to TMA, trimethylamine N-oxide (TMAO), dimethylamine (DMA) and monomethylamine (MMA). The second electrode responding to TMAO, DMA and MMA was prepared using Ps. aminovorans grown on TMAO. The difference in oxygen uptake was linearly related to the TMA concentration in the range of 5-26 microM. The minimum detectable level was 2.6 microM and the relative standard deviation was determined to be 14% for 16 repeated analyses. When operated and stored at 30 degrees C, the response of the system was stable for only 2 days. However, when the biosensor system was operated at 30 degrees C but stored overnight at 4 degrees C, the system was stable up to 20 days. The biosensor system was applicable for the determination of TMA in fish tissue extracts and the results compared well with those determined by HPLC.

Permeation and inhibition of polycystin-L channel by monovalent organic cations.

Polycystin-L (PCL), homologous to polycystin-2 (71% similarity in protein sequence), is the third member of the polycystin family of proteins. Polycystin-1 and -2 are mutated in autosomal dominant polycystic kidney disease, but the physiological role of PCL has not been determined. PCL acts as a Ca-regulated non-selective cation channel permeable to mono- and divalent cations. To further understand the biophysical and pharmacological properties of PCL, we examined a series of organic cations for permeation and inhibition, using single-channel patch clamp and whole-cell two-microelectrode voltage clamp techniques in conjunction with Xenopus oocyte expression. We found that PCL is permeable to organic amines, methlyamine (MA, 3.8 A), dimethylamine (DMA, 4.6 A) and triethylamine (TriEA, 6 A), and to tetra-alkylammonium cation (TAA) tetra-methylammonium (TMA, 5.5-6.4 A). TAA compounds tetra-ethylammonium (TEA, 6.1-8.2 A) and tetra-propylammonium (TPA, 9.8 A) were impermeable through PCL and exhibited weak inhibition on PCL (IC50 values>13 mM). Larger TAA cations tetra-butylammonium (TBA, 11.6 A) and tetra-pentylammonium (TPeA, 13.2 A) were impermeable through PCL as well and showed strong inhibition (IC50 values of 2.7 mM and 1.3 microM, respectively). Inhibition by TBA was on decreasing the single-channel current amplitude and exhibited no effect on open probability (NPo) or mean open time (MOT), suggesting that it blocks the PCL permeation pathway. In contract, TEA, TPA and TPeA reduced NPo and MOT values but had no effect on the amplitude, suggesting their binding to a different site in PCL, which affects the channel gating. Taken together, our studies revealed that PCL is permeable to organic amines and TAA cation TMA, and that inhibition of PCL by large TAA cations exhibits two different mechanisms, presumably through binding either to the pore pathway to reduce permeant flux or to another site to regulate the channel gating. These data allow to estimate a channel pore size of approximately 7 A for PCL.

Synthesis of stavudine amino acid ester prodrugs with broad-spectrum chemotherapeutic properties for the effective treatment of HIV/AIDS.

A series of prodrugs of stavudine were synthesized in an effort to enhance spectrum of chemotherapeutic properties for the effective treatment of HIV/AIDS. The 5'-OH function of stavudine was esterified with ciprofloxacin, norfloxacin, isoniazide, pyrazinamide, piperazine and dimethylamine acetic acid. The anti-HIV-1 activity of the esters was determined in CEM cell line and stavudine ester bearing piperazine acetic acid was found to be the most potent compound with a selective index of >15,723. Stavudine prodrug bearing ciprofloxacin and norfloxacin acetic acid showed 100% inhibition against Mycobacterium tuberculosis H(37)Rv at 6.25 microg/mL. The prodrugs also exhibited antibacterial activity against 24 pathogenic bacteria. In vitro hydrolysis of the various esters in human plasma indicated that these agents were relatively stable toward plasma esterases with t(1/2) ranging from 20-240 min.

Synthesis and SAR studies of diarylpyrrole anticoccidial agents.

2-(4-Fluorophenyl)-3-(4-pyridinyl)-5-substituted pyrroles were prepared and evaluated as anticoccidial agents in both in vitro and in vivo assays. Among the compounds evaluated, the dimethylamine-substituted pyrrole 19a is the most potent inhibitor of Eimeria tenella PKG (cGMP-dependent protein kinase). Further SAR studies on the side chain of the 2-pyrrolidine nitrogen did not enhance in vivo anticoccidial activity.

Regiospecific one-pot synthesis of pyrimido[4,5-d]pyrimidine derivatives in the solid state under microwave irradiations.

Electron rich 6-[(dimethylamino)methylene]amino uracil 1, undergoes [4+2] cycloaddition reactions with various in situ generated glyoxylate imine and imine oxides 6 to provide novel pyrimido[4,5-d]pyrimidine derivatives of biological significance, after elimination of dimethylamine from the (1:1) cycloadducts and oxidative aromatisation. This procedure provides a convenient method for the direct synthesis of pyrimido[4,5-d]pyrimidines in excellent yields when carried out in the solid state and under microwave irradiations.

7-(aryl/heteroaryl-2-ylethynyl)-4-phenylamino-3-quinolinecarbonitriles as new Src kinase inhibitors: addition of water solubilizing groups.

New 4-phenylamino-3-quinolinecarbonitriles with a 7-ethynyl group substituted by a pyridine, phenyl or thiophene ring containing basic water solubilizing groups were prepared and evaluated as Src kinase inhibitors. Of these new analogs, potent activity was observed with compounds having a (2,4-dichloro-5-methoxyphenyl)amino group at C-4, a methoxy or ethoxy group at C-6, and a pyridyl group bearing a dimethylamine or N-methylpiperazine on the ethynyl group at C-7.

Synthesis of novel sigma-receptor ligands from methyl alpha-D-mannopyranoside.

For the first time a monosaccharide (methyl alpha-D-mannopyranoside) has been used as starting material for the synthesis of novel sigma-receptor ligands. The hept-3-ulopyranoside dimethyl ketals 14 and 15 were obtained from the nitrile 7 via two synthetic routes. After selective hydrolysis of the ketone dimethyl acetal, various amino substituents were introduced into position 3. High sigma(1)-receptor affinity and selectivity was attained with equatorially arranged amino substituents in position 3 and a dichlorophenylacetamide moiety in position 7. The anomeric mixture of dimethylamines 26alpha/beta displayed the highest sigma(1)-receptor affinity (K(i)=21nM) within this small series of test compounds.

Arsenic(V) removal from aqueous solutions using an anion exchanger derived from coconut coir pith and its recovery.

The performance of a new anion exchanger (AE) prepared from coconut coir pith (CP), for the removal of arsenic(V) [As(V)] from aqueous solutions was evaluated in this study. The adsorbent (CP-AE) carrying dimethylaminohydroxypropyl weak base functional group was synthesized by the reaction of CP with epichlorohydrin and dimethylamine followed by treatment of hydrochloric acid. IR spectroscopy results confirm the presence of -NH(+)(CH(3))(2)Cl(-) group in the adsorbent. XRD studies confirm the decrease of crystallinity in CP-AE compared to CP, and it favours the protrusion of the functional group into the aqueous medium. Batch experiments were conducted to examine the efficiency of the adsorbent on As(V) removal. Maximum removal of 99.2% was obtained for an initial concentration of 1 mgl(-1) As(V) at pH 7.0 and an adsorbent dose of 2 gl(-1). The kinetics of sorption of As(V) onto CP-AE was described using the pseudo-second-order model. The equilibrium isotherms were determined for different temperatures and the results were analysed using the Langmuir equation. The temperature dependence indicates an exothermic process. Utility of the adsorbent was tested by removing As(V) from simulated groundwater. Regeneration studies were performed using 0.1N HCl. Batch adsorption-desorption studies illustrate that CP-AE could be used to remove As(V) from ground water and other industrial effluents.

Determination of aliphatic amines using N-succinimidyl benzoate as a new derivatization reagent in gas chromatography combined with solid-phase microextraction.

A simple, selective and sensitive approach was developed for the quantitation of aliphatic amines in lake water applying a new reagent (N-succinimidyl benzoate, SIBA), synthesized in the laboratory of the authors. Derivatization of the n-C1-C6 aliphatic monoamines and dimethylamine in aqueous solution with SIBA was followed by headspace solid-phase microextraction (SPME). Derivatives were identified by gas chromatography-mass spectrometry and determined by gas chromatography-flame ionization detection. Both derivatization and SPME conditions have been optimized. Derivatizations were performed in borate buffer (pH 8.8), at 60 degrees C for 22 min. SPME was carried out from saturated sodium chloride solution, at 80 degrees C for 60 min, desorption at 250 degrees C for 2 min. Detection limit of derivatized amines proved to be 0.13-7.2 nmol/l, while recovery of amines from lake water samples, in the concentration range of 100-200 microg/l, varied from 94.1 to 102.7%.

An evaluation of solid phase microextraction for aliphatic amines using derivatization with 9-fluorenylmethyl chloroformate and liquid chromatography.

The reliability of SPME combined with a chemical reaction for the analysis of short-chain aliphatic amines by liquid chromatography has been investigated. Different options to couple SPME and derivatization have been tested and compared: (i) derivatization of the analytes in solution followed by the extraction of the derivatives, (ii) extraction of the analytes and subsequent derivatization by immersing the SPME fibre onto a solution of the reagent, and (iii) extraction/derivatization of the analytes using fibres previously coated with the reagent. Methylamine (MA), dimethylamine (DMA) and trimethylamine (TMA) have been selected as a model of primary, secondary and tertiary amines, respectively. The analytes have been derivatized with the fluorogenic reagent 9-fluorenylmethyl chloroformate (FMOC), and the fibre coating was Carbowax-templated resin (CW-TR). The employment of fibres coated with FMOC to extract and derivatize the analytes was the best option, as compared with the other approaches tested the sensitivity was considerably improved. On the basis of these studies, a new procedure for the determination of MA, DMA and TMA in water is presented. To demonstrate the utility of the proposed conditions data on linearity, accuracy, repeatability and sensitivity are given. Results of the determination of the amines in tap, river and waste water are also presented.

An investigation into the antimicrobial mechanisms of action of two contact lens biocides using electron microscopy.

Polyquaternium-1 (PQ-1) and myristamidopropyl dimethylamine (MAPD) are biocides used commercially in a contact lens disinfecting solutions. Electron microscopy was used to provide further evidence on the mechanism(s) of action of these agents against a wide range of ocular pathogens including bacteria, fungi and protozoa. Both PQ-1 and MAPD caused multiple forms of damage to the organisms tested, evidenced by structural alterations, blebbing, leakage and cell destruction. The extent of damage and the selectivity against specific type of microorganisms was consistent with the antimicrobial activity of these agents. Although electron microscopy is a powerful tool, it has its limitations when used to examine the mode of action of biocides. Indeed, there was no evidence of gross structural alteration to Acanthamoeba castellani or Aspergillus fumigatus following treatment.

Adsorption of fluorescently labeled protein residues on poly(ethylene-co-acrylic acid) films modified with affinity functionalities.

Poly(ethylene-co-acrylic acid) (EAA) films were reacted with glycine, 12-aminododecanoic acid, aspartic acid, 5-aminoisophthalic acid, ethanolamine, diethylamine, dimethylamine, N-isopropylamine, and dimethylaminoethyleneamine to prepare EAA films with negatively charged, non-charged, hydrophilic, and hydrophobic functionalities. Attenuated total reflectance Fourier transform infrared spectroscopy, differential scanning calorimetry, and contact angle measurements were used to characterize the modified EAA films. Analyses revealed that the films were modified on the surfaces and also in the bulk; therefore, bulk properties such as cohesive energy density were changed even though the surfaces remained hydrophobic. Adsorption studies were performed for two fluorescently labeled protein residues, dansyl-L-phenylalanine (dansyl-F) and dansyl-L-glutamine (dansyl-Q), from pH 7.4 buffer solutions. The adsorption results revealed that dimethylaminoethyleneamine functionality gave the highest uptake among the functionalities studied, and adsorption was more favorable for dansyl-F than dansyl-Q. Adsorption behavior is discussed in terms of hydrophobic-hydrophobic (dispersion) interactions and Coulombic interactions.

Synthesis and evaluation of ethylene glycol 3-diethylamino-propionate methacrylate as a polymerizable amine coinitiator for dental application.

OBJECTIVE: The primary objective of this study was to synthesize and characterize ethylene glycol 3-diethylamino-propionate methacrylate (EGDPM) as a polymerizable coinitiator to replace the commercial amine coinitiator. The 2,2-bis[4-(2-hydroxy-3-methacryloxypropoxy)phenyl]-propane (Bis-GMA) and triethylene glycol dimethylacrylate (TEGDMA) resin mixture was polymerized by camphorquinone/EGDPM initiator system under visible light irradiation. The mechanical properties, water sorption and solubility of cured samples were also evaluated. METHODS: EGDPM was synthesized via Michael-Addition reaction and characterized using FTIR and 1H NMR spectroscopy. Photopolymerization kinetics of the dental resin mixtures were monitored by real-time IR (RTIR). The mechanical properties of cured samples were recorded by dynamic mechanical analyzer (DMA). And the water sorption and solubility of cured samples were detected according to ISO 4049. RESULTS: Both the double bond conversion and the rate of polymerization of the resin mixtures increased as increasing the concentration of EGDPM but were lower than that of ethyl-4-dimethylaminobenzoate (EDMAB) and 2-(dimethylamine)ethyl methacrylate (DMEM) as a coinitiator at some concentration. When it served as diluent, the final double bond conversion was comparable to that of EDMAB, and the rate of polymerization was higher than that of DMEM. The modulus and T(g) of the cured samples were very close. Water sorption and solubility of the samples were almost the same except that of EGDPM as diluent. CONCLUSIONS: EGDPM was synthesized by Michael-Addition reaction. It could be used as a potential coinitiator but not suitable as diluent for dental composite.

Synthesis and antiproliferative activity of triazenoindazoles and triazenopyrazoles: a comparative study.

Several triazenoindazoles and triazenopyrazoles were prepared transforming the appropriate aminoindazoles and aminopyrazoles in the corresponding diazonium salts which were reacted with dimethylamine, diethylamine and pyrrolidine. All the triazenes were tested for their antiproliferative activity against K562, HL60, L1210 and MCF7 cell lines. The biological data showed that the benzocondensation plays a positive role on the antiproliferative activity. The (1)H-NMR spectra showed that the rotational barrier around the N(2)-N(3) bond in the triazene group can be influenced both by the position of this group in the indazole nucleus and by the substitution pattern in the benzene moiety.

Role of 5-HT1A and 5-HT1B receptors in the antinociceptive effect of tramadol.

Tramadol, (1RS,2RS)-2-[(dimethylamine)-methyl]-1-(3-methoxyphenyl)-cyclohexanol hydrochloride, is an atypical centrally acting analgesic agent with relatively weak opioid receptor affinity and which, like some antidepressants, is able to inhibit the reuptake of serotonin (5-hydroxytryptamine, 5-HT) in the raphe nucleus. We have previously demonstrated that pindolol, a beta-adrenoceptor blocker/5-hydroxytryptamine(1A/1B) receptor antagonist, enhanced tramadol antinociception and that the selective 5-HT1A agonist 8-Hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) reduced it. These effects were related to the negative feedback control that regulates raphe region neurones. The current study examines the ability of the selective antagonist at somatodendritic 5-HT1A receptors, N-[2-[4-(2-methoxyphenyl)-1-piperazinyl] ethyl]-N-(2-pyridinyl) cyclohexane carboxamide (WAY100635, 0.8 mg/kg), the selective antagonist at terminal 5-HT1B receptors, N-[3-(2-dimethylamino) ethoxy-4-methoxyphenyl]-2'-methyl-4'-(5-methyl-1,2,4-oxadiazol-3-yl)-(1,1'-biphenyl)-4-carboxamide (SB216641, 0.1-0.8 mg/kg) and the selective agonist at 5-HT1B receptors, 1,4-tDihydro-3-(1,2,3,6-tetrahydro-4-pyridinyl)-5H-pyrrolo[3,2-b] pyridin-5-one (CP93129, 0.2-0.4 mg/kg), to modify the antinociceptive effect of 4-64 mg/kg of tramadol in the hot plate test in mice. The results show that 0.8 mg/kg of WAY100635 enhanced antinociceptive effect of tramadol while neither agonism nor antagonism at the 5-HT1B receptor modifies it significantly at the doses tested. These results account for involvement of the somatodendritic 5-HT1A receptors in the analgesic effect of tramadol and support the supraspinal interaction of serotonin and the opioid system in the regulation of pain.

Synthesis and in vitro antileukemic activity of new 4-triazenopyrazole derivatives.

Several new 4-(3,3-dimethyltriazeno)-5-benzamidopyrazole derivatives were prepared by reacting 4-diazo-5-benzamidopyrazole derivatives with dimethylamine. The compounds were tested at 10 microM for their vitro antileukemic activity against K562 (Human chronic myelogenous leukemia) and Raji (human Burkitt limphoma ) cell lines. Dacarbazine and methotrexate were used for comparative purpose. The 3-methyl-4-(3,3-dimethyltriazeno)-5-(substituted benzamido)pyrazoles, bearing the pyrazole nucleus free at 1 position, resulted more active than the 1-(substituted phenyl)-3-methyl-4-(3,3-dimethyltriazeno)-5-benzamidopyrazoles. Dacarbazine at 10 microM showed no activity in the above tests. The observed difference among Dacarbazine and the active 4-triazenopyrazoles migth be explained admiting that these last compounds, differently by Dacarbazine, did not follow a mechanism of action based on the cytochrome P-450 induced demethylation. The most active compound 2d showed growth inhibition values of 97.8 and 99.4% against K562 and Raji cell lines respectively. Methotrexate inhibition values at 0.2 microM against the above cell lines were 86.7 and 75.1% respectively.
