Synthesis of three disaccharides for the preparation of immunogens bearing immunodeterminants known to occur on glycoproteins.

p-Nitrophenyl 2-acetamido-3,6-di-O-benzyl-2-deoxy-beta-D-glucopyranoside was condensed with 2,3,4,6-tetra-O-benzyl-alpha-D-galactopyranosyl bromide, the product deprotected, and the disaccharide glycoside converted into p-trifluoroacetamidophenyl 2-acetamido-2-deoxy-4-O-beta-D-galactopyranosyl-beta- D-glucopyranoside. p-Nitrophenyl 3-O-benzoyl-4,6-di-O-benzylidene-alpha-D-mannopyranoside was condensed with 3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-beta-D-glucopyranosyl bromide, and the product was deprotected, to yield p-nitrophenyl 2-O-(2-acetamido-2-deoxy-beta-D-glucopyranosyl)-alpha-D-mannopyranoside. p-Nitrophenyl 2-acetamido-3,4-di-O-benzoyl-2-deoxy-beta-D-glucopyranoside was condensed with 2,3,4-tri-O-benzyl-alpha-L-fucopyranosyl bromide, and, after reduction, trifluoroacetylation, and deprotection, p-trifluoroacetamidophenyl 2-acetamido-2-deoxy-6-O-alpha-L-fucopyranosyl-beta-D-glucopyranoside was obtained.

Electrosyntheses of disaccharides from phenyl or ethyl 1-thioglycosides.

Constant current electrolyses of the glycosyl donors phenyl and ethyl 2,3,4,6-tetra-O-benzyl-1-thio-beta-D-glucopyranoside in dry acetonitrile in the presence of various primary and secondary sugar alcohols, performed in an undivided cell, gave beta-linked disaccharide derivatives selectively in good yields. Phenyl 2,3,4,6-tetra-O-benzoyl-1-thio-beta-D-glucopyranoside gave the beta-glucosides exclusively in good to moderate yields.

A convenient synthesis of beta-D-galactosyl disaccharide derivatives using the beta-D-galactosidase from Bacillus circulans.

beta-D-Gal-(1-->4)-beta-D-GlcNAc-OC6H4NO2-p (p-nitrophenyl N-acetyl-beta-lactosaminide) and beta-D-Gal-(1-->6)-beta-D-GlcNAc-OC6H4NO2-p (p-nitrophenyl N-acetyl-beta-isolactosaminide) were regioselectively synthesized from lactose and p-nitrophenyl 2-acetamido-2-deoxy-glucopyranoside, employing transglycosylation by the beta-D-galactosidase from Bacillus circulans and by controlling the concentration of organic solvent in the reaction system. The (1-->4)-linked disaccharide was formed exclusively when the concentration of organic solvent was high, whereas the (1-->6)-linked isomer was produced with a low concentration. Further utilization of the transglycosylation by the enzyme led to the regioselective formation of beta-D-Gal-(1-->4)-D-GalNAc and beta-D-Gal-(1-->4)-beta-D-GalNAc-OC6H4NO2-p. With the enzyme, beta-D-galactosyl transfer occurred preferentially at the O-4 position of GlcNAc and GalNAc, regardless of the configuration of the hydroxyl group.

Selective release of the disaccharide 2-acetamido-2-deoxy-3-O-(beta-D-galactopyranosyl)-D-galactose from epiglycanin by endo-N-acetyl-alpha-D-galactosaminidase.

Epiglycanin, the major glycoprotein of TA3-Ha mammary carcinoma ascites cells, was radiolabeled with tritium in the terminal D-galactose and 2-acetamido-2-deoxy-D-galactose residues. Alkaline-borohydride treatment, reported to release five O-glycosyl-linked chain types from epiglycanin, resulted in the cleavage of 98-99% of the radioactivity from the protein. Of this, 63% of the radioactivity from epiglycanin and 70% from asialoepiglycanin co-migrated with an authentic sample of 2-acetamido-2-deoxy-3-O-(beta-D-galactopyranosyl)-D-galactitol on a column of Bio-Gel P-6. Incubation of [3H]galactose-epiglycanin with endo-N-acetyl -alpha-D-galactosaminidase (Diplococcus pneumoniae), and fractionation of the mixture on a column of Bio-Gel P-4, gave only one oligosaccharide peak containing 62 and 70%, respectively, of the radioactivity of epiglycanin and asialoepiglycanin. This oligosaccharide comigrated with authentic 2-acetamido-2-deoxy-3-O-(beta-D-galactopyranosyl)-D-galactose (1) on columns of Bio-Gel P-2 and P-4 and on paper chromatograms. Results of experiments in which unlabeled epiglycanin was treated with enzyme and the products analyzed, by three different methods, suggested that 78-85% of 1 had been cleaved. Another enzyme, N-acetyl-alpha-D-galactosaminyl-oligosaccharidase from Clostridium perfringens, exhibited similar specificity and cleaved 65% of the radioactivity from ([3H]galactose)asialoepiglycanin, which was eluted from a Bio-Gel P-2 column as the disaccharide 1.

Protected glycosides and disaccharides of 2-amino-2-deoxy-D-glucopyranose by ferric chloride-catalyzed coupling.

The ferric chloride-catalyzed glycosylation of hydroxy compounds by protected 2-acylamino-2-deoxy-beta-D-glucopyranose 1-acetates is described. In addition to known glycosides from the reaction of alcohols with 2-acetamido-1,3,4,6-tetra-O-acetyl-2-deoxy-beta-D-glucopyranose (3), ally (and other alkyl) beta-glycosides were obtained from the N-benzoyl, N-phenoxyacetyl, N-methoxyacetyl, N-chloroacetyl, and N-phthaloyl congeners of 3. The latter compounds, except for the N-phthaloyl derivative, gave oxazolines in the absence of an alcoholic reactant. Compound 3 and the related N-benzoyl, N-chloroacetyl, N-acetyl-3,4,6-tri-O-benzyl, and N-acetyl-4-O-acetyl-3,6-di-O-benzyl derivatives were coupled to one or more protected sugars to form protected, beta-linked disaccharides. Coupling at the 6-positions of acceptors proceeded smoothly and gave 67-80% yields. For successful coupling at positions 3 and 4, long reaction times and multiple additions of glycosyl donor were required, and yields ranged from 60% to as low as 30%. 1,3,4,6-Tetra-O-acetyl-2-(chloroacetamido)-2-deoxy-beta-D- glucopyranose appeared to be the most reactive glycosyl donor in this series. The reaction of 2-methyl-(3,4,6-tri-O-acetyl-1,2-dideoxy-alpha-D-glucopyrano)[2,1- d]-2-oxazoline (derived from 3) with allyl alcohol was catalyzed by ferric chloride, and oxazolines were detected as intermediates in some of the glycosylations of protected sugars.

Chemical Phenomena↗

Enzymic synthesis of HexNAc-containing disaccharide glycosides.

The following disaccharide glycosides were obtained from the appropriate donor and acceptor glycosides by employing glycosidases from the mollusc Chamelea gallina as catalysts: alpha-D-Galp-OMe (N-acetyl-alpha-D- galactosaminidase), beta-D-GalpNAc-(1----3)-beta-D-Galp-OMe, beta-D-GlcpNAc-(1----3)-beta-D-Galp-OMe, and beta-D-GlcpNAc-(1----6)-alpha-D-Manp-OMe (N-acetyl-beta-D-hexosaminidase). The regioselectivity of the N-acetyl-beta-D-hexosaminidase-catalysed reactions depended on the anomeric configuration of the acceptor. Thus, alpha-D-Galp-OMe gave beta-D-GlcpNAc- (1----6)-alpha-D-Galp-OMe almost exclusively, whereas beta-D-Galp-OMe gave beta-D-GlcpNAc-(1----3)-beta-D-Galp-OMe and beta-D-GlcpNAc-(1----6)-beta-D-Galp-OMe in almost equal amounts. The isolation of the products by chromatography was straightforward.

Preparation of disaccharides having a beta-D-mannopyranosyl group from N-phthaloyllactosamine derivatives by double or triple SN2 substitution.

Condensation of 1,2,3,4,6-penta-O-acetyl-beta-D-galactopyranose with methyl 3,6-di-O-benzyl-2-deoxy-2-phthalimido-beta-D-glucopyranoside, followed by alkaline methanolysis, gave a derivative of lactosamine that has an unsubstituted beta-D-galactopyranosyl group. Tributyltin oxide-mediated allylation gave a good yield of the 3'-O-allyl (9) and a poor yield of the 3',6'-di-O-allyl ether (8). Protection of 9 at O-6' was achieved by reductive opening of the 4',6'-O-anisylidene derivative, to give the 6'-O-(4-methoxybenzyl) ether 15. Conversion of 8 and 15 to their 2',4'-bis(trifluoromethanesulfonates), followed by SN2 reaction with benzoate, gave the corresponding beta-D-mannopyranosyl disaccharides. However, the model methyl 3-O-allyl-beta-D-galactopyranoside and 9 were converted into beta-D-mannopyranosyl derivatives in better yield (52-55%) by a one-pot, triple SN2 substitution of the tris(trifluoromethanesulfonates).

Immobilisation of beta-D-galactosidase from Escherichia coli on cellulose beads and its use for the synthesis of disaccharide derivatives.

beta-D-Galactosidase, isolated from cloned E. coli, was immobilised on cellulose beads via oxidation with sodium periodate, activation by cyanuric chloride, or diazotisation. beta-D-Galactosidase immobilised via azo bonds showed the highest relative activity and thermostability, and was used for synthesis of disaccharide methyl glycosides.

Synthesis of beta-D-GlcA-(1----3)-beta-D-Gal disaccharides with 4- and 6-sulfate groups and 4,6-disulfate groups.

The sodium salts of the 6-sulfate 7, the 4-sulfate 10, and the 4,6-disulfate 12 of benzyl 3-O-(beta-D-glucopyranosyl uronate)-beta-D-galactopyranoside (5) have been synthesized. Methyl (2,3,4-tri-O-acetyl-1-bromo-1-deoxy-alpha-d-glucopyran)uronate (1) was coupled with benzyl 2-O-benzoyl-4,6-O-benzylidene-beta-D-galactopyranoside (2) to yield 3. The benzylidene acetal of 3 was hydrolyzed to give benzyl 2-O-benzoyl-3-O-[methyl (2,3,4-tri-O-acetyl-beta-D-glucopyranosyl)uronate]-beta-D-galactopyra noside (4). Compound 4 was utilized as a key intermediate to prepare the sulfated disaccharides 7,10, and 12. Direct sulfation of 4 with sulfur trioxide-trimethylamine for 2 days yielded the 6-sulfate 6. The 4,6-disulfate 11 was accessible by running the reaction under the same conditions for 14 days. The 4-sulfate 9 was obtained after protecting the 6-OH group of 4 by reaction with benzoyl imidazole to give the 6-benzoate 8, followed by sulfation under vigorous conditions. Treatment of the protected compounds 4, 6, 9, and 11 with aqueous sodium hydroxide in tetrahydrofuran gave the unprotected 5, 7, 10, and 12, respectively.

E.s.r. study of the alkaline degradation of disaccharides in methyl sulfoxide.

The degradation of a series of deoxy sugars and disaccharides in methyl sulfoxide-tetrabutylammonium hydroxide has been studied by e.s.r. spectroscopy. The generation of radicals and the types of radical formed are dependent on the anomeric configuration of non-reducing moieties and on the positions of the linkages and deoxy groups. Mechanisms for the pathways of degradation are proposed.

The synthesis is reported of methyl 4,4'-dithio-alpha-maltotrioside (12) and the spacer-modified disaccharide glycosides methyl 4-S-(4-alpha-D-glucopyranosylthio-2-hydroxybutyl)-4-thio-alpha -D-glucopyranoside (20) and methyl 4-S-[(1,5/4,6)- and (4,6/1,5)-4-alpha-D-glucopyranosylthio-5,6-dihydroxy-2- cyclohexen-1-yl]-4-thio-alpha-D-glucopyranoside (29a/b), which are analogues of methyl alpha-maltotrioside. The Ki values for alpha-amylase for these compounds were determined as were those of methyl alpha-maltotrioside and maltose.

NMR studies of some (1-->6)-linked disaccharide methyl glycosides.

NMR studies have been performed on the methyl glycosides of some (1-->6)-linked disaccharides. Observed J5,6pro-R and J5,6pro-S values indicate that, for the 6-substituted D-gluco- and D-galacto-pyranosides, the rotamer distribution around the C-5-C-6 bond deviates somewhat from that observed for the respective unsubstituted monosaccharide glycosides. There is also a difference between 6-O-alpha-D- or 6-O-beta-L- on the one hand and 6-O-beta-D- or 6-O-alpha-L-substituted glycosides on the other, with somewhat larger values for J5,6pro-R for the latter two indicating a higher proportion of the gauche-trans conformer. The glycosylation shifts observed for the signals from the 6-protons in the glycosidic linkage were dependent on the type of anomeric and absolute configuration of the glycosyl group. NOE measurements by irradiation of the anomeric proton indicated that sugars 6-substituted with alpha-D- or beta-L-glycosyl groups have highly populated conformations in which H-1 and H-6pro-S are proximal, and for beta-D- and alpha-L-glycosyl groups conformations in which H-1 and H-6pro-R are proximal.

Synthesis of O-[2-acetamido-2-deoxy-6-O-stearoyl- and -6-O-(2-tetradecylhexadecanoyl)-beta-D-glucopyranosyl]-(1-->4)-N- acetylnormuramoyl-L-alpha-aminobutanoyl-D-isoglutamine, lipophilic disaccharide analogues of MDP.

Silver triflate-promoted condensation of 3,4,6-tri-O-acetyl-2-deoxy-2- phthalimido-beta-D-glucopyranosyl bromide (1) with benzyl 2-acetamido-6-O-benzyl-2-deoxy-3-O- (methoxycarbonyl)methyl-alpha-D-glucopyranoside (4) afforded the key compound, benzyl 2-acetamido-6-O-benzyl-2-deoxy-3-O- (methoxy-carbonyl)methyl-4-O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-b eta-D- glucopyranosyl)-alpha-D-glucopyranoside (5), which after deprotection was transformed into acid 10. Condensation of 10 with the benzyl ester of L-alpha-aminobutanoyl-D-isoglutamine and deisopropylidenation of the product 11 afforded the benzyl ester of N-(2-O-[benzyl 2-acetamido-4-O-(2-acetamido-3-O-benzyloxymethyl-2- deoxy-beta-D-glucopyranosyl)-6-O-benzyl-2,3-dideoxy-alpha-D-glucopyra nosid-3- yl]glycoloyl)-L-alpha-aminobutanoyl-D-isoglutamine (12). Partial O-acylation of 12 and hydrogenolysis of protecting groups gave the 6-O-stearoyl- and 6-O-(2-tetradecylhexadecanoyl)-disaccharide-dipeptides 17 and 18, respectively. Pyrogenicity and adjuvant activity in cell-mediated immunity are reported.

An NMR spectroscopic and conformational study of 12 pseudo-disaccharides (D-glucopyranosyl-5a-carba-D- and -L-glucopyranoses).

NMR spectroscopic data for 12 pseudo-disaccharides of the general structure: (alpha or beta)-D-glucopyranosyl-(1-->chi)-5a-carba-(D or L)-glucopyranose, representing analogues of laminaribiose (beta-D-Glc p, chi = 3), cellobiose (beta-D-Glc p, chi = 4), and maltose (alpha-D-Glc p, chi = 4) are presented. The assigned NMR chemical shifts together with NOE difference measurements in association with calculations applying the HSEA force field combined with Monte Carlo simulations have been used to assess the conformational preferences of the investigated compounds. The results are correlated with general structural features involved in the interactions between monosaccharide units of oligosaccharides.

Synthesis of specifically deoxygenated disaccharide derivatives of the Shigella dysenteriae type 1 O-antigen.

The synthesis of methyl O-alpha-L-rhamnopyranosyl-(1-->2)-alpha-D-galactopyranosides specifically deoxygenated at position 2 (31), or 4 (21) of the rhamnopyranosyl residue was accomplished using methyl 3,4,6-tri-O-benzoyl-alpha-D-galactopyranoside (18) as the glycosyl acceptor. Phenyl thionocarbonate activation of the penta-O-benzoylated disaccharide precursor followed by Barton reduction and Zemplén transesterification gave 31, while 21 was obtained via condensation of the deoxygenated monosaccharide donor with 18, and subsequent debenzoylation of the product.

Synthesis of 1D-(1,3,5/2,4)-4-acetamido-5-amino-1,2,3-cyclohexanetriol and its incorporation into a pseudo-disaccharide.

The synthesis of the title compound and 1D-(1,3,5/2,4)-4-acetamido-5-amino-3-O-(beta-D-glucopyranosyluronic acid)- 1,2,3-cyclohexanetriol [sequence: see text] is described. Starting from methyl 2-acetamido-2-deoxy-alpha-D-glucopyranoside 2L-(2,4,5/3)-4-acetamido-3-benzoyloxy-2-benzyloxy-5- hydroxycyclohexanone [sequence: see text] was prepared via a series of transformations including the regioselective ring opening of the benzylidene acetal and the mercury(II)-catalyzed carbocyclic ring closure reaction of 5-enopyranoside. Stereoselective reduction of ketone 11 with NaBH(OAc)3 gave 1D-(1,2,4/3,5)-2-acetamido-3-O-benzoyl-4-O-benzyl-1,3,4,5- cyclohexanetetrol [sequence: see text] (88%), which was then converted into 1D-(1,3,5/2,4)-4-acetamido-5-azido-3-O-benzoyl-2-O- benzyl-1-O-pivaloyl-1,2,3-cyclohexanetriol [sequence: see text] through selective 5-OH protection, 1-O-mesylation, and subsequent azide displacement. Saponification and hydrogenation of this gave the title compound. Selective O-debenzoylation with 1.1 equiv of K2CO3 in MeOH gave 1D-(1,3,5/2,4)-4-acetamido-5-azido-2-O-benzyl-1-O- pivaloyl-1,2,3-cyclohexanetriol [sequence: see text] (73%). Glycosylation of this compound with methyl (2,3,4-tri-O-acetyl-alpha-D-glucopyranosyl bromide) uronate in Ch2Cl2, using silver triflate as the promoter, afforded 1D-(1,3,5/2,4)-4-acetamido-5-azido-2-O-benzyl-3-O-(methyl 2,3,4-tri-O-acetyl-beta-D-glucopyranosyluronate)-1-O- pivaloyl-1,2,3-cyclohexanetriol [sequence: see text] and subsequent hydrogenation of this compound gave the basic pseudo-disaccharide.
