In situ photoactivation of a caged phosphotyrosine peptide derived from focal adhesion kinase temporarily halts lamellar extension of single migrating tumor cells.

Focal adhesion kinase (FAK), a non-receptor tyrosine kinase, mediates integrin-based cell signaling by transferring signals regulating cell migration, adhesion, and survival from the extracellular matrix to the cytoplasm. Following autophosphorylation at tyrosine 397, FAK binds the Src homology 2 domains of Src and phosphoinositide 3-kinase, among several other possible binding partners. To further investigate the role of phosphorylated FAK in cell migration in situ, peptides comprising residues 391-406 of human FAK with caged phosphotyrosine 397 were synthesized. Although the caged phosphopeptides were stable to phosphatase activity, the free phosphopeptides showed a half-life of approximately 10-15 min in cell lysates. Migrating NBT-II cells (a rat bladder tumor cell line) were microinjected with the caged FAK peptide and locally photoactivated using a focused laser beam. The photoactivation of caged FAK peptide in 8-microm diameter spots over the cell body led to the temporary arrest of the leading edge migration within approximately 1 min of irradiation. In contrast, cell body migration was not inhibited. Microinjection of a non-caged phosphorylated tyrosine 397 FAK peptide into migrating NBT-II cells also led to lamellar arrest; however, this approach lacks the temporal control afforded by the caged phosphopeptides. Photoactivation of related phosphotyrosine peptides with altered sequences did not result in transient lamellar arrest. We hypothesize that the phosphorylated FAK peptide competes with the endogenous FAK for binding to FAK effectors including, but not limited to, Src and phosphoinositide 3-kinase, causing spatiotemporal misregulation and subsequent lamellar arrest.

Migration from India to Australia.

"The article examines the contemporary trends and future prospects of migration from India to Australia. The focus is on Indian Settlers and Temporary Entrants admitted to Australia for employment and Indian students admitted to Australia for higher studies. The volume of emigration for permanent residence during the early 1990s has made India one of the leading source countries of migration to Australia. A majority of Indians admitted as Settlers every year join the labor force. Recent data indicate that, among Indian Settlers, there is a preponderance of unsponsored Independent Skilled Migrants. Given the anticipated growth in the number of Indian students, the coming years are likely to witness a spurt in Skilled Temporary Workers from India."

Peasants in reserve: temporary West Indian labor in the U.S. farm labor market.

In the past 10 years, the British West Indies Temporary Alien Labor Program has received widespread judicial and legislative support and criticism. While sugar and apple producers who import West Indians argue that domestic labor is insufficient to harvest their crops, labor organizations and their supporters maintain that domestic labor is adequate. The resulting labor disputes focus primarily on the issue of whether or not West Indians are displacing US workers or undermining wage rates and working conditions. This article examines the relationships among legal issues surrounding the program, the US farm labor market, and the Jamaican peasantry. It argues that continued imports of foreign labor during times of high domestic unemployment, as well as the varied factors which underlie the continued willingness and ability of Jamaican peasant households to supply workers to US producers, can be most clearly understood from an international and historical perspective, rather than focussing on the needs and problems of any 1 nation.

Long-term immigration to the United States: new approaches to measurement.

"The United Nations has recommended the measurement of types of international migration using demographic criteria, including length of stay and purpose of travel. Information systems at the U.S. Immigration and Naturalization Service (INS) have the potential to provide a basis for documenting these demographic characteristics, in particular, length of stay of temporary migrants to the United States. This article analyzes these characteristics of selected categories of nonimmigrant aliens. The results of the analysis are used to produce series of estimates of alien immigration that conform more closely to the U.N. recommended definitions and better represent demographic concepts of long-term immigration. A strategy for measuring emigration of aliens from the United States using INS information systems is also described."

Labour circulation and the urban labour process.

The author investigates aspects of labor circulation, which he defines as "temporary movement between geographical areas for work or in search of work....[He attempts to determine] what roles have been played by labour circulation in the development of urban-industrial labour forces in the transition to industrial capitalism." Factors considered include the exploitation and oppression of labor migrants; the industrial-urban labor reserve; urban socioeconomic stratification and discrimination by age, sex, or race; the division of labor; and policy options.

Family formation and marital selectivity among Colombian and Dominican immigrants in New York City.

"Utilizing data from a 1981 survey of Dominican and Colombian immigrants to New York City, and from 1975 marriage certificates for the entire city, this article describes the extent of family formation in the U.S. and patterns of marital selectivity of recent Hispanic immigrants residing in New York City. A core goal of the analysis is the provision of indicators of the extent of and nature of integration processes at an early stage of the immigration." It is found first that "Hispanic immigrants in New York City are clearly long-term settlers, although not necessarily permanent ones. They are forming nuclear families in this country to a considerably greater extent than they are either transferring families from the origin country or residing as unattached temporary migrants. Second, a significant minority of marriages contracted in this country are with spouses of different national origin groups, indicating an openness in ethnic boundaries in the host society context."

Canine intestinal myoelectric activity after open versus laparoscopically assisted right hemicolectomy.

BACKGROUND: It is a common belief that a laparoscopic procedure results in a shorter duration of postoperative ileus compared with the equivalent open procedure. This study was undertaken to determine whether laparoscopically assisted right hemicolectomy in the dog results in a shorter duration of ileus compared with open right hemicolectomy. METHODS: Eight bipolar serosal electrodes (4 on the small bowel, 4 on the left colon) were implanted in each dog (n = 10). Three weeks after electrode implantation baseline recording was made for 5 days; then 5 dogs underwent laparoscopically assisted right hemicolectomy and 5 underwent open right hemicolectomy. Myoelectric activity was recorded continuously for 72 hours postoperatively. Tracings were analyzed for the time of reappearance, duration, migration velocity, and cycle length of phase 2, phase 3, and the migrating colonic complex. The criteria used for the resolution of postoperative ileus were the return of phase 2, phase 3, and the migrating colonic complex. RESULTS: All dogs had temporary loss of organized myoelectric activity postoperatively. The mean reappearance time (minutes +/- standard deviation) for phase 3 was 857 +/- 574 versus 761 +/- 600; the phase 2 reappearance time was 1,845 +/- 610 versus 1,590 +/- 668; and the migrating colonic complex reappearance time was 534 +/- 365 versus 572 +/- 552, open versus laparoscopically assisted right hemicolectomy, respectively. The times were not different (Wilcoxon rank sum test, P > 0.05). The time required for phase 3, phase 2, and the migrating colonic complex to attain preoperative configuration also was not different between the open and laparoscopically assisted group. CONCLUSION: Myoelectric resolution of postoperative ileus did not occur earlier in the dog undergoing laparoscopically assisted right hemicolectomy compared to the dog undergoing open right hemicolectomy. This data does not support the hypothesis that a laparoscopically assisted colectomy results in a shorter duration of postoperative ileus than the equivalent open procedure.

Gender, migration, risky sex, and HIV infection in China.

Gender differences in sexual behavior as a consequence of migration have been ignored in both the migration and the HIV literature in China. This study examines differences among temporary migrants in terms of sexual behavior and factors that make female migrants more vulnerable to the risk of acquiring HIV infection. Results suggest that the interplay of migration and gender renders female temporary migrants particularly vulnerable to engaging in casual and commercial sex. Although male temporary migrants do not differ from male nonmigrants in prevalence of casual and commercial sex, the prevalence rates of casual and commercial sex for female temporary migrants are found to be 14 and 80 times those for female nonmigrants, respectively. Female temporary migrants' higher unemployment rate and concentration in the service and entertainment sectors are keys to understanding differences in the prevalence of casual and commercial sex among temporary migrants according to sex. Policy measures to promote female temporary migrants' equal access to employment are urgently needed to improve their economic well-being and to reduce their risky sexual behavior.

Suppression of expulsion of Aspiculuris tetraptera in hydrocortisone and methotrexate treated mice.

Hydrocortisone treated male and female mice, given a primary infection with Aspiculuris tetraptera, did not reject the worms during the third week of infection. Mice given hydrocortisone during the first week of infection had elevated worm burdens on day 10, suggesting that some worm loss was encountered during the anterior migration in control mice. Furthermore, this temporary period of treatment was sufficient totally to suppress rejection and to allow the parasite to persist until day 28. Methotrexate also significantly delayed rejection, but larval growth was retarded in treated mice. These results, it is suggested, add strength to the hypothesis that the loss of A. tetraptera in a primary infection in mice, is an immunological phenomenon.

Multinucleation of skeletal muscle in vitro.

Healthy, mature, spontaneously contracting muscle was cultivated from explants of 13-day chick embryos for periods up to 4 months in the multipurpose chamber (Rose, 1954) using cellophane-strip technique (Rose et al., 1958) with silicone gaskets, Eagle's medium including 10 per cent horse serum reinforced with 300 mg-per cent of glucose, and the teased type of explant. This method provided optically ideal conditions for the study of muscle fibers with oil immersion, phase contrast time-lapse cinematography at 1 frame per minute without apparent damage for periods as long as 10 days. In no case was mitosis, amitosis, or nuclear "budding" observed in the course of muscle development. Multinuclear muscle fibers have been shown with cine technique to result from both myoblast fusion and polar extension of preformed (explanted) muscle tissue. Myoblast fusion was the only demonstrable way of giving rise to multinucleation. Nuclear membrane "wrinkling" was shown to be merely a temporary distortion that occurred during nuclear migration and rotation. It is suggested that this phenomenon may be responsible for numerous reports of amitosis in the genesis of muscle fibers. The histological development of new straps resulted from an orderly sequence of events. Included in these were polar extension, nuclear migration, rotation, and fixation. Following these events there was increased mitochondrial activity, myofibril formation, and cross-banding. Spontaneous contractions were seen throughout the entire course of differentiation in vitro but became more regular and stronger in the later stages.

Early technical and clinical results with retrievable inferior vena caval filters.

Deep venous thrombosis with or without subsequent pulmonary embolism is a common preventable cause of hospital death. Although anticoagulation is the accepted standard therapy for thromboembolic disease, in situations in which anticoagulation is contraindicated, interruption of the inferior vena cava (IVC) by means of percutaneous placement of a filter has become a widely used alternative. We report our initial experience with two retrievable IVC filters. Between July 2002 and April 2003, 13 patients (mean age 54 +/- 7 years; range 29-75 years) underwent percutaneous placement of either the Gunther Tulip (n = 5; Cook Inc., Bloomington, IN) or OptEase (n = 8; Cordis, Miami Lakes, FL) IVC filter. Five patients had filters placed prophylactically before major surgery. The remaining eight patients had had a contraindication to anticoagulation, and three had experienced a hemorrhagic complication as a result of anticoagulation following either a recently documented deep venous thrombosis (n = 3) or pulmonary embolism (n = 5). Filters were successfully placed in all 13 patients, with a duration of implantation ranging from 2 to 15 days. Retrieval was attempted in 12 patients (in 1 patient, permanent filtration was secondarily requested) and was achieved in 10 (84.6%) patients. In 2 patients, retrieval failure was due to device angulation within the vena cava precluding safe retrieval. In both instances, the device used was the Gunther Tulip filter. No patient developed symptomatic pulmonary embolism or insertion-site thrombosis following either filter deployment or removal. Trapped thrombus in the filters was seen in all patients. Retrieval required a mean of 6.8 minutes (range 5-10.2 minutes) of fluoroscopy time. Neither filter migration nor caval injury was observed. Temporary IVC filters are effective and are associated with a high retrieval success rate. Further study is warranted to determine the maximal duration of implantation and whether retrievable IVC filters should expand the indications for IVC filter placement.

[Immunomorphological study of time course of repair of corneal deep burn defects after stromal equivalent transplantation].

Early defects due to the afflux of monocytic phagocytes into the inflammatory focus, rapid resorption of detritis and normalisation of correlation between contents of matrix proteinases and their inhibitors, stimulation of stromal cornea keratinocytes migration and their proliferation characterize repair of postburn defects in transplantation of the stromal equivalent. Repair against the background of stromal equivalent transplantation is of a histotypical character: collagen gel organizes and serves the basis for migration of the cornea fibroblasts and temporary matrix for growing new epithelium, while restored basal membrane is a derivative of cornea epithelium.

Migrant-nonmigrant fertility differentials in Anhui, China.

"This analysis is designed to provide insights into the relation between migration and fertility in China. Survey data collected from Anhui Province in 1989 are used to evaluate whether fertility is higher or lower among the temporary migrants to urban places in comparison to permanent residents in those locations.... Our analysis will...assess the extent to which socioeconomic characteristics, family planning, and migration help to explain observed differences in the fertility of temporary migrants and permanent residents. In doing so, the basic question to be answered is whether temporary migrants have used their movement as a way to avoid official policies controlling fertility and, in so doing, have they contributed to a higher than average fertility in China?"

Occupational and spatial mobility of temporary Mexican migrants to the U.S.: a comparative analysis.

"U.S. job and spatial mobility are compared here for recent returnee migrants from two Mexican areas--Rio Grande, Zacatecas, in the interior; and Nueva Rosita-Muzquiz, Coahuila, near the U.S. border. Results suggest that the interior migrants fit a hierarchical migrant model: they move up the urban hierarchy from U.S. rural areas to towns and cities, experiencing substantial job mobility at first, but little after reaching the urban sector. Border migrants fit a shuttle migrant model: they return to the same job and place year after year, experiencing little or no spatial and occupational mobility, although they tend to hold somewhat higher status jobs."

The determinants and consequences of female migration in Thailand.

Female migration in Thailand, especially from rural areas to the Bangkok Metropolis, is an important migratory phenomenon which leads to an unfavorable population distribution. Generally the migration level in Thailand is low but many women from the rural areas, especially from the Northeast Region, move to Bangkok Metropolis to seek work. Findings reveal that the majority of migrants are young, single women. Women migrants find it relatively easy to participate in the Bangkok labor market. Service work is the main source of employment. For the temporary migrants, it was found that those from the Northeast Region tend to return to their hometown in higher proportion than those from other regions. This was particularly so among young women migrants.

The effect of ischemia of the dog's colon on transmural migration of bacteria and endotoxin.

The purpose of this investigation was to evaluate the effect of temporary ischemia created during the surgical preparation of the colon for resection on transmural migration of bacteria and passage of endotoxin through the ischemic wall of the canine colon. Eighteen dogs were used: in fourteen the colon was devascularized by ligating all marginal vessels. Aerobic and anaerobic cultures and washings for endotoxin assay were obtained from the surface of the bowel at intervals up to 6 hr after creating the ischemia. Peripheral and portal blood samples were also obtained at equal intervals for bacteriologic cultures and endotoxin assay. Bacterial transmural migration was examined in 10 dogs. In 6 of the dogs biologically marked bacteria were introduced into the colon via a rectal catheter before producing the ischemia. In 4 dogs radioactively labeled endotoxin was introduced into the colon in a similar fashion. In all the dogs, surface cultures, both routine and specific for the marked bacteria, were negative for the whole period of up to 6 hr after creation of ischemia. All portal and peripheral vein cultures were also negative. Transmural migration of endotoxin was investigated in 8 dogs, 4 of which served as controls and underwent a sham operation. In the 4 dogs in which the colon was devascularized endotoxin was discovered in peritoneal washings, and in portal and systemic blood samples, as early as 30 min after the preparation of the bowel was completed. In the control dogs endotoxin assays were negative throughout the experiment. In this model up to 6 hr of ischemia did not result in migration of bacteria through the ischemic wall. Endotoxin, however, entered the peritoneum and the blood very soon after producing the ischemia.
