[Continuous interpretation system for the cytophotometer "Amplival photometry"=-studies with pesticide combinations].

By means of complementary technique the cytophotometer "Amplival photonetrie" is converted in a continuous interpretation system. Thereby it is possible to move the slides by controlling step motors in alpha- and/or gamma-axis. The number of stepts and the speed of them can be predetermined. For the quantitative histochemical examination the sum of the extinctions of single steps is determined. therefore the arythmetical average can be calculated per arbitrary unit tissue. In examinations with combinations of pesticides the usefulness of the interpretation system is demonstrated.

[Anthralin erythema: effect of concentration, contact time, oxidation products and corticosteroids. Clinical, reflection photometry and electron microscopy studies].

A comparative study on psoriatic patients and healthy volunteers demonstrated that short-term treatment with Anthralin shows significantly less side-effects on healthy skin than long-term therapy. We could also prove that the oxidation products Chrysazin and Bianthron do not cause any irritations, and that a combined therapy with Anthralin and corticoids does not reduce the development of erythema.

[Single cell photometry analysis of nuclear DNA in treated undifferentiated prostate cancer and its clinical significance].

Based on 67 specimens from 20 conservatively treated undifferentiated prostate cancer patients cell nucleic acid analysis combined with a prospective clinical study was attempted using single cell scanning-cytophotometry. Regardless of therapy type a statistically significant difference in nuclear DNA-content was evident between successfully treated prostate cancers and therapy-resistant cancers. Good prognosis was suggested by a significant decline from aneuploid or polyploid to diploid nuclear-DNA-content with a narrow peak in 2c, especially within the first 12 weeks of treatment. Lack of DNA-content alteration or a DNA-frequency distribution change to the right were correlated with rapid clinical regression. Although all had undifferentiated prostate cancer morphology statistically significant differences in nuclear DNA-content were found. The prognostic value of this remains unknown.

A new approach to photometry of glycated hemoglobin in human blood.

This new approach to measurement of glycated hemoglobin (Hb A1) is a three-step procedure involving (a) conversion of oxyhemoglobin into NO-hemoglobin by sodium dithionite and sodium nitrite in the presence of inositol hexaphosphate; (b) incubation with haptoglobin, which preferentially binds glycated hemoglobin; and (c) determination of the hemoglobin/haptoglobin complex by its peroxidase activity in acid buffer. The procedure is suitable for automated analysis with instruments that are capable of adding a starting reagent. We describe preliminary adaptations for the Abbott VP and Hitachi 705 analyzers and demonstrate the correlation of results with Hb A1 concentrations determined by ion-exchange chromatography.

Reflectance digital matrix photometry.

We have designed and constructed a digital matrix photometer for quantitative measurement of reflected light of small chromophoric areas or colored spots. The areas are divided conceptually into small subunits in which the reflected light is measured. This is done through stepwise scanning with a photodiode array. The sum of the individual reflectance measurements is related to the total amount of an analyte in the spot. We have found that this technique makes possible accurate and precise quantitation of light-absorbing substances in small areas of different sizes, shapes, and internal irregularity measuring as little as 1 mm across.
