alpha-MSH and Pro-Leu-Gly-NH2 (PLG; MIF-1): influence on dopamine (DA) uptake in crude synaptosomal preparations from rat mediobasal hypothalamus (MBH) and caudate putamen (CP).

The uptake of tritiated dopamine [3H] (DMI insensitive DA uptake) by synaptosomal fractions isolated from rat mediobasal hypothalamus (MBH) and caudate putamen (CP) was measured in the presence of different concentrations of alpha-melanocyte stimulating hormone (alpha-MSH) and Pro-Leu-Gly-NH2 (PLG; MIF-1) which is an inhibitor of alpha-MSH release. Compared to control, [3H]DA uptake increased significantly when the synaptosomal fraction of CP was incubated with 0.1 and 1 microM of alpha-MSH and also when the rat was previously injected with alpha-MSH. A simultaneous reduction of endogenous dopamine content was observed. Kinetic studies suggest that the enhanced uptake induced by alpha-MSH 1 microM is the consequence of the rise in Vmax, without changes in the apparent km. The uptake of [3H]DA in hypothalamic (MBH) preparations on the other hand, was not modified by the presence of alpha-MSH. PLG did not have any significant effect on [3H]DA uptake either in the CP or in the MBH. alpha-MSH may act as a modulator of the dopaminergic nigrostriatal system and the results obtained incubating CP synaptosomes in its presence demonstrate a possible direct modulator action by alpha-MSH on the terminal area of the substantia nigra neurons.

The effects of MIF-I, beta-endorphin and alpha-MSH on d-amphetamine induced paradoxical behavioral thermoregulation: possible involvement of the dopaminergic system.

The neuropharmacological basis for d-amphetamine induced paradoxical behavioral thermoregulation remains unclear. This study examined thermoregulatory behavior of rats in a runway device that housed a heat source at one end and in which locomotion along the length of the runway could be observed. Sprague Dawley rats were pretreated with IP injections of saline, beta-endorphin, MIF-1, or alpha-MSH, with a repeat injection after 30 min. In a second experiment, d-amphetamine was administered as the repeat drug for all Ss. The results showed clear differences for heat-source-on vs. heat-source off. All peptides induced hypermotility, although no differentiated effects for the peptides on d-amphetamine induced paradoxical behavioral thermoregulation were found. These findings are discussed in light of the theoretical possibilities that: (a) a ceiling effect exists; (b) there are separate control systems for maintaining body temperature and another for behavioral thermoregulatory responses, and (c) other neurotransmitters may be involved in such induced paradoxical behavioral thermoregulation.

Chronic treatment with MIF-1 prevents the painful stimuli threshold elevation induced by neonatal handling in mice.

Chronic postnatal stressful handling results in a hyposensitivity to thermal nociceptive stimuli. This phenomenon is strongly affected by manipulations of the opioid system. In the present experiment, we report that chronic treatment with MIF-1 during the neonatal period prevents the behavioral alterations induced by handling while it is completely ineffective if injected acutely before antinociceptive testing by the tail flick test at 45 days of life.

Inhibition of the brain to blood transport system for enkephalins and Tyr-MIF-1 in mice addicted or genetically predisposed to drinking ethanol.

Enkephalin concentrations in the brain correlate inversely with ethanol intake and the predisposition of different strains of mice to drink. This and other evidence link ethanol ingestion, addiction, and withdrawal to opiate peptides. We studied the effect of these conditions on the saturable, stereospecific system that transports the enkephalins and Tyr-MIF-1 (a peptide with antiopiate action) out of the brain. The transport rate in mice physically dependent on ethanol was only 56% of the rate in control mice, but during withdrawal from ethanol transport rates increased to levels seen in controls. Transport rates were also lower in strains of mice previously determined to have lower enkephalin concentrations in the brain and to be predisposed to drinking ethanol. Acute intraperitoneal or intracerebroventricular injection of ethanol had minimal direct effects on the transport rate, suggesting that it is not ethanol itself, but those factors associated with addiction and the predisposition to drink ethanol, that altered transport. These studies raise the possibility that the inhibition of this system that transports enkephalins/Tyr-MIF-1 out of the brain might offer a new approach to the control of drinking and withdrawal from ethanol.

The specificity of nephritogenic antibodies: I. Evidence on anti-T-cell specificity in nephritogenic antibodies detected by cytotoxicity and MIF assays.

Heterologous antibodies directed to brushborder antigens of rat kidney tubules are nephritogenic in that an immediate immune complex glomerulopathy occurs after injection ot these antibodies into normal rats. Since previous observations suggested the presence of anti-T-cell specificity within these antibodies, we now compared the specificities of these antibodies with those of heterologous anti-rat thymocyte antibodies, using immunofluorescence (IF), cytotoxicity and migration inhibition (MIF) assays. The results suggest that anti-brushborder anti-serum contains anti-thymocyte specificities, while antithymocyte antiserum contains anti-brushborder specificity. These specificities could be removed selectively from both of the antisera by absorption with insoluble tissue extracts containing the appropriate antigens, i.e. thymocyte antigens or brushborder antigens respectively, indicating that two specificities are involved rather than only cross-reacting antibodies. Since it is unlikely for several other reasons that this feature is simply the result of an immunologic cross-reaction due to antibodies raised by immunization with purified kidney tissue antigens, this dual specificity of anti-brushborder antibody might play a role in the pathogenesis of experimental glomerulopathy.

Trace analysis of the MIF analogue pareptide in blood plasma by high-performance liquid chromatography and short-wavelength excitation fluorometry.

A high-performance liquid chromatographic procedure was developed and applied to analysis of the pharmacologically active MIF analogue pareptide in human plasma. The procedure involves formation of a fluorescent 7-chloro-4-nitrobenzyl-2-oxa-1,3-diazole (NBD-Cl) pareptide derivative followed by separation of the NBD derivative from plasma components on a 30-cm microparticle octadecylsilane bonded column. The separated derivative was quantitated using a short-wavelength excitation fluorometric detector. The detection limit of pareptide in plasma samples was 5 ng or 17 pmoles per ml of plasma. In the absence of plasma, the corresponding on-column detection limit was 0.5 pmoles.

Identification of differentially expressed genes in models of melanoma progression by cDNA array analysis: SPARC, MIF and a novel cathepsin protease characterize aggressive phenotypes.

Currently, the scale and consistency of changes of gene expression profiles in models of melanoma progression are largely unknown. Therefore, we investigated siblings of cell lines or malignant melanomas (MM), which have been selected by nude mouse passages for (a). increased tumorigenicity (local ECM-independent growth), (b). metastatic potential, or (c). selected for increase invasiveness using the Boyden chamber. cDNA array analysis surveying more than 27.000 transcripts per cell line showed that 1.5-2.8% of all detectable transcripts were consistently differentially regulated during selection process in those models. Using array analysis, we identified 33 individual transcripts that exhibited significant differential hybridization paralleling the increased aggressiveness of the selected progeny. Because some of those genes could play a significant functional role in the progression of MM, we additionally proved their regulative pattern using Northern blotting. Among others, progressive overexpression of osteonectin/SPARC, a angiogenesis, was found in the selected offspring from all three experimental models and may therefore be considered as a potential marker for aggressive MM as well a promising therapeutic target. We further show that the selection of MM cells for increased ECM-independent local growth was accompanied by overexpresssion of macrophage migration inhibiting factor (MIF), an important modulator of both cell cycle progression and angiogenesis, and cathepsin Z, a novel member of the family of matrix degrading proteinases.

Methionine-deficient diet extends mouse lifespan, slows immune and lens aging, alters glucose, T4, IGF-I and insulin levels, and increases hepatocyte MIF levels and stress resistance.

A diet deficient in the amino acid methionine has previously been shown to extend lifespan in several stocks of inbred rats. We report here that a methionine-deficient (Meth-R) diet also increases maximal lifespan in (BALB/cJ x C57BL/6 J)F1 mice. Compared with controls, Meth-R mice have significantly lower levels of serum IGF-I, insulin, glucose and thyroid hormone. Meth-R mice also have higher levels of liver mRNA for MIF (macrophage migration inhibition factor), known to be higher in several other mouse models of extended longevity. Meth-R mice are significantly slower to develop lens turbidity and to show age-related changes in T-cell subsets. They are also dramatically more resistant to oxidative liver cell injury induced by injection of toxic doses of acetaminophen. The spectrum of terminal illnesses in the Meth-R group is similar to that seen in control mice. Studies of the cellular and molecular biology of methionine-deprived mice may, in parallel to studies of calorie-restricted mice, provide insights into the way in which nutritional factors modulate longevity and late-life illnesses.

MIF-like activity in simian virus 40-transformed 3T3 fibroblast cultures.

Simian virus 40-transformed fibroblasts (SV3T3), as compared with their untransformed counterparts (3T3), elaborate a macromolecular product that inhibits macrophage migration and causes macrophages to aggregate and lose one type of cell coat material. I'he SV3T3 cells also lack this surface material relative to 3T3 cells. There may be a relationz between migration inhibition factor (MIF), the cell coat, and cell migration.

MIF production of lymphocytes from patients with rheumatoid arthritis with antigen-antibody complexes.

Immune complexes, human erythrocytes coated with human IgG antibody, inhibit the migration of lymphocytes from rheumatoid arthritis (RA) patients. No correlation could be observed between migration inhibition and Waaler-Rose titre (sheep cell agglutination titre) in inidvidual patients. Production of migration inhibitory factor (MIF) could be detected in 16 of 20 cultures of RA lymphocytes incubated with antigen-antibody complexes, when the supernates were tested with guinea pig macrophages. Only two supernates of healthy persons showed migration inhibition activity under these conditions. The results suggest that migration inhibition by antigen antibody complexes in RA represents a true reaction of delayed hypersensitivity.

Carrier-mediated transport of enkephalins and N-Tyr-MIF-1 across blood-brain barrier.

The saturable, carrier-mediated system capable of the brain-to-blood transport of small peptides with an N-terminal tyrosine was characterized. The rate of disappearance of intraventricularly injected iodinated peptide in the presence or absence of the inhibitor being tested was determined from formulas based on the residual radioactivity in the brains of mice after decapitation. The injection of 100 nmol/mouse of unlabeled N-Tyr-MIF-1 (TMIF) increased the half-time disappearance of 125I-TMIF (ITMIF) in the central nervous system (CNS) from 14.1 to 88.7 min (P less than 0.00005). Technetium, a substance transported out of the brain by the same system that transports iodine, was used as a control; the half-time disappearance of technetium pertechnetate was unaffected by unlabeled TMIF. With two related but distinct techniques, the maximum transport rate out of the CNS (Vmax) for TMIF was 0.266 nmol X g of brain per min (method 1) and 0.297 nmol X g-1 X min-1 (method 2), while the amount of unlabeled material needed to achieve 50% of Vmax (Km) was 15.2 nmol/g (method 1) and 15.1 nmol/g (method 2). The lack of effect of the tyrosinated fragments of TMIF as inhibitors indicates that TMIF is being transported in intact form. The Vmax for methionine enkephalin determined with labeled and unlabeled methionine enkephalin was 0.630 nmol X g-1 X min-1 and the Km was 24.95 nmol/g. Studies with the metabolic modulators furosemide, acetozolamide, reserpine, ouabain, and theophylline suggest that the system is sodium dependent and probably independent of ATPase.(ABSTRACT TRUNCATED AT 250 WORDS)

Genome comparison in the genus Mus: a study with B1, MIF (mouse interspersed fragment), centromeric, and Y-chromosomal repetitive sequences.

Using four repetitive sequences, we compared DNAs isolated from Mus caroli, M. cookii, M. hortulanus, M. musculus, M. pahari, M. saxicola, and M. spretus. Except for B1, these probes showed species-specific hybridization patterns. Mouse interspersed fragment (MIF) sequences were present in all species examined, but those defined by the 1.3-kb EcoR1 band were fewer in M. pahari and M. saxicola than in the other species. The Y-chromosomal probe showed male-specific accumulation only in M. hortulanus, M. musculus, and M. spretus, which are known to be closely related. The genetic difference between M. spretus and the other two species (M. hortulanus and M. musculus) was clearly demonstrated by a M. musuclus centromeric sequence that hybridized strongly to M. hortulanus and M. musculus DNA but was underrepresented in the genome of M. spretus. These results may suggest the usefulness of these repetitive sequences in the classification of Mus species that display only subtle morphological differences.

Synthetic MIF has no effect on beta-MSH and ACTH hypersecretion in Nelson's syndrome.

The effect of synthetic MIF (H-Pro-Leu-Gly-NH2) on beta-MSH secretion was studied in five patients with Nelson's syndrome and in one patient with Addison's disease. Two milligrams of the tripetide were injected intravenously (1 mg in an acute injection, followed by a 30-minute-infusion of 1 mg in 20 ml of saline solution). No consistent effect could be observed during the 90-minute period after the beginning of the infusion. In the same patients, LVP stimulation and dexamethasone suppression tests brought about significant changes in the plasma beta-MSH and ACTH levels.

Mastocytoma cell migration in vitro: inhibition by MIF-containing supernatants.

Supernatants with macrophage migration inhibition factor (MIF) activity were obtained from cultures of antigen-stimulated guinea pig and human lymphocytes, and from SV40-infected monkey kidney cells. The monkey and human but not guinea pig preparations were effective in inhibiting migration of mastocytoma cells as well as macrophages. This inhibition of migration was not associated with cytotoxicity and was reversible.

Soluble fragments of the low-affinity IgE receptor (CD23) inhibit the spontaneous migration of U937 monocytic cells: neutralization of MIF-activity by a CD23 antibody.

U937 monocytic cells were found to respond by diminished spontaneous migration when confronted with affinity-purified soluble fragments of the low-affinity receptor for IgE (FcER2/CD23). Unlike B lymphoma cells, U937 cells could not be activated to respond with enhanced DNA synthesis through their membrane-bound CD23 antigen by MHM6, a monoclonal antibody within the CD23 cluster. MHM6 did, however, effectively neutralize the U937-directed MIF (migration inhibition factor) activity contained within the soluble CD23 preparations. The findings not only suggest a role for soluble CD23 as a novel cytokine at sites of inflammation but also indicate different functions for the membrane-bound forms expressed on B cells and monocytes.
