Differences in FM response correlate with morphology of neurons in the rat inferior colliculus.

The response characteristics to linear frequency sweeps were studied in two groups of FM (frequency modulation) sensitive neurons in the rat inferior colliculus. 'FM specialized' cells responded to frequency sweeps but not to pure tones. 'Mixed' cells responded to both frequency sweeps and pure tones. FM specialized cells preferred faster and broader sweeps of higher intensity than did mixed cells and were more directionally selective. In addition, FM specialized cells were more sharply tuned to FM velocity and FM range and had longer response latencies. Physiologically identified FM cells stained intracellularly with horseradish peroxidase revealed differences in morphology correlating with the differences in their responses to tones. FM specialized cells had larger dendritic fields, more dendritic branching and more dendritic spines than did mixed cells. The findings are taken as evidence that the two groups of inferior colliculus neurons are both functionally and morphologically distinct.

Electrochemical metallization of self-assembled porphyrin monolayers.

Multifunctional sensor systems are becoming increasingly important in electroanalytical chemistry. Together with ongoing miniaturization there is a need for micro- and nanopatterning tools for thin electroactive layers (e.g. self-assembling monolayers). This paper documents a method for production of a micro-array of different metal-porphyrin monolayers with different sensor properties. A new method has been developed for the selective and local metallization of bare porphyrin monolayers by cathodic pulsing and sweeping. The metal-porphyrin monolayers obtained were characterized by cyclic voltammetry. It was shown that porphyrin monolayers can be metallized with manganese, iron, cobalt, and nickel by use of the new method. It is expected that all types of metal-porphyrin monolayers can be produced in the same manner.

The natural history of Drosophila ehrmanae.

Drosophila ehrmanae [Parsons P. A., and Bock I. R. (1977). Aust. J. Zool. 25: 249-268] is a desiccation-sensitive species of undisturbed southern Australian temperate-zone rainforests. In this habitat category, nine Drosophila species have been collected, all belonging to the dominant Australian subgenus, Scaptodrosophila. Adults of these species are collected by sweeping foliage in damp habitats so providing temperature/desiccation information on adaptive behavioral selection within microhabitats. Biochemical and metabolic analyses of two common species of this subgenus indicate that D. ehrmanae should be alcohol dehydrogenase null with low ADH activity, should not utilize ethanol and its derivatives as a major resource, and therefore should not be attracted to these metabolites. Ultimately, such species may offer possibilities for behavioral/genetic analyses.

An improved method for rapid measurement of accommodation and firing frequency of single motor units in man.

A stimulator is described producing trapezoid wave forms with accurately defined linear rise and fall at pre-determined slopes independent of the maximum current which is limited at the threshold for a motor unit recorded by discriminative electrodes. The stimulation threshold, measured in rheobase units, is directly proportional to the latency of response and is more accurately measured by sweep expansion (by computer). Thresholds to linearly rising currents of pre-selected slopes are rapidly measured and plotted on a grid calibrated to allow immediate measurement of the time-constant of accommodation.

[Three dimensional (3D-) clinostat and its operational characteristics].

Three dimensional clinostat has been developed for simulation of microgravity on ground. It has applied in many disciplines in gravitational biology. Outline of operational principle is described together with its mechanical design. Rotation around two independent axes makes direction of gravity vector to scan whole steric angle. Magnitude and direction of rotational angular velocity is selected randomly at a certain interval of time to avoid singularity in sweep trajectory of gravity vector. Methods for validation of the operation are presented to test randomness of motion and cancellation of gravity by clino-rotation. Concerns discussed are vibration originated in motor and pseudo-weak magnetic field generated on clinostat. Fluid flow induced by clino-rotation is pointed as another problem to be taken into account.

Threshold sensitivity and frequency specificity in auditory brainstem response audiometry.

Frequency-specific electric response audiometry can be performed on difficult to test young children if the child is sedated and proper choices are made of acoustic stimuli and recording parameters, although certain compromises are necessary. A very satisfactory sedative is secobarbital, administered intramuscularly in doses related to the weight of the child. As stimuli we recommend '2-1-2' tone bursts at 500, 1 000, 2 000, and 4 000 Hz: i.e., with a rise and fall of two periods and a plateau of one period of the modulated tone. A very robust and sensitive response that is not significantly modified by the sedation and is effective for all four frequencies is the P6-SN10 of the early brainstem sequence. To record this complex favorably requires a bandpass input filter of the Butterworth type with pass-band (at -3 dB) from 50 to 1 700 Hz and rejection rates of 24 dB/octave. With this combination, polarity of stimulus is unimportant and sweep time, rate of stimulation and number of responses averaged may be selected for convenience and simplicity. A routine that requires about an hour of testing time is described and the necessary correction factors are given for estimating a child's behavioral pure-tone thresholds. We believe that our threshold estimates are generally correct within 10 dB, and are sufficiently frequency-specific for proper selection of a hearing aid.

Application of fast Fourier transform to auditory evoked brainstem response.

The current application of fast Fourier transform (FFT) to the analysis of auditory evoked brainstem response (ABR) is reviewed under four categories: (1) digital filtering, which facilitates isolation of fast and slow components from the same ABR wave, is the most common use of FFT; (2) power spectral analysis: this seems significant in ABR for isolating and analysing slow, middle and fast components from the Fourier components around each peak of the power spectrum with a three-peak pattern by inverse fast Fourier transform; (3) cross correlation function shows the relationship between two signals being analysed from the viewpoint of their phase. Clinical applications are used in the diagnosis of multiple sclerosis and for automatic detection of ABR; and (4) phase spectral analysis: the synchrony measure method (Fridman, 1984) is a type of phase spectral analysis. In this method, the phase variances of selected Fourier components are calculated, from among 10 averaging groups of 200 sweeps in the same stimulating conditions, to determine the presence or absence of a response. The clinical application of this method to the automatic evaluation of ABR is discussed.

Resource utilization and pathways: meeting the challenge of cost containment.

The 1990s will bring sweeping changes with managed care and capitation. To address this cost/quality paradox, selective intensive care utilization is coupled with clinical pathways as an innovative change for all patients having cerebral revascularization (CVR) or femoral revascularization (FR). From January 1, 1991 through June 30, 1995, data were accumulated on 2023 procedures in 1524 patients. The study was based on 848 CVRs and 1175 FRs. Intensive care unit (ICU) observation was necessary in 73 patients (3.6%) for cardiac or hypertensive management. Twenty-six patients (1.2%) transported to a vascular surgical floor from the postanesthesia recovery room required return to an ICU for complications during hospitalization. There were nine strokes or transient ischemic attacks (0.4%) in the CVR group, four myocardial infarctions (0.2%), and five perioperative deaths (0.3%). In the FR group, there were 14 deaths (0.9%). Readmission during the perioperative period, 30 days, was necessary in 46 patients (3.1%). Financial cost analysis revealed the mean adjusted cost for CVR in 1990 adjusted to 1995 dollars was $7223. The institution of case management reduced this to $4490 (37.8 per cent reduction in total hospital costs). The cost for FR in 1990 dollars adjusted to 1995 was $14,332 reduced to $5541 (a 59 per cent reduction in total hospital costs). This study suggests the use of clinical pathways does not impair quality of care, leads to no higher morbidity or mortality, and can produce significant cost savings to a hospital.

Sweeping of the membranes versus uterine stimulation by oxytocin in nulliparous women. A randomized controlled trial.

The aim of this study was to evaluate whether sweeping of the membranes at term could shorten the length of pregnancy and reduce the incidence of postterm pregnancies. We randomly selected 104 nulliparas with uncomplicated pregnancy and gestational age between 281 and 287 days. Our patients were divided into three groups. Group A consisted of 34 women who were subjected to sweeping of the membranes. Uterine stimulation with oxytocin was applied in 35 women (group B), and 35 women (group C) were used as a control group. We had no significant reduction of the time interval from sweeping of the membranes until delivery (1.9 +/- 1.2 days), compared to that of group B (2.1 +/- 0.8 days) as well as that of the control group (2.5 +/- 0.9 days). The incidence of spontaneous labor in patients after sweeping of the membranes was greater (67.6%) when compared with oxytocin-stimulated patients and the control group (p < 0.05). Furthermore, a better Bishop score was noted in patients of group A. No statistically significant difference was noted in the mode of delivery between the groups, but sweeping of the membranes significantly decreased the incidence of postterm pregnancies (p < 0. 05). We concluded that sweeping of the membranes is an effective method for initiating labor in women with a gestational age between 40 and 41 weeks, thus reducing the need for induction.

Comparison of the use of single capillaries and coupled capillaries based on micellar electrokinetic chromatography (MEKC) and sweeping-MEKC modes.

The use of single capillaries (25 and 50 microm inner diameter (ID)) and coupled capillaries of different diameters (100-50 and 75-25 microm ID) based on micellar electrokinetic chromatography (MEKC) and sweeping-MEKC modes is compared and reported. Naphthalene-2,3-dicarboxaldehyde (NDA)-derivatized dopamine was selected as the model compound by examining the fluorescence intensity when a violet (410 +/- 7 nm, 2 mW) light-emitting-diode (LED) was used as the light source. When a single capillary (50 microm ID) was used, the detection limit for NDA-derivatized dopamine was determined to be 2.0 x 10(-7) M (Signal-to-nose ratio S/N = 3) based on the MEKC mode. This was improved to 4.0 x 10(-9) M when the sweeping-MEKC mode was applied. In addition, this can be further improved to 1.0 x 10(-9) M and 5.6 x 10(-10) M when 100-50 and 75-25 microm ID coupled capillaries are used. The use of the coupled capillary is also helpful for improving the separation efficiency. Based on the sweeping-MEKC mode, the number of theoretical plates (N) for the detected peaks were determined to be 6.3 +/- 2.7 x 10(5) by means of a single capillary (50 microm ID). This can be improved to 9.4 +/- 3.6 x 10(5) and 9.4 +/- 0.9 x 10(6) when the 100-50 and 75-25 microm ID coupled capillaries were applied.

A method to extract three-dimensional objects from three-dimensional NMR image data.

A method to extract shapes of bubble polyhedra from three-dimensional nuclear magnetic resonance image data of foam has been developed. The method consists of four steps: first, to compute series of maximum intensity projection images from the three-dimensional NMR image data of foam to visualize the network structure of the bubble edges; second, to measure the three-dimensional coordinates of vertices of the bubble polyhedra interactively on a CRT using the series of MIP images to obtain polyhedral edges; third, to construct polyhedral faces from the assembly of the edge data; and fourth, to construct bubble polyhedra from the assembly of the face data. The algorithm for construction of polygons and polyhedra is based on the selection of polygons or polyhedra from an enormous number of possible combinations of edges or faces under some simple selection rules. Since the computation time for the selection increased exponentially with the number of the edge data, a 'partial volume sweeping' method in which a cubic region which could accommodate any bubble polyhedron was swept over the three-dimensional image area, was developed to reduce the computation time. By using this method, it was demonstrated that the shapes of many bubble polyhedra could be extracted automatically from the assembly of the edge data within a practical computation time.

Electrochemical behaviors of diethylstilbestrol and its application in pharmacokinetics.

The electrochemical behaviors of diethylstilbestrol (DES) at a glassy carbon electrode were investigated by cyclic voltammetry, linear sweep voltammetry, and differential pulse voltammetry (DPV). Based on these, a sensitive and selective DPV method was developed for determination of DES. The linear response range of DES is 1.0 x 10(-4)-2.0 x 10(-6) mol/L and the detection limit was 8.0 x 10(-8) mol/L. The developed method has been used for the pharmacokinetics of DES in rabbit blood plasma.

A new process for drug loaded nanocapsules preparation using a membrane contactor.

In this paper, we describe a new process for the preparation of drug loaded nanocapsules using a membrane contactor which may be scaled up for industrial applications. Nanocapsules are prepared according to the nanoprecipitation method. The organic phase (solvent, polymer, oil, and drug) is pressed through the pores of an ultrafiltration membrane via the filtrate side. The aqueous phase (water and surfactant) circulates inside the membrane module, and sweeps away the nanocaspules forming at the pore outlets. Two model drugs are selected for the preparation of drug loaded nanocapsules: indomethacin and vitamin E. It is shown that indomethacin loaded nanocapsules with a mean diameter of 240 nm and vitamin E loaded nanocapsules with a mean diameter of 230 nm are obtained with a 150,000 daltons ultrafiltration membrane, a transmembrane pressure of 3 bar, and a crossflow rate of 1.7 m.s(- 1). High fluxes are also obtained (around 0.6 m3/h.m2), leading to the preparation of 1.8 10(- 3) m3 drug loaded nanocapsules in 8 min. The advantage of this membrane contactor compared to other processes for drug loaded nanocapsules preparation is shown to be its scale-up ability.

Complementary on-line preconcentration strategies for steroids by capillary electrophoresis.

Complementary on-line preconcentration strategies are needed when analyzing different classes of solutes in real samples by capillary electrophoresis (CE) with UV detection. The performance of three different on-line preconcentration (focusing) techniques under alkaline conditions was examined in terms of their selectivity and sensitivity enhancement for a group of steroids, including classes of androgens, corticosteroids and estrogens. Electrokinetic focusing of large sample injection plugs (up to 28% of effective capillary length or 22.1 cm) directly on-capillary can be tuned for specific classes of steroids based on changes in their mobility (velocity) using a multi-section electrolyte system in CE. A dynamic pH junction was applied for the selective resolution and focusing of weakly acidic estrogens using borate, pH 11.0 and pH 8.0 in the background electrolyte and the sample, respectively. Sweeping, using an anionic bile acid surfactant and neutral gamma-cyclodextrin (gamma-CD) under alkaline conditions (pH 8), resulted in focusing and separation of the moderately hydrophobic (non-ionic) classes of steroids, such as androgen and corticosteroids. Optimal focusing and resolution of all test steroids under a single buffer condition was realized by a dynamic pH junction-sweeping format using borate, pH 11.0 and bile acid surfactant with gamma-CD in the BGE, whereas the sample is devoid of surfactant at pH 8.0. The design of selective on-line focusing strategies in CE is highlighted by the analysis of microgram amounts of ethynyl estradiol derived from a female contraceptive pill extract using the dynamic pH junction method, which resulted in over a 100-fold enhancement in concentration sensitivity.

Determinants of sound location selectivity in bat inferior colliculus: a combined dichotic and free-field stimulation study.

This study of the neural representation of sound location in the bat Pteronotus parnellii describes how the peripheral and central components of its auditory system shape the horizontal and vertical spatial selectivity of single neurons in the inferior colliculus. Pteronotus extracts spatial information from the echoes of an emitted pulse composed of four constant-frequency harmonics (30, 60, 90, and 120 kHz), each terminated by a downward frequency sweep. To quantify the intensity cues available in the echo, cochlear microphonic response thresholds were used to measure the directional selectivity of the ear and the interaural intensity level disparities (IIDs) created between ears at standardized speaker positions in the bat's frontal sound field, at frequencies in the pulse spectrum. Speaker positions where thresholds were lowest were termed the sensitive area (SA) of the ear. Positions where IID values were greater than 10 dB were termed the difference area (DA). Ear directionality exhibited a pronounced frequency dependence, both in terms of the degree of directional selectivity and the position of the SA. At the 30-kHz harmonic of the pulse, the ear was broadly directional; the SA covered most of the lower half of the ipsilateral field. The ear was highly directional at the 60- and 90-kHz harmonics. Also, the vertical position of the SA changed dramatically between 60 and 90 kHz, from the horizontal midline at 60 kHz to 40 degrees below the midline at 90 kHz. The positions of the DAs also showed a pronounced frequency dependence. The 30-kHz DA was restricted to the extreme lateral part of the frontal sound field. The 60- and 90-kHz DAs were located in the same positions as the equivalent SAs and exhibited the same difference in vertical position. The DAs of the pulse harmonics differ in both their horizontal and vertical positions; the ears thus generate pronounced binaural spectral cues, which provide two-dimensional spatial information. In the inferior colliculus, a combined paradigm of closed-field dichotic stimulation, followed by free-field stimulation, was used to document the frequency tuning and binaural response properties of single neurons and to correlate these properties with the neuron's horizontal and vertical spatial selectivity in the frontal sound field. Where a neuron responded to free-field stimulation at the lowest intensity is termed its SA. A neuron's frequency tuning primarily influenced its degree of spatial selectivity and its sensitivity in the vertical plane, reflecting the directional properties of the external ears at the neuron's best frequency.(ABSTRACT TRUNCATED AT 400 WORDS)

Frequency-modulated radiofrequency pulses in spin-echo and stimulated-echo experiments.

Frequency-modulated rf pulses based on a linear frequency sweep are studied experimentally in spin-echo and stimulated-echo sequences. It turns out that these FM pulses generate a quadratic phase perpendicular to the selected slice and that the quadratic phases of the FM pulses in a spin-echo or stimulated-echo sequence may compensate each other if sweep velocity and gradients are suitably chosen. For a complete rephasing, a formula is given and supported by measured slice profiles. Slice profiles of excellent rectangularity are obtained. Finally, it is shown that with this technique images can be obtained using pulses of increased bandwidth and reduced peak power compared with the AM situation.

Effect of delayed inoculation of selective media in antenatal detection of group B streptococci.

OBJECTIVE: To determine accuracy of group B streptococcal culture swabs immediately inoculated after sampling compared with swabs transported to the laboratory and inoculated subsequently. METHODS: Lower genital tract specimens were collected by sweeping two rayon-tipped swabs across the lower vagina and rectum of gravidas at 35-37 weeks' gestation. One swab was placed immediately in selective growth medium (immediate inoculation group). The other was placed in standard transport media, sent routinely to the clinical laboratory, and transferred subsequently to selective growth medium within 2 hours (delayed inoculation group). RESULTS: Matched specimens were collected from 374 women during the study period. Ninety-three women had positive cultures, a colonization rate of 24.9%. Concordant results were found in 364 of 374 (97.3%). Six women had negative results by the immediate method but positive by the delayed method, and four women had positive immediate cultures but negative by the delayed method. There was no statistically significant difference between the two methods in the detection of positive cultures. CONCLUSION: Use of immediate inoculation instead of delayed inoculation in culturing group B streptococcus specimens does not result in an improved detection rate.

Evidence against the practicality and cost-effectiveness of a gram-positive coccal selective plate for routine urine cultures.

A total of 899 urine cultures were evaluated to assess the need for and cost-effectiveness of using a gram-positive coccal selective plate in the initial plating of urine cultures. Of these cultures, 437 were examined retrospectively and 462 were examined prospectively. Urines were quantitatively plated to three media: sheep blood sugar, MacConkey agar, and phenyl ethyl alcohol agar. Of all urine samples in both studies, 52% yielded no growth on any of the three media. Of all 899 urine cultures, there were only 5 cultures (less than 1%) in which a significant count of a gram-positive organism was detected only on the phenyl ethyl alcohol agar plate and not recoverable on the sheep blood agar plate. In each of these five instances, the need for the use of the selective plate occurred when a Proteus mirabilis strain swarmed over an enterococcus. The inclusion of a selective gram-positive coccal medium for initial plating of urine cultures is unnecessary and not cost-effective. When Proteus swarms on sheep blood agar, a sweep should be made with an inoculating loop from the sheep blood agar and streaked to phenyl ethyl alcohol agar or a similar gram-positive coccal selective medium.
