Amino acid sequences of ferredoxins from Scopolia japonica and Lycium chinense: their similarities to that of Datura arborea.

The complete amino acid sequences of [2Fe-2S] ferredoxins from Scopolia japonica and Lycium chinense have been determined by automated Edman degradation of the entire Cm-proteins and of the peptides obtained by enzymatic digestions. These two ferredoxins exhibited only 2-7 differences in the amino acid sequence when compared to the Datura-ferredoxins (D. stramonium, D. metel, and D. arborea), and especially only 2 or 3 differences compared to D. arborea. On the contrary, 8-19 differences were observed among the other solanaceous ferredoxins. This suggests that S. japonica and L. chinense are closely related taxonomically to Datura plants, especially to D. arborea.

Amino acid sequences of ferredoxins from Atropa belladonna and Hyoscyamus niger: their similarities to those in other tropane-alkaloid-containing plants.

The complete amino acid sequences of [2Fe-2S] ferredoxin from Atropa belladonna and Hyoscyamus niger have been determined by automated Edman degradation of the entire S-carboxymethylcysteinyl proteins and of the peptides obtained by enzymatic digestion. These two ferredoxins exhibited 1-8 differences in their amino acid sequences compared to those of other tropane-alkaloid-containing plants (Scopolia japonica, Datura stramonium, D. metel, and D. arborea), and only 1 or 4 differences compared to S. japonica and D. arborea. In contrast, 9-23 differences were observed among the other solanaceous ferredoxins. This suggests that tropane-alkaloid-containing plants are closely related taxonomically.

Antimuscarinic intoxication resulting from the ingestion of moonflower seeds.

OBJECTIVE: To report a case in which ingestion of moonflower seeds resulted in antimuscarinic intoxication. CASE SUMMARY: An 18-year-old man was found at a local convenience store hallucinating and incoherent. Upon presentation to the emergency department, his signs and symptoms included tachycardia, confusion, dilated pupils, and dry, flushed, hot skin. He was admitted to the intensive care unit. Hallucinations and symptoms resolved within 36-48 hours after hospitalization. The patient then reported that he had ingested moonflower seeds. He recovered and was released 4 days after admission. DISCUSSION: Based on the patient's description and clinical presentation, the moonflower seeds were believed to be Datura inoxia. This species of plant is similar to jimson weed, or Datura stramonium. These plants are known to contain high concentrations of anticholinergic substances; ingestion can result in anticholinergic intoxication. Signs and symptoms that commonly occur include hallucinations, tachycardia, dilated pupils, and disorientation. In our patient, use of the Naranjo probability scale indicated a possible relationship between the moonflower seed ingestion and the patient's signs and symptoms. CONCLUSIONS: Ingestion of the Datura species can result in severe toxicity. Each plant varies in the concentrations of alkaloid substances. For this reason, it is very important for individuals to become educated on the toxicities and potential risks associated with recreational use of these plants.

Confirmed Datura poisoning in a horse most probably due to D. ferox in contaminated tef hay.

Two out of a group of 23 mares exposed to tef hay contaminated with Datura ferox (and possibly D. stramonium) developed colic. The 1st animal was unresponsive to conservative treatment, underwent surgery for severe intestinal atony and had to be euthanased. The 2nd was less seriously affected, responded well to analgesics and made an uneventful recovery. This horse exhibited marked mydriasis on the first 2 days of being poisoned and showed protracted, milder mydriasis for a further 7 days. Scopolamine was chemically confirmed in urine from this horse for 3 days following the colic attack, while atropine could just be detected for 2 days. Scopolamine was also the main tropane alkaloid found in the contaminating plant material, confirming that this had most probably been a case of D. ferox poisoning. Although Datura intoxication of horses from contaminated hay was suspected previously, this is the 1st case where the intoxication could be confirmed by urine analysis for tropane alkaloids. Extraction and detection methods for atropine and scopolamine in urine are described employing enzymatic hydrolysis followed by liquid-liquid extraction and liquid chromatography tandem mass spectrometry (LC/MS/MS).

Evidence for the presence of N-CAM 180 on astrocytes from rat cerebellum and differences in glycan structures between N-CAM 120 and N-CAM 140.

Differences in the N-linked oligosaccharides on isoforms of the neural cell adhesion molecule (N-CAM) on astrocytes were found using a lectin, namely, Datura stramonium agglutinin (DSA). Integral proteins of astrocytes prepared from newborn rat cerebella were solubilized with Nonident P-40 and then separated into two fractions, an unbound fraction and a bound fraction, by a DSA-agarose column. Both fractions thus obtained were subjected to immunoblotting using an anti-N-CAM monoclonal antibody. In the DSA-bound fraction, N-CAM 180 was found as well as N-CAM 120 and N-CAM 140. On the other hand, N-CAM 180 was not detected in the unbound fraction, whereas N-CAM 120 and N-CAM 140 were. Furthermore, N-CAM 180 did not carry the HNK-1 epitope, whereas the other two isoforms did. Although the presence of N-CAM 180 on astrocytes was controversial until recently, the results shown here indicate that N-CAM 180 exists on rat astrocytes and exclusively carries a glycan structure reacting with DSA. This is the first demonstration of the production of N-CAM glycoforms carrying different oligosaccharides by a homogeneous astrocyte preparation. These results suggest that the glycosylation of each N-CAM isoform might be regulated independently. Whether each N-CAM with different glycans participates in different functions remains to be established.

Plant poisonings from herbal medication admitted to a Tunisian toxicologic intensive care unit, 1983-1998.

To collect data on Tunisian herbal toxicity we analysed 56 medical records of patients admitted to the toxicological intensive care unit from 1983 to 1998 following the ingestion of plants. The sex ratio of patients was 1:2, the mean age of patients was 26y (2 to 75y) and II species of plants were involved with Atractylis gummifera (32%), Datura stramonium (25%), Ricinus communis (9%), Nerium oleander (7%) and Peganum harmala (7%) most commonly infested. Poisonings involved neurological (91%), gastrointestinal (73%) and cardiovascular systems (18%). Treatment was mainly symptomatic. The only lethal cases of liver failure involved 16 Atractylis gummifera poisonings.

Differences of alpha3beta1 integrin glycans from different human bladder cell lines.

Expression as well as properties of integrins are altered upon transformation. Cell adhesion regulated by integrins is modulated by glycosylation, one of the most frequent biochemical alteration associated with tumorogenesis. Characterisation of carbohydrate moieties of alpha3beta1 integrin on the cultured human bladder carcinoma (T-24, Hu456, HCV 29T) and human normal ureter and bladder epithelium (HCV 29, Hu609) cell lines was carried out after an electrophoresis and blotting, followed by immunochemical identification of alpha3 and beta1 integrin chains and analysis of their carbohydrates moieties using highly specific digoxigenin-labelled lectins. In all the studied cell lines alpha3beta1 integrin was glycosylated although in general each subunit differently. Basic structures recognized in beta1 subunit were tri- or tetraantennary complex type glycans in some cases sialylated (T-24, HCV 29, HCV 29T) and fucosylated (Hu609, HCV 29T). Positive reaction with Phaseolus vulgaris agglutinin and Datura stramonium agglutinin suggesting the presence of beta1-6 branched N-linked oligosaccharides was found in cancerous cell lines (T-24, Hu456) as well as in normal bladder epithelium cells (Hu609). High mannose type glycan was found only in beta1 subunit from Hu456 transitional cell cancer line. On the other hand alpha3 subunit was much less glycosylated except the invasive cancer cell line T-24 where high mannose as well as sialylated tri- or tetraantennary complex type glycans were detected. This observation suggests that changes in glycosylation profile attributed to invasive phenotype are rather associated with alpha3 not beta1 subunit.

The pattern of poisonings with substance of abuse in Poland (1997-1998).

Substances of abuse have been known in many cultures and geographic zones, and they were used for different reasons, from religions up to hedonistic ones. Central Europe, a region of Poland has never been an area of religious use of narcotics, but lately a number of drug abusers is still growing up. The extent of the drug abuse problem in Poland is difficult to assess, because there is no monitoring system, which registers drug abusers. In this study, authors retrospectively analyzed the files of all drug abuse poisoned patients treated in 10 Polish Poison Centres; as well as the files of patients treated in other hospitals, but who were consulted by toxicologists from these units. During the last 10 years the number of patients treated for drug abuse intoxication has grown 10 times. The structure of used substance of abuse has also changed. In the early 90's opiates dominated. In 1997 all groups of drug abuses were noted, from natural or synthetic opiates, cocaine, LSD, amphetamine, up to Cannabis sativa derivatives (THC). We also revealed intoxications with hallucinogenic mushrooms and plant--Datura stramonium. The most common drugs used were opiates and amphetamine. THC were the third. Amphetamine and THC derivatives were the most often sources of intoxication in young people 15-25 years of age. The opiate poisonings were the commonest in young males, 20-40 years old.

Chemical control of perennial and annual weeds in herbicide resistant soybean crops.

In Romania, the first tests with Roundup Ready on soybean crops were performed in 1998, on 2 soil types: a) at Teleorman Station on chernozem containing 3.5% humus, 4.5% clay b) at Brăila Station placed in Danube Meadow on alluvial soil containing 3.90% humus and 46% clay. In every locality cultivated soybean cultivar S.2254 was resistant to glyphosate. During the three years of experiments (1998-2000) the crop of soybean was infested with various species of weeds (both annual and perennial) of which the most important are: Sorghum halepense (60-80%), Echinochloa crus-galli, Setaria glauca, Amaranthus retroflexus, Solarium nigrum, Yanthium italicum, Abutilon theoprasthi, Sinapis arvensis, Datum stramonium, Polygonum persicaria, Calystegia sepium, Cirsium arvense. In 3 years of experience the best weed control and the highest soybean production were obtained in the variants treated 2 times postemergent with Roundup Ready at a dose of 3 + 3 l/ha. Similar results were also obtained in the farms of the Academy of Agricultural Forestry Sciences, where GMO soybean was cultivated on 1500 hectares.

Carbohydrate moieties of N-cadherin from human melanoma cell lines.

Expression of N-cadherin an adhesion molecule of the cadherin family, in tumor cells is associated with their increased invasive potential. Many studies suggested the role of N-linked oligosaccharides as important factors that contribute to metastasis by influencing tumor cell invasion and adhesion. N-cadherin is a heavily glycosylated protein. We have analysed the carbohydrate profile of this protein synthesized in human melanoma cell lines: WM35 from the primary tumor site and WM239, WM9, and A375 from different metastatic sites. N-cadherin was immunoprecipitated with anti-human N-cadherin polyclonal antibodies. Characterisation of its carbohydrate moieties was carried out by SDS/PAGE electrophoresis and blotting, followed by immunochemical identification of the N-cadherin polypeptides and analysis of their glycans using highly specific digoxigenin or biotin labelled lectins. The positive reaction of N-cadherin from the WM35 cell line with Galanthus nivalis agglutinin (GNA), Datura stramonium agglutinin (DSA) and Sambucus nigra agglutinin (SNA) indicated the presence of high-mannose type glycans and biantennary complex type oligosaccharides with alpha2-6 linked sialic acid. N-cadherin from WM239, WM9, and A375 cell lines gave a positive reaction with Phaseolus vulgaris leukoagglutinin (L-PHA) and lotus Tetragonolobus purpureas agglutinin (LTA). This indicated the presence of tri- or tetra-antennary complex type glycans with alpha-fucose. In addition, N-cadherin from WM9 (lymphomodus metastatic site) and A375 (solid tumor metastatic site) contained complex type chains with alpha2-3 sialic acid (positive reaction with Maackia amurensis agglutinin--MAA). The results demonstrated that N-glycans of N-cadherin are altered in metastatic melanomas in a way characteristic for invasive tumor cells.

The role of some weeds as hosts of Capsicum viruses in the rift valley parts of Ethiopia.

Hot pepper (Capsicum annuum. L.) is a traditionally and economically important spice crop in Ethiopia. Chronic infection with viruses is reported as major constraint, that often force farmers to ban its production. However, epidemiological studies on these viruses are lacking in the country. Knowledge of the weed flora as virus reservoir is essential to reduce the spread of plant viruses. The potential role of some frequently and abundantly occurring weed species as host plants for Capsicum viruses was therefore investigated. Surveys were conducted during the 2000/2001 cropping- and off-season. Samples of various weed species revealing virus symptoms were collected from different districts. Collection was done from inside and at the border of pepper cropped and fallow fields. It was also performed along road sides and irrigation channels. The identity of the collected isolates was established based on serology, electron microscopy and host range studies. Moreover, seeds were collected from natural populations of the most prevalent weeds, namely Datura stramonium and Nicandra physalodes. They were grown under glass house conditions and tested for their ability to host viruses. To verify their role further, healthy seedlings from these weeds were artificially inoculated with all viruses isolated from weeds and Capsicum leaf samples. Natural infection of different Potyviruses with an average incidence of 33% in the weed samples were the most dominant. In addition, the occurrence of Cucumber mosaic virus, Tomato mosaic virus and Potato virus X in these weeds was demonstrated. However, none of the isolated viruses was found to be seed borne in the tested weed species and pepper crop (var.Mrkofanna) under glass house conditions. Recently, there was an explosion in population size of some solanaceous weeds, particularly Datura and Nicandra species in the area. The presence of infected weeds throughout the year means, that they are an important reservoir and source for secondary spread. It is suggested therefore, that a rigorous weed control is an important step to reduce the incidence of viruses infecting Capsicum in the area.

[Neurological syndromes linked with the intake of plants and fungi containing a toxic component (I). Neurotoxic syndromes caused by the ingestion of plants, seeds and fruits].

INTRODUCTION: A wide range of plants, seeds and fruits used for nutritional and medicinal purposes can give rise to neurotoxic symptoms. DEVELOPMENT: We review the neurological pathology associated with the acute or chronic consumption of plants, seeds and fruits in human beings and in animals. Of the plants that can trigger acute neurotoxic syndromes in humans, some of the most notable include Mandragora officinalis, Datura stramonium, Conium maculatum (hemlock), Coriaria myrtifolia (redoul), Ricinus communis, Gloriosa superba, Catharanthus roseus, Karwinskia humboldtiana and Podophyllum pelatum. We also survey different neurological syndromes linked with the ingestion of vegetable foodstuffs that are rich in cyanogenic glycosides, Jamaican vomiting sickness caused by Blighia sapida, Parkinson dementia ALS of Guam island and exposition to Cycas circinalis, Guadeloupean parkinsonism and exposition to Annonaceae, konzo caused by ingestion of wild manioc and neurolathyrism from ingestion of Lathyrus sativus, the last two being models of motor neurone disease. Locoism is a chronic disease that develops in livestock feeding on plants belonging to Astragalus and Oxytropis sp., Sida carpinifolia and Ipomea carnea, which are rich in swainsonine, a toxin that inhibits the enzyme alpha mannosidase and induces a cerebellar syndrome. CONCLUSIONS: The ingestion of neurotoxic seeds, fruits and plants included in the diet and acute poisoning by certain plants can give rise to different neurological syndromes, some of which are irreversible.

[Salem witches, flying brooms, and synthetic drugs].

As supplementary material to Health Education programs about synthetic drugs, the authors present a historical summary on LSD, stramonium and khat. "Tripis", Special K and other synthetic pills contain these substances and are being widely used by youths. The history of these main hallucinogenic active ingredients has a strong tie to the mythology of witchcraft and witches: a historically interesting time period bearing a large amount of religious intolerance. The objective of this review is to end the belief today's youth have that they are taking new substances which have no risks.

[Changes of alkaline phosphatase sugar chains in hepatocellular carcinoma tissue].

OBJECTIVE: To investigate the changes of sugar chain structures of alkaline phosphatase (ALP) in hepatoma tissue and its relation to the invasiveness of hepatocellular carcinoma (HCC). METHODS: The binding ratios of ALP from 9 normal liver tissues, 16 hepatoma tissues and 16 noncancerous tissues surrounding hepatoma were analysed by affinity chromatography on various lectin columns including leukoagglutinating phytohemagglutinin (L-PHA), lentil lectin (LCA), Datura stramonium agglutinin (DSA), erythroagglutinating phytohemagglutinin (E-PHA) and Sambucus nigra bark agglutinin (SNA). RESULTS: The binding ratios of ALP on L-PHA (22.94%+/-5.30%), DSA (55.97%+/-13.72%), LCA (38.16%+/-8.87%), E-PHA (11.56%+/-4.81%) and SNA (69.80%+/-13.71%) in HCC tissues were significantly increased (P<0.01) compared with that in normal liver tissues (L-PHA 5.89%+/-2.75%, DSA 36.20%+/-11.58%, LCA 17.90%+/-6.71%, E-PHA 5.38%+/-2.20%, SNA 57.32%+/-11.27%), respectively. t values between the two groups were 8.94, 3.64, 5.94, 3.62 and 2.32, respectively. L-PHA-binding ratio (25.84%+/-4.67%) of ALP in HCC with invasiveness was significantly higher than that (18.10%+/-3.64%) without invasiveness (t=3.71, P<0.01). CONCLUSION: The changes of ALP sugar chain structures occur in HCC tissue. b1-6 branching sugar chain structure of ALP is related to the invasiveness of HCC.

Therapy of Parkinson's disease.

The most disabling form of Parkinsonism is that occurring after encephalitis. It may occur in persons of any age. The results of surgical treatment, which has been used for the most part only for seriously handicapped patients, have been discouraging in general, although in a few isolated circumstances operation has been of dramatic benefit.The solanaceous alkaloids-atropine, stramonium and hyoscine-either in pure forms or in mixed extracts or tinctures - are the best established drugs at present for the treatment of the postencephalitic forms of Parkinsonism. They have not proven too helpful for patients in the older age group with paralysis agitans. The antihistaminic compounds, particularly Benadryl,(R) have been a very valuable addition. They are of greatest value for patients in the older age group. The newer synthetic compounds, Artane(R) and Panparnit,(R) are also valuable additions. Amphetamine and the related and subsequently produced agents in this group are very helpful for patients showing undue fatigue and lethargy. Tolserol(R) is proving helpful, particularly for patients with painful spasms of rigid muscles.

Study on the weed-crop competition for nutrients in maize.

Considering the effect of crop-weed competition the rate of weed growing, the competitiveness of the occurring weed species and the duration of competition are determining factors. Experiments were carried out on fields in order to collect data on the effect of early weed competition on maize, including the competition for nutrients and the possible rate of nutrient removal by weeds. From 7 sampling areas of the 9.2 ha field weeds and maize samples were collected 1 month after the sowing of maize. We determined the total numbers and the species numbers of weeds by plots. The removed plant species and maize were weighed then dried until the weight balance was reached. The samples were tested for N, P, K and Ca. Comparison was done with the weight and nutrient element content of maize plants taken from the treated, weed-free area. At the same time comparative analyses were made with the mass and nutrient contents of maize plants. There were 12 occurring weed species in this experiment. Based on the rate of weed cover the following species were dominant: Datum stramonium L., Cannabis sativa L., Amaranthus chlorostachis Willd., Chenopodium album L., Chenopodium hybridum L. Our experiments revealed that in the areas being likely to produce high weed populations and showing a considerable high nutrient removal by weeds, the competition between weed plants and maize occurs at an earlier stage of the vegetation period of maize than on fields with moderate weed populations. Weeds have utilised significant amount of nutrients which has been many fold of maize in case of unit area.

Differential glycosylation and cell surface expression of lysosomal membrane glycoproteins in sublines of a human colon cancer exhibiting distinct metastatic potentials.

Changes in the glycosylation of asparagine-linked oligosaccharides have been shown in various tumor cells, including human colon cancer. Attempts were made to elucidate the difference in Asn-linked oligo-saccharides attached to lysosomal membrane glycoproteins isolated from sublines of human colon carcinoma exhibiting high and low metastatic potentials in nude mice. Lysosomal membrane glycoproteins (lamp) 1 and 2 were immunoprecipitated from the cells after labeling with radioactive sugars, and the glycopeptides prepared were fractionated by serial lectin affinity chromatography employing immobilized concanavalin A, Datura stramonium agglutinin, and tomato lectin. Comparison of Asn-linked oligosaccharides from the different colonic carcinoma cells revealed the following features. First, the highly metastatic carcinoma cells express more poly-N-acetyllactosaminyl side chains with branched galactose residues than cells with low metastatic potential. Second, sialylation is more significant in the highly metastatic carcinoma cells than in the poorly metastatic ones. Conversely, N-acetyllactosamine units are less fucosylated in the highly metastatic cells than in poorly metastatic cells. These structural changes were apparently caused by the increase in sialyltransferase and the decrease in alpha 1----3 fucosyltransferase in the highly metastatic cells. The results also suggest that highly metastatic carcinoma cells express more sialyl Lex structures at the termini of poly-N-acetyllactosaminyl side chains than poorly metastatic carcinoma cells. Further, highly metastatic cells were found to express more lamp-1 and lamp-2 on the cell surface. These results were found to be correlated to the increased expression of sialyl Lex structures with high affinity binding of anti-sialyl Lex antibody on highly metastatic cells. Increased expression of sialyl Lex in the poly-N-acetyllactosamines of the cell surface may contribute to the metastatic behavior of the cells, assuming that this structure can serve as a better ligand for selectins present on endothelial cells and platelets.

[Establishment of a method to detect multiple antennae carcinoembryonic antigen (MA-CEA) and MA-CEA level in the sera of patients with malignant tumors].

AIM: To establish a rapid quantitative method to detect the serum multiple antennae carcinoembryonic antigen (MA-CEA), and to measure the MA-CEA levels in the sera of patients with malignant tumors. METHODS: Neuraminidase (NMD) was used to digest the sialic acid at terminals of sugar chains of MA-CEA, and then the datura stramonium agglutinin (DSA) and anti-CEA antibody were used to set up streptavidin-biotin complex (ABC) system EIA (ABC-EIA) for detecting serum MA-CEA levels of 239 patients with carcinomas of lung, stomach, liver, colon and ovary. The serum CEA level was determined by ELISA. RESULTS: The level of serum MA-CEA was remarkably elevated in the CEA-positive patients with lung cancer. The positive rate of MA-CEA was 14.6% in 75 patients with lung cancers, and 47.3% in 19 CEA-positive patients with lung cancers. The MA-CEA was not detected in the sera of other tumor patients. CONCLUSION: We have developed a method for detection of MA-CEA levels, which lays the foundation for the differential diagnosis of lung cancer and for research on the other multiple antennae tumor markers.
