Polarized effects of amiloride and 4,4'-diisothiocyano-stilbene-2,2'-disulfonic acid on ATP-induced contraction of trachea.

Polarity in the effects of amiloride and 4,4'-diisothiocyano-stilbene-2,2'-disulfonic acid (DIDS) in the guinea-pig isolated, perfused trachea was investigated to evaluate the roles of epithelial and airway smooth muscle Na+ and Cl- channels in the development of contractile responses to ATP. The blockers were applied to the mucosal (intraluminal) perfusing solution or to the serosal (extraluminal) bath before the second of two challenges with ATP (10(-4) M), which was added to the same bath as the blocker, or to the abluminal bath. In epithelium-intact tracheas, amiloride (10(-4) M) added to the extraluminal or intraluminal bath rapidly (1 min) and extensively inhibited contractions to extraluminally applied ATP (10(-4) M). In contrast, contractions to intraluminally applied ATP (10(-4) M) were relatively resistant to extraluminal and intraluminal amiloride (10(-4) M), in terms of the degree and onset of the inhibition. DIDS (10(-4) M) present in the extraluminal or intraluminal baths caused a slowly developing elevation of baseline tone. After a 30 min incubation, extraluminal DIDS potentiated responses to extraluminally added ATP, but intraluminal DIDS inhibited contractions to ATP added to the extraluminal and intraluminal baths. In contrast to the intact tracheas where there was no difference, the second response of epithelium-denuded preparations to intraluminally administered ATP was diminished. In rubbed tracheas the response to intraluminally added ATP was inhibited further by intraluminal amiloride but was potentiated by intraluminal DIDS. The results suggest that the effects of amiloride and DIDS were polarized across the tracheal wall and involved epithelial and smooth muscle ion channels.

Up-regulation of nuclear IGF-I receptor by short term exposure of stilbene estrogen, diethylstilbestrol.

In the present study we report the novel findings that IGF-I receptors are present in the nucleus based on the evidence from binding assay, detection of receptors by affinity labeling and Western blotting, and localization by immunofluorescence. The level of nuclear IGF-I receptors (nIGF-IRs) almost doubled in stilbene estrogen (diethylstilbestrol, DES) treated hamster kidneys compared to the controls. The binding constants Kd and Bmax of nIGF-IR of controls, 22.1 nM and 6.6 nmol/mg protein, respectively, were changed in response to DES treatment to 5.3 nM and 16.3 nmol/mg protein (P < 0.001), respectively. The enhanced level of IGF-IR in DES-treated nuclei was confirmed by both affinity labeling and Western blotting. These data suggest that IGFs may exert their biological effect through the IGF-I receptor present in the nucleus. Whether the up-regulation of nIGF-IR by exposure to a carcinogenic dose of DES may be a factor in the induction of renal cancer in Syrian hamsters, is not clear.

HCO(3)(-)-independent rescue from apoptosis by stilbene derivatives in rat cardiomyocytes.

Apoptosis of rat cardiomyocytes induced by staurosporine is prevented by a stilbene derivative (DIDS), which is a known blocker of both Cl(-)/HCO(3)(-) exchangers and Cl(-) channels. To clarify its target, staurosporine-induced activation of caspase-3, DNA laddering and cell death were examined in cultured rat cardiomyocytes. Removal of ambient HCO(3)(-), which minimizes the function of Cl(-)/HCO(3)(-) exchangers, failed to affect the preventive effect of DIDS on apoptosis. A carboxylate analog Cl(-) channel blocker, which does not block Cl(-)/HCO(3)(-) exchangers, also inhibited apoptotic events. Thus, rescue by DIDS of cardiomyocytes from apoptosis is mediated by blockage of Cl(-) channels.

The influence of sodium dodecyl sulfate/benzyl alcohol/H2O system on the photoisomerization of trans-stilbene.

Photoisomerization of trans-stilbene (TS) was investigated in sodium dodecyl sulfate (SDS)/benzyl alcohol (BA)/H(2)O systems in order to establish the relationship between the reaction yields and the compositions and structures of molecular organized assemblies. The results show that, in SDS/BA/H(2)O systems with the structures of oil in water microemulsions, bicontinuous microemulsions, and hexagonal liquid crystals, the reaction yields are much larger than those in benzyl alcohol, both with and without the photosensitizer 9,10-anthracenedicarbonitrile. However, SDS/BA/H(2)O water in oil microemulsion and lamellar liquid crystal can obviously catalyze the photoisomerization of TS only when the photosensitizer is not present. The yield is increased with the increase in H(2)O content or with the decrease in BA content.

Reduction of intracellular pH by inhibitors of natural killer cell activity, nicardipine, methyl 2-(N-benzyl-N-methylamino)ethyl-2,6-dimethyl-4-(2-isopropyl-pyrazolo[1, 5-a]pyridine-3-yl)-1,4-dihydro-pyridine-3,5-dicarboxylate (AHC-52), and 4,4'-diisothiocyano-2,2'-disulfonic acid stilbene (DIDS).

Our previous study showed that nicardipine and its structural analog, methyl 2-(N-benzyl-N-methylamino)ethyl-2,6-dimethyl-4-(2-isopropyl-pyrazolo[1,5 -a]pyridine-3-yl)-1,4-dihydro-pyridine-3,5-dicarboxylate (AHC-52), which is devoid of calcium channel blocking activity, were equally effective in inhibiting natural killer (NK) cell activity, perhaps through inhibition of P-glycoprotein. In this study, we confirmed this finding using a human NK-like cell line, YTN, which is highly cytotoxic to JY cells. The YTN cell-mediated cytotoxicity toward JY cells was inhibited by nicardipine and AHC-52 in a concentration-dependent manner, the concentrations required for 50% inhibition being 14 and 7 microM, respectively. We then examined by flow cytometry whether these reagents modulate the intracellular pH (pHi), since P-glycoprotein reportedly plays a role in pHi homeostasis, perhaps by altering chloride translocation. Both reagents reduced pHi at concentrations similar to those required for inhibition of the cytotoxicity. In addition, 4,4'-diisothiocyano-2,2'-disulfonic acid stilbene (DIDS), an inhibitor of anion exchangers, also inhibited NK cell activity, with an IC50 value of 160 microM, and reduced pHi at a similar concentration, although it is not a P-glycoprotein blocker. Thus, the inhibitory activities of nicardipine, AHC-52, and DIDS toward NK cell activity paralleled their lowering activities of pHi, suggesting the possibility that disregulation of pHi is related to inhibition of NK cell activity.

Cross-reaction of chalcone synthase and stilbene synthase overexpressed in Escherichia coli.

Chalcone synthase (CHS) and stilbene synthase (STS) are related plant polyketide synthases belonging to the CHS superfamily. CHS and STS catalyze common condensation reactions of p-coumaroyl-CoA and three C(2)-units from malonyl-CoA but different cyclization reactions to produce naringenin chalcone and resveratrol, respectively. Using purified Pueraria lobata CHS and Arachis hypogaea STS overexpressed in Escherichia coli, bisnoryangonin (BNY, the derailed lactone after two condensations) and p-coumaroyltriacetic acid lactone (the derailed lactone after three condensations) were detected from the reaction products. More importantly, we found a cross-reaction between CHS and STS, i.e. resveratrol production by CHS (2.7-4.2% of naringenin) and naringenin production by STS (1.4-2.3% of resveratrol), possibly due to the conformational flexibility of their active sites.

The inhibition of DNA repair capacity by stilbene estrogen in Leydig cells: its implications in the induction of instability in the testicular genome.

In this study, we examined the effect of stilbene estrogen, diethylstilbestrol (DES), on the DNA repair capacity of mouse Leydig cells using the host cell reactivation assay. Cells transfected with UV-damaged plasmids, undamaged plasmids, or no plasmids (sham treatment) were grown in serum, serum-free, or DES plus serum-free medium. The serum-grown cells which have a shorter cell cycle time (16h) showed a 40% decrease in DNA repair capacity when compared to serum-free cells with a longer cell cycle time (25h). A significant decrease in the DNA repair capacity of the Leydig cells exposed to DES was observed compared to untreated cells grown in a serum-free environment (P<0.05). The effect of DES on DNA repair in Leydig cells was dose dependent. We have recently shown that DES stimulates the growth of Leydig cells. Stimulatory growth of Leydig cells coupled with a decrease in DNA repair capacity by DES may allow the accumulation of mutagenic lesions in DNA. The mutagenic lesions may result from the attack of redox cycling products of DES and/or errors of replication. This, in turn, may produce alterations in the genome of Leydig cells resulting in genetically unstable cells that may serve as precursor cells for testicular carcinogenesis.

4-Hydroxy-2-pyrone formation by chalcone and stilbene synthase with nonphysiological substrates.

Valerophenone synthase (VPS) is a polyketide synthase that catalyzes the formation of the phloroglucinol derivatives in the synthesis of the bitter acids in hop (Humulus lupulus). The reaction uses isovaleryl-CoA or isobutyryl-CoA, but otherwise it is identical to that of the chalcone synthase in flavonoid biosynthesis. Our study showed that chalcone synthase can perform the function of VPS, but not perfectly, because the majority of the reactions terminated after two condensation reactions (products: 4-hydroxy-2-pyrone derivatives). The same experiments with stilbene synthase yielded exclusively the 4-hydroxy-2-pyrone derivatives, not the products expected from three condensation reactions. The results are discussed in the context of the functional diversity and evolution in the family of CHS-related polyketide synthases.

cis-Stilbene and (1 alpha,2 beta,3 alpha)-(2-ethenyl-3-methoxycyclopropyl)benzene as mechanistic probes in the Mn(III)(salen)-catalyzed epoxidation: influence of the oxygen source and the counterion on the diastereoselectivity of the competitive concerted and radical-type oxygen transfer.

cis-Stilbene (1) has been epoxidized by a set of diverse oxygen donors [OxD], catalyzed by the Mn(III)(salen)X complexes 3 (X = Cl, PF(6)), to afford a mixture of cis- and trans-epoxides 2. The cis/trans ratios range from 29:71 (extensive isomerization) to 92:8, which depends both on the oxygen source [OxD] and on the counterion X of the catalyst. When (1 alpha,2 beta,3 alpha)-(2-ethenyl-3-methoxycyclopropyl)-benzene (4) is used as substrate, a mechanistic probe which differentiates between radical and cationic intermediates, no cationic ring-opening products are found in this epoxidation reaction; thus, isomerized epoxide product arises from intermediary radicals. The dependence of the diastereoselectivity on the oxygen source is rationalized in terms of a bifurcation step in the catalytic cycle, in which concerted Lewis-acid-activated oxygen transfer competes with stepwise epoxidation by the established Mn(V)(oxo) species. The experimental counterion effect is attributed to the computationally assessed ligand-dependent reaction profiles and stereoselectivities of the singlet, triplet, and quintet spin states available to the manganese species.

Diastereoselective [2 + 2] photocycloaddition of stilbene to chiral fumarate. Direct versus charge-transfer excitation.

The selective excitation of the charge-transfer (CT) complex and the direct excitation of the substrate gave distinctly different product ratios and diastereomeric excesses (de's), as well as their temperature dependencies, in [2 + 2] photocycloaddition of (E)-stilbene to bis((R)-1-methylpropyl) fumarate, clearly demonstrating that the excited CT complex and the conventional exciplex differ in structure and reactivity. This conclusion is supported by the contrasting fluorescence behavior exhibited by the relevant excited species, particularly at low temperatures.

Selective endothelin A receptor antagonists. 4. Discovery and structure-activity relationships of stilbene acid and alcohol derivatives.

This publication describes the synthesis and optimization of a novel series of stilbene endothelin antagonists. Analysis of the SAR established for previous papers in this series prompted the design and synthesis of (Z)-4-phenyl-5-(3-benzyloxyphenyl)pent-4-enoic acid 3 which was found to be a moderately active inhibitor of the binding of [125I]ET-1 to ETA receptors with an IC50 of 6 microM. More interestingly, the intermediate compound (E)-2-phenyl-3-(3-benzyloxyphenyl)propenoic acid 5 was equiactive with 3. Optimization of 5 resulted in the preparation of (E)-2-phenyl-3-(2-cyano-5-(thien-3-ylmethoxy))phenylprope noic acid 18 (RPR111723) which had an IC50 in the binding assay of 80 nM on the ETA receptor and a pKB of 6.5 in the functional assay, measured on rat aortic strips. Reduction of the acid group of 5 gave the first nonacidic ETA antagonist in our series, (E)-2-phenyl-3-(3-benzyloxyphenoxy)prop2-enol 6 with an IC50 of 20 microM. Optimization of 6 resulted in the preparation of 2-(2-methylphenyl)-3-(2-cyano-5-(thien-3-ylmethyl)phenyl)pro p-2-enol 33 with an IC50 of 300 nM on the ETA receptor.

Novel synthesis of protected thiol end-capped stilbenes and oligo(phenylenevinylene)s (OPVs).

The first general procedures for preparation of thiol end-capped stilbenes and oligo(phenylenevinylene)s (OPVs) with tert-butyl- and acetyl-protected thiol termini have been developed. These reactions proceed via Br/Li exchange, McMurry, and Wittig-type reactions. The thiol functionality is protected against strong basic and acidic reaction conditions as a t-Bu sulfide. As a key point in the method, reprotection of the thiol group is accomplished by means of acetyl chloride and boron tribromide. The novel strategy forms the basis for stepwise introduction of 4-mercaptostyryl units in OPVs. The new mono-, di-, and trimercapto OPVs have potential applications as one, two, and three terminal molecular devices in gold nanoparticle clusters, self-assembled monolayers, and optoelectronic devices.

The photochemical addition of 2,2,2-trifluoroethanol to methoxy-substituted stilbenes.

The excited-state lifetime of the trans-stilbene chromophore in acetonitrile is prolonged by methoxy substituents in the meta positions. The long-lived singlet excited state of trans-3,5-dimethoxystilbene (trans-2d) is quenched upon the addition of 2,2,2-trifluoroethanol (TFE), and the Markovnikov ether is observed as the major product from steady-state irradiations. The results indicate that the reaction pathway proceeds through a carbocation intermediate.

Cis-Trans Isomerization and Oxidation of Radical Cations of Stilbene Derivatives.

Isomerization from cis stilbene derivatives (c-S (S = RCH=CHC(6)H(5): 1, R = C(6)H(5); 2, R = 4-CH(3)C(6)H(4); 3, R = 4-CH(3)OC(6)H(4) (= An); 4, R = 2,4-(CH(3)O)(2)C(6)H(3); 5, R = 3,4-(CH(3)O)(2)C(6)H(3); 6, R = 3,5-(CH(3)O)(2)C(6)H(3); 7, AnCH=C(CH(3))C(6)H(5); 8, AnCH=CHAn)) to trans isomers (t-S) and oxidation of S with O(2) were studied in gamma-ray radiolyses of c-S in Ar-saturated 1,2-dichloroethane (DCE) and of S in O(2)-saturated DCE, respectively. On the basis of product analyses, it is suggested that a smaller barrier to c-t unimolecular isomerization for c-3(*+)-5(*+) and 8(*+) than for c-1(*+), 2(*+), and 6(*+) due to the single bond character of the central C=C double bond for c-3(*+)-5(*+) and 8(*+) with a p-methoxyl group but not for c-1(*+), 2(*+), and 6(*+) without a p-methoxyl group because of the contribution of a quinoid-type structure induced by charge-spin separation. The isomerization proceeds via chain reaction mechanisms involving c-t unimolecular isomerization and endergonic hole transfer or dimerization and decomposition. The isomerization of c-3(*+) to t-3(*+) is catalyzed by addition of 1,4-dimethoxybenzene but terminated by triethylamine. The regioselective formation of 3d in oxidation of 3(*+) with O(2) is explained by spin localization on the beta-olefinic carbon in 3(*+). The results of product analyses are compared with the rate constants of the unimolecular isomerization and the oxidation for S(*+) measured with pulse radiolyses.

Dichroic properties of bis(benzoxazolyl)stilbene and bis(benzoxazolyl)thiophene dispersed into oriented polyethylene films: a combined experimental and density functional theory approach.

In this work, a combination of experimental and quantum mechanical investigations is performed for the study of dichroic absorption properties of melt-processed linear low-density polyethylene (LLDPE) oriented films containing < or =0.5 wt % of either 4,4'-bis(2-benzoxazolyl)stilbene (BBS) or 2,5-bis(5-tert-butyl-2-benzoxazolyl)thiophene (BTBBT). The data acquired reveal that the film optical anisotropy and the performances as linear polarizer are strongly dependent on the molecular structure of the chromophore. In particular, the rodlike structure of BBS favors the alignment of the dye along the drawing direction of the PE film, providing dichroic ratios as high as 100 and optical performances as linear polarizer close to the pseudo-affine deformation scheme. On the contrary BTBBT, although characterized by huge anisotropic potentialities, confers the oriented film very poor dichroism and is unsuitable for linear polarizer applications. This behavior is attributed to the more complex banana-shaped structure of BTBBT dye caused by the thiophene 2,5-functionalization that limits the molecule parallel orientation to the drawing direction.

Time-dependent density functional theory studies of the photoswitching of the two-photon absorption spectra in stilbene, metacyclophenadiene, and diarylethene chromophores.

Photochromophores such as cis-stilbene (1a), metacyclophenadiene (2a), and the diarylethene 3a undergo photoinduced conrotatory opening and closing of a central bond and are currently being sought out as potential candidates for media within 3D optical information storage devices. Strong molecular two-photon absorption (inducing the reversible photoisomerization) is a necessary feature for this application due to the need for high 3D spatial resolution. Here, the one- and two-photon absorption (OPA and TPA) characteristics of the open- and closed-ring isomers of 1-3 have been investigated using time-dependent density functional theory. It was determined that the excited states populated by two-photon absorption were nearly 1 eV higher in energy than the lowest energy excited state populated by one-photon absorption. The electronic structures of the TPA and OPA accessed states were then compared utilizing natural transition orbital analysis. There, it was found that states excited by OPA had pipi* character about the C-C framework associated with the bond formation/scission of the central C-C bond. In contrast, the states populated by TPA have pipi* character along the C-C skeletal periphery, including phenyl excitations. It is postulated that these differences in excited state electronic structure may lead to reaction pathways alternative to photoisomerization about the central C-C bond, impacting the utility of these compounds as 3D information storage media.
