Whole-plant water flux in understory red maple exposed to altered precipitation regimes.

Sap flow gauges were used to estimate whole-plant water flux for five stem-diameter classes of red maple (Acer rubrum L.) growing in the understory of an upland oak forest and exposed to one of three large-scale (0.64 ha) manipulations of soil water content. This Throughfall Displacement Experiment (TDE) used subcanopy troughs to intercept roughly 30% of the throughfall on a "dry" plot and a series of pipes to move this collected precipitation across an "ambient" plot and onto a "wet" plot. Saplings with a stem diameter larger than 10 cm lost water at rates 50-fold greater than saplings with a stem diameter of 1 to 2 cm (326 versus 6.4 mol H(2)O tree(-1) day(-1)). These size-class differences were driven largely by differences in leaf area and cross-sectional sapwood area, because rates of water flux expressed per unit leaf area (6.90 mol H(2)O m(-2) day(-1)) or sapwood area (288 mol H(2)O dm(-2) day(-1)) were similar among saplings of the five size classes. Daily and hourly rates of transpiration expressed per unit leaf area varied throughout much of the season, as did soil matrix potentials, and treatment differences due to the TDE were observed during two of the seven sampling periods. On July 6, midday rates of transpiration averaged 1.88 mol H(2)O m(-2) h(-1) for saplings in the "wet" plot, 1.22 mol H(2)O m(-2) h(-1) for saplings in the "ambient" plot, and 0.76 mol H(2)O m(-2) h(-1) for saplings in the "dry" plot. During the early afternoon of August 28, transpiration rates were sevenfold lower for saplings in the "dry" plot compared to saplings in the "wet" plot and 2.5-fold lower compared to saplings in the "ambient" plot. Treatment differences in crown conductance followed a pattern similar to that of transpiration, with values that averaged 60% lower for saplings in the "dry" plot compared to saplings in the "wet" plot and 35% lower compared to saplings in the "ambient" plot. Stomatal and boundary layer conductances were roughly equal in magnitude. Estimates of the decoupling coefficient (Omega) ranged between 0.64 and 0.72 for saplings in the three TDE treatment plots. We conclude that red maple saplings growing in the understory of an upland oak forest are responsive to their edaphic and climatic surroundings, and because of either their small stature or their shallow root distribution, or both, are likely to be impacted by precipitation changes similar to those predicted by global climate models.

Multielement composition of wines and their precursors including provenance soil and their potentialities as fingerprints of wine origin.

The influence of the provenance soil and vinification process on the wine multielemental composition was investigated. For this purpose, two different vineyards from the Douro wine district, Portugal, were selected. Monovarietal grapes from a 10 year old vineyard were used to produce a red table wine, in a very modern winery. Polyvarietal grapes from a 60-70 year old vineyard were used to produce a red fortified wine, similar to Port, through a traditional vinification process. The multielement compositions (Al, As, B, Ba, Be, Ca, Cd, Co, Cr, Cs, Cu, Fe, Ga, Hf, Li, Mn, Mo, Nb, Ni, Pb, Rb, Sb, Sc, Sr, Ti, Th, Tl, U, V, W, Y, Zn, Zr, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, and Lu) of soil, grape juices (prepared in the laboratory), and samples collected in the different steps of each winemaking process were measured. Inductively coupled plasma mass spectrometry was used, after suitable pretreatment of the samples (by UV irradiation for liquid samples and high-pressure microwave digestion for soil). Both vinification processes influenced the multielement composition of the wines. Most of the elements presented similar or even lower concentrations in the wine as compared to that observed in the respective grape juice, probably as a result of precipitation or coprecipitation with suspended particles during fermentation and/or wine aging. Evidence of effective contamination during grape pressing, fermentation, and/or fining of wines (depending on the element) was observed for Cd, Cr, Cu, Fe, Ni, Pb, V, and Zn in the fortified wine and Al, Cr, Fe, Ni, Pb, and V in the table wine. Nevertheless, significant correlations were obtained between the multielement composition of the wine and the respective grape juice (R = 0.997 and 0.979 for the fortified and table wines, respectively, n = 31, P < 0.01), as well as between that in the wine (median of the two studied wines) and the provenance soil (R = 0.994, n = 19, P < 0.01), for the set of elements determined in common in the different types of samples. These results are promising concerning the usefulness of the elemental patterns of both soil and wine as fingerprints of the origin of the studied wines. Nevertheless, more wines from the same and other wine districts must be studied in order to consolidate this conclusion. The multielement compositions of the studied wines were compared with those of wines of different characteristics and origins, as well as with the respective legal threshold limit values, when available. Relatively low metal levels, below their threshold limit values, were found in all cases.

Penicillium persicinum, a new griseofulvin, chrysogine and roquefortine C producing species from Qinghai Province, China.

A unique Penicillium isolate from Chinese soil with terverticillate penicilli and ellipsoidal to cylindrical smooth-walled conidia, produces, in addition to the common metabolite ergosterol, copious amounts of an unknown peach-red pigment and the following secondary metabolites: griseofulvin, dechlorogriseofulvin, lichexanthone, roquefortine C, roquefortine D, chrysogine, 2-pyrovoylaminobenzamide, 2-acetyl-quinazolin-4(3H)-one. This isolate, CBS 111235, is described as Penicillium persicinum sp. nov., which belongs to subgenus Penicillium section Chrysogena but is morphologically similar to P. italicum. On the basis of the production of secondary metabolites it resembles P. griseofulvum and P. coprophilum. Sequence data using part of the beta-tubulin gene showed that it is phylogenetically related to P. chrysogenum and P. aethiopicum in section Chrysogena with which it shares both secondary metabolites and ability to grow at 37 degrees C.

Ecophysiology of Acer rubrum seedlings from contrasting hydrologic habitats: growth, gas exchange, tissue water relations, abscisic acid and carbon isotope discrimination.

Eight red maple (Acer rubrum L.) provenances, four each from wet and dry sites, were grown under the same conditions and their physiological responses to soil water availability investigated. Under well-watered conditions, seedlings of wet-site provenances grew faster and had consistently higher net photosynthesis, leaf conductance, maximum carboxylation rate, maximum rate of coupled photosynthetic electron transport, apparent quantum use efficiency, light-saturated photosynthesis and dark respiration than seedlings of dry-site provenances. Under conditions of low soil water availability, only dry-site provenances responded with decreased osmotic potential at full hydration and at the turgor loss point; however, provenances from wet sites showed a smaller reduction in absolute growth rate, a greater reduction in gas exchange and a greater increase in abscisic acid concentrations than dry-site provenances.

Altering the axial light gradient affects photomorphogenesis in emerging seedlings of Zea mays L.

The axial (longitudinal) red light gradient (632 nanometers) of 4 day old dark-grown maize seedlings is increased by staining the peripheral cells of the coleoptile. The magnitude of increase in the light gradient is dependent solely on the light-absorbing qualities of the stain used. Metanil yellow has no effect on the axial red-light gradient, while methylene blue causes a large increase in this light gradient. These stains did not affect growth in darkness or the sensitivity of mesocotyl elongation to red light. However, mesocotyl elongation was altered for the dark-grown seedlings stained with methylene blue when these seedlings were transplanted, covered with soil, and permitted to emerge under natural lighting conditions. These observations are consistent with the idea that there is a single perceptive site below the coleoptilar node, and suggest that this perceptive site gives the actinic light which has traveled downward through the length of the shoot from an entry point in the plant tip region.

Selenium status of Christchurch infants and the effect of diet.

OBJECT: New Zealanders, because of a soil deficiency, have a low intake of selenium. To determine the impact of this on the infant population in Christchurch. METHODS: we have measured red cell and plasma selenium and the selenoenzyme, glutathione peroxidase, in 70 infants less than 12 months old and related these to age and diet. RESULTS: the infant population as a whole had mean plasma levels of selenium and glutathione peroxidase of 33 micrograms/L and 97 U/L compared with adult values of 74 micrograms/L and 150 U/L. Infant red cell levels of 0.30 mu g selenium and 9.0 U glutathione peroxidase per g haemoglobin were similar to those in adults. The selenium status of most breast fed infants after birth remained similar to that of cord blood. Mean plasma selenium and glutathione peroxidase levels in formula fed infants were about half those of breast fed infants, and their red cell selenium was also significantly lower. These did not increase until solids were introduced into the diet. The status of the infants reflected their diet, with the concentration of selenium in formulae being 3.9-5.2 micrograms/mL compared with a mean of 13.4 micrograms/mL in breast milk. CONCLUSIONS: since infants in more replete selenium areas show a gradual rise in blood selenium parameters after birth, this study suggests that formula fed and some breast fed infants in Christchurch receive an inadequate selenium intake. Consideration should be given to supplementing infant formulae and perhaps also the diet of pregnant and/or breast feeding mothers.

Nutrient accumulation in trees and soil following irrigation with municipal effluent in Australia.

Irrigation of tree crops is being evaluated as a method of land disposal of municipal effluent in Australia. A study was carried out from 1980-84 in which seven tree species were sprinkler-irrigated with effluent at an annual rate of 1191-1752 mm. Effective weed control and frequent irrigation resulted in good survival of all species (range 83-100%) at 12 months. Total productivity was estimated at age 4 years by measuring biomass of each species inclusive of litter and roots to a soil depth of 80 cm. Biomass production of the high-yielding species, flooded gum (Eucalyptus grandis) and Sydney blue gum (E. saligna), was around 10 kg m(-2). Percentage leaf mass of these species was small (8-9%) compared with 25% and 29% for the relatively slow-growing river she-oak (Casuarina cunninghamiana) and radiata pine (Pinus radiata). Accumulation of nutrients in the total biomass differed significantly between species and ranged from 34-54 g m(-2) for nitrogen, 4.0-10.4 g m(-2) for phosphorus, 2.1-12.2 g m(-2) for sodium, 22-34 g m(-2) for potassium, 12-61 g m(-2) for calcium and 4.7-9.3 g m(-2) for magnesium. River she-oak and river red gum (E. camaldulensis), because of their relatively large crown and litter masses, accumulated more nitrogen, phosphorus, potassium and calcium than flooded gum or Sydney blue gum. Chemical properties of soils (0-150 cm) were measured in 1980 and again in 1984. Irrigation significantly increased pH (by around 1 unit), throughout the profile. Concentrations of total phosphorus, and exchangeable sodium, calcium and magnesium were increased in the upper profile. Overall, soil chemical properties were not adversely affected by effluent irrigation over the 4-year period, though there was a trend towards more sodic conditions in the soil profile. Nutrient accumulation in soil occurred mainly in the 0-35 cm depth, coinciding with the main root zone of the trees. Renovation of the effluent was therefore estimated as the amount of each nutrient accumulated in the biomass (averaged over the seven species) plus soil (0-35 cm), expressed as a percentage of amount applied in irrigation over the 4 years; that is, nitrogen, 29%; phosphorus, 78%; sodium, 15%; potassium, 26%; calcium, 98% and magnesium, 54%.

[Lead, cadmium, copper and zinc content in vegetables, gooseberry fruit and soil from gardening plots of Lublin].

Lead, cadmium, copper and zinc contents in vegetables, fruits of gooseberries and in soil of Lublin gardening plots "Pionier" and "Podzamcze" both situated along heavy traffic streets, and "Pionier" additional to the close vicinity of automobile factory were determined by atomic absorption spectrophofometry. Statistical analysis of the results respected the streets or factory distances from the gardening plots, and the species of the vegetables. Levels of the determined elements in the most of the samples were lower than permitted by the Ministry of Health Regulation established in 1993. According to that Regulation a slight exceeding of zinc content in the red beetroots and in the overground parts of the leak were found. Lead content in the parsley root and in the dill was close to the tolerance limit. Both lead and cadmium of the soil of "Podzamcze" gardening plot significantly exceeded the levels considered as tolerable for unpolluted grounds, whereas the soil of "Pionier" contained lead close to limit of tolerance level.

Revegetation strategies for bauxite refinery residue: a case study of Alcan Gove in Northern Territory, Australia.

Alumina extraction from bauxite ore with strong alkali produces waste bauxite refinery residue consisting of residue sand and red mud. The amount and composition of refinery residue depend on the purity of the bauxite ore and extraction conditions, and differs between refineries. The refinery residue is usually stored in engineered disposal areas that eventually have to be revegetated. This is challenging because of the alkaline and sodic nature of the residue. At Alcan Gove's bauxite refinery in Gove, Northern Territory, Australia, research into revegetation of bauxite residue has been conducted since the mid-1970s. In this review, we discuss approaches taken by Alcan Gove to achieve revegetation outcomes (soil capping of refinery residue) on wet-slurry disposal areas. Problems encountered in the past include poor drainage and water logging during the wet season, and salt scalding and capillary rise during the dry season. The amount of available water in the soil capping is the most important determinant of vegetation survival in the seasonally dry climate. Vegetation cover was found to prevent deterioration of the soil cover by minimising capillary rise of alkalinity from the refinery residue. The sodicity and alkalinity of the residue in old impoundments has diminished slightly over the 25 years since it was deposited. However, development of a blocky structure in red mud, presumably due to desiccation, allows root penetration, thereby supplying additional water to salt and alkali-tolerant plant species. This has led to the establishment of an ecosystem that approaches a native woodland.

The action spectrum for maximal photosensitivity of germination.

Fruits of Garden Lettuce, imbibed in 0.01 M KNO3, were depleted of maternal active phytochrome B by saturating deep-red exposure and photosensitized by chilling for 1 week at 4 degrees C. Twenty saturated fluence-response curves for photoinduced germination were elaborated between 300 and 800 nm, using exposure periods from 6 to 600 s at 22.5 degrees C; there is linear and closely parallel regression in the logarithmic probability net. The reciprocals of the half-response fluences obtained gave the apparent conversion spectrum of the controlling pigments and this was corrected for the transmittance of the seed-coat. It is a phytochrome spectrum of Pr with photoconversion cross-sections of 1.2 x 10(9) and 4.5 x 10(3) m2 mol-1 at 666 and 800 nm, respectively. This means that for half-saturated germination of sensitized seed, fewer than 1 out of 200,000 phytochrome A molecules have to be photoconverted to Pfr, and no photo-reversibility by deep-red was found. Therefore, all spectral colours of nightly moon- or skylight should stimulate the germination of sensitized weed seeds if they are exposed at the soil surface between sequential tillage operations.

Lead contamination in Portuguese red wines from the Douro region: from the vineyard to the final product.

To quantify lead contamination in wines and to try to identify major lead sources, two winemaking processes were followed during one annual cycle of wine production. Two vineyards from the Douro Portuguese region and two types of wine, one red table wine, which has been produced in a very modern winery, and one red fortified wine (similar to Port), which has been produced by a traditional vinification process, were selected for this study. Aerosols from the vineyards atmosphere, vineyard soil, vine leaves, grapes, and samples from the intermediary and final wine product were collected. Suitable pretreatments, namely, high-pressure microwave assisted digestion (soil, leaves, and grapes) and UV-irradiation (grape juices and samples from the different steps of the vinification processes), were used. The samples were analyzed in terms of lead total concentration and respective isotope ratios by using inductively coupled plasma mass spectrometry and atomic absorption spectrophotometry with electrothermal atomization. It was observed that the major sources of lead were in the vinification system, the more traditional one introducing more lead than the modern one. For the fortified wine, the lead concentration increased from 4.7 microg L(-1), in the grape juice, to 17.2 microg L(-1), in the final product, while for the table wine the increase was from 4.1 to 13.1 microg L(-)(1). Therefore, only about 1/4 (fortified wine) and 1/3 (table wine) of the lead total content of the final products came from soil and atmospheric deposition. Therefore, it is expected that marked reductions of the lead content in the wines would occur if the sources of lead were removed from the tubes and containers used in the vinification system, particularly by using welding alloys and small fittings free of lead. The lead levels in the vine leaves (global mean of 0.43 microg g(dry leave)(-1)) and grapes (global mean of 35 ng g(dry grape)(-1)) were similar in both vineyards.

Ecotoxicological evaluation of three deicers (NaCl, NaFo, CMA)-effect on terrestrial organisms.

The use of chemical deicers such as sodium chloride (NaCl) has increased significantly during the past three decades. Deicers induce metal corrosion and alter the physicochemical properties of soils and water. Environmental damage caused by the use of NaCl has prompted government agencies to find alternative deicers. This article presents a comparative ecotoxicological study of three deicers on soil organisms. Sodium formiate (NaFo) and calcium-magnesium acetate (CMA) are the most interesting commercially available deicers based upon their characteristics and potential toxicity. Organisms used in this study were four species of macrophytes (cress (Lepidium sativum), barley (Ordeum vulgare), red fescue grass (Festuca rubra), Kentucky bluegrass (Poa pratensis)) and an invertebrate (Eisenia fetida). Using standardized and modified methods, the relative toxicity of deicers was CMA < NaFo congruent with NaCl. The results demonstrate that these chemicals could have similar impacts in terrestrial environments since similar quantities of NaFo and greater amounts of CMA are necessary to achieve the same efficiency as NaCl. The toxicity of the tested substances was lower in natural composted soil than in artificial substrate (silica or OECD soil), indicating decreased environmental bioavailability. The response of the organisms changed according to endpoint, species, and soil characteristics (artificial substrate as compared to natural organic soil). The most sensitive endpoint measured was macrophyte growth with Kentucky bluegrass being the most sensitive species.

Accumulation of 137Cesium and 90Strontium from abiotic and biotic sources in rodents at Chornobyl, Ukraine.

Bank voles (Clethrionomys glareolus) and laboratory strains of house mice (Mus musculus BALB and C57BL) were relocated into enclosures in a highly contaminated area of the Red Forest near the Chornobyl (Ukraine) Reactor 4 to evaluate the uptake rates of 137Cs and 90Sr from abiotic sources. Mice were provided with uncontaminated food supplies, ensuring that uptake of radionuclides was through soil ingestion, inhalation, or water. Mice were sampled before introduction and were reanalyzed every 10 d for 137Cs uptake. Levels of 90Sr were assessed in subsamples from the native populations and in experimental animals at the termination of the study. Uptake rates in house mice were greater than those in voles for both 137Cs and 90Sr. Daily uptake rates in house mice were estimated at 2.72 x 10(12) unstable atoms per gram (whole body) for 137Cs and 4.04 x 10(10) unstable atoms per gram for 90Sr. Comparable rates in voles were 2.26 x 10(11) unstable atoms per gram for 137Cs and 1.94 x 10(10) unstable atoms per gram for 90Sr. By comparing values from voles in the enclosures to those from wild voles caught within 50 m of the enclosures, it was estimated that only 8.5% of 137Cs was incorporated from abiotic sources, leaving 91.5% being incorporated by uptake from biotic materials. The fraction of 90Sr uptake from abiotic sources was at least 66.7% (and was probably much higher). Accumulated whole-body doses during the enclosure periods were estimated as 174 mGy from intramuscular 137Cs and 68 mGy by skeletal 90Sr in house mice over 40 d and 98 mGy from 137Cs and 19 mGy from 90Sr in voles over 30 d. Thus, uptake of radionuclides from abiotic materials in the Red Forest at Chornobyl is an important source of internal contamination.

Digging behavior of Solenopsis invicta workers when exposed to contact insecticides.

ABSTRACT Contact between ants and insecticides is a prerequisite for contact insecticides to be effective in the control of red imported fire ants, Solenopsis invicta Buren. Typically, passive contact occurs in the insecticide application process, but ants also may actively contact insecticides by digging in treated soil or walking on a treated soil surface. Laboratory experiments were conducted to determine whether fire ant workers would dig sand treated with contact insecticides in two different scenarios: (1) no-choice bioassays where insecticide-treated sand was the only available digging substrate, and (2) two-choice bioassays where nontoxicant sand was also available for digging. Eight insecticides that are currently registered in the United States for imported fire ant control were tested. They include acephate, bifenthrin, carbaryl, cyfluthrin, deltamethrin, gamma-cyhalothrin, permethrin, and pyrethrin. Workers dug the treated sand for every insecticide tested, even at concentrations up to 10 times of the lowest lethal concentration (LLC) which caused 100% mortality in a toxicity bioassay. However, generally, insecticides significantly reduced the digging effort, even at a concentration that did not cause any significant mortality in the toxicity bioassay.

Expression of pectinase activity among Aspergillus flavus isolates from southwestern and southeastern United States.

Aspergillus flavus is a widely distributed filamentous fungus that contaminates crops with the potent carcinogen aflatoxin. This species can be divided into S and L strains on the basis of sclerotial morphology. During crop infection, A. flavus can secrete a large array of hydrolytic enzymes. These include pectinase, which aids fungal spread through plant tissues. A survey of pectinase expression by soil isolates derived from different regions of the United States revealed geographic polymorphisms. Strain L isolates from Arizona produced moderate to high levels of a specific pectinase P2c, while S strain isolates produced variable amounts of P2c. In contrast, L strain isolates from southeastern U.S. yielded variable P2c production, while S strain isolates consistently expressed high P2c levels. These results were corroborated by pectinase surveys of additional collections of A. flavus from soil and cottonseed. Expression patterns for P2c and pectinmethylesterase were evaluated for a select number of isolates using an isoelectric focusing technique. Clear zone reactions from the pectinase plate assay corresponded to the presence of P2c, while red ring reactions corresponded to the lack of P2c. Commercial cottonseed infected by S strain isolates frequently contained aflatoxin, even when infected by S strain isolates that did not produce pectinase P2c. Thus, although P2c-lacking isolates have reduced invasiveness, these isolates still have sufficient pathogenicity to cause aflatoxin contamination.

Isolation, identification and analysis of antibacterial activity of soil streptomycetes isolates from north Jordan.

A total of 90 different Streptomyces isolates were recovered from 36 soil samples and assessed for their antibacterial activity. Nine isolates were identified by the absence of an aerial mycelium. The rest were grouped into six colour series, namely grey, white, yellow, green, red and polymorphic colours (pink, orange or violet) with total numbers of 29, 18, 14, 8, 3 and 9, respectively. The isolates (68%) showed a reverse side culture pigmentation, 30% produced melanin and 25% produced other soluble pigments. Isolates (48%) were characterized by flexuous spore chains, 21% with spiral and 10% for each of the rectus and retinaculum apertum arrangement. The antibiotic activity against a wide range of bacteria was exhibited by 54% of the isolates which were effective against Bacillus subtilis (57%), Staphylococcus aureus (47%), Escherichia coli (24%), Klebsiella spp (16%), and Shigella spp (12%). The lowest activity (8%) was exhibited against Pseudomonas spp and Salmonella spp. The antibacterial activity of the isolates was divided into four groups according to the diameter of the inhibition zone produced. Groups 3 and 4 with larger inhibition zones indicated their potential as a possible source of novel antibiotics.

Variation in polygalacturonase production among Aspergillus flavus isolates.

Pectinase production by Aspergillus flavus was determined by measuring clear zones formed around colonies stained with ruthenium red. Several isolates produced red zones instead of clear zones. Red zones were reproduced with pectinesterase and correlated with absence of specific polygalacturonases. Of 87 isolates tested, 15 produced red zones.

A simple spectrophotometric method for the determination of cobalt in industrial, environmental, biological and soil samples using bis(salicylaldehyde)orthophenylenediamine.

Bis(salicylaldehyde)orthophenylenediamine (BSOPD) has been proposed as new analytical reagent for the direct non-extractive spectrophotometric determination of cobalt. It reacts with cobalt in slightly acidic (0.0002-0.001 M H(2)SO(4)) 50% 1,4-dioxanic medium to form a red-orange chelate with a molar ratio 1:1. The reaction is instantaneous and the maximum absorbance was obtained at 458 nm and remains constant for over 24h. The average molar absorption coefficient and Sandell's sensitivity were found to be 1.109 x 10(4)l mol(-1)cm(-1) and 20 ng cm(-2) of Co(II), respectively. Linear calibration graph was obtained for 0.1-15 mg l(-1) of Co(II) with a correlation coefficient value of 0.995 for Co-BSOPD complex. Large excess of 44 cations, anions and complexing agents do not interfere in the determination. The method was successfully used in the determination of cobalt(II) from synthetic mixture and certified reference materials for the purpose of validating the method and the results of analyses were found to be excellent agreement with those of actual values. This developed method was also used for determination of cobalt in some environmental waters (potable and polluted), biological (blood and urine) and soil samples and solution containing both cobalt(II) and cobalt(III). The results of the proposed method for biological samples were comparable with AAS and were found to be in good agreement. The method has high precision and accuracy (s=+/-0.01 for 0.5 mg l(-1)).
