Ultrafast evolution of the excited states in the chlorophyll a/b complex CP29 from green plants studied by energy-selective pump-probe spectroscopy.

The energy transfer process in the minor light-harvesting antenna complex CP29 of green plants was probed in multicolor transient absorption experiments at 77 K using selective subpicosecond excitation pulses at 640 and 650 nm. Energy flow from each of the chlorophyll (Chl) b molecules of the complex could thus be studied separately. The analysis of our data showed that the "blue" Chl b (absorption around 640 nm) transfers excitation to a "red" Chl a with a time constant of 350 +/- 100 fs, while the 'red' Chl b (absorption at 650 nm) transfers on a picosecond time scale (2.2 +/- 0.5 ps) toward a "blue" Chl a. Furthermore, both fast (280 +/- 50 fs) and slow (10-13 ps) equilibration processes among the Chl a molecules were observed, with rates and associated spectra very similar to those of the major antenna complex, LHC-II. Based on the protein sequence homology between CP29 and LHC-II, a basic modelling of the observed kinetics was performed using the LHC-II structure and the Förster theory of energy transfer. Thus, an assignment for the spectral properties and orientation of the two Chl's b, as well as for their closest Chl a neighbors, is put forward, and a comparison is made with the previous assignments and models for LHC-II and CP29.

[Postoperative respiratory depression following ambulatory anesthesia for abortion. Evolution of the ventilatory response to CO2 following the use of propofol alone or in association with different opioids: comparison with midazolam in the same situation].

Sixty patients, undergoing minor gynaecologic procedures, were prospectively enrolled in this randomized study. They were anaesthetized with either propofol or midazolam supplemented or not with fentanyl or alfentanil. Respiratory depression was evaluated using repeated measurements of ventilatory response to CO2 (Read's method) during the preoperative period until the second postoperative hour. No respiratory depression was observed when propofol was used alone. By contrast, midazolam alone induced a significant respiratory depression during the first postoperative hour.

Unique pattern of convergent envelope evolution in simian immunodeficiency virus-infected rapid progressor macaques: association with CD4-independent usage of CCR5.

The rate of disease development in simian immunodeficiency virus (SIV) infection of macaques varies considerably among individual macaques. While the majority of macaques inoculated with pathogenic SIV develop AIDS within a period of 1 to 2 years, a minority exhibit a rapid disease course characterized by absence or transience of humoral and cellular immune responses and high levels of virus replication with widespread dissemination of SIV in macrophages and multinucleated giant cells. The goal of this study was to examine viral evolution in three SIVsmE543-3-inoculated rapid progressors to determine the contribution of viral evolution to the development of rapid disease and the effect of the absence of immune pressure upon viral evolution. PCR was used to amplify and clone the entire SIV genome from tissues collected at necropsy, and the course of viral evolution was assessed by env sequences cloned from sequential plasma samples of one rapid progressor (RP) macaque. The majority of sequence changes in RP macaques occurred in the envelope gene. Substitutions were observed in all three animals at specific conserved residues in envelope, including loss of a glycosylation site in V1/V2, a D-to-N/V substitution in a highly conserved GDPE motif, and a P-to-V/H/T substitution in the V3 loop analog. A cell-cell fusion assay revealed that representative env clones utilized CCR5 as a coreceptor, independent of CD4. The selection of specific substitutions in envelope in RP macaques suggests novel selection pressures on virus in such animals and suggests that viral variants that evolve in these animals may play a role in disease progression.

The role of heterarchical control in the evolution of central pattern generators.

The acceptance of the concept of central pattern generators (CPGs) led to the perception that descending inputs initiate stereotyped movements, such as locomotion, but play relatively minor roles after the movement begins. Sensory input could entrain the CPG, and the CPG was responsive to the proper inputs for switching, etc. Evidence is here presented that the influences of both descending and sensory inputs are two-way. Descending inputs are shown to be involved in an ongoing manner during locomotion, as it has been found that CPGs are phasically driving the same descending systems that themselves activate the CPGs. Similarly, sensory inputs are being actively processed by the CPG and, here again, produce a two-way interaction between sensory input and CPGs. Finally, mechanical factors are shown to be major contributors to the form of the movement. Thus, overall the CPG can only be considered as one of several contributors to any movement; all concurrently process the flow of information. Control is viewed as distributed, that is, as heterarchical as opposed to hierarchical. Because of the complexity of interaction between the levels of the system, it is argued that any change in the body will propagate through the system and effect the final output regardless of where the change originates. Thus, ontogenetic and phylogenetic changes must have influences felt throughout the entire system. Examples are presented to demonstrate that this is, indeed, the case. The effect of these changes will primarily be manifest in the interface between the layers, that is, in the interface between the descending and sensory inputs and the CPG, so that the changes can be adaptively accommodated on a moment by moment basis when necessary.

Evolution of Hodgkin's disease.

Hodgkin's disease is an oncogenic core disorder characterized by both mitotic and amitotic neoplastic multiplication, and is associated with collateral disorders such as lacunar formation and leukocytic infiltration. Research has demonstrated that Hodgkin's disease progresses stepwise, beginning with a reversible, biological stage during which Hodgkin and Reed-Sternberg cells are formed, followed by constitutive, but reversible Hodgkinogenic medical stage that leads to an irreversible, systemic and fatal proto-oncogenic stage. This disease results from collateral activation of cytokine and archaic oncogenes, suppression of DNA repair genes in multiple chromosomes. The variability of Hodgkin's disease manifestations has required antisynthetic (antimetabolites, radiotherapy), anti-viral (acyclovir) and anti-mitotic (vincristine, vinblastine) for different loci minores of treatment. Continued molecular biological research of the ancestral and prokaryotic oncogenes is recommended.

Viral evolution toward change in receptor usage: adaptation of a major group human rhinovirus to grow in ICAM-1-negative cells.

Major receptor group common cold virus HRV89 was adapted to grow in HEp-2 cells, which are permissive for minor group human rhinoviruses (HRVs) but which only marginally support growth of major-group viruses. After 32 blind passages in these cells, each alternating with boosts of the recovered virus in HeLa cells, HRV89 acquired the capacity to effectively replicate in HEp-2 cells, attaining virus titers comparable to those in HeLa cells although no cytopathic effect was observed. Several clones were isolated and shown to replicate in HeLa cells whose ICAM-1 was blocked with monoclonal antibody R6.5 and in COS-7 cells, which are devoid of ICAM-1. Blocking experiments with recombinant very-low-density lipoprotein receptor fragments and enzyme-linked immunosorbent assays indicated that the mutants bound a receptor different from that used by minor-group viruses. Determination of the genomic RNA sequence encoding the capsid protein region revealed no changes in amino acid residues at positions equivalent to those involved in the interaction of HRV14 or HRV16 with ICAM-1. One mutation was within the footprint of a very-low-density lipoprotein receptor fragment bound to minor-group virus HRV2. Since ICAM-1 not only functions as a vehicle for cell entry but has also a "catalytic" function in uncoating, the use of other receptors must have important consequences for the entry pathway and demonstrates the plasticity of these viruses.

The phylogenetic distribution of electroreception: evidence for convergent evolution of a primitive vertebrate sense modality.

Specializations for electroreception in sense organs and brain centers are found in a wide variety of fishes and amphibians, though probably in a small minority of teleost taxa. No other group of vertebrates or invertebrates is presently suspected to have adaptations for electroreception in the definition given here. The distribution among fishes is unlike any other sense modality in that it has apparently been invented, lost completely and reinvented several times independently, using distinct receptors and central nuclei in the medulla. There are so far no clearly borderline or transitional fishes, either physiologically or anatomically. We rather expect a few new electroreceptive taxa to be found. The evoked potential method and the newly validated central anatomical criteria provide two useful tools for searching. Although Myxiniformes probably lack electroreception, it is well developed in Petromyzoniformes and in all other non-teleost fishes except Holostei. Thus Elasmobranchia, Holocephala, Dipneusti, Crossopterygii, Polypteriformes and Chondrostei have the physiological and anatomical specializations in a common form consistent with a single origin in primitive vertebrates. Amphibian ancestors probably inherited the system from a stem similar to one of these and passed it on at least to the ambystomatoid and salamandroid urodeles, apparently after losing the kinocilium of the sense cell. The suggestion of electroreception in ichthyophid apodans from skin histology has not been confirmed physiologically, behaviorally or by brain anatomy. With respect to more advanced fishes the most parsimonious interpretation is that the entire system, peripheral and central was lost in ancestors of holostean and teleostean fishes and new systems reinvented in Siluriformes, in Gymnotiformes, in Xenomystinae and in Mormyriformes. These 4 taxa must represent at least two, and probably 3 or 4 independent inventions, presumably from mechanoreceptive lateral line organs and brain centers.

Karyotypic stability in chronic B-cell leukemia.

Twenty-one patients with B-cell chronic lymphocytic leukemia (B-CLL) have been followed for more than 2 years with serial cytogenetic studies, including 11 cases for more than 5 years and three others for more than 10 years. A chromosomally abnormal clone was present at the time of initial study in 10 of these patients, and neither these nor the 11 individuals with a normal karyotype had any cytogenetic evolution during the follow-up period, although clinical progression, requiring therapy, was observed in 13 cases. In an additional 12 B-CLL patients who had repeat chromosome studies but were followed for less than 2 years, two patients with advanced disease and multiple cytogenetic abnormalities developed minor additional karyotypic changes and died within 18 months, and two patients with a normal karyotype developed rapidly progressive disease associated with an emerging chromosomally abnormal clone and survived only 1 year. These results demonstrate that karyotypic evolution is rare in B-CLL. Its occurrence indicates a poor prognosis, but its rarity suggests that clinical progression in this disease is usually more dependent on other factors.

Chromosomal location of genes coding for endosperm proteins of Hordeum chilense, determined by two-dimensional electrophoresis of wheat-H. chilense chromosome addition lines.

The proteins of Hordeum chilense grain were resolved into 25 major components by two-dimensional electrophoresis. Their solubilities in aqueous alcohol solutions were determined to distinguish prolamin storage proteins from metabolic and structural proteins. The prolamins were divided into two groups, based on the presence or absence of intermolecular disulfide bonds determined by gel-filtration chromatography. Using an incomplete set of Chinese Spring wheat-H. chilense disomic addition lines, the structural genes of 21 of the 26 most dominant seed proteins were assigned to chromosomes. The great majority of the prolamin genes, including those coding for a high molecular weight (HMW) prolamin subunit, was present on chromosome 1 Hch. However, a small number of prolamin genes also occurred on chromosomes 5 Hch and 7 Hch. A minor protein, probably belonging to the nonstorage group of proteins, is coded by genes on 5 Hch. Various ditelosomic addition lines and ditelosomic and disomic substitution lines for chromosome 7 Hch were also analyzed by electrophoresis. This technique revealed that the genes for three major prolamins occur on the beta arm of chromosome 7 Hch and that a gene for a minor protein, also thought to be a prolamin, occurs on the alpha arm. These results are discussed in relation to the evolution of prolamin genes in the Triticeae.

Anaerobic digestion of alcohol sulfate (anionic surfactant) rich wastewater--batch experiments. Part II: influence of the hydrophobic chain length.

High-strength wastewater of some industries contains high concentrations of surfactants and readily biodegradable compounds like starch and other carbohydrates. Wastewater of this type is found in the textile wet-processing industry (e.g., cotton desizing). The anaerobic degradation process in such wastewater is inhibited due to the high surfactant content. Alcohol sulfate (AS) of increasing hydrophobic chain length (C8 to C18, surfactant) and soluble starch (size) were used as model compounds in a series of batch experiments with high loadings of the biomass (65 g AS/kg cell dry weight; 910 g starch/kg cell dry weight). The strongest inhibition of the hydrolysis, acidogenesis and acetogenesis was found in reactors containing medium chain ASs like dodecyl sulfate. Starch hydrolyzation rates decreased by nearly 90% in the presence of dodecyl sulfate. Decyl sulfate caused the strongest inhibition of the methanogenesis and a decrease of the methane evolution by 75% within 87 days experimental time. It was found that short as well as long chain ASs caused only minor inhibition phenomena. The presented data can be used as a basis for the selection of suitable surfactants that do not (or not seriously) inhibit the anaerobic digestion of industrial effluents.

An impact analysis of a flexible bat using an iterative solver.

Although technology has now infiltrated and prompted evolution in most mass participation sports, the advances in bat technology in such sports as baseball and cricket have been relatively minor. In this study, we used a simple finite element modelling approach to try to shed new light upon the underlying mechanics of the bat-ball impact, with a view to the future optimization of bat design. The analysis of a flexible bat showed that the point of impact that produced the maximum post-impact ball velocity was a function of the bat's vibrational properties and was not necessarily at the centre of percussion. The details of the analysis agreed well with traditional Hertzian impact theory, and broadly with empirical data. An inspection of the relative modal contributions to the deformations during impact also showed that the position of the node of the first flexure mode was important. In conclusion, considerable importance should be attached to the bat's vibrational properties in future design and analysis.

Assessment of genetic instability in melanocytic skin lesions through microsatellite analysis of benign naevi, dysplastic naevi, and primary melanomas and their metastases.

Microsatellite instability (MSI) is caused by replication errors due to deficient DNA mismatch repair and has been associated with tumour progression in various types of cancer. Controversial results have been reported concerning the frequency and significance of MSI in malignant melanoma. In this study, the time of onset and relative incidence of MSI were determined during the progression of melanocytic tumours, starting with benign melanocytic naevi. MSI was studied at 13 loci containing single, di- or trinucleotide repeat sequences and mapping to five different chromosomal locations. Tumours were classified as being low frequency MSI (L-MSI+) or high frequency MSI (H-MSI+) when either one or at least two marker loci, respectively, displayed mutant alleles in tumour DNA compared with the corresponding normal tissue DNA. None of the eight melanocytic naevi studied showed MSI, whereas a moderate frequency of H-MSI was detected in dysplastic naevi (one out of 11; 9%) and primary melanomas (six out of 56; 11%). The incidence of H-MSI was increased in melanoma metastases from the same patients (nine out of 42; 21%). In contrast to previously reported data showing higher rates of MSI in melanoma, genetic instability seems to be present in a minority of malignant melanoma lesions. However, our findings are consistent with the hypothesis that MSI may be sequentially induced during malignant evolution, contributing to the progression of a subset of melanocytic tumours.

Competition-driven network dynamics: emergence of a scale-free leadership structure and collective efficiency.

Using the minority game as a model for competition dynamics, we investigate the effects of interagent communications across a network on the global evolution of the game. Agent communication across this network leads to the formation of an influence network, which is dynamically coupled to the evolution of the game, and it is responsible for the information flow driving the agents' actions. We show that the influence network spontaneously develops hubs with a broad distribution of in-degrees, defining a scale-free robust leadership structure. Furthermore, in realistic parameter ranges, facilitated by information exchange on the network, agents can generate a high degree of cooperation making the collective almost maximally efficient.

Nucleotide sequence of a neurovirulent variant of the type 2 oral poliovirus vaccine.

Infectious cDNAs of the Sabin type 2 poliovirus vaccine virus and a vaccine-derived neurovirulent type 2 strain (P2/117) have been cloned in Escherichia coli. Nucleotide sequence analysis revealed that P2/117 differs from the vaccine strain by just 23 point mutations. Three occur in the 5' noncoding region. The remainder result in a total of 5 coding changes located in VP1, VP4, 2B, and 3D. The likely role of these mutations in the evolution to neurovirulence is discussed.

[Inflammatory optic neuropathies].

The article presents a study on 94 patients hospitalized in the Clinic of Ophthalmology of Cluj-Napoca, during 1993-1998, with optic neuritis. There are presented the clinical and functional aspects of optic neuritis, especially the importance of static perimetry, for the diagnosis, and her superiority, compared with the dynamic perimetry, in finding minor visual field defects in the diagnosis of the optic neuritis too. After treatment, 75% of the cases have a favorable evolution, but the static perimetry reveals sequela visual field defects at 85.3% of the eyes which suffered optic neuritis. According with this findings, static perimetry has a important role in the diagnosis of optic neuritis, in the revealing the functional sequela defects and in the monitoring of the patients.

[Identification of G6PD gene mutations in several nationalities from Yunnan province].

OBJECTIVE: To analyze the G6PD gene mutations in several nationalities from Yunnan province. METHODS: G6PD gene mutation of 29 samples from G6PD deficient patients were analyzed by using common and mis-matched primering PCR followed by restriction enzyme digestion. The samples were collected from Yunnan province among several nationalities. RESULTS: In these 29 cases, 20 were of two known mutations, including 19 of 1388 (G-->A) and 1 of 1376 (G-->T) mutation. The 7 cases of national minorities, including 3 of Yi ([symbol: see text]), 2 of Bai ([symbol: see text]), 1 of Dai ([symbol: see text]), and 1 of Naxi ([symbol: see text]), were all 1388 mutations. No report of the G6PD gene 1388 mutation in these ethnic groups was published excepting for Dai. CONCLUSION: The mutation probably had been prevalent before the emergence of these national minorities. The approximate rate of G6PD gene mutations provided in Yunnan province may, in some respects, help to interpret the molecular evolution.

The peopling of the Americans as viewed from South America.

Examination of data from Brazilian prehistoric remains, anthropometric variation and genetic polymorphisms of Brazilian or South American Indians in general do not furnish clues about the routes travelled by the first colonizers within the continent. Although the main contribution for the Amerindian gene pool should have originated in Siberia, the genetic data do not contradict the possibility of other, minor sources from the Pacific (Proto-Polynesians ?).
