Innate immune function of the human decidual cell at the maternal-fetal interface.

The aim of the study was to investigate the innate immune function of decidual cells from term unlabored pregnancies by determining Toll-like receptor presence and function. Using immunohistochemistry, reverse transcriptase polymerase chain reaction and NF-kappaB-luciferase plasmid transfection, Toll-like receptor presence and function was determined. Decidual cells express Toll-like receptors 1, 2, 4 and 6 which respond to lipopolysaccharide and peptidoglycan stimulation producing Interleukin-8. Decidual cells from women at term have innate immune function and are capable of producing Interleukin responses to bacterial ligands.

[Cellular immune function influence on postoperative patients with hepatocellular carcinoma and the clinical effect of TILs treatment].

OBJECTIVE: To investigate the cellular immune function influence on postoperative patients with hepatocellular carcinoma (HCC) and clinical effect of TILs, rIL-2 and cyclophosphamide treatment. METHODS: Eight postoperative HCC patients were treated with TILs, rIL-2 and cyclophosphamide. Six patients were infused from the hepatic artery, and the other 2 infused from the portal vein. RESULTS: The IL-2 NK activity, Tsubsets, CD4(+)/CD8(+) level in the peripheral blood of all patients increased and the IL-6, TNF level of most patients decreased (P > 0.05, P < 0.01). For 6 - 18 month follow-up after management, 6 patients with radical tumor resection showed no recurrence. One patient having palliative resection showed no remnant tumor enlargement. One patient with increased TNF level for 8 IU died from hepatic failure 8 months after chemotherapy. CONCLUSIONS: The TILs, rIL-2 and cyclophosphamide treatment of postoperative HCC patients is effective in improving antitumorgenic immune function, preventing tumor recurrence, and restraining remnant tumor growth.

Short and long restraint differentially affect humoral and cellular immune functions.

The aim of this work was to examine the effect of different periods of restraint on the humoral and cellular immune functions in adult male rats. Short restraint stress (2 h over 2 consecutive days) enhanced the primary serum antibody response to sheep red blood cells. The enhancement of this humoral response was dependent on the restraint period, since long restraint stress (6 h over 4 days) failed to modify this response. Short and long restraint decreased both the number of lymphocytes and the T-lymphocyte response to Con A stimulation in the peripheral blood. Neither 2 h over 2 days nor 6 h over 4 days modified the splenic lymphoproliferative response to Con A stimulation, but restraint stress progressively decreased the number of mononuclear splenic cells. Both periods of restraint significantly increased plasma concentration of corticosterone, however plasma prolactin levels were significantly lower after 4 days of restraint but not after short restraint (2 h over 2 days). These results indicate that although some immune functions can be increased after acute or short stress, long stress has an immunosuppressive effect, above all on the cellular immunity which is more susceptible to this effect than the humoral response.

Opioid peptides, receptors, and immune function.

The studies have clearly demonstrated that binding sites for opioid peptides, beta-endorphin, and methionine-enkephalin exist on T lymphocytes. beta-Endorphin appears to be immunodepressant, whereas methionine-enkephalin is immunostimulant. Both in vitro and in vivo studies have shown that methionine-enkephalin can influence some immune functions. Since in vitro modification of immune function requires very low concentrations, it is reasonable to believe that methionine-enkephalin plays a physiological role in the immune system. Although not well established, methionine-enkephalin appears to activate T lymphocytes via opioid receptors and triggers a series of intracellular signals leading to the activation of receptors for interleukin-2 (IL-2), OKT10, and active sheep T red blood cell receptors. Methionine-enkephalin enhances the activity of NK cells and induces the production of IL-2, which in turn may recruit and activate other T-cell subsets like CD3 and CD4. Methionine-enkephalin also enhances mitogen-induced proliferation of lymphocytes. Since preliminary studies with methionine-enkephalin in ARC patients have provided beneficial effects by the improvements in their symptoms, it will be worthwhile to extend these observations to a larger number of patients with ARC and AIDS. Finally, it appears that some endogenous opioid peptides and their analogs, in addition to methionine-enkephalin, may provide therapeutic benefits not only in ARC and AIDS but also in other immunodeficient states.

[Impact of PBDE-209 exposure during pregnancy and lactation on immune function of offspring rats].

OBJECTIVE: To assess the potential hazard of environmental deca-brominated diphenyl ether (PBDE-209) exposure to the immune function of the offspring rats. METHODS: The parental Wistar rats were exposed to PBDE-209 administered intragastrically during pregnancy and lactation, and the development of the immune organs and changes in T lymphocyte subset and their proliferation, NK cell surface markers CD161 and serum immunoglobulins (IgM and IgG) were observed. RESULTS: Significant differences in the weight of the immune organs were noted between the exposure group and control group. In the exposure group, the percentage of T lymphocyte subset CD3(+), CD4(+), CD8(+), CD4(+)CD8(+), the percentage of NK cell surface markers CD16 1 and CD4(+)/CD8(+) ratio were lowered, while the percentage of CD4(-)CD8(-) cells increased. T lymphocyte subset proliferation in the exposure group did not show obviously changes, but compared with the control group, the IgM level in the exposure group was significantly lowered. No significant differences were observed in IgG levels between the exposure and control groups. CONCLUSION: Continuous exposure to high-dose PBDE-209 in female rats during pregnancy and lactation results in possible adverse effect on the immune function of the offspring rats.

Effects of supplemental fructooligosaccharides plus mannanoligosaccharides on immune function and ileal and fecal microbial populations in adult dogs.

The goal of this study was to examine whether supplemental fructooligosaccharides (FOS) plus mannanoligosaccharides (MOS) influenced immune function and ileal and fecal microbial populations of adult dogs. Eight adult dogs surgically fitted with ileal cannulas were used in a crossover design. Dogs were fed 200 g of a dry, extruded, kibble diet twice daily. At each feeding, dogs were dosed with either 1 g sucrose (placebo) or 2 g FOS plus 1 g MOS orally via gelatin capsule. Fecal, ileal, and blood samples were collected at the end of each 14-d period to measure microbial populations and immune characteristics. Treatment least squares means were compared using the GLM procedure of SAS. Supplementation of FOS plus MOS increased fecal bifidobacteria and fecal and ileal lactobacilli concentrations. Dogs fed FOS plus MOS also tended to have lower blood neutrophils and greater blood lymphocytes vs placebo. Serum, fecal, and ileal immunoglobulin concentrations were unchanged by treatment. Supplementation of FOS plus MOS beneficially altered indices of gut health by improving ileal and fecal microbial ecology. Supplementation of FOS plus MOS also altered immune function by causing a shift in blood immune cells.

Changes of immune function in patients with liver cirrhosis after splenectomy combined with resection of hepatocellular carcinoma.

OBJECTIVE: To study the changes of immune function in liver cirrhosis patients after splenectomy combined with resection of hepatocellular carcinoma (HCC). METHODS: Sixteen patients with HCC associated with liver cirrhosis were divided into two groups: splenectomy combined with hepatectomy (splenectomy group n=7) and hepatectomy (non-splenectomy group, n=9). T lymphocyte subsets such as CD4, CD8, CD4/CD8 and Th lymphocyte cytokines such as interferon gamma (IFN-gamma), IL-2, IL-10 in 7 ml peripheral venous blood before operation and 2 months after operation were examined and compared between the two groups. RESULTS: There was no significant difference in pre-operative CD4, CD8, CD4/CD8, IL-2, IFN-gamma, IL-10 levels in the two groups. Two months after operation, the levels of CD4 (38.2%+/-3.7%), CD4/CD8 (1.7+/-0.3), IFN-gamma (104.4+/-14.9 pg/ml), IL-2 (98.6+/-18.6 pg/ml) were increased and those of CD8 (23.7+/-3.7 pg/ml), IL-10 (55.5+/-11.2 pg/ml) levels were decreased in the splenectomy group. The levels of CD4 (32.5%+/-4.0%), CD4/CD8 (1.1+/-0.1), IFN-gamma (70.5+/-12.6 pg/ml), IL-2 (80.9+/-13.5 pg/ml) in the non-splenectomy group were much lower than those in the splenectomy group, but the levels of CD8 (29.4%+/-4.0%), IL-10 (89.4+/-10.0 pg/ml) in the non-splenectomy group were significantly higher than those in the splenectomy group (P<0.05). CONCLUSIONS: Splenectomy combined with hepatectomy for HCC patients associated with liver cirrhosis does not decrease but promote the recovery of T lymphocyte subsets and Th1/Th2 cytokines from imbalance and improve anti-tumor immune function of the patients.

Reduced immune function and malnutrition in the elderly.

An important observation in elderly subjects is their susceptibility to infection associated with a decline in host immune function. Nutrition is also an important factor that influences host defense against infection. We, therefore, evaluated the relationship between nutritional status in 155 healthy subjects ranging in age from 20 to 99 years and various immunological parameters, including the phagocytic and bactericidal activities of neutrophils and monocytes, superoxide production and chemotaxis of neutrophils, lymphocyte subsets, blastoid transformation and serum immunoglobulins. Aging was associated with increased phagocytic activity of neutrophils but not bactericidal activity, superoxide production or chemotaxis of neutrophils. Aging was also associated with a significant decrease in the number of lymphocytes as well as a decline in mature T cells and helper/inducer T cells but with increased numbers of activated T cells, suppressor T cells and natural killer cells. In addition, blastoid transformation in response to phytohemagglutinin (PHA) and concanavalin A (Con A) was significantly reduced in aged subjects. A poor nutritional status was noted in individuals 60 years of age or older. The nutritional status did not influence neutrophil function but correlated significantly with the number of lymphocytes and degree of blastoid formation with PHA and Con A stimulation. Our results suggest that the cell-mediated immunity in elderly subjects is reduced as a result of malnutrition, and that improvement of the nutritional status may enhance the immune function, likely contributing to their successful aging.

[Beta-endorphin involved in the regulation of humoral immune function of rats during acute hypoxia].

In order to investigate the role of beta-endorphin in the regulation of humoral immune function of rats during acute hypoxia, the effects of beta-endorphin on mitogen-induced spleen lymphocyte DNA synthesis and hemolysin formation as well as IgG production to chicken egg albumen were observed. It was found that after rats received icv injection of beta-endorphin (1 ng/rat), the T-lymphocyte DNA content, the hemolysin-forming capacity of SRBC-sensitized rats and the IgG level were reduced significantly compared with the control (icv normal saline). Similar suppressive effects on immune function could be found after rats were exposed to 7 km altitude in a hypobaric chamber for 48 h. Pretreatment with icv injection of naltrexone, the immunosuppressive effects of acute hypoxia were partially blocked. Icv administration of beta-endorphin produced an increase in splenic cathcholamines, similar to those of hypoxia treated group (7 km for 12 h). All the above findings suggest that beta-endorphin may modulate the immune response to hypoxia stress via opioid receptor and its inhibitory action may be mediated by activating sympathetic nervous system.

[Effect of milkvetch injection on immune function of children with tetralogy of Fallot after radical operation].

OBJECTIVE: To evaluate the effect of milkvetch injection (MI) on immune function of children with tetralogy of Fallot (TOF) after radical operation. METHODS: Forty-children with TOF were divided into two groups, the 20 in the control group treated with conventional treatment alone and the 20 in the treated group treated with conventional treatment plus 15 ml of MI every 12 hrs for 14 days. Changes of immunoglobulin, complements, lymphocyte phenotypes and cytokines were observed. RESULTS: In the treated group, the abnormally increased levels of IgG, IgM, C3, C4, CD8+ and CD19+ began to lower at lst-2nd week after treatment, and basically restored to the levels of normal at 3rd-4th week; while the decreased levels of IgA, CD3+, CD4+, CD4+/CD8+ ratio, CD3+/HLA-DR+ and CD3+/CD16+ -CD56+ raised gradually from the 1st week and restored to normal range at 2nd-3rd week. The IL-6 and tumor necrosis factor-alpha (TNF-alpha) levels in the plasma and supernatant, produced in vitro by peripheral blood mononuclear cells (PBMC) decreased gradually at 1st week and restored to the normal level at 3rd-4th weeks. The different value before and after treatment of the above-mentioned indexes in the treated group were superior to those in the control group (P<0.05 or P<0.01). CONCLUSION: MI could significantly improve the immune function of children with TOF after radical operation.

[Determination of the immune function of the erythrocyte and leucocyte and the activity of IL-4 and IFN-gamma in patients with allergic rhinitis].

OBJECTIVE: To investigate the immune function of erythrocyte (RBC) and leucocyte (WBC), and variety of the interleukin-4 (IL-4) and interferon-gamma (IFN-gamma) among patients with allergic rhinitis. METHODS: IL-4 and IFN-gamma secreted by peripheral blood mononuclear cells (PBMC) were measured and compared with immune adhesive and BA-ELISA method. RESULTS: Results showed the RBC and WBC immune adhesive function and IFN-gamma were significantly decreased than of the control, and IL-4 activity were significantly higher than the controls (P < 0.01) in patients with perennial allergic rhinitis. CONCLUSION: These results suggest that RBC and WBC the immune function and IL-4 and IFN-gamma may play an important role in allergic rhinitis.

The effects of pre-emptive epidural sufentanil on human immune function.

BACKGROUND: Surgical stress and general anaesthesia suppress immune functions, including natural killer cell (NK) activity. This suppression could be attributed, at least in part, to the effect of opiates. METHODS: Twenty patients undergoing abdominal hysterectomy received epidural sufentanil (50 microg) either before (pre-emptive) or at the end (control group) of surgery. Post-operative pain relief was provided using sufentanil from a patient-controlled epidural analgesia (PCEA) system. Systemic immunity was assessed by determining leucocyte counts, NK cell counts and activity, lymphocyte response to mitogen stimulation, and secretion of pro-inflammatory cytokines. RESULTS: In the pre-emptive group there was a significant decrease in NK activity on the first and third post-operative day (P < 0.05) compared with baseline values and on the third postoperative day (P < 0.05) compared with the control group. The number of total leucocytes and neutrophiles increased in both groups post-operatively, but no differences were found in the levels of mononuclear lymphocyte populations or in their mitogen responses. Interleukin-6 (IL-6) concentration increased in both groups after the operation. In addition, at the end of the surgery the IL-6 level was greater in the control group than in the pre-emptive group. Interleukin-1 (IL-1) levels had decreased significantly at the end of surgery and 4 h later compared with baseline levels in the pre-emptive, but not in the control group. CONCLUSIONS: Pre-emptive epidural sufentanil during combined propofol and isoflurane anaesthesia had minor effects on the immune response after hysterectomy. The lower production of pro-inflammatory cytokines (IL-1, IL-6) in the pre-emptive group compared with the control group is beneficial, but its clinical importance remains to be determined.

[Effect of zinc on immune function in mice spleen lymphocytes].

OBJECTIVE: In order to study the effects of different levels of zinc nutrition status on the immune function of mice spleen lymphocytes. METHODS: The experiment included two periods: zinc depleted period and repletion period. In the depleted period, mice were divided into 3 dose groups: zinc deficient group (ZD, 5.2mg/kg), zinc pair-fed group (PZ) and normal zinc group (NZ, 25.6 mg/kg). In the repletion period, mice were divided into 3 dose groups: ZD group, DZ-NZ group and PZ-NZ group. It was measured some biomarkers of immunity to assess the zinc nutritional status, including lymphocytes amount, lymphocytes proliferation index and IL-2 activity. RESULTS: The lymphocytes counts of ZD group spleen showed significant decline at seventy days fed zinc deficiency diets. The counts of ZD-NZ group were increased after 22 days of taking normal zinc diet. Zinc deficiency could suppressed the proliferation of the spleen lymphocytes. The spleen lymphocytes proliferation of the ZD-NZ group showed a moderate but non-significant increase compared with the ZD group. Zinc deficiency could decrease IL-2 production. The production of IL-2 of ZD group was lower than those of ZP (P < 0.05), IL-2 production increased following zinc supplement. CONCLUSION: It is found that zinc status can affect the immune function. Production of IL-2 in spleen lymphocyte is affected evidently by zinc.

[Effect of anticancer polypeptide from Buthus Martensii venom on immune function in the H22-bearing mice].

OBJECTIVE: To observe the effect of anticancer polypeptide from Buthus Martensii Venom (APBMV) on Immune function in the H22-bearing mice. METHODS: The MTT colorimetric method, homolysin assay, lymphocyte transformation test, delayed hypersensitivity assay and WBC-count of peripheral blood were used in this study. RESULTS: APBMV could obviously augment NK activity, promote proliferation of lymphocytes induced by Con A, potentiate the response of DTH induced by DNCB, antagonize the decrease of WBC in peripheral blood induced by 5-Fu in the H22-bearing mice. CONCLUSION: APBMV can obviously increase immune function in the H22-bearing mice and antagonize hypoimmunity immunodeficiency or immunodeficiency induced by chemotherapy or the tumor.

Stress and immune function in diabetes mellitus.

Type I diabetic men were found to have a greater number of stresses, more perceived stress per episode, greater anxiety, poorer immune function, and less metabolic control than Type II diabetic men. Degree of family support and compliance with the medical regimen did not differ. Correlation of perceived stress with immune function and metabolic control showed that Type I had more correlates than Type II, with more perceived stress associated with more depressed immune responses.

Effects of cypermethrin and methyl parathion mixtures on hormone levels and immune functions in Wistar rats.

To study interaction and dose-related effects of mixed cypermethrin and methyl parathion on endocrine and immune functions, 120 Wistar rats were divided randomly into six groups of ten male and ten female rats, respectively, at the age of 2 months. All groups were force-fed every 2 days for 30 days with cypermethrin 0.0, 8.0, 0.0, 8.0, 1.8, 0.4 mg/kg bw and methyl parathion 0.0, 0.0, 0.23, 0.23, 0.0518, 0.0115 mg/kg bw. Controls received vehicle solvent only. Body weight gain and organ weights were measured. Serum or blood were used to test luteinizing hormone, follicle stimulating hormone, estradiol (E(2)), testosterone, thyroid hormones (T(3), T(4), TSH), IgG, IgA, rate of neutrophil phagocytosis and of lymphocyte transformation. The effects on relative weights of ovaries and adrenals, IgA and rate of lymphocyte transformation were antagonistic interaction; the effect on estradiol was synergistic in female, whereas addictive in male rats; and the other indices indicated addictive interaction. Organ weights were similar in exposed and control animals except for adrenal (heavier in exposed rats, P<0.01). Serum levels of FSH and estradiol were higher in exposed groups than in controls (P<0.01). IgG levels were lower in exposed rats than in controls (P<0.01), and IgA levels were higher in exposed females than in controls (P<0.01). Lymphocyte transformation rates were lower (P<0.01) and neutrophil phagocytosis rates were higher (P<0.01) in exposed rats than in controls. Our results showed that exposure to low-dose mixtures of cypermethrin and methyl parathion may affect hormone levels (especially estradiol) and immune function in rats, and the NOAELs of combined compounds were located at 1/600 LD(50).

[The use of Chinese traditional medicines to improve impaired immune functions in scald mice].

For the purpose of searching for immunomodulators, this experiment studied the effects of 6 kinds of Chinese traditional herbs on the restoration of the suppressed immune functions in scald mice, including cell-mediated, humoral and non-specific immunity. All control non-treated scald mice showed definite depression of immune functions in various degrees. Polygonum cuspidatum, Taraxacum officinale and Oidenlandia diffusa (wild) roxb showed immunomodulating effects as measured with 3 immunological parameters. But the effects varied according to the dosage of drugs. Rehmannia glutinosa gave definite improving effects on cell-mediated and non-specific immunity, but no significant effect on humoral immunity, while Gui Ling Gao only showed some effect on humoral immunity.

Effects of vitamin C supplementation on performance, iron status and immune function of weaned piglets.

Two experiments were conducted to evaluate the effects of vitamin C supplementation on performance, iron status and immune function of pigs during the 21-day post-weaning period. In experiment one, 48 crossbred pigs (Chester White x Large White x Yorkshire), weaned at 30 days of age and weighing 7.7 +/- 0.9 kg, were allotted to diets containing either 0 or 300 mg/kg vitamin C. In experiment two, 96 crossbred pigs (Chester White x Large White x Yorkshire), weaned at 20 +/- 2 days and weighing 7.1 +/- 0.5 kg, were allotted to diets containing 0.75 or 300 mg/kg vitamin C. Six replicate pens were assigned to each treatment in experiment one while experiment two had eight replicates. All pens housed two barrows and two gilts. In both experiments, no improvement (P > 0.05) in growth rate, feed intake or feed conversion was observed as a result of vitamin C supplementation. Plasma iron concentration increased (P < 0.10) with increased vitamin C in the diet while free and total iron binding capacity were unaffected by treatment. There were no differences in the intradermal response to the mitogen phytohemaggutinin used as an indicator of cellular immunity (P > 0.05). In trial 2, the plasma levels of the immunoglobulin IgG showed a linear (P = 0.07) increase with increasing levels of vitamin C and the same trend was noted in trial 1. Antibody titers to bovine serum albumin also tended to increase in both trials but the increases were not statistically significant. In conclusion, the overall results of these experiments indicate that weanling pig performance is not improved as a result of vitamin C supplementation. Whether or not vitamin C plays a role in stimulating humoral immune function in pigs requires further study since the results of our experiments do not completely rule out the possibility that such a role exists.
