The influence of endotracheally administered bleomycin on hydroxyproline content and collagenase and elastase activity in rat lungs.

The effect of endotracheal administration of bleomycin (10 mg x kg-1) on hydroxyproline content, and on collagenase and elastase activities in rat lungs was investigated. A significant increase in lung tissue hydroxyproline content on 7th day after bleomycin administration was observed. Similar rise in this amino acid level in bronchoalveolar lavage was observed, however, on 14th day after the drug administration. Collagenase activity was lowered by 17% and 36%, respectively. No changes in elastase activity were found. These data suggest that bleomycin cause the lung fibrosis by restraining the collagenase activity.

[N-acetyl-beta-D-glucosaminidase and diurnal excretion of hydroxyproline in chronic viral hepatitis].

In 37 patients with chronic active hepatitis, in 14 with chronic persistent hepatitis and in 11 with normal histological pattern of the liver the serum activity of N-acetyl-beta-glucosaminidase and the 24-hour excretion of hydroxyproline with urine were determined. The 24-hour excretion of hydroxyproline was similar in all groups of patients, while the activity hepatitis and higher than in those of chronic persistent hepatitis (36.9 +/- 12.9 i.u. per 100 ml versus 22.4 +/- 8.2 i.u. per 100 ml, p less than 0.05). In patients with high serum NAGL activity cirrhosis developed significantly more frequently.

Effect of cimetidine on the mucosal hydroxyproline content of rat gastric lesions induced by compound 48/80.

Extensive gastric mucosal lesions developed in rats after the administration of compound 48/80 at a dose of 0.75 mg/kg daily for 3 days. The hydroxyproline content of gastric mucosa was significantly increased 4 h after the last injection of compound 48/80, and subsequently decreased as the lesions healed. Treatment with cimetidine (CAS 51481-61-9) at a dose of 50 mg/kg (twice a day for 3 days) did not affect either the development or the healing of the compound 48/80-induced gastric lesions. However, the increase in mucosal hydroxyproline content was significantly reduced by treatment with cimetidine. This fact suggests that cimetidine might have an inhibitory effect on the regeneration of collagen in gastric mucosa.

Relationship between bone uptake of 99mTc-pyrophosphate and hydroxyproline in blood and urine.

In a group of hospital patients with various diseases, the urinary hydroxyproline-to-creatinine ratio showed a significant correlation (r = 0.63; p less than 0.001) with the 5-hr bone-to-soft-tissue ratio for 99mTc-pyrophosphate uptake. In patients on chronic hemodialysis, a similar correlation was found between the 5-hr bone-to-soft-tissue ratio and hydroxyproline levels in plasma and serum. The findings suggest that 99mTc-pyrophosphate binding by bone is related to collagen metabolism.

A correlation between muscular strength and hydroxyproline concentration in human patellar tendon.

The connective tissue of the knee is frequently injured by athletes, especially those involved in contact sports. It would be important in the prevention of injury as well as the strategy of physical fitness training to know whether the connective tissue is modified in response to athletic stress or training. The potential modification investigated was variability in the concentration of hydroxyproline, a post-translationally modified imino acid found principally in collagen protein. A correlation was sought between this variability and the leg strength parameters torque (expressed as ft-lbs), torque/body weight and work (expressed as ft-lbs). In a preliminary study of five subjects, no correlation was found between hydroxyproline concentration of the patellar tendon and any of the leg strength parameters. These results suggest that this modification of the collagen in connective tissue does not occur in response to athletic stress or training, but rather, for the small number of subjects investigated, appeared to be relatively constant across a range of leg strengths.

[Effect of early local post-irradiation reaction on total and protein-bound hydroxyproline levels in the blood and urine of patients with laryngeal cancer].

In 52 laryngeal cancer patients treated by radiotherapy the serum and urine level of hydroxyproline was tested in connection with the intensity of local postirradiation reactions. The serum and urine level were tested by colorimetric method of Proctor and Udenfriend in Tomaszewski modification. In patients with second and third steps of local reaction intensity there was an elevated level of hydroxyproline only in urine. No correlation was observed between the serum level and local reaction.

The importance of urinary hydroxyproline and serumal osteocalcin in the evaluation of post-menopausal osteoporosis.

Forty-nine women aged 49 to 65 who had been in menopause for more than fifteen years were selected on the basis of densitometric data and clinical symptomatology. Twenty-four of these suffered from severe backaches and had a bone mineral content (B.M.C.) level inferior by at least three standard deviations to the average level according to the standards of our laboratory; these women were diagnosed with severe osteoporosis. The remaining twenty-five subjects were asymptomatic with a B.M.C. between one and two SD from the average; these were diagnosed with moderate asymptomatic osteopenia. A control group was formed by ten apparently healthy women of childbearing age. The women with severe osteoporosis showed a significantly lower level of serumal osteocalcin (Student t Test: P less than 0.05) than the control group, while no significant difference was observed between the latter and the women with moderate asymptomatic osteopenia. The level of hydroxyproline was significantly higher (Student t Test: P less than 0.01) in both groups of menopausal women than in the control group. This data shows that the increase in the level of urinary hydroxyproline characterizes menopause independent of the severity of bone loss. On the other hand, low levels of osteocalcin mainly appear in menopausal women who show severe skeletal demineralization. As a result, we feel that the urinary hydroxyproline/osteocalcin ratio is shown to be useful in both evaluating the risk of post-menopausal osteoporosis and monitoring it.

Increase in urinary excretion of phosphates, hydroxyproline, and cAMP during acetate desoxycorticosterone + saline treatment in the rat.

We investigated the urinary excretion of phosphates, hydroxyproline, and cAMP in rats treated with desoxycorticosterone acetate (DOCA) associated with a hypersaline diet. We measured the three substances in groups with or without parathyroid or thyroid glands, or both. Urinary phosphate excretion increased greatly during the first weeks of treatment in normal rats and in rats autografted with parathyroids and lacking thyroid, and increased slightly in the groups without parathyroids. Hydroxyproline and cAMP had the same pattern; their excretion was greatest during week 2 of treatment in all groups, but these phenomena were more intense in animals with parathyroids. These fact support the conclusion that the parathyroid glands are stimulated during the start of mineralocorticoid-saline treatment.

[Predicting the course of wound healing after appendectomy using the hydroxyproline test].

A test for free hydroxyproline in blood plasma, urine, and wound exudate was conducted in a group of children who were operated on for acute catarrhal and destructive forms of appendicitis in an uncomplicated or complicated post-operative wound process. The changes in the concentration of the biochemical index were most regular in the wound exudate. Free hydroxyproline increased after the operation, reached highest values in 48 hours, and then reduced coming close to the control values 5-6 days after. In wound complications it increased three- to four-fold as compared to the control values, which could serve as an informative test in predicting the course of the postoperative wound process in children.

A supplement to alkaline phosphatase fractionations: utilization of gamma-glutamyl transpeptidase and hydroxyproline assays.

Fractionation of serum alkaline phosphatase by either biochemical or electrophoretic techniques is often insufficient to determine the source of the elevated serum enzyme. In 31 unselected patients with high serum alkaline phosphatase a simultaneous determination of serum gamma glutamyl transpeptidase and urinary hydroxyproline excretion rates was performed, in addition to urea denaturation and L-phenylalanine inhibition tests. Of 25 patients with definite diagnosis, the urea denaturation test correctly identified the source of the elevated serum alkaline phosphatase (ALP) in 16 (64 percent). The serum gamma-glutamyl transpeptidase (GGTP) alone predicted the correct diagnosis in 64 percent of the cases and the urinary hydroxyproline (HOP) alone in 69 percent. When all three tests were performed simultaneously, the combination of results identified the source of the ALP in 88 percent of the cases. It is believed that this combination offers better sensitivity and specificity than any of the three tests used individually.

Trypsin cleaves lysylproline in a hydroxyproline-rich glycoprotein from Zea mays.

Although trypsin is highly specific for lysyl and arginyl bonds, some peptide bonds, such as lysylproline, are generally trypsin-resistant, with rare exceptions as reported here. Trypsin cleaved a specific Lys-Pro bond in the chymotryptic peptide: Thr-Hyp-Ser-Hyp-Lys-Pro-Hyp-Thr-Pro-Lys-Pro-Thr-Hyp-Hyp-Thr-Tyr isolated from a Zea mays hydroxyproline-rich glycoprotein (HRGP). The daughter peptides, Thr-Hyp-Ser-Hyp-Lys-Pro-Hyp-Thr-Pro-Lys and Pro-Thr-Hyp-Hyp-Thr-Tyr, show cleavage of only one of the two Lys-Pro bonds in the parent peptide. From these and other data we suggest that there are two prerequisites for Lys-Pro cleavage: First, an extended helix characteristically present in proline or hydroxyproline-rich proteins; second, flexibility in two residues flanking the Lys-Pro bond.

Influence of eicosapentaenoic acid and vitamin E on hepatic hydroxyproline content in rabbits fed cholesterol-rich diet.

The effect of eicosapentaenoic acid (EPA) and vitamin E on hepatic hydroxyproline content, as an index of collagen was examined in rabbits receiving cholesterol rich diets for a period of 45 days. Rabbits were divided as control (A) and cholesterol fed groups (B, C, D). Group C received 80 mg. of EPA and group D received 100 IU of vitamin E daily in addition to the cholesterol rich diet (2% w/w) which was solely given to group B. The maintenance of rabbits on high cholesterol diets resulted in significantly increased liver cholesterol concentrations. This effect was most pronounced in rabbits receiving cholesterol alone. Hepatic triglyceride levels remained unchanged in all cholesterol-fed rabbits, but total phospholipid levels in liver significantly decreased in EPA and vitamin E supplemented rabbits. An interesting finding was the increase in hepatic hydroxyproline content in rabbits following the administration of EPA and vitamin E to cholesterol rich diet.

Fasting urinary hydroxyproline: creatinine ratios in silicosis.

Fasting urinary hydroxyproline: creatinine (HOP:C) ratios were measured in 74 patients with suspected (borderline) silicosis (10), simple silicosis (46) and complicated silicosis (9 silicotuberculosis, 4 massive fibrosis, and 5 combined silicotuberculosis and massive fibrosis) and in 18 healthy subjects (controls). There was no statistically significant difference in urinary HOP:C ratios between control subjects (mean 13.8, SD 3.6) and suspected silicotics (mean 18.0, SD 7.6); however, urinary HOP:C ratios were significantly higher in both simple silicosis (mean 25.0, SD 9.9, p less than 0.001) and complicated silicosis (silicotuberculosis and progressive massive fibrosis) (mean 28.6, SD 11.3; p less than 0.001). Urinary HOP:C ratios appeared to show a graduated increase to their highest levels in category 2 silicosis and thereafter remained constant or declined slightly in category 3 silicosis and massive fibrosis. The results support the suggestion that urinary hydroxyproline might be useful as an indicator of disease progression in established silicosis. However, further longitudinal studies are needed to confirm its predictive value.

Hyperpurification of paired helical filaments reveals elevations in hydroxyproline content and a core structure related peptide fragment.

We have subjected conventionally-purified Alzheimer's paired helical filaments (PHF) to electrophoresis in a Tris/borate/SDS buffer and obtained the separation of PHF core protein(s) (PHFi) from solubilized PHF-associated proteins (PHFs). Electron microscopy revealed intact PHF structures before and after this separation, but no evidence of any other structures in the PHFi fraction. The percent mass of hydroxyproline and glycine increased in the PHFi fraction after 4.5 hr of electrophoresis to account for 5.8% and 13.6% of the total mass, respectively. ELISA data confirmed that our PHFi and PHFs fractions were reactive with several putative PHF-specific antibodies. These data suggest that inappropriate hydroxylation of proline residues occurs in precursor PHF protein(s), resulting in the polymerization and subsequent insolubility of PHF in brain regions affected with Alzheimer's disease. Neurofibrillary tangles (NFT), one of the primary neuropathological features of Alzheimer's disease, are comprised of cytosolic bundles of uniform proteins which microscopically appear to be paired helical filaments (PHF). PHF are thought to be responsible for the cellular necrosis associated with the clinical symptoms of Alzheimer's disease (Dayan, 1970; Hirano and Zimmerman, 1962). Optical reconstruction of PHF has recently indicated that the true structure is more accurately described as a twisted ribbon of 30 A in the axial direction (Wischik et al., 1988). Immunological studies have suggested that tubulin (Grundke-Iqbal et al., 1979), microtubule associated proteins (Grundke-Iqbal et al., 1986; Kosik et al., 1986; Wood et al., 1986; Ksiezak-Reding et al., 1987), intermediate filaments (Yen et al., 1983), neurofilaments (Anderton et al., 1982), and ubiquitin (Mori et al., 1987; Perry et al., 1987), form part of the PHF core protein. To date, however, no study has been able to definitively show that any one of these purported PHF components is contained in the PHF core structure, rather than being non-covalently bound proteins or co-purifying contaminants. The present study extends previously published purification procedures, resulting in an isolated stable PHF core structure which is indistinguishable from previously published descriptions of PHF. The amino acid composition (Bidlingmeyer et al., 1984) of the PHF core structure appears to include hydroxyproline, an amino acid not commonly found in cytosolic proteins. We also demonstrate that some monoclonal antibodies previously raised against semi-purified PHF recognize determinants which are not related to the PHF core structure but rather recognize PHF-associated proteins or contaminants of the purification process.

The determination of urinary 3-trans-hydroxyproline (3 OHP). II. Normal values in neonates, infants and preschool children.

3-trans-hydroxyproline is a hydroxyproline isomer present in collagens. It is more abundant in basement membrane collagen (collagen type IV) where it can be found in a relation of 12 per 1000 amino acid residues. This implicates its probable use as a marker substance for collagen type IV metabolism. Up to now only hydrolyzed urine samples were examined for the presence of this amino acid, thus indicating total urinary excretion. We are reporting the free urinary 3 OHP content of neonates, infants and preschool children. Neonates (n = 23) showed mean excretion of 0.363 micrograms/mg creatinine (SD +/- 0.127), SEM 0.026, min. 0.197, max. 0.598. Infants (n = 11) showed mean excretion of 1.501 micrograms/mg creatinine (SD +/- 0.468), SEM 0.141, min. 0.908, max. 2.553. Preschool children (n = 16) had a mean excretion of 1.442 micrograms/mg creatinine (SD +/- 0.637), SEM 0.159, min. 0.318, max. 2.342. As determined by statistical calculations, neonates differed from infants and preschool children significantly (p less than 0.0005). Infants did not differ from preschool children, however (p less than 0.4). The reason for the low urinary excretion of 3 OHP in the neonatal period could be explained by the low enzyme activities which are physiologically observed in this developmental period in general and in the special case of a presumably low activity of the 3-prolyl-hydroxylase. Infants and preschool children presented levels 5 times higher than their neonatal mates, probably reflecting the increased 3-prolyl-hydroxylase activity which in turn is a marker of collagen synthesis.

[Ascorbic acid and hydroxyproline levels in the serum and granulation tissue of rats with aseptic and infected wounds].

Hydroxyproline and ascorbic acid were measured in wound tissues and ascorbic acid was determined in the blood sera of 230 mature male Wistar rats with aseptic and infected surface wounds on days 1-10 (daily), 12, and 15. The principal morphologic characteristics of the granulation tissue were assessed by semiquantitative methods. An infected wound is characterized by increased hydroxyproline levels in the granulation tissue and elevated ascorbic acid concentration in the blood serum. The granulation tissue ascorbic acid level augments during the first nine days in case of an aseptic wound and during twelve days in case of an infected wound. Morphologic and biochemical correlations are indicative of a relationship between the ascorbic acid level on the one hand and granulation tissue vascularization and fibroblast proliferation on the other.

[Urinary excretion of hydroxyproline in patients with lesions of the peripheral nervous system].

Seventy-one urinary total hydroxyproline determinations were carried out by the Prockop-Udenfried method in 61 patients with central nervous system damage (Bell's palsy, pareses of small muscles or muscle groups caused by damage to peripheral nerves or nerve roots, brachial plexus injury and motor polyneuropathy) and in 45 controls. Significantly raised urinary hydroxyproline excretion was found in cases of brachial plexus injury and severe motor polyneuropathy. This excretion in subgroups of Bell's palsy and damage to single nerves or roots was not significantly different from that in controls.
