The reentry hypothesis: the putative interaction of the frontal eye field, ventrolateral prefrontal cortex, and areas V4, IT for attention and eye movement.

Attention is known to play a key role in perception, including action selection, object recognition and memory. Despite findings revealing competitive interactions among cell populations, attention remains difficult to explain. The central purpose of this paper is to link up a large number of findings in a single computational approach. Our simulation results suggest that attention can be well explained on a network level involving many areas of the brain. We argue that attention is an emergent phenomenon that arises from reentry and competitive interactions. We hypothesize that guided visual search requires the usage of an object-specific template in prefrontal cortex to sensitize V4 and IT cells whose preferred stimuli match the target template. This induces a feature-specific bias and provides guidance for eye movements. Prior to an eye movement, a spatially organized reentry from occulomotor centers, specifically the movement cells of the frontal eye field, occurs and modulates the gain of V4 and IT cells. The processes involved are elucidated by quantitatively comparing the time course of simulated neural activity with experimental data. Using visual search tasks as an example, we provide clear and empirically testable predictions for the participation of IT, V4 and the frontal eye field in attention. Finally, we explain a possible physiological mechanism that can lead to non-flat search slopes as the result of a slow, parallel discrimination process.

On the development of ectopic eyes in Drosophila melanogaster produced by the mutation extra eye (ee).

The mutation ee often produces an ectopic eye on the vertex that is a mirror image partial duplication of the normal eye on the ipsilateral side of the head. The pattern of the duplication and a clonal analysis by mitotic recombination indicate that the duplications are of dorsal eye and orbital structures. Large ectopic eyes (more than 100 ommatidia) and their surrounding bristles may be produced without cuticular deficiencies. The penetrance of ee is temperature dependent with penetrance higher (72%) at 25 degrees and 29 degrees than at 19 degrees (43%). Temperature shift experiments show two temperature-sensitive periods: one at midembryogenesis, the other at mid-first larval instar. Microscopic examination of ee late-second and third instar imaginal cephalic discs show no indication of growth of the extra tissue needed to produce the duplication until after mid-third instar. This was confirmed by cell counts of ee and wild-type discs. There is no evidence of differential cell death in the two types of discs at this stage, although much earlier cell death is postulated. Tests for cell autonomy of the mutation by the production of morphogenetic clones suggest nonautonomy. Formation of pattern duplications by mutant genes is discussed in terms of cell death that eliminates whole developmental compartments, restricted cell death that occurs within a compartment, extensive cell death within a compartment and proliferative growth unassociated with cell lethality.

Draize rabbit eye test compatibility with eye irritation thresholds in humans: a quantitative structure-activity relationship analysis.

Draize rabbit eye test scores, as modified maximum average score (MMAS), for 68 pure bulk liquids were adjusted by the liquid-saturated vapor pressure P. These 68 adjusted scores, as log (MMAS/P), were shown to be completely equivalent to eye irritation thresholds (EIT), expressed as log (1/EIT), for 23 compounds in humans. Thus, for the first time the Draize eye test in rabbits for pure bulk liquids is shown to be perfectly compatible with eye irritation thresholds in humans. The total data set for 91 compounds was analyzed by the general solvation equation of Abraham. Values of log (MMAS/P) or log (1/EIT) could be fitted to a five-parameter equation with R2 = 0.936, SD = 0.433, AD = 0.000, and AAD = 0.340 over a range of 9.6 log units. When divided into a training set of 45 compounds, the corresponding equation could be used to predict the remaining 46 compounds in a test set with AD = -0.037 and AAD = 0.345 log units. Thus, the 91-compound equation can now be used to predict further EIT values to around 0.4 log units. It is suggested that the mechanism of action in the Draize test and in the human EIT involves passive transfer of the compound to a biophase that is quite polar, is a strong hydrogen bond base, a moderate hydrogen bond acid, and quite hydrophobic. The biophase does not resemble water or plasma, but resembles an organic solvent such as N-methylformamide.

Corneal preparation of eye bank eyes for experimental surgery.

PURPOSE: To evaluate the efficacy of dextran in balanced salt solution (BSS) as a preparation of eye bank corneas for experimental surgeries. METHODS: We used 12 eye bank eyes that were unsuitable for transplant. The corneas were removed from the globe and affixed to a Hanna artificial anterior chamber. Four concentrations of dextran-BSS (10%, 15%, 20%, and 30% [+/-0.56%]) were used to dehydrate the corneas, and corneal thickness was assessed with an ultrasonic pachymeter (+/-5 microm) at varying time-intervals over 3 hours. RESULTS: The corneas were thinned to an average thickness of 477+/-54 microm, 430+/-54 microm, 406+/-61 microm, and 391+/-52 microm at average times of 75+/-0 minutes, 85+/-23 minutes, 60+/-12 minutes, and 45+/-0 minutes for the 10%, 15%, 20%, and 30% concentrations, respectively. Corneal thickness was stabilized in both a 15% and a 20% solution. The data was fit to exponential curves until a minimum value was reached, after which linear regression analysis was used to determine the slopes of the data. The slopes for the 15% and 20% concentrations were not significantly different from 0 (p = 0.5 and p = 0.4, respectively). The slopes for each set of data from the 10% and 30% concentrations were significantly different from each other (p = 0.010 and p = 0.001, respectively). CONCLUSIONS: A solution of 20% dextran in BSS is effective for dehydrating eye bank corneas to a quasi-normal physiologic thickness and for maintaining the thickness for a sufficient amount of time. Both the posterior and anterior sides of the cornea should be exposed to the solution for 60 minutes.

Discharge of frontal eye field neurons during eye movements in unanesthetized monkeys.

Single unit activity was recorded from the frontal eye fields (area 8) in unanesthetized monkeys seated in a primate chair with the head restrained. The frontal eye field units were identified by antidromic response to stimulation of the cerebral peduncle. The findings indicate that most of the neurons discharge only after initiation of eye movements. These cells showed steady discharge when the eyes were immobile and oriented in a specific direction.

Localisation of SDF-1 and its receptor CXCR4 in retina and choroid of aged human eyes and in eyes with age related macular degeneration.

AIM: To examine the immunolocalisation of stromal cell derived factor 1 (SDF-1) and its receptor CXCR4 in aged control human donor eyes and eyes with age related macular degeneration (AMD). METHODS: Postmortem eyes from eight aged control donors (mean age 79.8 years) and from 12 donors with AMD (mean age 83.9 years) were cryopreserved and sectioned through the macular region. SDF-1 and CXCR4 were localised using streptavidin alkaline phosphatase immunohistochemistry and then sections were bleached. Three independent masked observers scored the immunohistochemical reaction product. RESULTS: In aged control retinas, SDF-1 immunoreactivity was most intense in inner photoreceptor matrix (IPM). CXCR4 showed a similar pattern of immunostaining, but was more prominent in inner segments of photoreceptors. In aged control and AMD choroid, SDF-1 and CXCR4 localisations were most prominent in retinal pigment epithelial (RPE) cells and choroidal stroma. However, the intensity for SDF-1 was significantly reduced in RPE (p < 0.0001) and choroidal stroma (p < 0.05) in late AMD eyes. SDF-1 and CXCR4 immunoreactivities were weak or nearly absent in disciform scars with choroidal neovascularisation (CNV). Circulating cells, presumably leucocytes, were most intensely positive for CXCR4. CONCLUSIONS: These results show that changes in distribution and relative levels of SDF-1/CXCR4 were not evident in early AMD. This suggests that SDF-1/CXCR4 may not contribute to the formation of CNV in AMD, in that CXCR4+ cells were not incorporated into neovascularisation. However, the examples of CNV studied were within disciform scars, so the authors cannot comment on the role of SDF-1/CXCR4 in the early stages of CNV formation.

Eye position and electromyographic observation of squint eyes under general anesthesia.

The position of eyes under general anesthesia was measured in different groups of nonparalytic strabismus. Almost all patients with esotropia or hyperactive inferior oblique muscle showed divergent eye positions while patients with exotropia showed no consistent trend. The eye position of patients with lid ptosis without squint, which served as a control group, were divergent in all cases. Electromyographic observation of the medial rectus muscle under the same condition of anesthesia revealed that muscle discharge disappeared almost completely at the stage at which the eyes were in a well-stabilized position.

Visual field accuracy and eye movement direction: a child's eye view.

Post-exposural eye movements were studied in 32 adults and 24 7-yr.-old children. Stimuli were binary figures exposed tachistoscopically in both visual fields simultaneously. The data showed significant correlations between direction of eye movement and locus of recognition for both children and adults. No significant differences were found in frequencies of eye movements of children and adults. The data are interpreted in terms of the facilitative effects of post-exposural eye movements upon perception for both groups.

The Tehran Eye Study: research design and eye examination protocol.

BACKGROUND: Visual impairment has a profound impact on society. The majority of visually impaired people live in developing countries, and since most disorders leading to visual impairment are preventable or curable, their control is a priority in these countries. Considering the complicated epidemiology of visual impairment and the wide variety of factors involved, region specific intervention strategies are required for every community. Therefore, providing appropriate data is one of the first steps in these communities, as it is in Iran. The objectives of this study are to describe the prevalence and causes of visual impairment in the population of Tehran city; the prevalence of refractive errors, lens opacity, ocular hypertension, and color blindness in this population, and also the familial aggregation of refractive errors, lens opacity, ocular hypertension, and color blindness within the study sample. METHODS DESIGN: Through a population-based, cross-sectional study, a total of 5300 Tehran citizens will be selected from 160 clusters using a stratified cluster random sampling strategy. The eligible people will be enumerated through a door-to-door household survey in the selected clusters and will be invited. All participants will be transferred to a clinic for measurements of uncorrected, best corrected and presenting visual acuity; manifest, subjective and cycloplegic refraction; color vision test; Goldmann applanation tonometry; examination of the external eye, anterior segment, media, and fundus; and an interview about demographic characteristics and history of eye diseases, eye trauma, diabetes mellitus, high blood pressure, and ophthalmologic cares. The study design and eye examination protocol are described. CONCLUSION: We expect that findings from the TES will show the status of visual problems and their causes in the community. This study can highlight the people who should be targeted by visual impairment prevention programs.

Video-based eye tracking: our experience with Advanced Stimuli Design for Eye Tracking software.

We present an independent, flexible, and easily programmable software program for generating a wide set of visual stimuli paradigms in eye-movement studies. The software, called ASTIDET (Advanced Stimuli Design for Eye Tracking), has been interfaced in real time with a high speed video-based eye tracking system in order to obtain a reliable measurement of saccades. Two saccadic paradigms have been tested (gap and memory guided tasks) in 10 normal subjects. The preliminary results confirm that ASTIDET is a user-friendly software and can be interfaced with a video-based eye-tracking device in order to obtain reliable measurement of saccades.

Studies on eye irritation caused by chemicals in rabbits--1. A quantitative structure-activity relationships approach to primary eye irritation of chemicals in rabbits.

A quantitative structure-activity relationship (QSAR) approach has been applied to the analysis of the relation between the structural features of chemicals and the primary eye irritation in rabbits. One hundred thirty one heterogeneous chemicals including medicals, pesticides, detergents and organic solvents were used in this study. The eye irritation ratings were made in three classes on the basis of the recovery time of corneal and conjunctival damages. Thirty six descriptors were used to describe the molecules. To correlate eye irritation ratings with the descriptors, a QSAR model was formulated by the adaptive least-squares method. A three-class discrimination was made as follows; class I included 23 chemicals which induced the damages recovering within 24 hr, class II included 64 chemicals which induced the damages persisting for more than 24 hr but recovering within 21 days, class III included 44 chemicals which induced the damages not recovering within 21 days. The discriminant function included 18 descriptors. The accuracy in classifying the chemicals was 86.3% in the recognition and 74.0% in the leave-one-out prediction. These results suggest that QSAR analysis is valuable to predict the primary eye irritation of chemicals.

Studies on eye irritation caused by chemicals in rabbits--II. Structure-activity relationships and in vitro approach to primary eye irritation of salicylates in rabbits.

Structure-activity relationships and in vitro evaluation of eye irritation potential of salicylates in rabbits were studied. The primary eye irritation potential of ten salicylates was evaluated according to Draize method. The effects of chemicals on model protein and lipid were investigated in vitro. The effects of chemicals on the protein could be detected by the production of aggregates of human serum gamma-globulin (HSG) and a good correlation was obtained between the ability of salicylates to produce aggregation of HSG and the potential of corneal irritation. The effects on the lipid could be detected by the adhesion potential of chemicals on lipid membrane and a linear correlation was not obtained between the adhesionary effects of salicylates on lipid membrane and the potential eye irritation. The corneal irritation and protein aggregation potential of salicylates were correlated with the acid dissociation constant more closely than octanol/water partition coefficient. The destruction of alpha-helix of proteins in corneal surface by salicylates were observed from the nondestructive structural analysis of corneal surface by Fourier Transform (FT)-IR spectroscopy. These results suggest that eye irritation caused by salicylates are mainly the results of denaturation of proteins in ocular tissue and that the effects on protein depend on the dissociation potential of molecules.

Distributed but convergent ordering of corticostriatal projections: analysis of the frontal eye field and the supplementary eye field in the macaque monkey.

The degree of parallel processing in frontal cortex-basal ganglia circuits is a central and debated issue in research on the basal ganglia. To approach this issue directly, we analyzed and compared the corticostriatal projections of two principal oculomotor areas of the frontal lobes, the frontal eye field (FEF) and the supplementary eye field (SEF). We first identified cortical regions within or adjacent to each eye field by microstimulation in macaque monkeys and then injected each site with either 35S-methionine or WGA-HRP conjugate. We analyzed the corticostriatal projections and also the interconnections of the pairs of cortical areas. We observed major convergence of the projections of the FEF and the SEF within the striatum, principally in the caudate nucleus. In cross sections through the striatum, both projections were broken into a series of discontinuous input zones that seemed to be part of complex three-dimensional labyrinths. Where the FEF and SEF projection fields were both present, they overlapped patch for patch. Thus, both inputs were dispersed within the striatum but converged with one another. Striatal afferents from cortex adjacent to the FEF and the SEF did not show convergence with SEF and FEF inputs, but did, in part, converge with one another. For all pairs of cortical areas tested, the degree of overlap in the corticostriatal projections appeared to be directly correlated with the degree of cortical interconnectivity of the areas injected. All of the corticostriatal fiber projections observed primarily avoided immunohistochemically identified striosomes. We conclude that there is convergence of oculomotor information from two distinct regions of the frontal cortex to the striatal matrix, which is known to project into pallidonigral circuits including the striatonigrocollicular pathway of the saccadic eye movement system. Furthermore, functionally distinct premotor areas near the oculomotor fields often systematically projected to striatal zones adjacent to oculomotor field projections, suggesting an anatomical basis for potential interaction of these inputs within the striatum. We propose that parallel processing is not the exclusive principle of organization of forebrain circuits associated with the basal ganglia. Rather, patterns of both convergence and divergence are present and are likely to depend on multiple functional and developmental constraints.

Prediction of severe eye injuries in automobile accidents: static and dynamic rupture pressure of the eye.

The purpose of this paper is to determine the static and dynamic rupture pressures of 20 human and 20 porcine eyes. This study found the static test results show an average rupture pressure for porcine eyes of 1.00 +/- 0.18 MPa while the average rupture pressure for human eyes was 0.36 +/- 0.20 MPa. For dynamic loading, the average porcine rupture pressure was 1.64 +/- 0.32 MPa, and the average rupture pressure for human eyes was 0.91 +/- 0.29 MPa. Significant differences are found between average rupture pressures from all four groups of tests (p = 0.01). A risk function has been developed and predicts a 50% risk of globe rupture at 1.02 MPa, 1.66 MPa, 0.35 MPa, and 0.90 MPa internal pressure for porcine static, porcine dynamic, human static, and human dynamic loading conditions, respectively.

[The nature and results of the treatment of eye burns based on data from the Chernovtsy Province Eye Trauma Center].

The character and results of eye burn treatment have been studied in 226 patients (307 eyes) aged from 10 to 80 years. The analysis of the character of eye burns has shown that the overwhelming majority of them were chemical ones (80.54%), of them 50% being alkaline. As to degree of severity, burns of moderate degree predominated (58.74%). The greatest part of the sufferers had everyday-life burns (67.7%). Results of treatment depended, mainly, on the severity of the burn. By the results of treatment, the patients were divided into 4 groups: group I included patients with total restoration of corneal transparency, who can return to their former labor activity; group-II patients with partially lost visual functions on account of remained, moderately expressed cicatricial opacifications of the cornea, who in future require repeated courses of resolving therapy; group-III patients with a remarkable loss of visual functions on account of coarse cicatricial opacifications of the cornea, who in future require surgical treatment--keratoplasty, keratoprosthesing and other surgical interventions; group IV-patients with total restoration of corneal transparency after treatment, but with insufficiently high visual acuity because of associated changes in the optic media and the eye fundus (beginning cataract, macular dystrophy, etc.), who require dispensary observation and treatment of associated diseases.

Corneal flattening by shallow circular trephination in human eye bank eyes.

Recent reports documenting central keratometric changes after removal of failed epikeratoplasty lenticules, compared to preoperative keratometry measurements, suggest that the annular corneal wound alters corneal curvature. Central corneal steepening has also been reported following circular and hexagonal keratotomy. We performed standard epikeratoplasty trephination with a Hessburg-Barron suction trephine followed by a peripheral lamellar spreading keratotomy on seven human eye bank eyes to determine the effect of these incisions on corneal topography. In seven human eye bank eyes, the mean acute central keratometric flattening from the shallow trephine incision was 2.81 D (SD 2.28, P = .017), with no significant change in keratometry due to peripheral lamellar spreading (P = .916). Computerized numeric and three-dimensional graphic analysis of the keratographs demonstrated this central topographic flattening. Further studies are needed to investigate the reversibility of host corneal changes induced by epikeratoplasty procedures.

Changes in the contrast sensitivity of an amblyopic eye after losing the other normal eye.

A man aged 17 had an amblyopic left eye with central fixation and lost his normal right eye by an accident. After the accident, spontaneous improvement of the visual acuity of the amblyopic left eye was rapid. Just after the accident, overall contrast sensitivity of the left eye was considerably less than that of a normal adult. Improvement of the contrast sensitivity was recorded first at low spatial frequencies and then at high spatial frequencies. However, the contrast sensitivity at medium spatial frequencies remained less than that of a normal adult. These results suggest that lateral inhibitory processes may be relatively undeveloped or receptive fields do not develop to be as small as those of normal adults.
