Influence of aspirin and/or anabolic steroid (nandrolone decanoate) on experimental metastasis formation.

The influence on transpulmonary passage by an inhibitor of platelet aggregation, aspirin, and/or anabolic steroid, (Nandrolone decanoate), was investigated in syngeneic CBA mice. An increase of extrapulmonary metastases was used as a measure of increased redistribution of tumor cells from lungs to other organs. Aspirin alone did not reduce metastasis formation in lungs and did not significantly increase transpulmonary passage of tumor cells. The anabolic steroid increased the metastasis crop only in the lungs. However, treatment with the two compounds together increased the metastases in lungs and extrapulmonary organs. The results disclosed that in this system inhibition of platelet aggregation did not reduce metastasis formation but in combination with another drug, an anabolic steroid, it increased the number of experimental metastases.

The social value of fluphenazine decanoate.

The Medical Research Council (England) conducted a double blind placebo controlled trial of fluphenazine decanoate as a form of maintenance therapy for schizophrenic out-patients. The results clearly demonstrated the clinical and social value of the drug in minimizing positive psychotic features, in preventing relapse and thereby alleviating distress amongst the parents of single patients and the wives of married patients. The drug allows the patients to fulfill normal social role expectations.

Haloperidol decanoate (Janssen) in the treatment of sexual deviations.

Haloperidol decanoate inj. (Janssen) á 150 mg was evaluated during long-term (max. 4.2 years) treatment of 30 sexual offenders. We proved that the drug was well tolerated also by non-psychotic patients by using of low doses (37.5 mg) with intervals between injections 6 weeks. Extrapyramidal side effects were on minimal level. "Side" effect on sexual functions and appetence turned to be the main effect of the drug. The influence on sexual reactivity was objectively examined by electrocapacitative phalloplethysmograph (Figar). There were detected the latency of initial phase of the PPG curve and prolongation of the incremental slope. The decremental slope of the PPG curve was shorter.

Haloperidol decanoate (Janssen) in the treatment of schizophrenia.

Haloperidol decanoate inj. (Janssen) á 150 mg was evaluated during long-term (max. 4 years) treatment of 24 schizophrenics. The drug was very good tolerated locally and generally. Only lower doses of drug was sufficient to keep stabilized clinical state during the maintenance therapy (37.5-75 mg) with intervals 4-6 weeks. The occurrence of side effects gradually decreased similarly as a necessity to use antiparkinsonics. The maintenance treatment helped the patients to resume their social and working positions - 68% restarted working.

Serum neuroleptic levels during reduced dose fluphenazine decanoate maintenance therapy.

Forty-one remitted and chronically psychotic schizophrenic out-patients completed a two-year clinical trial during which they were assigned, on the basis of their clinically determined maintenance dosages, to one of two reduced, fixed-dose fluphenazine decanoate (FD) regimens: 35 mg/4 wks (19 patients) or 10 mg/4 wks (22 patients). Eighty-one percent of chronically psychotic patients, who represented 74% of the high dose group, relapsed, in comparison with only 38% of remitted patients (p < .001), who represented 86% of the low dose group. During this study serum neuroleptic levels were assessed, using the radioreceptor assay, before the administration of each FD injection and whenever a patient relapsed. Overall, 334 serum neuroleptic activity measurements were performed. Serum neuroleptic levels were detectable in all patients and were higher, although not significantly, in the 35 mg/4 wks group. The dichotomous clinical outcome of chronically psychotic and remitted patients occurred within the framework of essentially similar serum neuroleptic levels. These findings suggest that: 1) serum neuroleptic levels can be monitored during low dose FD treatment, 2) the poor maintenance therapy outcome of chronically psychotic patients cannot be accounted for by inadequate neuroleptic bioavailability, 3) a majority of remitted FD maintained patients retain their clinical response at serum neuroleptic levels lower than those initially attained at steady state.

A model for studies of dermal surface epithelization: with observations on the effects of dexamethasone and nandrolone decanoate.

Experiments were designed to study the events of cell proliferation and migration during epithelization of an intact dermal surface in man. Epidermis was removed by prolonged suction and epithelization was followed by serial skin biopsies. DNA-synthesis and migration of labelled cells were followed by radioautography. Migration and cell proliferation were not found to be exclusive events in this model. Dexamethasone applied locally in a cream base was found to delay epithelization and nandrolone decanoate was found to counteract this to some extent. The model can be used in studies on effects of drugs on events involved in epithelization.
