Determination of non-ortho polychlorinated biphenyls in environmental Standard Reference Materials.

The concentrations of three non-ortho ("coplanar") polychlorinated biphenyls, 3,3',4,4'-tetrachlorobiphenyl (IUPAC PCB 77), 3,3',4,4',5-pentachlorobiphenyl (IUPAC PCB 126), and 3,3',4,4',5,5'-hexachlorobiphenyl (IUPAC PCB 169), were determined in five NIST Standard Reference Materials (SRMs) of environmental and biological interest. The measured levels were approximately between (0.2 to 1.3) ng/g in SRM 1588a (Organics in Cod Liver Oil), (0.3 to 9) ng/g in SRM 1944 (New York/New Jersey Waterway Sediment), (0.2 to 0.4) ng/g in SRM 1945 (Organics in Whale Blubber), (1 to 18) ng/g in SRM 2974 (Organics in Freeze-dried Mussel Tissue [Mytilus edulis]), and (0.1 to 0.4) ng/g in candidate SRM 1946 (Lake Superior Fish Tissue). PCB 169 was present at < 0.1 ng/g in SRMs 1944 and 2974.

Capillary gas chromatography combined with atomic emission detection for the analysis of DDT and metabolites.

Capillary gas chromatography with atomic emission detection (GC-AED) was evaluated for the determination of DDT and metabolites in biological samples. Utilizing chlorine-selective detection at 479 nm, DDT, DDE, and DDD were quantified down to the 20 ng/g (fat weight) level (S/N = 10) in cod liver oil, while the detection, limit was 6 ng/g (S/N = 3). With splitless injection performed in the pressure programmed mode, DDT and related compounds were quantified based on a single chlorine calibration curve (universal calibration). The quantitative data obtained by GC-AED were in excellent accordance with similar results from capillary gas chromatography with electron capture detection (GC-ECD), while the procedure for calibration was simplified with the former technique.

Diet and risk of cutaneous malignant melanoma: a prospective study of 50,757 Norwegian men and women.

The relationship between dietary habits and subsequent risk of cutaneous malignant melanoma (CMM) was studied in 25,708 men and 25,049 women aged 16-56 years attending a Norwegian health screening in 1977-1983. Linkage to the Cancer Registry of Norway and the Central Bureau of Statistics of Norway ensured a complete follow-up until December 31, 1992. Diet was recorded through a semi-quantitative food-frequency questionnaire at the time of screening, and 108 cases of CMM were identified during follow-up. Use of cod liver oil supplementation and intake of polyunsaturated fat were associated with significant increased risk and drinking coffee with significant decreased risk of CMM in women. Adjusting for height, body mass index, body surface area, education, smoking or occupational or recreational physical activity did not change the results. No significant association was found between the incidence of CMM and any of the dietary factors in men. Important aspects are residual confounding by sun exposure and social class, as well as concern with multiple comparisons.

Estimation of essential fatty acid requirements of common carp larvae using semi-purified artificial diets.

Two trials were conducted with duplicate groups of (first feeding) carp larvae fed artificial dry diets based on casein and dextrin over 21 or 25 days. One control diet based on yeast was also tested. Survival, growth and fatty acid profiles of larvae were studied. In trial 1, (n-3) fatty acid requirement was estimated using diets supplemented or not with methyl linolenate or cod liver oil. After 21 days, the best survival and growth were observed in larvae fed the unsupplemented diet [(n-3) fatty acid level: 0.05%]. Survival and growth were not improved by higher levels of (n-3) fatty acids. In trial 2, (n-6) fatty acid requirement was estimated using diets with graded levels of methyl linolenate or peanut oil. After 25 days, the best survival and growth were obtained with diets supplemented with 0.25% methyl linolenate (total (n-6) fatty acid level: 1%) or with 1.25% peanut oil (total (n-6) fatty acid level: 0.89%). Survival and growth were not improved by higher levels of (n-6) fatty acids. Fatty acid composition of carp reflected that of the diets and also showed that carp larvae are capable of elongating and desaturating linolenic acid and linoleic acid in longer chain fatty acids.

Endothelium-dependent relaxation in response to aggregating platelets in porcine femoral veins and its modulation by diet.

The present study examined the protective role of the venous endothelium against aggregating platelets and its modulation by diet. Yorkshire pigs were fed a regular chow (control pigs), 2% high-cholesterol diet (for 10 weeks, cholesterol-fed pigs), and regular chow plus cod-liver oil (30 ml/day for 4 weeks, oil-fed pigs). Endothelium-dependent responses were examined in vitro in rings of femoral veins in the presence of the inhibitor of cyclooxygenase indomethacin. In control pigs, aggregating platelets, serotonin, and ADP caused endothelium-dependent relaxations. The platelet-induced relaxations were attenuated by methiothepin (a combined 5-HT1 and 5-HT2 serotonergic blocker) or apyrase (an ADPase and ATPase) and were abolished by the combination of the two agents. In quiescent rings, platelets caused contractions, which were reduced in the presence of endothelium; the contractions were prevented by ketanserin (a 5-HT2 serotonergic blocker) or methiothepin but not by R 68 070 (a thromboxane A2 receptor blocker) or dazoxiben (a thromboxane-synthetase blocker). In cholesterol-fed pigs, the platelet-induced relaxations were not altered, whereas in oil-fed pigs, the endothelium-dependent relaxations to platelets, serotonin, and ADP were augmented. Platelet-induced contractions were significantly reduced in rings with endothelium from oil-fed pigs, whereas the contractions were comparable in rings without endothelium among the three groups. Endothelium-dependent relaxations in response to the calcium ionophore A23187, direct relaxations in response to sodium nitroprusside, and direct contractions in response to potassium chloride were comparable among the three groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Thromboxane A3 (TXA3) is formed in human platelets after dietary eicosapentaenoic acid (C20:5 omega 3).

Platelet-rich plasma of subjects, who had ingested cod liver oil containing 10% eicosapentaenoic acid (C20:5 omega 3), the precursor of trienoic prostanoids, was stimulated ex vivo with collagen. Formation of thromboxane B3, the hydrolysis product of non-aggregatory thromboxane A3, from endogenous eicosapentaenoic acid was demonstrated by combined capillary gas chromatography-mass spectrometry. Concomitantly platelet aggregation in platelet-rich plasma upon low doses of collagen and associated thromboxane B2 formation from endogenous arachidonic acid were reduced. We conclude that both the formation of inactive thromboxane A3 as well as the reduction of thromboxane A2 may contribute to the reduced platelet reactivity after dietary eicosapentaenoic acid.

[Correction of disorders of cardiac electric stability in post-infarction cardiosclerosis using a diet enriched with polyunsaturated fatty acids].

In a 14-20 days after feeding of rats with "eikonole" (cod-liver oil, deduced from skeletal muscle and enriched by eicosapentaenic and docosahexaenic fatty acids). Myocardial infarction by Selie has been induced and in a 30 days of the same feeding, contractile function and heart electrical stability in situ have been investigated. Eikonole increased heart fibrillation threshold by 50%, decreased the frequency of spontaneous extrasystole in 3 times and ectopic activation (extrasystole), induced by n. vagus stimulation, in 2.5 times. Eikonole increased the intensity of structures' functioning in the state of relative physiological rest, but did not influence essentially on pressure and the velocity of myocardial contraction and weakening.

Distribution and mobility of omega 3 fatty acids in rainbow trout fed varying levels and types of dietary lipid.

The availability of essential fatty acids in fish neutral lipid to tissue phospholipids was determined under conditions of adequate and inadequate essential fatty acid intake as well as during fasting. Juvenile rainbow trout were fed a semi-purified diet containing varying levels of cod liver oil, with or without supplementary olein. Fatty acid analysis indicated that in all treatments the neutral lipid pool was not turned over during feeding but was enhanced by exogenous or endogenously synthesized fatty acids. Fish that received diets devoid of essential fatty acids maintained virtually all of the docosahexenoic acid originally present in each lipid pool. Fish fed diets containing essential fatty acids deposited them in proportion to the dietary levels. After a 4-week fast, no change was noted in the relative levels of fatty acids in neutral lipid indicating that all fatty acids in neutral lipid were catabolized equally--including essential fatty acids. During fasting there was a selective retention of docosahexenoic and linoleic acids in the phospholipid pool.

Preparation of fatty acid methyl esters for gas-chromatographic analysis of marine lipids: insight studies.

Assays for fatty acid composition in biological materials are commonly carried out by gas chromatography, after conversion of the lipid material into the corresponding methyl esters (FAME) via suitable derivatization reactions. Quantitative derivatization depends on the type of catalyst and processing conditions employed, as well as the solubility of said sample in the reaction medium. Most literature pertinent to derivatization has focused on differential comparison between alternative methods; although useful to find out the best method for a particular sample, additional studies on factors that may affect each step of FAME preparation are urged. In this work, the influence of various parameters in each step of derivatization reactions was studied, using both cod liver oil and microalgal biomass as model systems. The accuracies of said methodologies were tested via comparison with the AOCS standard method, whereas their reproducibility was assessed by analysis of variance of (replicated) data. Alkaline catalysts generated lower levels of long-chain unsaturated FAME than acidic ones. Among these, acetyl chloride and BF(3) were statistically equivalent to each other. The standard method, which involves alkaline treatment of samples before acidic methylation with BF(3), provided equivalent results when compared with acidic methylation with BF(3) alone. Polarity of the reaction medium was found to be of the utmost importance in the process: intermediate values of polarity [e.g., obtained by a 1:1 (v/v) mixture of methanol with diethyl ether or toluene] provided amounts of extracted polyunsaturated fatty acids statistically higher than those obtained via the standard method.

Dietary lipid requirements of adult lobsters, Homarus americanus (M.E.).

The effects of several dietary lipids on adult American lobster (Homarus americanus) were assessed over a 10 month feeding period. Cod liver oil (CLO) resulted in greater precent weight gains, feed conversion, percent edible meat and higher serum protein and hemocyte counts than either corn oil (CO) or hydrogenated coconut oil (HCO). These differences were probably due to an essential fatty acid (EFA) requirement by the lobster for linolenic series omega3 or other fatty acids present in CLO. The linoleic or omega6 fatty acids of CO appeared to have some sparing effect on several of the EFA deficiency symptoms. It was found that 5% CLO was optimal for mean percent weight gain, molt incidence, feed conversion and hemocyte counts of lobsters. Further increases in dietary CLO to 10% and 15% resulted in no significant improvement of any of the condition indices used. There was a decrease in serum protein and calcium when lobsters were fed a non-saponifiable sterol deficient diet. The addition of 1% cholesterol to the diet raised the serum protein, but resulted in even a greater decrease in the serum calcium level.

[Clinical evaluation of aiconol side effects].

In the study of 164 patients with atherogenic dyslipidemia the authors studied side effects of aiconol, concentrated cod liver oil. Its lipid component contains not less than 18% of omega-3 polyunsaturated fatty acids (PUFA). All the patients were divided into 4 groups: 51 subjects of group 1 received aiconol in the form of oil, 35 subjects of group 2 had protein dietetic caviar prepared from animal proteins with addition of 3% aiconol, 38 subjects of group 3 were given protein-oil emulsion containing 20% aiconol and 40 patients of group 4 took gelatin aiconol in capsules. Side effects of the above food ingredients enriched with omega-3 PUFA were found to be of metabolic nature. These manifested most evidently in patients on oil aiconol incorporating both maximal doses of omega-3 PUFA and the largest amounts of inert constituents acting as additional fat load on the liver and pancreas. Group 1 patients also suffered from aggravated chronic gastrointestinal, hepatic and pancreatic disorders. Because of reduced quantities of omega-3 PUFA in the diets of group 2, 3 and 4 patients, they displayed much less severe side effects.

The association between parathyroid hormone, vitamin D and bone mineral density in 70-year-old Icelandic women.

Parathyroid hormone (PTH) may be an important determinant of cortical bone remodeling in the elderly. Vitamin D status is one of the determining factors in this relationship. The aim of this study was to quantify the relationship between serum PTH, vitamin D and bone mineral density (BMD) in elderly women in Reykjavik (64 degrees N), where daily intake of cod liver oil is common and mean calcium intake is high. In PTH correlated inversely with 25(OH)D (r = -0.26, p<0.0 1). In multivariate analysis PTH correlated inversely with whole body BMD (mostly cortical bone) (R2 = 2.2%, p = 0.04) but not with the lumbar spine BMD, reflecting more cancellous bone. No association was found between 25(OH)D levels and BMD at any site in univariate or multivariate analysis. Osteocalcin, a measure of bone turnover, was negatively associated with BMD and this association remained significant when corrected for PTH levels. In summary, in this fairly vitamin D replete population with high calcium intake, PTH was negatively associated with total body BMD. We infer that suppression of PTH may reduce cortical bone loss, but other factors are likely to contribute to age-related bone remodeling and osteoporosis.

Quantitation of volatiles and nonvolatile acids in an extract from coffee beverages: correlation with antioxidant activity.

The antioxidant activities of a commercial brewed coffee were investigated by measuring malonaldehyde (MA) formation from oxidized cod liver oil using a gas chromatographic method (MA-GC assay) and a thiobarbituric acid method (TBA assay). The highest antioxidant activity obtained by the MA-GC assay was from regular whole brewed coffee (97.8%) at a level of 20%, and the highest antioxidant activity obtained by the TBA assay was from decaffeinated whole brewed coffee (96.6%) at a level of 5%. Among 31 chemicals identified in a dichloromethane extract, guaiacol, ethylguaiacol, and vinylguaiacol exhibited antioxidant activities, which were comparable to that of alpha-tocopherol. Among nine chlorogenic acids (three caffeoylquinic acids, three feruloylquinic acids, and three dicaffeoylquinic acids) identified, 5-caffeoylquinic acid contained the greatest amount both in regular (883.5 microg/mL) and in decaffeinated (1032.6 microg/mL) coffees; it exhibited 24.5% activity by the MA-GC assay and 45.3% activity by the TBA assay at a level of 10 microg/mL. Caffeic and ferulic acids showed moderate antioxidant activities in both assays.

Long-term patency of regenerated neoaortic wall following the implant of a fully biodegradable polyurethane prosthesis: experimental lipid diet model in pigs.

The degradation of a polyurethane vascular prosthesis, the time course and characteristics of tissue ingrowth and prosthesis replacement, and the incidence of thrombosis have been investigated in 50 young pigs. A 6-7 cm long cone-shaped prosthesis was implanted in the infrarenal aorta of 50 young growing pigs which were sacrificed at intervals from 60 to 365 days. Ten animals were controls, 25 were given aspirin 10 mg/kg of body weight/day, and 15 were fed with cod liver oil. Aortography, done at 30, 60, and 90 days, showed 100% of implants were thrombosed in the surviving controls, 55% of the aspirin group had patent grafts, and 100% of animals in the lipid diet group had patent grafts. At final graft retrieval, the aspirin group showed only three patencies, while in the lipid diet group seven out of nine animals were patent. The laboratory and morphological studies indicated that all patent prostheses were lined with tissue that resembled the intima of native aorta with a layer of smooth muscle cells which appeared complete at 180 days. From these data we conclude that the biodegradable polyurethane vascular prosthesis is reliable for experimental implants in a pig model. The lipid-rich diet of polyunsaturated fatty acids potentiates long-term patency, perhaps by preventing platelet aggregation and thrombosis, and allowing the growth of a neoendothelium and neomedia within the lumen of the prosthesis, which slowly degrade towards fatty tissue and form neoadventitia.

Polybrominated diphenyl ether congeners and toxaphene in selected marine standard reference materials.

Polybrominated diphenyl ether (PBDE) congeners and components of the complex mixture toxaphene are stable in the environment and readily bioaccumulated into wildlife and human tissues. PBDEs are presently used in large quantities worldwide as flame retardants in textiles, furniture, computer equipment, and cables. Toxaphene is a complex mixture of chlorinated bornanes and bornenes that was the most heavily used pesticide in the United States until it was banned in 1982; however, some countries continue to use toxaphene. The National Institute of Standards and Technology has quantified PBDE congeners and toxaphene in several available Standard Reference Materials (SRMs) using methods of gas chromatography with electron impact mass spectrometry (GC-EI-MS) and GC negative chemical ionization (NCI) MS, respectively. SRM 1588a Organics in Cod Liver Oil and SRM 1945 Organics in Whale Blubber were examined for PBDE congeners 47, 99, 100, 153, and 154, total toxaphene, and toxaphene congeners 26, 50, and 62. SRM 1946 Lake Superior Fish Tissue was also examined for total toxaphene and toxaphene congeners. The sum of the PBDE congeners (mean, (1 SD) wet basis) for SRM 1945 was 150 ng g(-1) (7 ng g(-1)). The concentration of PBDE 47 in SRM 1588a was 82.7 ng g(-1) (2.8 ng g(-1)). Other PBDEs were detected in SRM 1588a but were not quantified due their low levels. The total toxaphene (wet mass basis) was 1,210 ng g(-1) (127 ng g(-1)), 1,960 ng g(-1) (133 ng g(-1)), and 3,980 ng g(-1) (248 ng g(-1)) in SRMs 1945, 1946, and 1588a, respectively. The values for PBDEs and toxaphene determined in the SRMs, while not certified, indicate that the SRMs will be suitable control materials for PBDE and toxaphene analyses.

[Ultrastructure and morphogenesis of ceroid pigment. I. Phagocytosis and formation of lipid-containing lysosomes in Kupffer Cells after intravenous injection of unsaturated lipids (author's transl)].

Wistar rats were injected intravenously with cod liver oil emulsion. The phagocytosis of these lipids by the Kupffer cells as well as the formation of lipid-containing lysosomes were studied electron microscopically within a period of 5 min to 12 hrs. The emulsified lipids are incorporated into the cytoplasm by membrane-vesiculation. The lipid-containing phagocytic vacuoles (lipophagosomes) were transformed into secondary lysosomes (lipophagolysosomes). This change occurs in two phases. Immediately after phagocytosis the lipophagosomes are provided with lysosomal enzymes by fusion with pre-existent lysosomes (dense bodies) of the Kupffer cells. These are mainly the so-called small homogenous dense bodies which are probably primary lysosomes of the type of "storage granules". In the second phase which begins 15-30 min after the injection; the fusion of lipophagosomes with newly produced primary lysosomes within the Golgi apparatus is predominant. Therefore, the enzyme supply of the phagocytic vacuoles does not occur at once but in small amounts during a longer period. The lipids slowly change into an amorphous electron-dense mass 12 hrs after the injection. Obviously, these changes are the first step to the formation of ceroid pigment.

[Does supplementation of essential fatty acids influence the visual acuity and cognitive function of term infants?].

BACKGROUND: Norwegian health authorities recommend supplementation of essential fatty acids such as cod liver oil to term infants from four weeks of age. The essential fatty acids are seen as contributing to optimal development of the visual acuity and cognitive functions. This study reviews recent research on this issue. MATERIAL AND METHODS: We assessed six randomized controlled trials, published in 1998 and 1999, of which two evaluated visual function and four cognitive development. Outcome were measured by a number of standardized tests. We compared the results from our review with results from a Cochrane review which evaluated six studies published from 1995 to 1997. RESULTS: Only one of the tests used in the visual acuity studies showed significant effect at three out of the four age-related points in time at which the test was administered. Only one of the tests used in the cognitive development studies showed significant effect. INTERPRETATION: The studies reviewed here are in accordance with previous research and do not provide evidence for the claim that supplementation with essential fatty acids as such is significant in order to achieve optimal visual function and cognitive development in healthy term infants.

Serum enriched with n-3 polyunsaturated fatty acids inhibits procoagulant activity in endothelial cells.

Nine healthy male volunteers (mean age 24.3 years, range 21-27 years) ingested 25 ml cod liver oil (CLO) daily for 8 weeks. Blood samples were collected before and after the intervention period. A slight increase in total serum cholesterol (+ 12.2%, P less than 0.01) and high density lipoprotein cholesterol (+ 16.9%, P less than 0.01) were calculated to have a non-significant effect on the high/low density lipoprotein ratio. Polyunsaturated fatty acids in serum were displaced from the n-6 family to the n-3 family, reflected by an eight-fold increase of the eicosapentaenoic (20:5 n-3)/arachidonic (20:4n-6) acid ratio. Human umbilical vein endothelial cells were grown in a medium to which 30% sterile filtered serum, collected before and after CLO supplementation for 72 h, was added. After 2 h exposure to lipopolysaccharides the thromboplastin activity in endothelial cultures incubated with serum enriched with n-3 fatty acids was 43% lower than in cultures containing serum collected before the intervention (1.86 +/- 0.48 10(-3)/10(6) cells vs 3.26 +/- 0.85 10(-3)/10(6) cells). It is suggested that serum prepared from subjects given n-3 fatty acids may support the thromboresistence of endothelial cells.
