Cardiovascular expression of the mouse WNK1 gene during development and adulthood revealed by a BAC reporter assay.

Large deletions in WNK1 are associated with inherited arterial hypertension. WNK1 encodes two types of protein: a kidney-specific isoform (KS-WNK1) lacking kinase activity and a ubiquitously expressed full-length isoform (L-WNK1) with serine threonine kinase activity. Disease is thought to result from hypermorphic mutations increasing the production of one or both isoforms. However, the pattern of L-WNK1 expression remains poorly characterized. We generated transgenic mice bearing a murine WNK1 BAC containing the nlacZ reporter gene for monitoring L-WNK1 expression during development and adulthood. We observed previously unsuspected early expression in the vessels and primitive heart during embryogenesis, consistent with the early death of WNK1(-/-) mice. The generalized cardiovascular expression observed in adulthood may also suggest a possible kidney-independent role in blood pressure regulation. The second unsuspected site of L-WNK1 expression was the granular layer and Purkinje cells of the cerebellum, suggesting a role in local ion balance or cell trafficking. In the kidney, discordance between endogenous L-WNK1 and transgene expression suggests that either cis-regulatory elements important for physiological renal expression lie outside the BAC sequence or that illegitimate interactions occur between promoters. Despite this limitation, this transgenic model is a potentially valuable tool for the analysis of spatial and temporal aspects of WNK1 expression and regulation.

The bacterial artificial chromosome (BAC) library of the narrow-leafed lupin (Lupinus angustifolius L.).

The narrow-leafed lupin possesses valuable traits for environment-friendly agriculture and for the production of unconventional agricultural products. Despite various genetic and environmental studies, the breeding of improved cultivars has been slow due to the limited knowledge of its genomic structure. Further advances in genomics require, among other things, the availability of a genomic DNA library with large inserts. We report here on the construction of the first DNA library cloned in a BAC (bacterial artificial chromosome) vector from diploid Lupinus angustifolius L. cv. Sonet. The high molecular weight DNA used for its preparation was isolated from interphase nuclei that were purified by flow cytometry. The library comprises 55,296 clones and is ordered in 144x384-well microtitre plates. With an average insert size of 100 kb, the library represents six haploid genome equivalents. Thanks to the purification of the nuclei by flow cytometry, contamination with chloroplast DNA and mitochondrial DNA was negligible. The availability of a BAC library opens avenues for the development of a physical contig map and positional gene cloning, as well as for the analysis of the plant's genome structure and evolution.

Mouse molecular cytogenetic resource: 157 BACs link the chromosomal and genetic maps.

We have established a collection of strong molecular cytogenetic markers that span the mouse autosomes and X chromosome at an average spacing of one per 19 Mb and identify 127 distinct band landmarks. In addition, this Mouse Molecular Cytogenetic Resource relates the ends of the genetic maps to their chromosomal locations. The resource consists of 157 bacterial artificial chromosome (BAC) clones, each of which identifies specific mouse chromosome bands or band borders, and 42 of which are linked to genetic markers that define the centromeric and telomeric ends of the Whitehead/MIT recombinational maps. In addition, 108 randomly selected and 6 STS-linked BACs have been assigned to single chromosome bands. We have also developed a high-resolution fluorescent reverse-banding technique for mouse chromosomes that allows simultaneous localization of probes by fluorescence in situ hybridization (FISH) with respect to the cytogenetic landmarks. This approach integrates studies of the entire mouse genome. Moreover, these reagents will simplify gene mapping and analyses of genomic fragments in fetal and adult mouse models. As shown with the MMU16 telomeric marker for the trisomy 16 mouse model of Down syndrome, these clones can obviate the need for metaphase analyses. The potential contribution of this resource and associated methods extends well beyond mapping and includes clues to understanding mouse chromosomes and their rearrangements in cancers and evolution. Finally it will facilitate the development of an integrated view of the mouse genome by providing anchor points from the genetic to the cytogenetic and functional maps of the mouse as we attempt to understand mutations, their biological consequences, and gene function.

[Construction of a deep coverage rice BAC library and identification of clones associated with disease-resistant genes].

A BAC library for IRBB56, an accession pyramiding Xa4, xa5 and xa13 three bacterial blight resistance genes, was constructed. The library contains 55,296 clones with an average insert size of 132 kb. Based on a haploid genome size of 450 Mb, the coverage of the library was about 14 genome equivalents that make it one of the most comprehensive BAC libraries available in rice and provide 99.99% possibility to isolate any interested rice genes or sequences in the library. To determine the representation of organelle DNA homologues in the library, the library was screened with three different chloroplast genes and four mitochondrial genes, respectively. Results from this screening showed that less than 1% of clones in the library contain organelle genomic DNA homologues. Then, DNA markers on three different chromosomes linked to Xa4, xa5, and xa13, and a PCR fragment of rice UROD gene, were used to screen the library resulting in a range of 11-106 hits that will promote the isolation of these genes. The deep coverage and the large insert size of the library will facilitate physical mapping, isolation, and cloning of rice genes.

Melon bacterial artificial chromosome (BAC) library construction using improved methods and identification of clones linked to the locus conferring resistance to melon Fusarium wilt (Fom-2).

Utilizing improved methods, two bacterial artificial chromosome (BAC) libraries were constructed for the multidisease-resistant line of melon MR-1. The HindIII library consists of 177 microtiter plates in a 384-well format, while the EcoRI library consists of 222 microtiter plates. Approximately 95.6% of the HindIII library clones contain nuclear DNA inserts with an average size of 118 kb, providing a coverage of 15.4 genome equivalents. Similarly, 96% of the EcoRI library clones contain nuclear DNA inserts with an average size of 114 kb, providing a coverage of 18.7 genome equivalents. Both libraries were evaluated for contamination with high-copy vector, empty pIndigoBac536 vector, and organellar DNA sequences. High-density filters were screened with two genetic markers FM and AM that co-segregate with Fom-2, a gene conferring resistance to races 0 and 1 of Fusarium wilt. Fourteen and 18 candidate BAC clones were identified for the FM and AM probes, respectively, from the HindIII library, while 34 were identified for the AM probe from filters A, B, and C of the EcoRI library.

The early effects of Ontario's Administrative Driver's Licence Suspension law on driver fatalities with a BAC > 80 mg%.

BACKGROUND: On November 29, 1996, Ontario introduced an Administrative Driver's Licence Suspension (ADLS) law, which required that anyone charged with driving with a blood alcohol concentration (BAC) over the legal limit of 80 mg% or failing to provide a breath sample would have their licence suspended for a period of 90 days at the time the charge was laid. This study evaluates the early effects of Ontario's ADLS law on alcohol-involved driver fatalities. METHODS: Interrupted time series analysis with ARIMA modelling was applied to the monthly proportion of drivers killed in Ontario with a BAC over 80 mg% for the period Jan. 1, 1988 to Dec. 31, 1997. RESULTS: A significant intervention effect was found, with ADLS being associated with an estimated reduction of 17.3% in the proportion of fatally injured drivers who were over the legal limit. CONCLUSION: These data provide an early indication that the law resulted in some success in reducing alcohol-related driver fatalities.

Graduation from a zero to .05 BAC restriction in an Australian graduated licensing system: a difficult transition for young drivers?

In Victoria, Australia, 21-26-year-old drivers are over-represented in alcohol-related fatal crashes. At this age drivers generally transfer from intermediate to full licensure, correspondingly experiencing an increase in legal BAC limit from zero to.05. This study used telephone surveys to compare drinking-driving profiles of 21-26-year-old fully-licensed, 18-20-year-old intermediate and 31-40-year-old experienced drivers ( N congruent with 700) to identify potential crash-risk factors. High driving and drinking exposure was a likely contributor to the over-involvement of male 21-26-year-olds, but not females. A tendency by 21-26-year-olds to rely on other drivers was both successful and unsuccessful in avoiding drink-driving. When driving themselves, there was a tendency to count or space drinks - less successful than abstaining from alcohol. The need to return home, convenience and lack of transport options were common reasons for drink-driving. Fear of crashes/injuries, licence loss and detection/arrest were common motivators to avoid drink-driving. Findings suggest the increase in BAC limit is a difficult transition for some drivers and may contribute to their crash involvement.

[Application of BAF-BAC process in advanced treatment of secondary effluent of refinery processing factory].

To find a new advanced technology for wastewater reuse in refinery processing factory, a pilot test using BAF-BAC process was carried out. The results revealed that when the COD concentration of the influent was less than 130 mg/L and BAF filtration rate was lower than 4.24 m/h, the average effluent COD concentration of BAF-BAC process was less than 50 mg/L, average turbidity was 4.46 NTU. At the same time this process has some effective removal rate on ammonia-nitrogen.

New genomic resources for the honey bee(Apis mellifera L.): development of a deep-coverage BAC library and a preliminary STC database.

We have constructed a bacterial artificial chromosome (BAC) library for a European honey bee strain using the cloning enzyme HindIII in order to develop resources for structural genomics research. The library contains 36,864 clones (ninety-six 384-well plates). A random sampling of 247 clones indicated an average insert size of 113 kb (range = 27 to 213 kb) and 2% empty vectors. Based on an estimated genome size of 270 Mb, this library provides approximately 15 haploid genome equivalents, allowing >99% probability of recovering any specific sequence of interest. High-density colony filters were gridded robotically using a Genetix Q-BOT in a 4 x 4 double-spotted array on 22.5-cm2 filters. Screening of the library with four mapped honey bee genomic clones and two bee cDNA probes identified an average of 21 positive signals per probe, with a range of 7-38 positive signals per probe. An additional screening was performed with nine aphid gene fragments and one Drosophila gene fragment resulting in seven of the nine aphid probes and the Drosophila probe producing positive signals with a range of 1 to 122 positive signals per probe (average of 45). To evaluate the utility of the library for sequence tagged connector analysis, 1152 BAC clones were end sequenced in both forward and reverse directions, giving a total of 2061 successful reads of high quality. End sequences were queried against SWISS-PROT, insect genomic sequence GSS, insect EST, and insect transposable element databases. Results in spreadsheet format from these searches are publicly available at the Clemson University Genomics Institute (CUGI) website in a searchable format (http://www.genome.clemson.edu/projects/stc/bee/AM__Ba/).

Study of the myenteric and submucous plexuses after BAC treatment in the intestine of rats.

A morphological and quantitative study in the ileal and colonic myenteric and submucous plexuses of rats after BAC denervation was performed. Four groups were employed: SI--ileum control; CBI--denervated ileum; SC--colon control; and CBC--denervated colon. We used the Myosin-V immunohistochemistry technique to study the myenteric and submucous plexuses. In the submucous plexus of the ileum and colon there was not a significant decrease in the number of neurons/mm2 and of ganglia/mm2. The denervation of the myenteric plexus in the group CBI was 44.7% and in the group CBC, 68.3%. In the myenteric plexus there was also a significant decrease in the number of ganglia/mm2 (13.8% in group CBI and 52.14% in group CBC) and in the number of neurons/ganglion (33.9% in group CBI and 39.6% in group CBC). The morphological analyses showed that there was an alteration in the shape of the ganglia of the ileal and colonic myenteric plexus. The area of the cell bodies had a significant increase both in the myenteric and the submucous plexus in groups CBI and CBC. These data demonstrate that the BAC treatment causes morphologic and quantitative changes in the myenteric plexus and quantitative changes in the cell body area of the submucous plexus.

[Effect of Bac. megaterium H on tumor induction by methylcholanthrene].

It is shown that Bac megaterium H which has cross-reacting antigens with malignant tumour tissues may influence the time of appearance and growth of tumours, induced by 20-methylcholanthrene in BALB/c mice. The strengthening of tumour growth rate was most distinctly displayed if Bac. megaterium H was injected once 7 days before the methylcholanthrene inoculation.

[Protein composition of crystals (delta-endotoxin) of different serotypes of Bac. thuringiensis].

The crystals of entomopathogenic protein from Bac. thuringiensis contain admixtures of proteinases adhering to their surfaces. A newly developed technique of protease inactivation allowed to estimate the true protein composition of the crystals of various strains of Bac. thuringiensis. It was shown that the crystals of all strains (with the exception of V and VIII) are composed of only one protein with molecular weights of 145,000, 135,000 and 130,000, depending on the strain. The crystals of serotype VIII and the majority of the V serotype strains have two proteins with molecular weights of 135,000 and 130,000. A method for estimation of the protein composition of crystal without their preliminary isolation from a crystal--spore mixture is proposed.

[Effect of MEA on DNA degradation in permeable irradiated Bac. stearothermophilus cells].

It was shown that DNA-degrading activity of permeable, intact and gamma-irradiated cells of Bac. stearothermophilus decreased under the effect of beta-mercaptoethylamine (MEA). MEA decreased also a DNAase activity, in a crude acellular extract of Bac. stearothermophilus, and activities of S1-nuclease and DNAase I. The data obtained prompt an assumption that MEA has an inhibitory action on the activity of endonucleases irrespective of their substrate specificity.

BAC array CGH reveals genomic aberrations in idiopathic mental retardation.

Array using 2,173 BAC clones covering the whole human genome has been constructed. All clones spotted were confirmed to show a unique signal at the predicted chromosomal location by FISH analysis in our laboratory. A total of 30 individuals with idiopathic mental retardation (MR) were analyzed by comparative genomic hybridization using this array. Three deletions, one duplication, and one unbalanced translocation could be detected in five patients, which are likely to contribute to MR. The constructed array was shown to be an efficient tool for the detection of pathogenic genomic rearrangements in MR patients as well as copy number polymorphisms (CPNs).

Distant regulatory elements in a Sox10-beta GEO BAC transgene are required for expression of Sox10 in the enteric nervous system and other neural crest-derived tissues.

Sox10 is an essential transcription factor required for development of neural crest-derived melanocytes, peripheral glia, and enteric ganglia. Multiple transcriptional targets regulated by Sox10 have been identified; however, little is known regarding regulation of Sox10. High sequence conservation surrounding 5' exons 1 through 3 suggests these regions might contain functional regulatory elements. However, we observed that these Sox10 genomic sequences do not confer appropriate cell-specific transcription in vitro when linked to a heterologous reporter. To identify elements required for expression of Sox10 in vivo, we modified bacterial artificial chromosomes (BACs) to generate a Sox10betaGeoBAC transgene. Our approach leaves endogenous Sox10 loci unaltered, circumventing haploinsufficiency issues that arise from gene targeting. Sox10betaGeoBAC expression closely approximates Sox10 expression in vivo, resulting in expression in anterior dorsal neural tube at embryonic day (E) 8.5 and in cranial ganglia, otic vesicle, and developing dorsal root ganglia at E10.5. Characterization of Sox10betaGeoBAC expression confirms the presence of essential regulatory regions and additionally identifies previously unreported expression in thyroid parafollicular cells, thymus, salivary, adrenal, and lacrimal glands. Fortuitous deletions in independent Sox10betaGeoBAC lines result in loss of transgene expression in peripheral nervous system lineages and coincide with evolutionarily conserved regions. Our analysis indicates that Sox10 expression requires the presence of distant cis-acting regulatory elements. The Sox10betaGeoBAC transgene offers one avenue for specifically testing the role of individual conserved regions in regulation of Sox10 and makes possible analysis of Sox10+ derivatives in the context of normal neural crest development.

Imprinted expression of neuronatin from modified BAC transgenes reveals regulation by distinct and distant enhancers.

Neuronatin (Nnat) is an imprinted gene that is expressed exclusively from the paternal allele while the maternal allele is silent and methylated. The Nnat locus exhibits some unique features compared with other imprinted domains. Unlike the majority of imprinted genes, which are organised in clusters and coordinately regulated, Nnat does not appear to be closely linked to other imprinted genes. Also unusually, Nnat is located within an 8-kb intron of the Bc10 gene, which generates a biallelically expressed, antisense transcript. A similar organisation is conserved at the human NNAT locus on chromosome 20. Nnat expression is first detected at E8.5 in rhombomeres 3 and 5, and subsequently, expression is widespread within postmitotic neuronal tissues. Using modified BAC transgenes, we show that imprinted expression of Nnat at ectopic sites requires, at most, an 80-kb region around the gene. Furthermore, reporter transgenes reveal distinct and dispersed cis-regulatory elements that direct tissue-specific expression and these are predominantly upstream of the region that confers allele-specific expression.
