Heterocyclic nucleoside analogues by cycloaddition reactions of 1-vinylthymine with 1,3-dipoles.

1,3-Dipolar cycloaddition of 1-vinylthymine to azides, nitrile oxides, nitrones and nitronates has been investigated as a route to heterocyclic nucleoside analogues in which the nucleoside ribose moiety has been replaced by an alternative heterocycle. Reaction of 1-vinylthymine with highly reactive nitrile oxides affords 1-(isoxazolin-5-yl)thymine products in excellent yield at room temperature. The less reactive nitrone dipoles undergo cycloaddition to 1-vinylthymine at elevated temperature to afford 1-(isoxazolidin-5-yl)thymine cycloadducts in good-to-moderate yields, but show a tendency to eliminate thymine from the cycloaddition products over long reaction times. Azide cycloadditions to 1-vinylthymine proceed only under forcing conditions to which the fragile triazoline products are unstable.

Bond strength of six soft denture liners processed against polymerized and unpolymerized poly(methyl methacrylate).

The bond strength of six commercial soft denture liners was evaluated by a two-phase tensile test. The soft denture liners investigated were VinaSoft, Prolastic, Flexor, Molloplast-B, Novus, and SuperSoft. The samples were fabricated by processing them (1) against polymerized poly(methyl methacrylate), and (2) against unpolymerized poly(methyl methacrylate). The soft denture liners were processed according to the manufacturers recommendations. The samples were tested using an Instron Universal Testing Machine. The mode of failure, adhesive or cohesive, was also recorded. The bond strength when processed against unpolymerized poly(methyl methacrylate) ranged from 0.48 to 2.60 MPa, and when processed against polymerized poly(methyl methacrylate) the bond strength ranged from 0.94 to 2.56 MPa. A two-way analysis of variance (P = .05) revealed a significant increase in bond strength when the liners were processed against polymerized poly(methyl methacrylate), except for Novus, which had no change, and VinaSoft, which decreased. The Tukey interval between materials was .22 and between methods of polymerization was .08. Four of the six liners investigated demonstrated increased bond strength when processed against polymerized poly(methyl methacrylate). It was concluded that bonding can be influenced by the processing method.

3-Methoxy-4-(2-nitrovinyl)phenyl glycosides as potential chromogenic substrates for the assay of glycosidases.

Selective glycosidation of 2,4-dihydroxybenzaldehyde with either 2,3,4, 6-tetra-O-acetyl-alpha-D-glucopyranosyl bromide, 2-acetamido-3,4,6-tri-O-acetyl-alpha-D-glucopyranosyl chloride, or 2,3,4,6-tetra-O-acetyl-alpha-D-galactopyranosyl bromide afforded the corresponding 4-O-glycosyl derivatives. Subsequent O-methylation, O-deacetylation, and condensation with nitromethane afforded the appropriate beta-glycoside of 3-methoxy-4-(2-nitrovinyl)phenol. The phenol is highly coloured at alkaline pH so that these glycosides may be suitable as chromogenic substrates for the assay of glycosidases.

Enhanced removal of p-chloroaniline from aqueous solution by a carboxylated polymeric sorbent.

In the present study a carboxylated styrene-divinylbenzene (St-DVB) polymeric sorbent (CSPS) was prepared for enhanced removal of p-chloroaniline from aqueous solution. A commercial styrene-divinylbenzene polymeric sorbent Amberlite XAD-4 was selected as the reference sorbent to evaluate the performance of CSPS. Characterization of CSPS was determined by infrared spectroscopy and pore size distribution. A p-chloroaniline sorption enhancement on CSPS at capacity about twice of Amberlite XAD-4 was observed, which was mainly attributed to the carboxyl group added on the polymeric matrix. Different pH-dependent sorption property of p-chloroaniline onto XAD-4 and CSPS was observed partly due to the role of carboxyl group. Sorption isotherm of p-chloroaniline on CSPS could be represented by Freundlich model reasonably, and that on XAD-4 was more suitable for Langmuir model. Kinetic studies demonstrated that the uptake of p-chloroaniline on CSPS and XAD-4 followed the pseudo-second order model. Moreover, breakthrough curves on CSPS further demonstrated its better performance towards p-chloroaniline removal from aqueous solution. Complete regeneration of the spent sorbent CSPS was achieved by dilute hydrochloric acid for its repeated use, implying its potential application in associated chemical wastewater treatments.

Unexpected fragmentation of beta-substituted meso-tetraphenylporphyrins induced by high-energy collisional activation.

The protonated molecules and radical cations of meso-tetraphenylporphyrins with beta-pyrrolic substituents, when formed by fast atom bombardment (FAB) and subjected to high-energy collisions, give rise to unexpected fragment ions. The reaction involves hydrogen migration from the ortho position of the phenyl ring to the a atom of the substituent, with formation of an intramolecular, six-membered ring. The process is analogous to condensed-phase cyclizations described for the same type of compounds. The fragmentation requires the presence of a double bond in the substituent group attached to the pyrrolic ring. A rearrangement process involving anchimeric assistance by the phenyl group (analogous to an ortho effect) is proposed for the formation of these ions.

Stereoselective synthesis of vinylsilanes by a gold(I)-catalyzed acetylenic sila-cope rearrangement.

Cationic tri-tert-butylphosphinegold(I) serves as a catalyst in the sila-Cope rearrangement of acetylenic allylsilanes. When phenol is employed as a nucleophile, the reaction allows for the stereoselective synthesis of vinylsilanes. Alternatively, use of methanol as a nucleophile leads to cyclic vinylsilanes, which can be viewed as latent vinylsilanes that are revealed on treatment with a mild Lewis acid. Thus, both of these reagents serve as useful reagents for stereoselective synthesis of trisubstituted olefins through transition-metal-catalyzed cross-coupling reactions.

Metalated diblock and triblock poly(ethylene oxide)-block-poly(4-vinylpyridine) copolymers: understanding of micelle and bulk structure.

The paper provides new insights into the structure of Pt-containing diblock and triblock copolymers based on poly(ethylene oxide) (PEO) and poly(4-vinylpyridine) (P4VP), using a combination of atomic force microscopy (AFM), X-ray diffraction (XRD), transmission electron microscopy (TEM), and anomalous small-angle X-ray scattering (ASAXS). Parallel studies using methods contributing supplemental structural information allowed us to comprehensively characterize sophisticated polymer systems during metalation and to exclude possible ambiguity of the data interpretation of each of the methods. AFM and TEM make available the determination of sizes of the micelles and of the Pt-containing micelle cores, respectively, while a combination of XRD, TEM, and ASAXS reveals Pt-nanoparticle size distributions and locations along with the structural information about the polymer matrix. In addition, for the first time, ASAXS revealed the organization of Pt-nanoparticle-filled diblock and triblock copolymers in the bulk. The nanoparticle characteristics are mainly determined by the type of block copolymer system in which they are found: larger particles (2.0-3.0 nm) are formed in triblock copolymer micelles, while smaller ones (1.5-2.5 nm) are found in diblock copolymer micelles. This can be explained by facilitated intermicellar exchange in triblock copolymer systems. For both systems, Pt nanoparticles have narrow particle size distributions as a result of a strong interaction between the nanoparticle surface and the P4VP units inside the micelle cores. The pH of the medium mainly influences the particle location rather than the particle size. A structural model of Pt-nanoparticle clustering in the diblock PEO-b-P4VP and triblock P4VP-b-PEO-b-P4VP copolymers in the bulk was constructed ab initio from the ASAXS data. This model reveals that nearly spherical micellar cores of about 10 nm in diameter (filled with Pt nanoparticles) aggregate forming slightly oblate hollow bodies with an outer diameter of about 40 nm.

An ab initio study on the allene-isocyanic acid and ketene-vinylimine [2 + 2] cycloaddition reaction paths.

The reaction paths of [2 + 2] cycloadditions of allene (H2C=C=CH2) to isocyanic acid (HN=C=O) and ketene (H2C=C=O) to vinylimine (H2C=C=NH), leading to all the possible 14 four-membered ring molecules, were investigated by the MP2/aug-cc-pVDZ method. In the two considered reactions, the 2-azetidinone (beta-lactam) ring compounds were predicted to be the most stable thermodynamically in the absence of an environment. Although 4-methylene-2-azetidinone is the most stable product of the ketene-vinylimine cycloaddition, its activation barrier is higher than that for 4-methylene-2-iminooxetane by ca. 6 kcal/mol. Therefore, the latter product can be obtained owing to kinetic control. The activation barriers in the allene-isocyanic acid reactions are quite high, 50-70 kcal/mol, whereas in the course of the ketene-vinylimine cycloaddition they are equal to ca. 30-55 kcal/mol. All the reactions studied were found to be concerted and mostly asynchronous. Simulation of the solvent environment (toluene, tetrahydrofuran, acetonitrile, and water) by using Tomasi's polarized continuum model with the integral equation formalism (IEF-PCM) method showed the allene-isocyanic reactions remained concerted, yet the activation barriers were somewhat higher than those in the gas phase, whereas the ketene-vinylimine reactions became stepwise. The larger the solvent dielectric constant, the lower the activation barriers found. The lowest-energy pathways in the gas phase and in solvent were confirmed by intrinsic reaction coordinate (IRC) calculations. The atoms in molecules (AIM) analysis of the electron density distribution in the transition-state (TS) structures allowed us to distinguish pericyclic from pseudopericyclic from nonplanar-pseudopericyclic types of reactions.

One-pot, three-component synthesis of linearly substituted homoallylic alcohols via allyl(isopropoxy)dimethylsilane.

[reaction: see text] A two-step approach that involves the synthesis of vinylsilane from allyl(isopropoxy)dimethylsilane (6) and the subsequent Pd-catalyzed cross-coupling of the resulting vinylsilane is applied in developing a novel diversity-oriented, three-component synthesis to homoallylic alcohols of common structure 1. Upon treatment of 6 with s-BuLi, the silylallylmetal 5 is generated and reacted with carbonyl compounds to produce functionalized vinylsilanes, which can couple with aryl/vinyl halides in the presence of palladium catalyst to afford 1.

[Environmental carcinogens: mechanisms of action and occurrence. Some aspects (author's transl)].

New aspects on the mechanism of action of known environmental carcinogens are described. These compounds are not active as such, but are bioactivated in the mammalian metabolism via chemically reactive intermediates to electrophilic reactants, forming with information-bearing biopolymeres covalent bonds. This change in the genetic code is in agreement with the mutation hypothesis of carcinogenesis. Aflatoxin B1, N-nitroso compounds and benzo(a)pyrene are taken as examples. "Threshold levels", e.g. no-effect-levels derived from animal experiments with all their inherent limitations, are compared with data from human exposure to benzo(a)pyren, aflatoxine, N-nitroso compounds and vinyl chloride. The difficulties of such risk evaluations are discussed.
