Cell cycle progression and chromosome segregation in mammalian cells cultured in the presence of the topoisomerase II inhibitors ICRF-187 [(+)-1,2-bis(3,5-dioxopiperazinyl-1-yl)propane; ADR-529] and ICRF-159 (Razoxane).

Certain bis(2,6-dioxopiperazine) derivatives, which include ICRF-187 [(+)-1,2-bis(3,5-dioxopiperazinyl-1-yl]propane; ADR-529) and its racemic compound ICRF 159 (Razoxane), have been investigated as antineoplastic agents. In addition, ICRF-187 is currently under intense study as an agent to ameliorate the cardiac toxicity of anthracycline therapy. These agents have recently been identified as inhibitors of topoisomerase II. We studied the effects of ICRF-187 and ICRF-159 on the progression of cultured epithelial cells through M phase. Beginning approximately 1.5 h after drug addition, chromosome condensation was significantly inhibited. Cells entered and progressed through M phase at near normal rates, but the lack of complete chromosome separation during anaphase resulted in catastrophic effects on normal chromosome distribution. Immunolabeling with Crest autoimmune sera, which recognizes centromere proteins, and with MPM-2 monoclonal antibody, which recognizes mitotic phosphoproteins, indicated that the centromeres of the chromosomes assembled a normal metaphase array in the presence of ICRF-187 and ICRF-159. Centromere separation in anaphase was initiated normally but was not completed because the chromatid arms failed to disengage from each other. Massive chromosome bridges were formed, and the chromatin mass became trapped in the cleavage furrow leading to its unequal distribution to the daughter cells. In many cases, all the chromatin was pushed into one of the two dividing cells. It is likely that previous studies, based on flow cytometry, indicating that bis(2,6-dioxypiperazine) derivatives cause an accumulation of cells with a 4N DNA content, reflect the incomplete segregation of chromosomes in mitosis rather than a block in G2 of the cell cycle as had been proposed.

Phenylmercury and its mobilization in the organism by a metal complex-forming substance: 2,3-dimercapto-1-propane sodium sulfonate.

Workers handling dressing machines for seed treatment with the product Agronal, containing a phenylmercury chloride fungicide, were exposed to high concentrations of phenylmercury dust in the working environment. Urine analyses for mercury result in concentration of up to 0.1 mg Hg/l of urine. After administration of a complex-forming substance-Unitol (2,3-dimercapto-1-propane sodium sulfonate)-a higher urinary excretion of mercury occurred. The amount of mercury excreted confirmed its deposit in the organism. It was speculated that subjective complaints by workers handling dressing machines could be caused by high exposure to phenylmercury. This suggestion cannot, however, be fully accepted because the complaints were not necessarily specific for mercury only, but could also have been caused by factors of nontoxic origin, such as stress at the workplace, discontent with work and environmental hygiene conditions, conflicts and alcoholism. Most probably, it was a case of interpotentiation of the effects of toxic and non-toxic nature.

[Cutaneous cryosurgery in family medicine: dimethyl ether-propane spray versus liquid nitrogen].

OBJECTIVE: To compare the efficacy, tolerance and safety of two types of cryotherapy, performed by family physicians, for benign cutaneous lesions: low freezing (-59 degrees C) with dimethyl either-propane cryogenic spray (DMEP) and intense freezing (196 degrees C) with conventional liquid nitrogen (LN). DESIGN: A randomised, multi-centered, controlled clinical trial, with single-blind assessment. SETTING: Three primary care teaching teams in the Community of Madrid. PATIENTS AND OTHER PARTICIPANTS: Ten MIR from family & community medicine intervened. There were 124 patients, who had 174 benign cutaneous lesions, suitable for cryotherapy. There were 3 voluntary withdrawals, none because of an adverse reaction. Interventions, in each case there was local application for a standard time of the randomised agent. Control-group intervention, 81 cases: swab soaked in LN. Study-group intervention, 93 cases: swab saturated with DMEP spray. Maximum of three freezings per case, at weekly intervals. MEASUREMENTS AND MAIN RESULTS: A doctor made a blind assessment of the results (elimination, adverse reaction, aesthetic result) 15 days after treatment. CONCLUSIONS: No clinically relevant differences between the efficacy, tolerance and safety of the two cryogenic agents used in primary care were found. The low freezing of DMEP was sufficient for the cryotherapy of benign lesions.

Antitumor activity and nephrotoxicity of a novel platinum complex, [1,3-bis(diphenylphosphino)propane](trans-1-dach)platinum(I I) dinitrate.

We have developed a new class of platinum complex [Pt(trans-l-dach)(1,3-bis(phosphino)propane)] dinitrate (KHPC-001) with potent antitumor activity and low nephrotoxicity, confirmed in vitro and compared in vivo with cisplatin, KHPC-001 or cisplatin was intraperitoneally injected on days 1, 5, and 9 into P388-bearing mice and the antitumor effects were compared. In vitro cytotoxicity, Pt accumulation, and DNA cross-link index were measured in P388 and LLC-PK1 cells after treatment with KHPC-001 or cisplatin. Twenty mg/kg (below one-tenth of LD50) of KHPC-001 had stronger antitumor effects than 2 mg/kg (about one-fifth of LD50) of cisplatin and cured 2 out of 6 mice without any toxicity. While the cytotoxicity of KHPC-001 and cisplatin were similar on P388 mouse leukemia cells, this new compound was much less cytotoxic to a kidney-derived line, LLC-PK1. This lower toxicity on the kidney cells was based on its low accumulation, causing less DNA crosslinking. KHPC-001 is a unique third-generation platinum complex with potent antitumor activity and low nephrotoxicity.

Metabolism of 2(R,S)-1,2-bis(nicotinamido)propane, a new agent with anti-vasospasm activity, in rats and rabbits.

1. The metabolic fate of the new Anti-vasospasm Substance (AVS), 2(R,S)-1,2-bis(nicotinamido)propane (CAS 79455-30-4), was studied using 14C-labelled drug in rats and rabbits by thinlayer chromatography, mass spectrometry and nuclear magnetic resonance. 2. More than 75% of the radioactivity was observed in the urine when 14C-AVS was given intravenously to rabbits and rats, showing that the major route of excretion of AVS and its metabolites is via the kidney. 3. Marked species differences were observed in the metabolism of AVS in rats and rabbits. In rabbits, the major metabolites were 6- or 6'-monopyridone (23.5% of dose), and there were a few minor metabolites such as the mono N- or N1-oxide of two pyridine rings. In rats, however, only approximately 5% of the radioactivity was due to metabolites, mainly the N-oxide. 4. Formation of AVS monopyridone by rabbit liver cytosol was much higher than in rats, and was markedly inhibited by the aldehyde oxidase inhibitor, menadione. The difference between rats and rabbits in oxidase activity giving the AVS monopyridone metabolite correlated well with that measured by the general assay method for aldehyde oxidase. These results suggest that the species difference in AVS metabolism between rats and rabbits is mainly due to the difference in aldehyde oxidase activity, which is involved in formation of the monopyridone.

[Effects of 1-(2,6-dimethylphenoxy)-2-(3,4-dimethoxy phenylethylamino) propane hydrochloride on cytosolic free calcium concentration of rabbit platelets].

The effects of 1-(2, 6-dimethylphenoxy)-2- (3,4-dimethoxyphenylethylamino) propane hydrochloride (DDPH) and verapamil on cytosolic free calcium concentration of rabbit platelets were investigated. The results showed that the elevation of cytosolic free calcium concentration ([Ca2+]i) of rabbit platelets following ionophore A23187 was significantly inhibited by DDPH and verapamil. The rates of inhibition with DDPH and verapamil were 58% and 78%, respectively, with no significant difference between the two groups (P > 0.05).

Encephalopathy and peripheral neuropathy following carbon monoxide poisoning from a propane-fueled vehicle.

Carbon monoxide poisoning is the leading cause of lethal poisonings in the United States. A growing number of recent reports indicate it is likely underdiagnosed as a cause of headaches, fatigue, dizziness, and other neurologic complaints. The mechanisms by which carbon monoxide exerts toxicity, especially delayed toxicity, are poorly understood. A recent case with a 5 year delay in diagnosis illustrates many of the unique and puzzling aspects of carbon monoxide toxicity.

An emission-controlled field desorption and electron impact spectrometry study of some N-substituted propane and butane sultams.

It is shown that the production of field desorption mass spectra with an emission-controlled emitter heating device is a useful tool in obtaining information about molecular structures. Since the structurally significant signals are sometimes missing in the corresponding electron impact spectra but not in the field desorption spectra the use of field desorption mass spectrometry appears to be more advantages. This will be especially true for substances that are thermally labile, unvolatile or unstable upon electron impact (both at high and low electron energy), the latter being the case for some of the compounds presently investigated which are derivatives of a drug with antitumour and antiepileptic effects. Further, emission-controlled desorption improves the reproducibility of the spectra obtained and thus appears to be a prerequisite for quantification in biomedical and pharmacokinetic studies.

Mutagenicity of halogenated propanes and their methylated derivatives.

1,2,3-Tribromopropane, 1,2,3-trichloropropane, and 1,2-dibromo-3-chloropropane are mutagenic in strains TA1535 and TA100 of Salmonella typhimurium, but only in the presence of rat liver S9 mix. This requirement for metabolic activation was unexpected for an alkyl halide and thus suggested the metabolic formation of the 2-keto derivatives (di-haloacetone). The 2-methyl derivatives of the halopropane compounds did not induce a doubling of revertants compared to controls. It was demonstrated that none of these compounds is converted into a secondary material that could be determined as structurally different by gas chromatography. These observations suggest that lack of mutagenicity of the methylated compounds is a manifestation of a steric effect.
