Blood samples from 30 patients with myocardial ischemia and 25 healthy donors were irradiated with He-Ne laser. ATP release was measured by the luciferin-luciferase test system on a PICA lumo-aggregometer (Chrono-Log Corporation, USA). There was no correlation between lectin-induced ATP release and cell aggregation. Neutrophils obtained from donors and patients differed in the number of lectin receptors, including mannose and sialic acids on the cellular surface. Following laser irradiation, WGA- and SBA-induced aggregation was decreased, while Con-A-induced aggregation was increased.
To clarify the changes in cellular reactivity in response to single irradiation of blood with He-Ne laser, the authors studied neutrophil aggregation and neutrophil ATP release induced by stimulation with lectins diverse in carbohydrate specificity. Blood samples from 30 healthy donors were investigated using PICA lumoaggregometer (Chrono-Log Corporation, USA) and concanavalin A, PHA-L, WGA, SBA as aggregation inductors. He-Ne laser radiation in most cases results in reduced neutrophil capacity for aggregation. Nevertheless, it activates ATP release under stimulation with lectins of various carbohydrate specificity. It follows that mechanisms of lectin-induced reactions, aggregation of neutrophils and ATP release, are different.

The paper provides the results of endoscopic and morphologic follow-up of healing of extensive corrosive gastric ulcer in He-Ne laser therapy of 34 patients (512 gastric biopsies). Laser radiation of ulcer reduces frequency of stenosis in healing outcome as well as treatment duration, creates conditions for healing without leukocytic fusion of necrotic tissues. Effectivity of laser therapy is determined by the time of its start and regularity.

Venous blood from 10 patients with dilated cardiomyopathy was irradiated with a laser in vitro. The control group consisted of 20 healthy donors. The neutrophil granulocytes were separated at gradient centrifugation. Alterations of neutrophils manifested with an increase of specific cytoplasmic granules number, thickening of submembrane actin, cell configuration changes with a relative increase of their surface. Laser irradiation of the blood resulted in destruction of the altered (less resistant) cells while morphometric parameters of the remaining cells approaches those of donor cells. Thus, low-intensity laser irradiation results in the renewal of the neutrophil population in patients with dilated cardiomyopathy and normalization of structural-functional changes in the circulating neutrophil population.

BACKGROUND AND OBJECTIVE: Numerous reports suggest that low-power laser irradiation (LPLI) is capable of affecting cellular processes in the absence of significant thermal effect. The objective of the present study was to determine the effect of LPLI on secretion of vascular endothelial growth factor (VEGF) and proliferation of human endothelial cells (EC) in vitro. STUDY DESIGN/MATERIALS AND METHODS: Cell cultures were irradiated with single different doses of LPLI (Laser irradiance from 0.10 to 6.3 J/cm(2)) by using a He:Ne continuous wave laser (632 nm). VEGF secretion by smooth muscle cells (SMC) and fibroblasts was quantified by sandwich enzyme immunoassay technique. The endothelial cell proliferation was measured by Alamar Blue assay. VEGF and transforming growth factor beta (TGF-beta) expression by cardiomyocytes was studied by reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: We observed that (1) LPLI of vascular and cardiac cells results in a statistically significant increase of VEGF secretion in culture (1.6-fold for SMC and fibroblasts and 7-fold for cardiomyocytes) and is dose dependent (maximal effect was observed with LPLI irradiance of 0.5 J/cm(2) for SMC, 2.1 J/cm(2) for fibroblasts and 1.05 J/cm(2) for cardiomyocytes). (2) Significant stimulation of endothelial cell growth was obtained with LPLI-treated conditioned medium of SMC (maximal increase was observed with LPLI conditioned medium with irradiance of 1.05 J/cm(2) for SMC and 2.1 J/cm(2) for fibroblasts. CONCLUSIONS: Our studies demonstrate that low-power laser irradiation increases production of VEGF by SMC, fibroblasts, and cardiac myocytes and stimulates EC growth in culture. These data may have significant importance leading to the establishment of new methods for endoluminal postangioplasty vascular repair and myocardial photoangiogenesis.
