Effects of cryopreservation on human lymphocyte functions: comparison of programmed freezing method by a direct control system with a mechanical freezing method.

The effects of cryopreservation with a new-type programmed freezer (TNP-82) were examined on natural killer (NK) activity and cell recovery of lymphocytes. This instrument is controlled by a sensor which directly detects the temperature in the samples, enabling easy setting of the temperature curve. Comparing results obtained by the new method to those from a less sophisticated mechanical method, cell recovery after cryopreservation was improved (95.3%) in the new compared to the older (about 80%) method. Furthermore, NK activity, percentage of Leu7+ cells and other lymphocyte functions were unaltered.

An electron microscopic and spectroscopic study of murine epiphyseal cartilage: analysis of fine structure and matrix vesicles preserved by slam freezing and freeze substitution.

Newborn mice epiphyseal growth plates were preserved by slam freezing/freeze substitution and examined by conventional electron microscopy, stereopsis, high voltage electron microscopy, and electron spectroscopic imaging (ESI). To illustrate the improved ultrastructure of this cryogenic procedure, conventional, aqueously fixed growth plates were included showing collapsed hypertrophic chondrocytes surrounded by a depleted and condensed extracellular matrix. In contrast, the cryogenically prepared epiphyses contain chondrocytes and extracellular matrix vesicles both in direct contact with proteoglycan filaments retained in an expanded state. ESI is an electron microscopic technique which enables the direct localization of atomic elements superimposed over fine structural details. This technique was used to examine the colocalization of calcium and phosphorus within matrix vesicles and within their associated extracellular environments. Matrix vesicles appeared in three distinct diameter ranges. The integrity of the matrix vesicles was examined at various stages of mineralization and also within the mineralized zone of provisional calcification.

Freezing mouse blastocysts. The influence of the preparations prior to freezing on the survival rate of the blastocysts.

A good survival rate in culturing mouse blastocysts can be obtained in Ovum Culture Medium, enriched with 20 per cent inactivated Foetal Bovine Serum or Sheep Serum under air. The transfer of fresh blastocysts gives the best results if the recipients are on day 3 of the pseudo-pregnancy, but with 20 hours' cultured blastocysts it is better to use recipients on day 4. Exposure to 1.5 M DMSO has no harmful effect, provided that the DMSO is added at 5 degrees C in 6 steps and is removed, again in 6 steps, at 35 degrees C. The crystallization of the medium containing the embryos at -5 degrees C to -6 degrees C doet not appear te have a harmful influence on culture results of the blastocysts.
