The biological sensitivity of cultured non-rheumatoid human synovial cells (NRSCs) and rheumatoid synovial cells (RSCs) was examined in terms of the ability of insulin to stimulate the uptake of alpha-aminoisobutyrate (AIB). NRSCs, like numerous fibroblastic lines, were sensitive to physiological concentrations of the hormone: half-maximal stimulation was obtained with (4 X 10(-10) M) insulin, while maximum transport was found with a 60-90 min association time. On the contrary, although the basal transport was similar in RSCs, insulin was totally unable to accelerate AIB transport in these cells. Inflammatory processes lead to an insulin resistance which most likely involves a post-receptor step at the cellular level.

The preparation, characterization, and molecular and crystal structures of succinimido 2-(tosylamino)isobutyrate, C(15)H(18)N(2)O(6)S, are described. The succinimido ring is nearly orthogonal to the ester group.
A peptide antibiotic has been isolated from Trichoderma reesei QM 9414. Although crystalline and uniform in TLC, this antibiotic could be resolved by HPLC into 3 sequence analogues. The close relationship to alamethicin was proved by chemical and spectroscopic methods, and the formation of ion-conducting pores in lipid bilayers.
The title acid, 3-amino-2-methylpropanoic acid monohydrate, C4H9NO2.H2O, crystallized in the centrosymmetric space group Pbca in the zwitterionic form. The three H atoms on N, which are involved in hydrogen bonding, are ordered. The three intermolecular N-H...O hydrogen bonds have N...O distances ranging from 2.758 (2) to 2.809 (2) A and N-H...O angles ranging from 149 (2) to 171 (1) degrees. The two intermolecular O-H...O hydrogen bonds have O...O distances 2.739 (2) and 2.755 (2) A, and O-H...O angles 170 (2) and 175 (2) degrees. Each acid molecule and its associated water molecule are directly hydrogen bonded to five acid molecules and two water molecules; the structure comprises two subsets of molecules which are not cross-linked by these hydrogen bonds. Through basic second-level graphs, approximately two-thirds of the hydrogen-bonding patterns are finite and one-third are chains; there is a single ring pattern, which occurs about a center of symmetry.
High excretion of BAIB is determined by an incompletely recessive gene jointly with other familial factors. The parameters estimated from a Japanese sample are: gene frequency = 0.6, displacement between homozygotes = 2.1 sigma, dominance = 0.2, and polygenic heritability = 0.1. Supplementary tests support this mixed model and (unlike classical analysis) give no evidence of segregation disturbance. Complexity of BAIB inheritance should be allowed for in population comparisons and linkage studies, which can be performed reliably under the mixed model.
A molecular mechanical study has been carried out to determine the most favorable conformation of poly(Aib) both in solution and in the solid state. An energetic approach to the packing has been carried out by studying the stability of pairs of poly(Aib) chains. The study reveals a higher stability of a 3(10)-helix when forming part of a dimer whereas in isolated molecules the alpha-helix structure seems to be more stable depending on the length of the chain and dielectric constant of the environment.
Active transport of A.I.B. from mother to fetus was studied. This was done in the intact animal and using the isolated placenta, artificially perfused at both sides. It was shown that A.I.B. is actively accumulated in the placental cells. An estimate of the kinetic constants is given. It is shown that this accumulation takes place predominantly from the maternal side of the placenta. A.I.B. that has been accumulated is cleared to the maternal and fetal circulation in equal amounts. So the netto active transport from mother to fetus is brought about by an unequal distribution of carriers, the maternal side being most active.
A 78-year-old man was admitted to our hospital with anemia and jaundice. Hematological studies revealed hyperchromic macrocytic anemia, and biochemical studies revealed findings of hemolysis. The folic acid level was low and megaloblasts were observed in the bone marrow. From these findings, the patient was diagnosed as having megaloblastic anemia due to folic acid deficiency. This patient had been a heavy alcohol drinker in the habit of drinking alcohol without meals. He began to eat regular meals in the hospital, and the anemia and jaundice improved gradually. Since liver cirrhosis was suspected, amino acid analysis of the urine was performed, and abnormal excretion of beta-amino-isobutyric acid (BAIB) was found. According to the amount of BAIB excreted, the Japanese population can be divided into low and high BAIB excretors comprising 65% and 36%, respectively. BAIB is also considered to reflect dissimilation of thymine. The present patient was included in the high excretion group because of the abnormally high urine level of BAIB, which was considered to be caused by ineffective hematopoiesis in the bone marrow as a result of his megaloblastic anemia. For this reason, dissimilation of thymine was considered to have been active in this patient.
