The stereochemistry of NADH utilization by the flavoenzyme monooxygenase orcinol hydroxylase.

Ribbons et al. (Ribbons, D.W., Ohta, Y., and Higgins, I.J. (1972) in Molecular Basis of Electron Transport, Miami Winter Symposic Series (Schultz, J., and Cameron, B.F., eds) Vol. 4, pp. 251-274, Academic Press, New York) presented a preliminary report that the flavoenzyme monooxygenase orcinol hydroxylase shows mixed type 4R, 4S stereospecificity with respect to dihydronicotinamide oxidation when resorcinol and m-cresol were used as substrate analogs. With the natural substrate orcinol, 4R chirality was maintained. In kinetic isotope experiments reported here, we demonstrate in fact that orcinol hydroxylase maintains 4R stereospecificity with respect to dihydronicotinamide oxidation with all three substrates, orcinol, resorcinol, and m-cresol. Deuterium and tritium kinetic isotope effects were detected under Vmax conditions with (4R)-[4-2H]-, and (4R)-[4-3H]NADH for all three substrates. No isotope effect was observed with (4S)-[4-2H]NADH and tritium labilization from assays with (4S)-[4-3H]-NADH was negligible in all cases.

Cutaneous penetration of some hairdyes in the hairless rat.

The cutaneous penetration of certain number of oxidation dyestuffs for hair [p-aminophenol, 2,4-diaminoanisole, 2,4-diaminophenoxyethanol, resorcinol, 3-acetylamino-2,6-dimethylphenol and the indamine: N-(4'-hydroxyphenyl)-3-amino-6-methylbenzoquinoneimine], either alone or when formulated with other dyestuffs, and under various experimental conditions, has been evaluated in the hairless rat using C14 labeled molecules. It has been found that: Dyestuffs having indamine structures, formed by oxidation of their precursors (bases, couplers), for all practical purposes do not cross the cutaneous barrier. No fixation of those dyestuffs investigated (resorcinol, 2,4-diaminoanisole, 2,4-diaminophenoxyethanol) which do pass through the cutaneous barrier is found in either liver or thyroid of the experimental animals, 4 days after their topical application. The amount of the hairdye 3-acetylamino-2,6-dimethylphenol which penetrates the cutaneous barrier is constant for quantities of hairdye solution greater than or equal to 20 mg/cm2 of skin. The type and composition of the excipient used in the formulation of hairdyes plays an important role in their penetration.

Utilization of some phenolic compounds by Azotobacter chroococcum and their effect on growth and nitrogenase activity.

Azotobacter chroococcum MH1 was grown in a mannitol and nitrogen free medium supplemented with p-hydroxybenzoic acid, resorcinol, catechol or vanillic acid as a sole carbon source. Growth and nitrogenase activity of p-hydroxybenzoic acid were supported by 8, 6 and 4 mM of p-hydroxybenzoic acid, resorcinol and catechol, respectively. The generation time of 1.71 h in p-hydroxybenzoic acid did not differ from a generation time of 1.64 h, when grown in mannitol. The compound p-hydroxybenzoic acid was utilized rapidly. However, the decomposition of other phenolic compounds tested proceeded slowly. These results suggested that phenolic compounds released during biodegradation of plant wastes could be utilized as carbon sources for both growth and nitrogen fixation of Azotobacter chroococcum.

Contact dermatitis from polyvinyl chloride identification bands.

One case of allergic contact dermatitis caused by polyvinyl chloride identification bands is reported. Patch tests in this case were positive for three of four bands tested, resorcinol monobenzoate, and benzoyl peroxide. Manufacturers could not confirm the presence of resorcinol monobenzoate and benzoyl peroxide in their respective bands. It is concluded that the patient's reaction was allergic and due to an unknown chemical(s) in the bands. Additives may comprise a significant portion of polyvinyl chloride bands. These components may be leachable, and thus lead to contact sensitization with subsequent allergy. Although patch testing is an excellent first step in determining the offending allergen, specialized chemical analysis may be required for exact identification.

A new type of calixarene: octahydroxypyridin.

A new type of calixarene has been synthesised. Like resorcinol and a number of resorcinol derivatives, 2,6-dihydroxypyridine has been proven to form cyclic tetramers in moderate yields with a number of aliphatic and two aromatic aldehydes in acidic media. The problem of the formation of configurational isomers can be reduced by increasing the reaction temperature and time. With electron-rich aromatic aldehydes, 2,6-dihydroxypyridine unexpectedly yields deep-coloured acyclic quinoid systems. The octahydroxypyridine[4]arenes may have a high potential as receptors for molecular recognition and self organisation.

Development of choline and acetylcholine Pt microelectrodes.

Choline (Ch) and acetylcholine (Ach) microenzyme sensors were developed based on the immobilization of choline oxidase (ChO) and acetylcholinesterase (AchE) at the tip of a 25-micron Pt wire sealed in glass. Several immobilization procedures were tested, including code-position of the enzyme/s with an electropolymer and cross-linking with glutaraldehyde. The various electropolymers used were 1,2-diaminobenzene, resorcinol, 4-hydroxybenzenesulfonic acid, and a combination of two polymers, 1,2-diaminobenzene and resorcinol. An inner membrane constructed from cellulose acetate (CA) was deposited prior to immobilization with glutaraldehyde. The analytical characteristics of the microelectrodes, including optimization of immobilization procedures, calibration curves, pH response curves, stability, and selectivity toward possible electroactive compounds found in the brain extracellular fluid, were determined. The best microelectrodes were prepared by cross-linking the enzymes with glutaraldehyde on top of the inner CA membrane. The responses are linear in the concentration range 5.0 x 10(-7)-1.0 x 10(-4) M Ch and 5.0 x 10(-7)-9.3 x 10(-5) M Ach. The time to reach 95% steady-state current was 15-20 s. The CA-coated Ch microelectrodes were useful for measurement of changes in Ch concentration in artificial brain extracellular fluid.

Developmental regulation of a glycolipid epitope on actively extending growth cones and central and peripheral projections in the medicinal leech.

The monoclonal antibody, A2B5, that recognizes vertebrate gangliosides also recognizes embryonic cells in the medicinal leech (Hirudo medicinalis) in a spatially and temporally regulated way. Furthermore, A2B5-positive glycolipids could be isolated from embryonic leeches. Early in development A2B5 labeled axon tracts within the central nervous system coincident with the initial growth of axons, while later in embryogenesis, A2B5 also labeled the forming peripheral nerves. This central and peripheral staining disappeared during the latter third of embryogenesis. A2B5 also labeled the growing tips of an identified myo-organizing cell. This cell, the C-cell, projects an array of parallel processes and exhibits a discrete transition in the rate of growth cone extension (J. Jellies, and W. B. Kristan, Jr. 1991. Dev. Biol. 148, 334-354.). A2B5 failed to recognize the relatively non motile growth cones of the C-cell during early embryogenesis. The C-cell growth cones began to exhibit A2B5 labeling as they became more rapidly extending and this labeling persisted throughout the later motile phase of C-cell growth. In addition to its widespread distribution on embryonic (but not mature) cellular processes, A2B5 also labeled the mitotic profiles of dividing cells in all tissues. Thus, the A2B5 epitope may be presented intracellularly, or both intra- and extracellularly. When glycolipids were extracted from embryonic leeches, partitioned by elution from a silicic acid column, and analyzed using high-performance thin-layer chromatography, at least one of the major glycolipid bands was resorcinol-positive, consistent with presentation of sialic acid residues. The fraction enriched for the resorcinol-positive band was recognized by A2B5 on dot blots, as was ganglioside GQ1b. While potential mechanisms remain to be examined, on the basis of the distinct distribution of the A2B5 epitope and our finding of A2B5-positive glycolipids in leech embryos, we suggest that complex polar glycolipids may play a role in the extension of cellular projections in the medicinal leech.

Fractal Analysis of Mercury Porosimetry Data in the Framework of the Thermodynamic Method.

The thermodynamic method for fractal analysis is applied to mercury intrusion data. The results for representative commercial carbon black samples and a series of resorcinol-formaldehyde (RF) freeze-dried gels for different values of the molar ratio of resorcinol (R) to catalyst (C) are discussed in relation with the type of behavior exhibited by the samples during mercury porosimetry measurements. The obtained surface fractal dimensions are compared with those derived previously from the small-angle X ray scattering (SAXS) and the nitrogen adsorption. Copyright 1999 Academic Press.

An improved method for the measurement of total lipid-bound sialic acids after cleavage of alpha 2,8 sialic acid linkage with Vibrio cholerae sialidase in the presence of cholic acid, SDS and Ca2+.

In the measurement of total lipid-bound sialic acids involving periodic acid oxidation, as in the periodate-resorcinol assay, the inner sialic acids of disialoglycolipids (such as GD3 and GD2) are not involved because their alpha 2,8 ketosidic linkages are resistant to periodic acid oxidation, even after acid/enzyme hydrolysis or alkali pretreatment. However, the sialic acids from these glycolipids can be recovered completely after cleavage of alpha 2,8 linkages by V. cholerae sialidase in the presence of cholic acid, sodium dodecyl sulphate and calcium. Interestingly, removal of calcium or detergent(s) or both significantly minimizes the sialidase action on the disialyl residues of these gangliosides. Therefore, we recommend sialidase (Vibrio cholerae) pretreatment of the glycolipids in the presence of cholic acid, SDS and Ca2+ for complete recovery of sialic acids from di- and polysialogangliosides and for accurate measurement of total lipid-bound sialic acids by periodate-resorcinol assay.

Generation of Hydrogen Peroxide, Superoxide Anion and the Hydroxyl Free Radical from Polyphenols and Active Benzene Metabolites: Their Possible Role in Mutagenesis.

Benzene is strongly suspected of being an animal and human carcinogen, but the mechanisms by which it induces tumors of lymphoid and hematopoietic organs are unknown. Production of active oxygen species from benzene metabolites [hydroquinone (HQ), catechol and 1,2,4-benzenetriol (1,2,4-BT) and related polyphenols (resorcinol, pyrogallol and phloroglucinol)] are investigated. Pyrogallol and 1,2,4-BT can produce H(2)O(2), O(-)(2) and (.)OH simultaneously, and have powerful mutagenic potential. Resorcinol and phloroglucinol cannot produce all of the active oxygen species, and show no mutagenic effects. Catechol can produce H(2)O(2), but cannot produce O(-)(2) and (.)OH, and has no mutagenic activity. These data strongly support the hypothesis that benzene metabolites can cause mutagenicity via the generation of oxygen radicals. Although HQ produces H(2)O(2) only, and less than produced by pyrogallol and 1,2,4-BT, the mutagenicity of HQ is higher. The results indicate that HQ may act via another mechanism to cause mutagenicity. In the presence of trace metal ions, the reactivity of polyphenols is increased. The biological significance of these phenomena are investigated and discussed. Copyright 1994 S. Karger AG, Basel

The bromination of acetone. application to multicomponent kinetic determinations.

A very simple kinetic method is proposed for the resolution of mixtures of acetone and a second component. It is based on the reaction, at constant temperature, between bromine and acetone. This reaction can be regarded, under certain conditions, as pseudo-zero-order on the bromine concentration. To show the possibilities of the method, mixtures of acetone (between 2 and 20 x 10(-4) mol L(-1)) and either hydroquinone (between 0.4 and 2.2 x 10(-4) mol L(-1)) or resorcinol (between 0.1 and 10.7 x 10(-4) mol L(-1)) have been used with the concentration ratios [acetone]:[hydroquinone] ranging between 0.5 and 50 and [acetone]:[resorcinol] ranging between 2.8 and 200. No systematic errors were found to exist (90% confidence level) and random errors were mainly under 5%. The method can be extended to mixtures of acetone and a second component whose reaction with bromine is fast (phenols, aromatic amines, etc.).

Contribution to the Detection and Identification of Oxidation Metabolites of Nonylphenol in Sphingomonas sp. strain TTNP3.

Sphingomonas sp. strain TTNP3 has been previously described as a bacterium that is capable of degrading the technical mixture of nonylphenol (NP) isomers and also the 4(3',5'-dimethyl-3'-heptyl)-phenol single isomer of NP. Until recently, 3,5-dimethyl-3-heptanol was the only reported metabolite of 4(3',5'-dimethyl-3'-heptyl)-phenol. A short time ago, the detection of an intracellular metabolite resulting from the oxidation of 4(3',5'-dimethyl-3'-heptyl)-phenol which was identified as 2(3,5-dimethyl-3-heptyl)-benzenediol has been reported. A decisive element for this identification was the occurrence of some slight differences with the two most probable metabolites i.e. 4(3',5'-dimethyl-3'-heptyl)-resorcinol and 4(3',5'-dimethyl-3'-heptyl)-catechol. These facts led us to hypothesise some NIH shift mechanisms explaining the formation of 2(3',5'-dimethyl-3'-heptyl)-benzenediol. In the present work, we describe the steps that led to the detection of these metabolites in the intracellular fraction of Sphingomonas sp. strain TTNP3. The formation of analogous intracellular metabolites resulting from the degradation of the technical mixture of NP is reported. To further elucidate these degradation products, studies were carried out with cells grown with 4(3',5'-dimethyl-3'-heptyl)-phenol as sole carbon source. The description of the syntheses of reference compounds, i.e. 4(3',5'-dimethyl-3'-heptyl)-resorcinol and 4(3',5'-dimethyl-3'-heptyl)-catechol and their comparative analyses with the intermediates of the degradation of 4(3',5'-dimethyl-3'-heptyl)-phenol are presented.

An unexpected side reaction in the guaiacol assay for peroxidase.

In routine guaiacol assays for thyroid peroxidase and lactoperoxidase employing a newly purchased bottle of guaiacol from Aldrich Chemical Co., we were surprised to find the formation of a blue color instead of the expected amber color classically associated with this assay. This was observed also with horseradish, myelo-, and cytochrome c peroxidase. The blue color (Amax approximately 650 nm) was not formed with guaiacol reagents obtained from two other chemical companies, nor was it seen with a bottle of old Aldrich guaiacol that had been in use in the laboratory for more than 10 years. In the present investigation we provide evidence that formation of the blue color is closely associated with the presence of a low concentration of catechol (approximately 0.5 mol%) in the new Aldrich guaiacol reagent. Catechol itself, even in much higher concentration, is a very weak donor for peroxidase, forming a light pink color. The blue color in Aldrich new guaiacol is not formed to the exclusion of 470-nm-absorbing product(s). Formation of the latter is, however, inhibited, and use of Aldrich new guaiacol for assay leads to low values for peroxidase activity. Other dihydroxyphenols (resorcinol and hydroquinone) do not mimic the action of catechol in formation of the blue color. Resorcinol is a very potent inhibitor of peroxidation of guaiacol. Possible schemes are proposed for formation of the products that may be associated with the amber and blue colors.

Ganglioside alterations in stimulated murine macrophages.

A two-dimensional thin-layer chromatographic technique has been used to separate and display gangliosides from murine peritoneal macrophages in different functional states. Resident macrophages have a relatively simple ganglioside pattern with about 15 resorcinol-positive spots. Gangliosides from resident cells contained mostly (90%) N-glycolylneuraminic acid. Thioglycolate-elicited and Corynebacterium parvum-activated macrophages have much more complex patterns with about 40 resorcinol-positive spots. Although ganglioside sialic acid content of stimulated macrophages was only slightly higher than that of resident cells, it consisted of nearly equal amounts of N-acetyl- and N-glycolylneuraminic acid. The shift in the ganglioside sialic acid type and the expression of different gangliosides in macrophages upon stimulation may help explain some of the differences in function and responsiveness noted in these macrophage populations.

Self-catalyzed inactivation of cytochrome P-450 during microsomal metabolism of cannabidiol.

When cannabidiol (CBD) was incubated with hepatic microsomes of mice in the presence of an NADPH-generating system, a significant decrease of cytochrome P-450 content was observed by measuring its carbon monoxide difference spectra. The decrease of cytochrome P-450 by CBD required NADPH and molecular oxygen. The effect was partially inhibited by SKF 525-A but not by various scavengers of active oxygen species, superoxide anion, hydroxyl radical and singlet oxygen. The incubation of CBD with hepatic microsomes did not affect total heme but decreased significantly free sulfhydryl contents in the microsomes. The derivatives of CBD modified in the resorcinol moiety, CBD-monomethyl- and dimethylethers, almost lost the effect on cytochrome P-450, whereas those modified in the terpene moiety, 8,9-dihydro- and 1,2,8,9-tetrahydro-CBDs exhibited some potency to inactivate cytochrome P-450. The inactivation of cytochrome P-450 by CBD and related compounds led to the inhibition of hepatic microsomal p-nitroanisole O-demethylase and aniline hydroxylase activities. These results suggest that the resorcinol moiety of CBD plays some role in the inactivation of cytochrome P-450 by the cannabinoid.

Characterization of cannabidiol-mediated cytochrome P450 inactivation.

Cannibidiol (CBD) has been shown to impair hepatic drug metabolism in several animal species and to markedly inhibit mouse hepatic microsomal delta 1-tetrahydrocannabinol (THC) metabolism by inactivating specific cytochrome P450s (P450) belonging to the 2C and 3A subfamilies. Elucidation of the mechanism of CBD-mediated P450 inhibition would be clinically very important for predicting its effect on metabolism of THC and the many other clinically important drugs known to be metabolized by P450s 2C and 3A. CBD-mediated inactivation of mouse hepatic microsomal P450s did not decrease hepatic microsomal heme content. However, [14C]CBD was found covalently bound to microsomal protein in an approximately equimolar ratio to P450 loss. Immunoprecipitation of microsomal protein with antibodies raised against either P450 2C or 3A revealed that approximately equal amounts of [14C]-CBD were bound to each of these P450s after CBD-mediated inactivation. Furthermore, this specific P450 binding was equivalent to P450 loss and accounted for nearly all of the microsomal [14C]CBD irreversible binding. Although > 80% of the enzyme activities attributed to P450s 2C and 3A were inactivated by CBD at the anticonvulsant dose of 120 mg/kg, P450 2C was approximately 3-fold more sensitive than P450 3A at the lower CBD doses tested. CBD analogs were synthesized in order to elucidate the chemical pathways for CBD-mediated P450 inactivation in vivo. Oxidations at allylic carbon positions or saturation of either the exocyclic double bond or both double bonds of the terpene moiety did not markedly affect the inhibitory properties of the analogs. Methylation of both phenolic groups of the resorcinol moiety completely blocked the P450-inhibitory properties of this analog, revealing the involvement of a free hydroxyl group in the inactivation process. Rotation of the resorcinol moiety in abnormal-CBD did not impair the inhibitory properties of the analog, suggesting that the position of the hydroxyl group relative to the terpene ring is unimportant. Further studies are required to fully understand the chemical basis of CBD-mediated P450 inactivation.

The effect of merulinic acid on biomembranes.

Merulinic acid (heptadecenylresorcinolic acid, resorcinolic acid) is one of the members of resorcinolic lipids, the natural amphiphilic long-chain homologues of orcinol (1,3-dihydroksy-5-methylbenzene). In the present study, membrane properties of merulinic acid were investigated. Merulinic acid exhibits strong haemolytic activity against sheep erythrocytes (EH50 of 5+/-2 microM) regardless of the form of its application-direct injection into the erythrocyte suspension or injection as merulinic acid-enriched liposomes. The lysis of erythrocytes induced by merulinic acid was inhibited by the presence of divalent cations. The effectivity of the protection of erythrocytes was highest for Zn2+ and weakest for Mn2+. Merulinic acid at low concentrations also exhibits the ability for protection of cells against their lysis in hypoosmotic solutions. This protective effect is significant as, at 10 microM concentration of merulinic acid, the extent of osmotically induced cell lysis is reduced by approximately 40%. Merulinic acid induces increased permeability of liposomal vesicles. This effect was shown to be dependent on the composition of liposomal bilayer and it was stronger when lipid bilayer contained glycolipids (MGDG and DGDG) and sphingomyelin. Changes of TMA-DPH and NBD-PE fluorescence polarization show that the degree of merulinic acid incorporation into liposomal membrane is not very high. The polar "heads" of the molecules of investigated compounds are localized on the level of fatty acid's ester bonds in phospholipid molecules. Merulinic acid caused the increased fluorescence of the membrane potential fragile probe. This indicated an alteration of the surface charge and a decrease of the local pH at the membrane surface. This effect was visible in both low- and high-ionic strength environment. Merulinic acid causes also a decrease in activity of the membrane-bound enzyme acetylcholinesterase.

Catechol sensor using poly(aniline-co-o-aminophenol) as an electron transfer mediator.

In this work, poly(aniline-co-o-aminophenol) (copolymer) was used as an electron transfer mediator in the electrochemical oxidation of catechol due to its reversible redox over a wide range of pH. The experimental results indicate that the anodic peak potential of catechol at the copolymer electrode is lower than that at the platinum electrode in a solution consisting of catechol and sodium sulfate with pH 5.0, and the activation energy for the electrochemical oxidation of catechol at the copolymer electrode is low (23.6 kJ mol(-1)). These are strong evidence for the electrocatalytic oxidation of catechol at the copolymer electrode. The -OH group on the copolymer chain plays an important role in the electron transfer between the copolymer electrode and catechol in the solution. Based on the catalytic oxidation, the copolymer is used as a sensor to determine the concentration of catechol. The response current of the sensor depends on the concentration of catechol, pH, applied potential and temperature. At 0.55 V (versus saturated calomel reference electrode (SCE)) and pH 5.0, the sensor has a fast response (about 10s) to catechol and good operational stability. The sensor shows a linear response range between 5 and 80 microM catechol with a correlation coefficient of 0.997. It was found that phenol and resorcinol cannot be oxidized at the copolymer electrode at potentials < or =0.55 V, so controlling the sensor potential affords a good way of avoiding the effect of phenol and resorcinol on the determination of catechol.
