Electrochemical partial fluorination of organic compounds. 80. Synthesis of cyclic alpha-arylthio-alpha-monofluorophosphonate esters.

[Reaction: see text]. Seven-membered cyclic alpha-monofluorophosphonate esters such as 2-allyloxy-3-fluoro-3-phenylthio-4,7-dihydro-[1, 2]-oxaphosphinine-2-oxide were successfully synthesized in moderate total yield as 41% from open-chain allyl phosphonates having an alpha-arylthio group as an electroauxiliary using an alternative sequence of anodic fluorination and ring-closing olefin metathesis (RCM). On the other hand, in an attempt to synthesize an eight-membered analogue, a different type of seven-membered fluorinated cyclic product was formed predominantly by the RCM reaction between the allyloxy groups.

Pharmacological effects of novel quinone compounds, 6-(fluorinated-phenyl)amino-5,8-quinolinediones, on inhibition of drug-induced relaxation of rat aorta and their putative action mechanism.

Two 6-(fluorinated-phenyl)amino-5,8-quinolinedione derivatives, OQ21 and OQ1, were newly synthesized as potent inhibitors of endothelial-dependent vasorelaxation. The purpose of the present study was to investigate the effects of OQ21 and OQ1 on different types of vasorelaxation and to pursue their action mechanisms. For acetylcholine both compounds, at a low concentration (0.1 microM), reduced the maximal response with increase of EC(50) values. OQ21 is a novel quinone compound and showed a more potent and efficacious inhibitory effect on acetylcholine-induced relaxation of rat aorta than that of LY83583 (6-anilino-5,8-quinolinedione). Relatively high concentrations (1 microM) of OQ21 and OQ1 inhibited the sodium nitroprusside-induced relaxation of endothelium-denuded ring, producing rightward shifts of the curve for sodium nitroprusside without altering the maximal response. They also prevented acetylcholine and sodium nitroprusside-induced elevations of cyclic GMP. In addition, OQ21 and OQ1 (1 microM) significantly decreased (52-72%) the sensitivity of L-arginine-induced relaxation of precontracted endothelium-denuded aortic rings from lipopolysaccaride-treated (20 mg/kg, i.p.) rats. The inhibitory effect of OQ21 on endothelium-dependent vasodilation was enhanced by N(G)-nitro-L-arginine, which inhibits nitric oxide synthase (NOS) by binding the oxygenase domain of the enzyme, but not by diphenylendiodonium, which inhibits NOS by binding to the reductase domain of the enzyme. Treatment of blood vessels with OQ21 or OQ1 showed a significant increase in chemiluminescence output, which was prevented by adding superoxide dismutase, suggesting that superoxide generation is involved in the action mechanism for OQ21. Present results indicate that a novel naphthoquinone compound, OQ21, potently inhibits endothelial NOS, possibly by interacting with the reductase domain of the enzyme, which leads to induce superoxide formation. The new benzoquinone compounds, OQ21 and OQ1, inhibit not only endothelium-dependent vasorelaxation but also endothelium-independent relaxation induced by exogenous NO generated from a nitrovasodilator via the reduction of cyclic GMP. They also reduced L-arginine-induced vasorelaxation in endotoxin-treated rats, indicating their possession of inhibitory effect on inducible NOS.

Synthesis and evaluation of fluorine-18 labelled compounds for imaging of bacterial infections with pet.

Syntheses of no carrier added (n.c.a.) 6-fluoro-1,4-dihydro-1-cyclopropyl-4-oxo-7-[4-[18F]fluoro-phenacyl-1-piperacinyl]-chinolincarboxylic acid ([18F]COPCA) and n.c.a. 4-[18F]fluoro-benzoyl-ubiquicidin 29-41 ([18F]UBI 29-41) are described. [18F]COPCA was synthesised within 120 min with a radiochemical yield of 9-12%. [18F]UBI 29-41 was synthesised within 150 min with a radiochemical yield of 15-20%. Both compounds had a specific activity of more than 35 GBq/micromol. The biological activity was verified by measuring its binding to Staphylococcus aureus bacteria. Specific binding was found for [18F]UBI 29-41 (12-17%), whereas no specific binding for [18F]COPCA was found.

[Effect of accumulating fluorine and sulphur compounds on the content of chlorophyll in pine needles (Pinus sylvestris L.) from some areas of southern Poland in 2001-2003].

Concentrations of F(-), S and chlorophyll in needles of pines growing near metallurgical works and power plants were determined. A statistically significant positive correlation between F(-) (in the range of 4.32 to 21.98 microg F(-)/g d.m.) and chlorophyll and a negative correlation between S (in the range of 1094 to 2150 microg S/g d.m.) and chlorophyll was found. This result suggests an antagonistic interaction between F(-) and S in pine needles.

A study of C-F.M+ interaction: alkali metal complexes of the fluorine-containing cage compound.

The C-F.M(+) interaction was investigated by employing a cage compound 1 that has four fluorobenzene units. The NMR ((1)H, (13)C, and (19)F) spectra and X-ray crystallographic analyses of 1 and its metal complexes showed clear evidence of the interaction. Short C-F.M(+) distances (C-F.K(+), 2.755 and 2.727 A; C-F.Cs(+), 2.944 and 2.954 A) were observed in the crystalline state of K(+) subset 1 and Cs(+) subset 1. Furthermore, the C-F bond lengths were elongated by the interaction with the metal cations. By calculating Brown's bond valence, it is shown that the contribution of the C-F unit to cation binding is comparable or greater than the ether oxygen in the crystalline state. Representative spectroscopic changes implying the C-F.M(+) interaction were observed in the NMR ((1)H, (13)C, and (19)F) spectra. In particular, (133)Cs-(19)F spin coupling (J = 54.9 Hz) was observed in the Cs(+) complex.

Large primary kinetic isotope effects in the abstraction of hydrogen from organic compounds by a fluorinated radical in water.

Isotope effects have been measured for the abstraction of hydrogen from a series of organic substrates by the perfluoro radical, Na+ -O3SCF2CF2OCF2CF2*, in water. Both primary and secondary deuterium isotope effects were measured, with the primary isotope effects ranging in value from 4.5 for isopropanol to 19.6 for acetic acid. The values for the alpha- and beta-secondary deuterium isotope effects were 1.06 and 1.035, respectively. It was concluded that tunneling contributes significantly to the production of the observed, large primary kinetic isotope effects in these C-H abstraction reactions.

Fluorine-19 NMR spectroscopy and imaging investigations of myocardial perfusion and cardiac function.

Fluorine-19 nuclear magnetic resonance images of rabbit hearts have been obtained with the administration of a fluorinated contrast compound. The use of fluorine-labeled compounds allows differences in tissue perfusion to be demonstrated between hearts with and without coronary artery ligation. In postischemic reperfusion studies, regional hypoperfusion is also demonstrated by 19F imaging. The results of in vivo rabbit studies show that 19F NMR can be used to monitor the rate of blood pool and extravascular space dilution of a single bolus injection of either a water soluble or emulsified fluorine-labeled agent.

Global biomonitoring of perfluorinated organics.

The environmental distribution of fluorinated organic compounds (FOCs) has been less well described than the other halogenated hydrocarbons such as chlorinated and brominated compounds. This is despite the fact that FOCs have been used in a wide variety of products and applications for more than 50 years. FOCs are resistant to hydrolysis, photolysis, microbial degradation, or metabolism by vertebrates due to the high energy of carbon-fluorine bond. In particular, perfluorinated (fully fluorinated) compounds (PFCs) have the potential to persist in the environment. But, until recently, the extent and magnitude of environmental distribution of PFCs was unknown. Recent development of an analytical technique for PFCs using high performance liquid chromatography-negative ion electrospray tandem mass spectrometry (HPLC-ESMSMS) permitted the survey of PFCs in livers and blood plasma of wildlife on a global scale.
