[Benzopyrene (BaP) implants into the glottic mucous membrane of hamsters].

For testing the effect of benzo (a) pyrene (BaP) on glottical epithelium European hamsters were used. With laryngofissure hamsters larynges had been opened and a mixture of tricaprilin, beewax and 1 mg BaP was implanted into a pocket formed by mucous membrane of one vocal fold. Compared to typical histology in smoker's larynx, squamous metaplasia and dysplasia and in one case papillary polyps could be described. No squamous cell carcinomata was found.

[Correlation of the 3,4-benzopyrene content in grain, grist products and baked bread].

The content of 3,4-benz (a) pyrene (BP) in wheat corn, grist products and baked bread was studied by the fluorescent-spectral technique under different content of the carcinogen in the initial corn. The content of BP in grist products correlated, within the range of experimental errors, with its content in corn. The content in flour was 30-60% of the total content in grist products. The bread baked from the dough that included only flour, yeast and water had much less BP (with reference to dry weight) as compared with that in the initial flour. Provided the dough was supplemented with KBrO3 the bread contained twice as much BP unlike that to which KBrO3 had not been added.

[Effect of the combined and separate administration of phenobarbital and 3-methylcholanthrene on liver benzopyrene hydroxylases in "sensitive" and "resistant" inbred mice].

Combined induction of liver monooxygenases with 3-methylcholanthrene (3-MC) and phenobarbital (PB) has been studied in experiments on DBA/2 and C57B/6 mice. As is known, PB increases the number of intracellular receptor sites for polycyclic hydrocarbons. When DBA/2 mice resistant to the induction were pretreated with PB, there were no signs of induction common to those induced by 3-MC. In similar experiments with the sensitive C57BL/6 mice, PB and 3-MC produced additive effects on the induction of liver monooxygenases.

[Comparison of fenoterol (Berotec) with the antiallergic benzopyrene compound PR-D 92 and a placebo in regard to the cutaneous immediate reaction in allergic patients (author's transl)].

After discussion of today's knowledge of the mechanism of allergic reactions with particular regard to the adenylyl cyclase system a report is given on an experimental study in which Fenoterol (Berotec), PR-D 92 (Bisbenzopyran) as well as placebo were repeatedly applied to defined areas of the forearm of 30 patients with known pollen allergies. The expansion of the allergy-induced erythemae and weals was exactly measured. A comparison of the sizes showed approximately 50% inhibition of the reaction following application of Berotec, the difference being highly significant compared with PR-D 92 and placebo. There was no statistical difference between the last two.

[Benzopyrene metabolism in cultured liver cells of human embryo].

The hepatic cells of human embryos cultivated as a monolayer retain their ability to metabolize the carcinogenic hydrocarbon benz (a) pyrene (BP). The intensity of BP metabolism is higher in the "young" cultures of hepatic cells than in fibroblast cultures obtained from the same embryo. At later terms the former becomes equal for both tissues as a result of a decreased metabolic activity of hepatic cells.

Metabolism and specific benzopyrene metabolite modification of DNA in early S by human lung epithelial and fibroblast cells leading to the expression of an abnormal phenotype.

Examination of the high pressure liquid chromatographic profiles of ethyl acetate extractable benzo [a] pyrene (B(a)P)-metabolites from human lung fibroblast and type II epithelial cells after S phase entry indicated that B(a)P-7,8-diol and 9,10-diol species were produced following the oxygenation of B(a)P. These metabolites were detected intracellularly and in the extracellular growth medium. Both cell types appeared to release extracellularly, elevated amounts of the B(a)P-7,8-diol species. It was interesting to note of the 4 pmol of oxygenated metabolites localized intracellularly, in the fibroblast, that we identified two major metabolites, B(a)P-9,10 and -7,8-diol species. Lung epithelial cells metabolize intracellular B(a)P extensively, greater than or equal to 93% of the parent B(a)P. No tetrols were detected intracellularly or extracellularly in the treated fibroblast cells. The treated epithelial cells produced both tetrols and sulfate conjugates. The extent of observed modification of early S phase nuclear DNA of lung epithelial cells was 7.5 +/- 4.9 adducts per 10(6) bases and 4.2 +/- 2.7 adducts per 10(6) bases in lung fibroblasts. The major adduct formed in both cell types was 7 beta-BPDE-I-dG. Under conditions for transformation, both the B(a)P treated lung epithelial cells and lung fibroblasts treated in early S with either B(a)P or BPDE-I yielded populations that exhibited properties of anchorage independent growth and cellular invasiveness. Metabolism and the presence internally of metabolites did not correlate with the extent of modification of DNA in early S.
